Babesia microti: Breaking the culture barrier.

Babesia microti poses a significant threat to human health, underscoring the need for an improved in vitro culture system to reduce reliance on animal models and support drug and vaccine screening. Key challenges include the tropism of preferential host red blood cells (RBCs), parasite metabolic needs, culture medium formulation, and optimization of the microaerophilic environment (oxygenation).

Babesia ovata: Taxonomy, phylogeny and epidemiology.

Babesia ovata, which is transmitted by Haemaphysalis longicornis, is an intraerythrocytic protozoan parasite of cattle. Based on its morphology, B. ovata is classified as a large-type Babesia. The developmental stages of B. ovata have been described both in cattle and the tick vector. In infected adult female ticks, the parasite is transovarially transmitted to the tick eggs. The sexual reproduction of B. ovata has been demonstrated in the tick midgut. The diagnostic tools that are currently available for the specific detection of B. ovata in cattle include microscopy and polymerase chain reac

Babesia divergens-like infection, Washington State.

Most reported U.S. zoonotic cases of babesiosis have occurred in the Northeast and been caused by Babesia microti. In Washington State, three cases of babesiosis have been reported previously, which were caused by WA1 (for "Washington 1")-type parasites. We investigated a case of babesiosis in Washington in an 82-year-old man whose spleen had been removed and whose parasitemia level was 41.4%. The complete 18S ribosomal RNA gene of the parasite was amplified from specimens of his whole blood by polymerase chain reaction. Phylogenetic analysis showed the parasite is most closely related, but no

Molecular diagnosis and phylogenetic analysis of Babesia bigemina and Babesia bovis hemoparasites from cattle in South Africa.

BACKGROUND: Babesia parasites, mainly Babesia bovis and B. bigemina, are tick-borne hemoparasites inducing bovine babesiosis in cattle globally. The clinical signs of the disease include, among others, anemia, fever and hemoglobinuria. Babesiosis is known to occur in tropical and subtropical regions of the world. In this study, we aim to provide information about the occurrence and phylogenetic relationship of B. bigemina and B. bovis species in cattle from different locations in nine provinces of South Africa. A total of 430 blood samples were randomly collected from apparently healthy cattle

Macrophages are critical for cross-protective immunity conferred by Babesia microti against Babesia rodhaini infection in mice.

Although primary infection of mice with Babesia microti has been shown to protect mice against subsequent lethal infection by Babesia rodhaini, the mechanism behind the cross-protection is unknown. To unravel this mechanism, we investigated the influence of primary infection of mice with nonlethal B. microti using different time courses on the outcome of subsequent lethal B. rodhaini infection. Simultaneous infections of mice with these parasites resulted in rapid increases in parasitemia, with 100% mortality in BALB/c mice, as observed with control mice infected with B. rodhaini alone. In con

Babesia canis canis, Babesia canis vogeli, Babesia canis rossi: differentiation of the three subspecies by a restriction fragment length polymorphism analysis on amplified small subunit ribosomal RNA genes.

The parasites Babesia canis and Babesia gibsoni (phylum Apicomplexa) are responsible for canine babesiosis throughout the world. Babesia canis was previously described as a group of three biologically different subspecies, namely B. canis canis, B. canis vogeli, and B. canis rossi. We report partial sequences of small subunit ribosomal RNA gene (ssu-rDNA) of each subspecies amplified in vitro with primers derived from a semi-conserved region of the ssu-rDNA genes in other Babesia species. The polymerase chain reaction combined with a restriction fragment length polymorphism analysis, using Hin

Babesia vulpes and Babesia banethi infections in red foxes: molecular and morphological detection with clinical and cytological findings.

Red foxes are widespread carnivorans acting as reservoirs of Babesia vulpes , which is highly prevalent in foxes, yet its clinical impact remains poorly understood. The description of Babesia banethi raised knowledge gaps regarding pathogenicity and life cycle. This study investigated the occurrence of Babesia spp. in red foxes from southern Italy. Overall, 54 red foxes were clinically examined, sampled for hemato-biochemical analyses, and screened for Babesia spp. using microscopy and PCR. Necropsies were performed on 20 animals. Ticks were collected, identified, and dissected for cytological

The first genetic study of Babesia caballi, Theileria equi, and Babesia occultans in Dermacentor spp. in Russian Siberia.

Equine piroplasmosis (EP) is a worldwide infection of Equides caused mainly by Theileria equi and Babesia caballi. Recently, these agents have been found in horses in the Republic of Altai. To identify probable vectors for EP agents in Western Siberia, 443 adult questing Dermacentor spp. (199 D. silvarum, 236 D. nuttalli, and 8 D. marginatus) were collected from vegetation in four districts of the Republic of Altai and examined for the presence of Piroplasmida DNA. Three Piroplasmida species were identified. Babesia caballi was found in ten (2.3%) Dermacentor spp., including eight D. silvarum

Effect of protease inhibitors on the intraerythrocytic development of Babesia microti and Babesia duncani, the causative agents of human babesiosis.

Human babesiosis is a malaria-like, tick-borne infectious disease with a global distribution. Babesiosis is caused by intraerythrocytic, apicomplexan parasites of the genus Babesia. In the United States, human babesiosis is caused by Babesia microti and Babesia duncani. Current treatment for babesiosis includes either the combination of atovaquone and azithromycin or the combination of clindamycin and quinine. However, the side effects of these agents and the resistance posed by these parasites call for alternative approaches for treating human babesiosis. Proteases play several roles in the c

Inhibitory effect of cyclophilin A from the hard tick Haemaphysalis longicornis on the growth of Babesia bovis and Babesia bigemina.

Haemaphysalis longicornis is known as one of the most important ticks transmitting Babesia parasites in East Asian countries, including Babesia ovata and Babesia gibsoni, as well as Theileria parasites. H. longicornis is not the natural vector of Babesia bovis and Babesia bigemina. Vector ticks and transmitted parasites are thought to have established unique host-parasite interaction for their survival, meaning that vector ticks may have defensive molecules for the growth control of parasites in their bodies. However, the precise adaptation mechanism of tick-Babesia parasites is still unknown.

Development of a rapid immunochromatographic test for simultaneous serodiagnosis of bovine babesioses caused by Babesia bovis and Babesia bigemina.

With the objective of developing a simpler diagnostic alternative, a rapid immunochromatographic test (BoiICT) was constructed for the simultaneous detection of Babesia bovis- and Babesia bigemina-specific antibodies using B. bovis recombinant merozoite surface antigen-2c and B. bigemina recombinant rhoptry-associated protein-1. The BoiICT selectively detected specific antibodies to B. bovis and B. bigemina. All sera from cattle infected with other protozoan parasites (i.e., Cryptosporidium parvum, Neospora caninum, and Theileria orientalis) showed negative results in the BoiICT. The relative

Immunity to Babesia in mice. III. The effects of corticosteroids and anti-thymocyte serum on mice immune to Babesia rodhaini.

BALB/c mice, immunized against Babesia rodhaini by an amicarbalide controlled infection, were exposed to selective immunosuppressive treatment with corticosteroids and anti-thymocyte serum (ATS) respectively. Hydrocortisone acetate, 100 mg/kg, given i.p. six times during the three weeks after challenge inoculation caused a rising parasitaemia and high mortality (6/7). Dexamethasone in the drinking water at 20 mg/l or 10 mg/l for 22 days had a similar suppressive effect on the protection against B. rodhaini. Mortality, 100% at the dose rate of 20 mg/l and 50% at 10 mg/l, occurred both in challe
