Diaphragm development and congenital diaphragmatic hernia.

Advances in the understanding of normal diaphragm embryogenesis have provided the necessary foundation for novel insights into the pathogenesis of congenital diaphragmatic hernia (CDH). Although diaphragm formation is still not completely understood, we have identified key structures and periods of development that are clearly abnormal in animal models of CDH. The pleuroperitoneal fold (PPF) is a transient structure which is the target for the neuromuscular component of the diaphragm. The PPF has been shown to be abnormal in multiple animal models of Bochdalek CDH; specifically, a malformation

Characterization of a novel thermostable nuclease homolog (NucM) in a highly divergent Staphylococcus aureus clade.

A thermostable nuclease homologue (NucM) in an animal-associated divergent clade of Staphylococcus aureus in sub-Saharan Africa has a highly divergent nucleotide sequence compared to those of the classical nuc1 and nuc2 genes of S. aureus. Its deduced amino acid sequences, tertiary structures, and nuclease activities, however, are similar.

ALS mutations do not alter perineuronal net formation in human stem cell-derived motor neurons.

Perineuronal nets (PNNs) are extracellular matrix structures that stabilise synaptic inputs and play a role in regulating neuronal plasticity. Although PNN dysregulation is observed in several neurological disorders, their relevance to amyotrophic lateral sclerosis (ALS) remains unclear. In particular, the extent to which PNN alterations reported in ALS animal models are motor neuron (MN)-intrinsic is unknown. We investigated whether human pluripotent stem cell-derived MNs form PNN-like structures in vitro, and whether ALS-associated mutations alter this process. We show that PNN-like structur

Longitudinal changes in gray and white matter microstructure during epileptogenesis in pilocarpine-induced epileptic rats.

PURPOSE: Temporal lobe epilepsy is associated with tissue abnormalities of several gray and white matter structures that are reproduced in animal models. Few longitudinal studies have focused on the identification of structural differences during epileptogenesis. The diffusion tensor model is a useful tool for evaluating cell death, gliosis, and axonal plasticity in epileptic subjects. This study aimed to evaluate temporal tissue changes after experimental status epilepticus in an animal model of chronic temporal lobe epilepsy. METHODS: Systemic pilocarpine-induced status epilepticus in adult

Development and implementation of an animal-inclusive Community-Led Total Sanitation program for rural Mali.

BACKGROUND: Community-Led Total Sanitation (CLTS) does not formally include animal feces management, despite its goal to prevent fecal matter from entering the environment. By integrating animal feces management into an existing community-level program to safely manage human feces, greater reductions in exposure to feces may be achieved, supporting improvements to both human and animal health. METHODS: We adapted the CLTS intervention to be animal-inclusive and piloted the resulting intervention in twenty-five villages; ten villages received an integrated human and animal feces CLTS program (

Unveiling Pathological Lipid Droplet Accumulation of Dopaminergic Neurons in Parkinson's Disease via a Super-Retentive Fluorescent Probe.

Lipid droplets are highly dynamic organelles in nerve cells and are essential for the function of the central nervous system. The abnormal accumulation of lipid droplets in dopaminergic neurons, found in Parkinson's disease (PD) cells and animal models, presents as a promising target for PD diagnosis. However, previously reported fluorescent probes are unsuitable for in vivo long-term tracking of lipid droplets, limiting their applications in animal models of PD. Here, we developed three lipid droplet-targeted probes, named s-CBTA, b-CBTA, and LD-b-PBTA, which share similar chemical structures

The use of magnetic resonance imaging to better define hoof pathology in the reticulated giraffe (Giraffa camelopardalis reticulata).

A 22-yr-old bull giraffe (Giraffa camelopardalis reticulata) with severely altered hoof conformation in both forelimbs was presented for necropsy following acute mortality. Due to multiple challenges that prevented safe immobilization, corrective hoof trimming procedures were never performed on this animal. To better define the extent of the damage of the soft tissue structures and bone within the hoof, a magnetic resonance imaging (MRI) system was used to obtain images of these structures. The MRI study found evidence of severe osteolysis, phalangeal fractures of both forelimbs, and tenosynov

Centrosomes and cilia in human disease.

Centrioles are microtubule-derived structures that are essential for the formation of centrosomes, cilia and flagella. The centrosome is the major microtubule organiser in animal cells, participating in a variety of processes, from cell polarisation to cell division, whereas cilia and flagella contribute to several mechanisms in eukaryotic cells, from motility to sensing. Although it was suggested more than a century ago that these microtubule-derived structures are involved in human disease, the molecular bases of this association have only recently been discovered. Surprisingly, there is ver

Idiopathic REM sleep behaviour disorder in the development of Parkinson's disease.

Parkinson's disease is a progressive neurodegenerative disorder associated with Lewy body disease pathology in central and peripheral nervous system structures. Although the cause of Parkinson's disease is not fully understood, clinicopathological analyses have led to the development of a staging system for Lewy body disease-associated pathological changes. This system posits a predictable topography of progression of Lewy body disease in the CNS, beginning in olfactory structures and the medulla, then progressing rostrally from the medulla to the pons, then to midbrain and substantia nigra, l

Impact of bronchopulmonary dysplasia on brain and retina.

Many premature newborns develop bronchopulmonary dysplasia (BPD), a chronic lung disease resulting from prolonged mechanical ventilation and hyperoxia. BPD survivors typically suffer long-term injuries not only to the lungs, but also to the brain and retina. However, currently it is not clear whether the brain and retinal injuries in these newborns are related only to their prematurity, or also to BPD. We investigated whether the hyperoxia known to cause histologic changes in the lungs similar to BPD in an animal model also causes brain and retinal injuries. Sprague Dawley rat pups were expose

Comparative study on the distribution of Pacinian corpuscles in the pancreas.

BACKGROUND: Pacinian corpuscles (PCs) are pressure- and vibration-sensitive mechanoreceptors found in hairless skin, external genitalia, joints, ligaments, lymph nodes, prostate, bladder, etc. While they are documented in the pancreas of cats, their presence in the normal pancreas remains speculative. PURPOSE: The present study therefore investigated the distribution of PCs in the normal human pancreas and compared the findings with those in several other animal species. METHODS: The study subjects included 74 human cadaver specimens, 3 Cynictis penicillata, 2 Saguinus mystaxs, 1 Felis domes

Lessons from the Barn to the Operating Suite: A Comprehensive Review of Animal Models for Fetal Surgery.

The International Fetal Medicine and Surgery Society was created in 1982 and proposed guidelines for fetal interventions that required demonstrations of the safety and feasibility of intended interventions in animal models prior to application in humans. Because of their short gestation and low cost, small animal models are useful in early investigation of fetal strategies. However, owing to the anatomic and physiologic differences between small animals and humans, repeated studies in large animal models are usually needed to facilitate translation to humans. Ovine (sheep) models have been use

From sound waves to molecular and cellular mechanisms: Understanding noise‑induced hearing loss and pioneering preventive approaches (Review).

Noise-induced hearing loss (NIHL) is a significant and urgent global public health concern, arising from prolonged exposure to elevated levels of noise. This auditory impairment harms delicate inner ear structures, particularly the essential hair cells transmitting auditory signals to the brain. Recognized by the World Health Organization as a major contributor to worldwide hearing loss, NIHL requires a comprehensive examination of its molecular and cellular mechanisms. Animal models emerge as indispensable tools for unraveling these intricacies, allowing researchers to simulate and study the

Identification of an animal sucrose transporter.

According to a classic tenet, sugar transport across animal membranes is restricted to monosaccharides. Here, we present the first report of an animal sucrose transporter, SCRT, which we detected in Drosophila melanogaster at each developmental stage. We localized the protein in apical membranes of the late embryonic hindgut as well as in vesicular membranes of ovarian follicle cells. The fact that knockdown of SCRT expression results in significantly increased lethality demonstrates an essential function for the protein. Experiments with Saccharomyces cerevisiae as a heterologous expression s

A cross-species framework for classifying sound-movement couplings.

Sound and movement are entangled in animal communication. This is obviously true in the case of sound-constituting vibratory movements of biological structures which generate acoustic waves. A little less obvious is that other moving structures produce the energy required to sustain these vibrations. In many species, the respiratory system moves to generate the expiratory flow which powers the sound-constituting movements (sound-powering movements). The sound may acquire additional structure via upper tract movements, such as articulatory movements or head raising (sound-filtering movements).

[Management of dog bite trauma of the external genital organs].

Animal bites to the scrotum are rare, but potentially serious. In the light of a personal case, the authors discuss the management of these lesions. An animal bite comprises a triple infectious risk: bacterial, tetanus and rabies. Surgical exploration is indicated for assessment of the structures involved and debridement. The morbidity is related to the severity of the bite and the waiting time before consultation, and complications are rare. Orchidectomy is sometimes necessary, as in the case reported here, and the patient must be warned about this possibility. Antibiotic therapy is empirical

Threshold-distance measures from electrical stimulation of human brainstem.

Threshold current levels for electrical stimulation of a single human brainstem via an auditory prosthesis are compared with postmortem measures of the distance between the electrode and stimulated structures. The results compare well with the summary of threshold-distance measures from animal experiments compiled by Ranck. The correspondence between the human and animal data gives confidence that the extent of current spread (distance to stimulable neural units) can be well estimated from the current level at threshold for 200 microseconds/phase biphasic pulses. This is of particular interest

Experimental tympanosclerosis following infection with Streptococcus pyogenes and vitamin D3 intoxication.

A rat animal model was used to study the ultrastructure of submucosal calcifications induced in the middle ear following inoculation with Streptococcus pyogenes and high doses of parenteral vitamin D3. The morphological changes present in affected animals resembled the classical picture of tympanosclerosis. While calcification occurred about bacterial remnants and myelin structures, the most important calcification centers were lysosomal and non-lysosomal matrix vesicles in the extracellular spaces. These formed band-like calcifications close to the basal membrane without affecting the epithel

Incidence and anatomy of gaze-evoked nystagmus in patients with cerebellar lesions.

BACKGROUND: Disorders of gaze-holding--organized by a neural network located in the brainstem or the cerebellum--may lead to nystagmus. Based on previous animal studies it was concluded that one key player of the cerebellar part of this gaze-holding neural network is the flocculus. Up to now, in humans there are no systematic studies in patients with cerebellar lesions examining one of the most common forms of nystagmus: gaze-evoked nystagmus (GEN). The aim of our present study was to clarify which cerebellar structures are involved in the generation of GEN. METHODS: Twenty-one patients with

Behavioural and neural-system analyses of the actions of anxiolytic drugs.

Anti-anxiety drugs (including benzodiazepines, barbiturates and alcohol) have a distinctive profile of behavioural action in animal species ranging from goldfish to chimpanzee. This profile may be summarised as a blockade of three kinds of reaction (behavioural inhibition, preparation for vigorous action, and increased attention to the environment) in response to any of three kinds of stimuli (novelty, stimuli associated with punishment, or stimuli associated with frustrative nonreward). On this basis, one may postulate a 'behavioural inhibition system' in the brain, responsible for organising
