The endothelial cells of the capillary network in the brain, in conjunction with pericytes and astrocyte end feet, form the blood-brain barrier (BBB). This highly selective cellular barrier helps to protect and maintain brain homeostasis. This barrier function must be circumvented to deliver therapeutics to the brain, a growing need owing to the increasing incidence in neurological disorders in progressively aging populations. Animal models may exhibit species-specific differences in response, ultimately leading to problems with predicting efficacy of treatments in humans, and conventional in

Evaluating skin sensitization is crucial for the safety of cosmetics and pharmaceuticals. Regulatory bans on animal testing have spurred the development of New Approach Methodologies (NAMs), Integrated Testing Strategies (ITS), and Next Generation Risk Assessment (NGRA). Defined Approaches (DAs) that combine various NAMs have been developed to assess skin sensitization hazards and potency, as recognized by the OECD. However, deriving a quantitative point of departure (PoD) through NAMs in accordance with NGRA remains challenging. This study evaluated the skin sensitization hazard and potency o

Consumer product manufacturers utilise a spectrum of alternative ocular irritation assays, as these tests do not require the use of live animals. Despite their usefulness, no regulatory-accepted assay assesses the reversibility of ocular damage, a key criterion of GHS ocular classification, like the rabbit eye test (i.e., Draize Rabbit Eye Test [DRET]) . The Porcine Corneal Opacity Reversibility Assay (PorCORA), an ex vivo intact corneal tissue culture model, predicts the reversibility of damage by ocular irritants. Inclusion of the damage reversibility endpoint in the PorCORA supplements othe

Until now, the Draize test on rabbits has been the only test performed to anticipate ocular toxicity of pharmaceutical compounds, mainly irritation. The OECD is urging the scientific community to develop and validate alternative methods to reduce the need for animal testing. Since the models and tests used cannot reflect the entire biologic response, it is necessary to combine them into integrated approaches to testing and assessment (IATA) to obtain robust data. IATAs, along with adverse outcome pathways (AOP) that encompass molecular cascades and key events, require the best combinations of

In a regulatory context, skin sensitization hazard and risk evaluations of manufactured products and their ingredients (e.g. cosmetics) are mandatory in several regions. Great efforts have been made within the field of 21st Century Toxicology to provide non-animal testing approaches to assess the skin allergy potential of materials (e.g. chemicals, mixtures, nanomaterials, particles). Mechanistic understanding of skin sensitization process through the adverse outcome pathway (AOP) has promoted the development of in vitro methods, demonstrating accuracies superior to the traditional animal test

INTRODUCTION: Alternative models are tools to replace and reduce the number of animals used in biomedical sciences, for either research or tests of industrial products. Several new alternative models have been developed in the most diverse fields. Their implementation has led to significant advances in the dermatological cosmetic industry, enabling chemical and molecular screening without animal use. However, limitations remain, particularly regarding tissue microenvironment complexity and systemic metabolic responses.
OBJECTIVES: The objective of this viewpoint is to present the existing alt

It is widely recognized that the ability of chemicals to sensitize, and the potency of those chemicals that are sensitizers, is related to their ability to covalently modify protein in the skin. With the object of putting non-animal-based prediction of skin sensitization on a more quantitative footing, a recent paper describes the development of the kinetic Direct Protein Reactivity Assay (kDPRA), in which a matrix of peptide depletion values for different reaction times and test chemical concentrations is generated and analyzed so as to derive a reactivity parameter, logkmax, which is used to

This paper offers a Bayesian Value-of-Information (VOI) analysis for guiding the development of non-animal testing strategies, balancing information gains from testing with the expected social gains and costs from the adoption of regulatory decisions. Testing is assumed to have value, if, and only if, the information revealed from testing triggers a welfare-improving decision on the use (or non-use) of a substance. As an illustration, our VOI model is applied to a set of five individual non-animal prediction methods used for skin sensitisation hazard assessment, seven battery combinations of t

Alternatives to mammalian testing are highly desirable to predict the skin sensitisation potential of agrochemical active ingredients (AI). The GARD assay, a stimulated, dendritic cell-like, cell line measuring genomic signatures, was evaluated using twelve AIs (seven sensitisers and five non-sensitisers) and the results compared with historical results from guinea pig or local lymph node assay (LLNA) studies. Initial GARD results suggested 11/12 AIs were sensitisers and six concurred with mammalian data. Conformal predictions changed one AI to a non-sensitiser. An AI identified as non-sensiti

Veterinary vaccines contribute to improved animal and human health and welfare by preventing infectious diseases. However, testing necessary to ensure vaccine effectiveness and safety can involve large numbers of animals and significant pain and distress. NICEATM and ICCVAM recently convened an international workshop to review the state of the science of human and veterinary vaccine potency and safety testing, and to identify priority activities to advance new and improved methods that can further reduce, refine and replace animal use. Rabies, Clostridium sp., and Leptospira sp. vaccines were

Serum is commonly used as a supplement to cell culture media. It provides a broad spectrum of macromolecules, carrier proteins for lipoid substances and trace elements, attachment and spreading factors, low molecular weight nutrients, and hormones and growth factors. The most widely used animal serum supplement is fetal bovine serum, FBS. Since serum in general is an ill-defined component in cell culture media, a number of chemically defined serum-free media formulations have been developed in the last two decades. Besides modern cell biological advances in cell and tissue culture and efforts

New approach methodologies (NAMs) originated in chemical hazard assessment, where two forces operate together: the scale of characterizing many substances, and a long-standing ethical and legal commitment to replacing animal testing. Drug development is weighted differently. It is a translational and decision problem applied to small sets of compounds at pharmacologically active exposures. As NAMs capabilities have expanded, the two domains have been conflated, and framing NAMs against animal studies as a binary choice risks undermining both. The so-called translatability crisis is not a singl

Cosmetic ingredients must be toxicologically assessed to determine their skin sensitizing potential. The in vitro human cell line activation test (h-CLAT; OECD TG 442E) addresses the activation of dermal dendritic cells by analyzing specific protein expression after exposure of THP-1 cells to the test chemical. According to the protocol, FITC-labeled antibodies are used for protein detection. However, some chemicals show strong autofluorescence at FITC-specific wavelengths so that antibody-specific signals cannot be distinguished appropriately from autofluorescence background. This leads to in

Research aimed at nonanimal approaches to provide the relevant information needed for the effective assessment of skin sensitization, for both hazard characterization and risk assessment purposes, is currently an area of high activity, stimulated by regulatory initiatives related to chemicals used in consumer products. The ability of a chemical to react covalently with protein or peptide nucleophiles in the skin is recognized as the key determinant in determining sensitization potency, and initiatives to develop peptide reactivity assays to replace animal testing have been undertaken recently.

Three selected photosensitizers (PS) with potential applications in PDT (ZnPcS2, TMPyP, PPIX) were evaluated for skin irritation, phototoxicity and sensitization using exclusively non-animal test methods. The experimental approach was based on OECD TG with protocol modifications to accommodate the specific requirements of photosensitizers testing. Reconstructed human epidermis (RhE) based assays were performed using adapted protocols derived from OECD TG 439 and OECD TG 498, while skin sensitization was assessed using methods based on OECD TG 442C and OECD TG 442D. TMPyP and PPIX showed no evi

Assessment of potential impacts of chemicals on the environment traditionally involves regulatory standard data requirements for acute aquatic toxicity testing using algae, daphnids, and fish (e.g., Organisation for Economic Co-operation and Development [OECD] test guidelines 201, 202, and 203, respectively), representing different trophic levels. In line with the societal goal to replace or reduce vertebrate animal testing, alternative bioassays were developed to replace testing with fish: the fish cell line RTgill-W1 acute toxicity assay (OECD test guideline 249) and the zebrafish embryo acu

The effects of ten test chemicals on thyroid sodium-iodide symporter (NIS), thyroid peroxidase (TPO), and deiodinases (DIOs) type I, II, and III were evaluated and compared in in vitro rat and human systems. Test chemicals known to directly affect TH levels in vivo were confirmed to effectively inhibit at least one of the tested in vitro endpoints, without significant disparities between species, and the test compounds known to not affect thyroid function were found ineffective. Interestingly, iodide transport blocker 5, a potent non-competitive iodine uptake inhibitor, exhibited effects beyon

The hazards and potency of skin sensitizers are traditionally determined using animal tests such as the local lymph node assay (LLNA); however, significant progress has been made in the development of non-animal test methods addressing the first three mechanistic key events of adverse outcome pathway in skin sensitization. We developed the epidermal sensitization assay (EpiSensA), which is a reconstructed human epidermis-based assay, by measuring four genes related to critical keratinocyte responses during skin sensitization. Four in vitro skin sensitization test methods (EpiSensA, direct pept
