AIMS: Several neurodegenerative and neuromuscular disorders are characterised by storage of polyglucosan, consisting of proteins and amylopectin-like polysaccharides, which are less branched than in normal glycogen. Such diseases include Lafora disease, branching enzyme deficiency, glycogenin-1 deficiency, polyglucosan body myopathy type 1 (PGBM1) due to RBCK1 deficiency and others. The protein composition of polyglucosan bodies is largely unknown.
METHODS: We combined quantitative mass spectrometry, immunohistochemical and western blot analyses to identify the principal protein components of

Glycogen storage disease type IV (GSD IV; Andersen disease) is caused by a deficiency of glycogen branching enzyme (GBE), leading to excessive deposition of structurally abnormal, amylopectin-like glycogen in affected tissues. The accumulated glycogen lacks multiple branch points and thus has longer outer branches and poor solubility, causing irreversible tissue and organ damage. Although classic GSD IV presents with early onset of hepatosplenomegaly with progressive liver cirrhosis, GSD IV exhibits extensive clinical heterogeneity with respect to age at onset and variability in pattern and ex

Although some lactic acid bacteria species harbor genes encoding α-glucanotransferases (4,6-α-GTase and 4,3-α-GTase), the technological and functional implications of these enzymes within sourdough ecosystems remain poorly understood. This study investigated the effects associated with α-GTase-positive and α-GTase-negative strains of Limosilactobacillus reuteri and L. fermentum on sourdough microbiota, bread quality, and starch digestibility. Starter culture inoculation steered the assembly of the sourdough microbiota, resulting in stable acidification and altered microbial community structure

Resistant starch (RS) is a dietary fiber that escapes small-intestinal digestion and reaches the colon, offering inherent colon-targeting potential, bioactive protection, and prebiotic benefits. However, native RS often shows limited mechanical strength and high hydrophilicity, driving the development of modified and composite delivery systems. This review summarizes the functional advantages of RS and evaluates representative RS-containing delivery systems according to carrier architecture and the functional role of RS, including particulate carriers, Pickering emulsions, hydrogels, nanoparti

Glycogen storage disease type IV (GSD IV), also called Andersen disease, or amylopectinosis, is a highly heterogeneous autosomal recessive disorder caused by a glycogen branching enzyme (GBE, 1,4-alpha-glucan branching enzyme) deficiency secondary to pathogenic variants on GBE1 gene. The incidence is evaluated to 1:600 000 to 1:800 000 of live births. GBE deficiency leads to an excessive deposition of structurally abnormal, amylopectin-like glycogen in affected tissues (liver, skeletal muscle, heart, nervous system, etc.). Diagnosis is often guided by histological findings and confirmed by GBE

Inflammation is a major mediator of CKD progression and is partly driven by altered gut microbiome and intestinal barrier disruption, events which are caused by: urea influx in the intestine resulting in dominance of urease-possessing bacteria; disruption of epithelial barrier by urea-derived ammonia leading to endotoxemia and bacterial translocation; and restriction of potassium-rich fruits and vegetables which are common sources of fermentable fiber. Restriction of these foods leads to depletion of bacteria that convert indigestible carbohydrates to short chain fatty acids which are importan

Legumes are increasingly used as functional ingredients in plant-based foods. However, their compact endogenous structure restricts techno-functional properties. Conventional fractionation methods that isolate proteins further impair these properties and generate co-products. Solid-state fermentation (SSF), which involves growth of microorganisms on moist solid legumes for in situ enzymatic modification, is a gentle and sustainable strategy to enhance techno-functionality. However, the mechanistic understanding of structural changes underpinning functional modifications in legume ingredients r

Water management during grain filling plays a critical role in determining rice quality, but its coordinated regulation of eating and aroma quality in aromatic rice remains unclear. In this study, two indica cultivars, Nongxiang 32 and Yuzhenxiang, were subjected to three irrigation regimes: continuous flooding (CF), alternate wetting and drying (AWD), and saturated soil irrigation (SSI). Our results showed that 2-acetyl-1-pyrroline (2AP) content exhibited the highest level under AWD and the lowest under SSI. Starch pasting properties were improved under AWD and SSI, as reflected by higher pea

Rice (Oryza sativa L.) is an excellent source of starch, which is composed of amylopectin and amylose. Resistant starch (RS) is a starch product that is not easily digestible and absorbed in the stomach or small intestine and instead is passed on directly to the large intestine. Cereals high in RS may be beneficial to improve human health and reduce the risk of diet-related chronic diseases. It has been reported through chemical mutagenesis and RNA interference studies that starch branching enzymes (SBEs) play a major role in contributing to higher levels of RS in cereal crops. In this study,

BACKGROUND: African star apple (Chrysophyllum albidum) is a traditonal fruit, which is predominant in tropical regions with the fruit parts consumed by the populace and used in folklore to manage diabetes. However, the likely activity mechanism is still undetermined. The current study examined and compared the inhibitory abilities of African star apple fruit parts on selected key enzymes related to diabetes mellitus in the pancreas tissue of rat.
METHODS: Inhibitory effect of aqueous extract (1:10 w/v) of African star apple fruit parts (pulp, cotyledon, seed coat and pulp coat) on the activit

Toxoplasma gondii, the causative agent of toxoplasmosis, is an obligate intracellular protozoan that can infect a wide range of vertebrate cells. Here, we describe the cytotoxic effects of the dinuclear iron compound [Fe(HPCINOL)(SO4)]2-μ-oxo, in which HPCINOL is the ligand 1-(bis-pyridin-2-ylmethyl-amino)-3-chloropropan-2-ol, on T. gondii infecting LLC-MK2 host cells. This compound was not toxic to LLC-MK2 cells at concentrations of up to 200 μM but was very active against the parasite, with a 50% inhibitory concentration (IC50) of 3.6 μM after 48 h of treatment. Cyst formation was observed a

APBD is a rare disorder most often affecting adults of Ashkenazi Jewish origin due to partial deficiency of the glycogen brancher enzyme (GBE). It is characterized by progressive involvement of both the central and peripheral nervous systems and deposition of amylopectin-like polyglucosan bodies. There have been no metabolic derangements that might suggest effective therapy nor have there been any clinical improvements for control of its relentless progression. The APBD patients, in this study, experienced stabilization of disease progression, and limited functional improvement in most patient

A severe, progressive myopathy developed in an 11-year-old, phosphofructokinase (PFK)-deficient, male, English Springer Spaniel dog. Results from a routine neurological examination were normal. Examination of histologic sections of skeletal muscle revealed large accumulations of material in some myofibers. These deposits were pale, basophilic, somewhat flocculent, and slightly granular with hematoxylin and eosin stain. Most fascicles examined in sections of limb and trunk muscles were affected to some degree, with up to 10% of muscle fibers being involved. Deposits stained strongly with period

Pylorus ligation changed the morphology and location of gastric lesions induced by water-immersion stress (WI stress) in rats; linear lesions in the corpus mucosa disappeared, and punctate lesions appeared in both the corpus and antrum, in association with the loss of H+ and the gain of Na+ in the gastric contents. Oral administration of antipeptic drugs such as amylopectin sulfate and sulfated glyptide or porcine pepsin significantly prevented or aggravated the lesions, respectively, although another type of elongated lesion appeared in response to high doses of antipeptic drugs. These antipe

Polysaccharides as immunomodulators are increasingly explored in preclinical studies, showing potential applications for preventing or treating different diseases. Among them is starch, an α-glucan formed by amylose and amylopectin chains. Given their abundance in nature, physicochemical characteristics, and applicability in pharmacy, they are versatile molecules that offer important biotechnological and biomedical advantages. Most studies about starch immunostimulant properties focus on modified-soluble and particulate α-glucans. However, little research has been done on the immunostimulant p

Glycogen, a branched polymer of glucose, functions as an energy reserve in many living organisms. Abnormalities in glycogen metabolism, usually excessive accumulation, can be caused genetically, most often through mutation of the enzymes directly involved in synthesis and degradation of the polymer leading to a variety of glycogen storage diseases (GSDs). Microscopic visualization of glycogen deposits in cells and tissues is important for the study of normal glycogen metabolism as well as diagnosis of GSDs. Here, we describe a method for the detection of glycogen using a renewable, recombinant

alpha-Amylase activity was screened in the peel, as waste fruit, of 13 species and cultivars of Egyptian citrus. The species Citrus sinensis cv. Abosora had the highest activity. alpha-Amylase AI from Abosora peel was purified to homogeneity using anion and cation-exchange, and gel filtration chromatographies. Molecular weight of alpha-amylase AI was found to be 42 kDa. The hydrolysis properties of alpha-amylase AI toward different substrates indicated that corn starch is the best substrate. The alpha-amylase had the highest activity toward glycogen compared with amylopectin and dextrin. Potat

Glycogen storage disease type IV (GSD-IV), also known as Andersen disease or amylopectinosis (MIM 23250), is a rare autosomal recessive disorder caused by a deficiency of glycogen branching enzyme (GBE) leading to the accumulation of amylopectin-like structures in affected tissues. The disease is extremely heterogeneous in terms of tissue involvement, age of onset and clinical manifestations. The human GBE cDNA is approximately 3-kb in length and encodes a 702-amino acid protein. The GBE amino acid sequence shows a high degree of conservation throughout species. The human GBE gene is located o

This study focused on black rice starch as the research subject, constructing different ultrasonic power-based black rice starch-resveratrol (BRS-RE) composite systems to explore the correlation between structural changes, physicochemical properties, and in vitro digestion behavior. Ultrasound increased the complexation index (CI) to a maximum of 80.38 % at 400 W. Particle size initially increased and then decreased with ultrasonic power, indicating power-dependent molecular reorganization. FT-IR and Raman analyses indicated changes in the hydrogen-bonding environment and short-range order, wh

Barley is classified as malt or non-malt barley according to its end use, yet the role of starch characteristics in determining malt quality remains poorly understood. In this study, we systematically compared starch content, composition, granule structure, and thermal properties between six malt and six non-malt barley varieties. Compared with non-malt barley, malt barley exhibited significantly higher total starch (TS) content, a lower amylose/amylopectin (Am/Ap) ratio, and a larger area of large starch granules (LSGs), features that may collectively enhance the contact surface for hydrolyti
