Six components of the mammalian 2-oxo acid dehydrogenase complexes have previously been identified as M2 autoantigens in primary biliary cirrhosis. In this report, we present data showing that both polypeptide-specific and cross-reacting antibodies are present in patients' sera. Antibodies reacting with E2 of the pyruvate dehydrogenase complex cross-react with protein X but not with any other mammalian antigen. The main immunogenic region on protein X has been localized to within its single lipoyl domain. Polypeptide-specific antibodies bind to E1 alpha and E1 beta of the pyruvate dehydrogenas

Quantitative results of Mycobacterial BCG antigen, immunoglobulin and complement in rat skin lesions during the evolution of the granuloma reveal peak values of these factors at 49 days. The combination of antigen, Ig and complement, present extracellularly and in polymorphs at this time, indicates immune complex formation at an antigen-antibody ratio which may approximate to equivalence. This development coincides with mass lysis of host macrophages, and is followed by a sharp reduction in the antigen load. At 8 mth, surviving bacilli are coated with antibody and complement, and sequestrated

It has been claimed that human anti-VIII:C antibodies do not form stable complexes with factor VIII and this fact has hampered in the past the isolation of such antibodies. In this study the purification of human anti-VIII:C antibodies appearing in haemophiliac patients following replacment therapy has been achieved using two different systems. In a liquid phase system, purified human factor VIII was mixed with IgG from a haemophilic patient with a high titre antibody. Specific anti-VIII:C antibodies were recovered following filtration of the antigen-antibody complexes on Biogel A-5m, dissocia

BACKGROUND: Arboviral infections are characterized by heterogeneity in inflammatory responses and clinical outcomes. While antibodies are essential for protection, antibody-antigen immune complexes may modulate innate immune signaling through Fc receptor engagement. The extent to which immune complex signaling contributes to coordinated cytokine responses in human arboviral infection remains incompletely defined.
METHODS: We conducted a mechanistic immunology study nested within a prospective cohort of patients with acute arboviral infection in southern India. Using paired ex vivo stimulation

The Mac-ELISA technique is based on the capture of IgM antibodies by the formation of an antigen-antibody complex in biological samples. IgM anti-yellow fever antibodies are captured by anti-IgM present on the polystyrene surface of the wells of the assay plate. The steps of the technique consist of stages (serum, antigen and anti-flavivirus conjugate - 6B6C-1), with incubation intervals at 37°C and washing. When adding the substrate in the next step, positive reactions (those containing the antigen-antibody complex) are blue in color by the action of the TMB substrate on the peroxidase enzyme.

Determining the structural basis of antigen recognition by antibodies and T cell receptors (TCRs) provides critical insights into effective immune targeting and can inform design of biotherapeutics and vaccines. Accurate computational modeling of antibodies and TCRs in complex with their targets poses a major challenge for predictive methods, including AlphaFold, which is generally accurate for modeling protein complexes but has shown limited success for immune recognition. In this study we assessed the performance of AlphaFold2, AlphaFold3, increased sampling protocols, and related deep learn

Mutation-induced changes in binding free energy (ΔΔG) at antibody-antigen interfaces are important for antibody optimization, mutational scanning, and viral immune escape assessment. However, computational prediction remains challenging because antibodies and antigens have distinct sequence backgrounds, mutation effects are often localized at interfaces, and related complexes may remain across training and evaluation partitions. We present AbAgMut-GNN as a task-oriented paired graph framework that coordinates established sequence and geometric learning components around explicit comparison of

Serial blood samples were obtained from 21 homosexuals who had developed symptomatic primary infection with human immunodeficiency virus (HIV) after a median incubation time of 14 days. During the first two weeks after the onset of illness HIV antigen (p24) was detected in the blood by enzyme linked immunosorbent assay (ELISA). During the second and third weeks after the onset of illness p24 antibody was detected by Western blot assay and antigen concentrations rapidly decreased to undetectable values. Dissociation of antigen-antibody complexes showed complexed antigen during the phase of decl

The samples of sera and pleural fluid from sick children have been analyzed by means of EIA techniques. To detect the time course of antibody production, the antigenic preparations of pneumococci (monovalent capsular polysaccharides, polyvalent polysaccharide vaccine and complex pneumococcal antigen) have been used. Antibody response observed in the forms of pneumococcal infection, studied in this investigation, has proved to be highly variable. It is expedient to determine antibodies to polysaccharide antigens not earlier than on days 10-12 from the beginning of the disease. But, besides the

Sera from 34 children with recurrent parotitis were measured by indirect immunofluorescence technique for antibody levels to several Epstein-Barr virus (EBV) antigens. IgG antibodies to EBV-capsid antigen (VCA) and EBV-associated nuclear antigen, often at high titres, were found in 29 of these patients, of whom 20 also had IgA antibody to VCA. Antibody level to early antigen complex (R and D) rose in 19 patients, of whom 18 had antibody to the R component alone. The abnormal patterns of EBV antibodies persisted for 3-14 months during and after the illness in 8 patients. These observations sugg

The serum markers of hepatitis B virus (HBV) infection are complex, mainly including HBV surface antigen (HBsAg) and antibody to HBsAg, HBV e antigen (HBeAg) and antibody to HBeAg, antibody to HBV core antigen. The natural history of chronic HBV infection is complex and variable and greatly influenced by the age of infection, the level of HBV replication, HBV genotype and mutations, presence of HBeAg, ALT level, coinfection with HCV and/or HDV, host immune status. Four stages of chronic HBV infection are the immune tolerant phase, the immune clearance phase, the low or non-replicative phase, a

An assay specific for immune complexes containing Entamoeba histolytica antigens has been developed. Rabbit antibody specific for human gamma globulin is attached to a solid phase (nitrocellulose). During the first incubation step polyethylene glycol-precipitated immune complexes are bound to the rabbit antibody attached to the solid phase. In the second incubation step a radiolabelled rabbit antibody specific for Entamoeba histolytica combines with E. histolytica antigens in the complexes. Quantitation of the radiolabel provides a direct measurement of the level of specific immune complexes.

PURPOSE: The aim of this study was to show that complex antibody patterns against retinal antigens in sera of patients with glaucoma, found in previous studies, are autoantibodies against human antigens.
METHODS: Sera of 179 patients were collected at the Department of Ophthalmology (University of Mainz, Germany): non-glaucomatous control patients (n=45), primary open-angle glaucoma (n=45), ocular hypertension (n=44), and normal tension glaucoma patients (n=45). The sera were tested against Western blots of human retinal antigens. IgG antibody patterns were analyzed by multivariate statistica

INTRODUCTION: The antigen uptake receptor DEC205 has been extensively studied in dendritic cells (DCs). By mediating antigen delivery to major histocompatibility complex (MHC) class II processing compartments, DEC205 facilitates efficient antigen presentation, a property that has been widely exploited for targeted antigen delivery to DCs.
AREAS COVERED: This review summarizes studies investigating various molecular strategies employing anti-DEC205 conjugates, as well as conjugates targeting other DC-specific receptors, to induce either immune activation or immune tolerance in animal models an

From a panel of monoclonal antibodies to Schistosoma mansoni, one that is specific to a shared cercarial and schistosomular antigen, and does not react with other major parasite species including Fasciola hepatica, was selected for use as an antigen-capture layer in a sandwich ELISA for detection of specific circulating immune complexes in the blood of S. mansoni-infected subjects from Sudan. The test, which identifies immune complexes of only a single antibody-bound antigen, is developed using human Ig class- and IgG subclass-specific enzyme (HRP)-antibody conjugates. Antigen-antibody interaction is crucial in autoimmune disease pathogenesis, as multiple sclerosis and neuromyelitis optica. Given that, autoantibodies are essential biomolecules, of which the myelin oligodendrocyte glycoprotein (MOG) can figure as a target. Here we combined Molecular Dynamics (MD), Steered Molecular Dynamics (SMD), and Atomic Force Microscope (AFM) to detail MOG recognition by its specific antibody. The complex model consisted of the MOG external domain interacting with an experimental anti-MOG antibody from the Protein Data Bank (1PKQ). Computational data demonstrated thirtee

Pemphigus foliaceus and pemphigus vulgaris are skin diseases in which antibodies against the cell surface of keratinocytes destroy the adhesion between epidermal cells, producing blisters. Patients with pemphigus foliaceus have antibodies to a complex of three polypeptides of 260, 160, and 85 kd (the foliaceus complex), whereas patients with pemphigus vulgaris have antibodies to a complex of 210-kd, 130-kd, and 85-kd polypeptides (the vulgaris complex). The 160-kd polypeptide of the foliaceus complex has been identified as desmoglein, a desmosomal glycoprotein. We suspected that the 85-kd comp
