Docosahexaenoic Acid Modulates Autophagy and Confers Neuronal Resilience under Hypoxia-Reoxygenation Stress.

Hypoxia dramatically worsens neuronal death after injuries and strokes, as neurons rely heavily on oxidative phosphorylation for energy. This study investigated the effects of docosahexaenoic acid (DHA) in reducing cell death in nerve cells. Neuron-like cells, nerve growth factor-differentiated PC12 (NGFDPC12) cell cultures, with or without DHA pretreatment, were exposed to hypoxic conditions (0.5% O₂) for 12 to 48 h, followed by reoxygenation. Results showed that hypoxia caused a significant increase in the stress-response gene HIF-1α, with mRNA levels rising 5.47- and 6-fold at 24 and 48 h,

Differential myocardial responses to indobufen and aspirin across clinical and experimental models of myocardial infarction.

BACKGROUND: Aspirin remains the cornerstone of antiplatelet therapy for acute myocardial infarction (AMI); however, its use is limited in patients with aspirin intolerance, hypersensitivity, or high bleeding risk. Indobufen, a reversible cyclooxygenase-1 inhibitor, has emerged as a potential alternative, yet its cardioprotective effects beyond platelet inhibition and the underlying mechanisms remain incompletely understood. METHODS: Clinical serum samples were collected from patients with ST-segment elevation myocardial infarction (STEMI) receiving aspirin or indobufen therapy. In parallel, m

Abnormal Cholesterol Metabolism and Lysosomal Dysfunction Induce Age-Related Hearing Loss by Inhibiting mTORC1-TFEB-Dependent Autophagy.

Cholesterol is a risk factor for age-related hearing loss (ARHL). However, the effect of cholesterol on the organ of Corti during the onset of ARHL is unclear. We established a mouse model for the ARHL group (24 months, n = 12) and a young group (6 months, n = 12). Auditory thresholds were measured in both groups using auditory brainstem response (ABR) at frequencies of 8, 16, and 32 kHz. Subsequently, mice were sacrificed and subjected to histological analyses, including transmission electron microscopy (TEM), H&E, Sudan Black B (SBB), and Filipin staining, as well as biochemical assays such

Enhancing endometrial receptivity: the roles of human chorionic gonadotropin in autophagy and apoptosis regulation in endometrial stromal cells.

Inadequate endometrial receptivity often results in embryo implantation failure and miscarriage. Human chorionic gonadotropin (hCG) is a key signaling molecule secreted during early embryonic development, which regulates embryonic maternal interface signaling and promotes embryo implantation. This study aimed to examine the impact of hCG on endometrial receptivity and its underlying mechanisms. An exploratory study was designed, and endometrial samples were obtained from women diagnosed with simple tubal infertility or male factor infertile (n = 12) and recurrent implantation failure (RIF, n =

Abnormal activation of MAPKs pathways and inhibition of autophagy in a group of patients with Zellweger spectrum disorders and X-linked adrenoleukodystrophy.

BACKGROUND: Zellweger spectrum disorders (ZSD) and X-linked adrenoleukodystrophy (X-ALD) are inherited metabolic diseases characterized by dysfunction of peroxisomes, that are essential for lipid metabolism and redox balance. Oxidative stress has been reported to have a significant role in the pathogenesis of neurodegenerative diseases such as peroxisomal disorders, but little is known on the intracellular activation of Mitogen-activated protein kinases (MAPKs). Strictly related to oxidative stress, a correct autophagic machinery is essential to eliminated oxidized proteins and damaged organel

Flutamide induces uterus and ovary damage in the mouse via apoptosis and excessive autophagy of cells following triggering of the unfolded protein response.

Intrauterine exposure to flutamide not only causes abnormal development of the reproductive organs in male offspring, but also damages ovaries and uteri. The unfolded protein response (UPR) is believed to play an important role in embryo development and teratogenic processes. In the present study, pregnant mice were administered either flutamide (300mg kg-1 day-1, p.o.) on an equivalent volume of soybean oil (control) on Days 12-18 of gestation. Eight weeks after birth, female offspring in the flutamide-treated group had a lower bodyweight and lower ovarian and uterine weights, but there was n

Integrated transcriptomic analysis identifies lysosomal autophagy-related genes in sarcopenia.

Sarcopenia (SARC) presents considerable challenges to the quality of life for the elderly and exerts a significant economic strain on healthcare systems. Contemporary therapeutic methods, such as physical activity, dietary supplementation, and medication, frequently demonstrate restricted effectiveness and significant individual variation. This study seeks to identify new therapeutic targets through the analysis of lysosomal autophagy-related differentially expressed genes (LARDEGs) associated with SARC. Differentially expressed genes(DEGs) related to SARC were identified through an extensive

Thymoquinone enhances kidney function in obesity-induced renal impairment by targeting redox balance, inflammation, autophagy, and apoptosis.

The prevalence of kidney impairment is associated with the consumption of a high-fat diet (HFD) and obesity via the dysregulation of various biological processes. Thymoquinone (TQ) is an active component of black cumin with antioxidant and anti-inflammatory effects. The present study examined the ability of TQ to mitigate kidney injury and assessed the interconnected biological mechanisms that improve kidney function and structure. Adult male rats were fed a HFD for 12 weeks. HFD-fed rats were orally administered TQ from week 9 to week 12. TQ improved kidney function by decreasing the levels o

Proteomics and Metabolomics Reveal Novel Impacts of Choline Supply on Calf Hepatocytes Experiencing Accumulation During a Fatty Acid Challenge.

Background/Objectives: Exposure to high and sustained levels of non-esterified fatty acids (NEFA) in the peripartal period is the main cause of fatty liver disease in dairy cows. Rumen-protected choline is often fed as part of the nutritional management of peripartal cows, with in vivo and in vitro data indicating positive effects of this nutrient on alleviating liver lipid accumulation. Although hepatic molecular mechanisms associated with choline supply have been studied using a target gene, protein, or metabolite approach, application of high-throughput technologies could vastly enhance fun

circ_SIRT1 upregulates ATG12 to facilitate Imatinib resistance in CML through interacting with EIF4A3.

Imatinib is the current gold standard for patients with chronic myeloid leukemia (CML). However, the primary and acquired drug resistance seriously limits the efficacy. To identify novel therapeutic target in Imatinib-resistant CML is of crucial clinical significance. CircRNAs have been demonstrated the essential regulatory roles in the progression and drug resistance of cancers. In this study, we identified a novel circRNA (circ_SIRT1), derived from the SIRT1, which is up-regulated in CML. The high expression of circ_SIRT1 is correlated with drug resistance in CML. Knockdown of circ_SIRT1 reg

Role of AMPKα2 in regulating the IRE1α-JNK pathway in metabolic dysfunction-associated fatty liver disease.

OBJECTIVES: Over 25% of the global population is affected by metabolic dysfunction-associated fatty liver disease (MAFLD), yet its pathogenesis remains unclear. Endoplasmic reticulum stress (ERS) may be involved in the onset and progression of MAFLD. Adenosine 5'-monophosphate-activated protein kinase α2 (AMPKα2), a key regulator of hepatic energy metabolism, may influence MAFLD development via ERS modulation. This study aims to investigate the role of AMPKα2 in a high-fat diet-induced MAFLD mouse model and its regulatory effect on the inositol-requiring enzyme 1 alpha (IRE1α)-c-Jun N-terminal

Macrophage TRIM21 knockout inhibits septic acute lung injury by downregulating autophagy regulator protein ubiquitination.

Acute lung injury (ALI) caused by sepsis is a fatal disease characterized by an systemic inflammatory response to invading pathogens. Inducing macrophage macroautophagy/autophagy is a critical strategy to combat the inflammatory response in septic ALI. The E3 ubiquitin ligase TRIM21 plays important roles in autophagy. However, the mechanism connecting macrophage TRIM21-associated autophagy to ALI development remains unclear. Therefore, this study was aimed to investigate the role of macrophage TRIM21 in septic ALI in human and mice. TRIM21 levels were significantly increased in the macrophages

STZ-induced gestational diabetes exposure alters PTEN/AKT/mTOR-mediated autophagy signaling pathway leading to increase the risk of neonatal hypoxic-ischemic encephalopathy.

Exposure to gestational diabetes mellitus (GDM) during pregnancy has significant consequences for the unborn baby and newborn infant. However, whether and how GDM exposure induces the development of neonatal brain hypoxia/ischemia-sensitive phenotype and the underlying molecular mechanisms remain unclear. In this study, we used a late GDM rat model induced by administration of streptozotocin (STZ) on gestational day 12 and investigated its effects of GDM on neonatal brain development. The pregnant rats exhibited increased blood glucose levels in a dose-dependent manner after STZ administration
