Serological studies of babesiosis and anaplasmosis of cattle.

Bos indicus-crossbred calves exposed to natural Babesia bovis infection in wet and dry tropical environments in northern Queensland were tested for antibodies using the indirect haemagglutination (IHA) test. There was evidence of maternal antibodies suggesting endemicity of B. bovis in the 2 environments. Within one year 35.3 to 94.6% of the calves had acquired B. bovis infection. The highest titres and most rapid conversion rates were observed just after the wet season when the tick population was at its peak. Clinical disease was not observed. A third group of Bos indicus-cross cattle of var

First Report on Molecular Characterization of Anaplasmosis in Small Ruminants in Pakistan.

Anaplasmosis is caused by a Gram-negative obligate intracellular bacterium of the genus Anaplasma with the pathogen having a zoonotic impact. The study aimed to estimate the prevalence of anaplasmosis in Pakistan, to unravel the association of potential risk factors, and to investigate the effect on hematological parameters in affected small ruminants. A total of 150 (n = 75 sheep; n = 75 goats) blood samples were initially screened microscopically and then subjected to PCR targeting the amplification of the 16S rRNA gene fragment of Anaplasma. The PCR-based positive samples were then processe

Molecular screening of piroplasms and Anaplasmataceae agents in Hyalomma dromedarii ticks from camels over different seasons in Egypt.

Piroplasmosis, a disease of domestic and wild animals, is caused by tick-borne protozoa of the genera Babesia and Theileria, while anaplasmosis is caused by tick-borne bacteria of genera Anaplasma. Hyalomma dromedarii is the most dominant tick species infesting camels in Egypt and act as a vector of piroplasms, Anaplasma, Rickettsia and Ehrlichia spp. The available information concerning the detection of these pathogens in H. dromedarii infesting camels is limited. The present study aimed to evaluate the status of these pathogens in H. dromedarii ticks over four seasons of a year, in addition

An enzyme linked immunosorbent assay to diagnose Babesia bovis infection in cattle.

The optimum gel filtration fraction from lysate of Babesia bovis infected erythrocytes was determined for use as an antigen in an ELISA to diagnose B. bovis infection in cattle. Of four enzyme labels tested, horseradish peroxidase was the most suitable. The assay is both sensitive and specific in detecting antibody for 2-4 years after a single infection. False positive reactions were obtained only with some sera from some Anaplasma marginale infected cattle.

Bovine anaplasmosis: elimination of the carrier state with injectable long-acting oxytetracycline.

Long-acting oxytetracycline was administered intramuscularly (20 mg/kg) at 3-day intervals to each of 16 naturally infected Anaplasma marginale carrier beef cows; 8 of the 16 cows were given 3 injections and the other 8 cows were given 4 injections. Four cows seropositive for A marginale served as untreated controls. Rapid card agglutination and complement fixation tests were performed 5 times during the 130-day posttreatment period. All treated cows became negative to both serologic procedures, whereas control cows remained seropositive. Three splenectomized calves that received pooled blood

Survey for selected pathogens in Philippine deer (Rusa marianna) from Guam, Marianna Islands, USA.

Philippine deer (Rusa marianna), native to the Philippine Islands, were introduced to Guam in the late 1700's. Dense populations have become established throughout the island where they cause damage to native plant communities resulting in habitat degradation. In addition, cervids can serve as reservoirs for important pathogens of livestock and people. From February-March 2015, blood, tissue and ectoparasite samples were collected from 132 free-ranging Philippine deer on Guam. Data from 10 deer sampled in 1997 were also analyzed. Deer were negative for antibodies to many of the pathogens asses

Advances in the diagnosis of some parasitic diseases by monoclonal antibody-based enzyme-linked immunosorbent assays.

Advances in diagnostic assays for parasitic diseases include the use of monoclonal antibodies (MAbs) in antigen capture and competitive inhibition enzyme-linked immunosorbent assays (C-ELISA). Antigen capture ELISAs for Anaplasma marginale and Cryptosporidium parvum provide direct detection of these parasites during clinical disease, and the C-ELISA format has been adapted for detection of anti-Babesia equi, anti-A. marginale and anti-bluetongue virus antibodies. False-positive results may occur when antigen preparations in other ELISA formats are contaminated with Escherichia coli, erythrocyt

The effect of natural exposure to Anaplasma and Babesia infections on native calves in an endemic area of Colombia.

The age at which native calves first became infected with Anaplasma marginale and Babesia bigemina and the effect of the infections as characterised by anaemia was determined by the biweekly examination of newly born calves on four ranches located in an endemic area of anaplasmosis and babesiosis on the north coast of Colombia. All of the calves examined became infected with A. marginale at 4-24 weeks of age and with B. bigemina at 2-34 weeks of age. The mean age at first infection with both haemoparasites was 11 weeks. A significant decrease occurred in the mean packed cell volumes of the cal

The twin-arginine translocation pathway in α-proteobacteria is functionally preserved irrespective of genomic and regulatory divergence.

The twin-arginine translocation (Tat) pathway exports fully folded proteins out of the cytoplasm of Gram-negative and Gram-positive bacteria. Although much progress has been made in unraveling the molecular mechanism and biochemical characterization of the Tat system, little is known concerning its functionality and biological role to confer adaptive skills, symbiosis or pathogenesis in the α-proteobacteria class. A comparative genomic analysis in the α-proteobacteria class confirmed the presence of tatA, tatB, and tatC genes in almost all genomes, but significant variations in gene synteny an

Concurrent infections with vector-borne pathogens associated with fatal hemolytic anemia in a cattle herd in Switzerland.

Bovine anaplasmosis is a vector-borne disease that results in substantial economic losses in other parts of the world but so far not in northern Europe. In August 2002, a fatal disease outbreak was reported in a large dairy herd in the Swiss canton of Grisons. Diseased animals experienced fever, anorexia, agalactia, and depression. Anemia, ectoparasite infestation, and, occasionally, hemoglobinuria were observed. To determine the roles of vector-borne pathogens and to characterize the disease, blood samples were collected from all 286 animals: 50% of the cows were anemic. Upon microscopic exam

Clinical chemistry of anaplasmosis: blood chemical changes in infected mature cows.

The clinical chemical changes induced by Anaplasma marginale infection were determined in 16 adult, intact cows infected with either of 2 virulent isolates and in 8 others treated with a live sheep-attenuated A marginale vaccine and were compared with the clinical chemical analyses in 7 noninfected controls. Blood samples from the cows were analyzed for various serum components and during clinical disease. Cows infected with the 2 virulent isolates had maximum erythrocyte parasitemias (0.5% to 66%) between 1 and 11 days of patency (DP); 7 of the 16 infected animals died within 5 to 12 DP. Cows

Emerging perspectives in the research of bovine babesiosis and anaplasmosis.

The Babesia bovis and B. bigemina apicomplexan protozoa in conjunction with the rickettsia Anaplasma marginale are intraerythrocytic pathogens that are responsible for the most prevalent and costly tick borne diseases (TBD's) of cattle worldwide. These organisms are historically associated as they can cause clinically related hemolytic diseases in cattle, are all transmitted by Rhiphicephallus (Boophilus) ticks, and share an uncanny ability to evade the immune systems of the vertebrate hosts, causing persistent disease. In addition, acute babesiosis and anaplasmosis can be prevented quite effe
