Colonic Engyodontium fungus triggers neutrophil antimicrobial activity to suppress Lactobacillus johnsonii-derived glutamic acid-maintained Tregs.

Isolating commensal fungi from mouse intestines has been challenging, limiting our understanding of their role in intestinal immune homeostasis and diseases. Using an Fc fusion protein of the C-type lectin receptor Dectin-2, we successfully purified the commensal Ascomycota fungus Engyodontium sp. from mouse feces. Engyodontium enhances the antimicrobial activity of colonic neutrophils via the CARD9 pathway and exacerbates colitis by impairing the colonization of intestinal Lactobacillus johnsonii WXY strain. L. johnsonii produces high levels of l-glutamic acid by expressing the glutaminase-en

Talaromyces spp. are tunable producers of light-activated secondary metabolites with anticancer potential.

Photodynamic therapy (PDT) is a promising treatment strategy for resistant or non-surgical tumors. To develop photosensitizers tailored to specific cancer types, new chemical scaffolds are needed. Macrofungi have recently been identified as a potent source of such compounds, particularly dimeric anthraquinones. However, the sources previously described (i.e., dermocyboid Cortinarii) cannot be cultivated in a biotechnologically applicable way. Thus, in this proof-of-concept study, we explored Talaromyces species (Ascomycota) for their potential to produce similar light-activated metabolites. Si

Low-Concentration Oxidants for Mitigating Continuous Cropping Obstacles: Improving Soil Properties and Microbial Functionality.

Continuous cropping obstacles (CCO) are common and severe in ginger cultivation. The use of oxidants to improve CCO-affected soils remains insufficiently explored. This study evaluated the effects of urea hydrogen peroxide (UHP), sodium persulfate (PS), and peracetic acid (PAA) on soil microbial communities and physicochemical properties. In the short term, oxidant treatments, especially PAA, significantly enriched beneficial taxa (e.g., Firmicutes, Actinobacteriota, Bacillales, Ascomycota) while suppressing the pathogenic Cercozoa (- 38.1%), except under UHP. All oxidants significantly increa

Integrated molecular networking-guided discovery of antimicrobial secondary metabolites from Thyronectria pinicola.

Using an LC-MS/MS-based molecular networking-guided approach, three previously undescribed chlorinated resorcylic acid lactone derivatives (5-7), two new cleistanthane-type diterpenoids (11-12) and a new dimeric derivative (13), together with seven known metabolites, were isolated from a cereal culture of Thyronectria pinicola (Ascomycota: Hypocreales). T. pinicola was isolated from a fruiting body of the devils'tooth mushroom, Hydnellum peckii (Basidiomycota, Thelephorales). The structures of the isolated compounds were elucidated using 1D and 2D NMR spectroscopy and HRMS, and their absolute

Taxonomic Reevaluation of Blackwellomyces Isolates Preserved in Korean Agricultural Culture Collection and Description of Two New Species.

Blackwellomyces is a genus of entomopathogenic fungi in the family Cordycipitaceae (Hypocreales, Sordariomycetes, Ascomycota) that primarily infects arthropod larvae, particularly those of Lepidoptera. Despite its ecological significance, this genus remains poorly studied in Korea. During the re-identification of Korean Cordyceps sensu lato isolates preserved in the Korean Agricultural Culture Collection (KACC), 12 strains were found to belong to the genus Blackwellomyces. Accordingly, this study aimed to accurately identify and clarify the taxonomic status of these isolates. The 12 Blackwello

Bridging the knowledge gap: From poly(butylene adipate-co-terephthalatebutylene) degradation to CO2-generating mineralization under the synergistic effect of bacteria and fungi.

Poly(butylene adipate-co-terephthalate) (PBAT) is a promising polymer with excellent mechanical properties and biodegradability. However, knowledge gaps between its degradation and mineralization processes in soil hampers its environmental impact and application potential. In this study, we elucidated the degradation process of PBAT, starting with the degradation of high-molecular-weight polymers into 30 intermediates, before ultimately mineralized into CO2. Bacteria and fungi drove the degradation and mineralization of these intermediates. We discovered that PBAT was synergistically degraded

Microbial Life History Mediates the Drought-Induced Decrease in Wood Decomposition in Subtropical Forests.

Progressive intensification of drought regimes worldwide threatens wood decomposition - a critical biogeochemical process in forest carbon cycling. However, the interactions between microbial strategies, wood traits and microclimate in regulating wood decomposition remain poorly understood under drought conditions. In a throughfall exclusion experiment (control 35% and 70% rainfall reduction) across 12 tree species, we found that drought-induced reductions in wood CO2 efflux rates were primarily driven by wood traits (density and carbon reduction) and shifts in fungal K/r-strategies. Survival of Icelandic airborne microbes towards simulated atmospheric stress factors.

Surface microbes are aerosolized into the atmosphere by wind and events such as dust storms, wildland fires, and volcano eruptions. Only microbial cells that survive the various atmospheric stressors during their transportation will deposit and colonize new environments. These stressors include desiccation, oxidative stress, solar radiation, osmotic shock, and freeze-thaw cycles. In this paper, we specifically studied the survival of representative microbial model strains isolated from the atmosphere over pristine volcanic landscapes to understand their potential to successfully disperse to no

Secondary Metabolites from Fungi Microsphaeropsis spp.: Chemistry and Bioactivities.

Microsphaeropsis, taxonomically classified within the kingdom fungi, phylum Ascomycota, subphylum Deuteromycotina, class Coelomycetes, order Sphaeropsidales, and family Sphaeropsidaceae, exhibit a ubiquitous distribution across various geographical regions. These fungi are known for their production of secondary metabolites, characterized by both structural novelty and potent biological activity. Consequently, they represent a significant reservoir for the advancement of novel pharmaceuticals. In this paper, a systematic review was present, marking the analysis of secondary metabolites synthes

Lucimycin, an antifungal peptide from the therapeutic maggot of the common green bottle fly Lucilia sericata.

We report the identification, cloning, heterologous expression and functional characterization of a novel antifungal peptide named lucimycin from the common green bottle fly Lucilia sericata. The lucimycin cDNA was isolated from a library of genes induced during the innate immune response in L. sericata larvae, which are used as therapeutic maggots. The peptide comprises 77 amino acid residues with a molecular mass of 8.2 kDa and a pI of 6.6. It is predicted to contain a zinc-binding motif and to form a random coil, lacking β-sheets or other secondary structures. Lucimycin was active against f

The Cbp1 protein is a peptidorhamnomannan adhesin that contributes to the Sporothrix schenckii virulence.

Sporothrix schenckii pathogenesis has been extensively studied to date, but several proteins of unknown function remain to be assigned to the infectious process. Previously, proteomic data showed that one protein was highly abundant (SPSK_02764) in cell wall peptidorhamnomannan, but no obvious homologs or sequence features hinted at the activity or function of this protein. In this work, in vitro analysis showed that this protein is highly glycosylated and is an adhesin with specificity for type-I and type-II collagen, renaming this protein as Collagen Binding Protein 1 or Cbp1. Yeast-like cel

Characterization of the fungal microbiota (mycobiome) in healthy and dandruff-afflicted human scalps.

The human scalp harbors a vast community of microbial mutualists, the composition of which is difficult to elucidate as many of the microorganisms are not culturable using current culture techniques. Dandruff, a common scalp disorder, is known as a causative factor of a mild seborrheic dermatitis as well as pityriasis versicolor, seborrheic dermatitis, and atopic dermatitis. Lipophilic yeast Malassezia is widely accepted to play a role in dandruff, but relatively few comprehensive studies have been reported. In order to investigate fungal biota and genetic resources of dandruff, we amplified t

Fulminating fungal sinusitis caused by Valsa sordida, a plant pathogen, in a patient immunocompromised by acute myeloid leukemia.

We describe a case in which a patient immunocompromised by acute myeloid leukemia experienced acute fulminating invasive fungal sinusitis followed by pneumonia and then death. Though the microbiology of the pneumonia could not be directly investigated, nasal lesions revealed fungal mycelium. Valsa sordida was consistently cultured from a biopsied sample. The fungus was identified to the genus level based on morphology in culture and DNA sequence homology, and then was placed at species level by means of phylogenetic analysis of the nuclear ribosomal internal transcribed spacer region. The fung

Diabetic foot exacerbates gut mycobiome dysbiosis in adult patients with type 2 diabetes mellitus: revealing diagnostic markers.

Type 2 diabetes mellitus (T2DM) is globally recognized as a significant health concern, with diabetic foot (DF) identified as a severe long-term complication that can lead to tissue death or amputation. The discovery of the impact of mycobiota, a diverse group of multicellular eukaryotes in the gut microbiome, on the onset of endocrine disorders holds great significance. Therefore, this research aimed to examine variations in fungal mycobiome and identify potential biomarkers for T2DM and T2DM-DF. Fecal and blood samples were collected from 33 individuals with T2DM, 32 individuals with T2DM-DF
