Bacteroides pyogenes is naturally found in the oral microbiome of cats and dogs and hence exposure, especially bites from these animals, is a major risk factor for human infections. B pyogenes is known to cause infections that persist despite antibiotic treatment and can have serious clinical outcomes. We present a novel case of complex lung abscesses associated with B pyogenes infection. A 55 year old man presents with a 3-month history of productive cough, night sweats, and 5 kg weight loss. An initial chest radiograph revealed mass-like opacities in the right upper lobe (RUL), right middle

Anaerobic bacteria of the genus Bacteroides are a large group of commensal microorganisms that colonize the human and animal digestive tract. The genus Bacteroides and the closely related genus Parabacteroides include the Bacteroides fragilis group (BFG) of potentially pathogenic bacteria which are frequently isolated from patients with anaerobic infections. The aim of this study was to assess the antimicrobial resistance of environmental strains of the Bacteroides fragilis group. Strains were isolated from human feces, hospital wastewater, influent (UWW) and effluent (TWW) wastewater from a w

An 82-year-old Japanese man, who presented with a fever and abdominal pain, was admitted to our hospital. According to enhanced computed tomography images, the probable diagnosis was abdominal aortic mycotic aneurysm. Eight sets of blood cultures obtained from the patient were negative. Despite administering treatment with vancomycin and ceftriaxone, the aneurysm progressively enlarged. He underwent open debridement surgery and in situ replacement because of an aneurysmal rupture. Bacteroides fragilis was isolated from the tissue culture of the aortic wall. Metronidazole was administered and d

BACKGROUND: To review the presentation of Bacteroides fragilis (B fragilis) spondylodiscitis.
METHODS: Two investigators independently searched the published English, Spanish and French languages literature through September 2005 using MEDLINE (1966-2005). We included all reported cases of vertebral osteomyelitis or spondylodiscitis caused by B fragilis, not related to sacral decubitus ulcers, in adults (age 16 yr and above). A third author independently reviewed all articles and extracted data for accuracy. The final pool of eligible publications included 11 articles, publication dates rangi

During a 22-year period, 13 patients with hematologic diseases developed bacteremia caused by the Bacteroides fragilis group, with a frequency which remained almost unchanged. Nine patients (69%) had polymicrobial infections. Acute leukemia was the most common underlying disease. The lower intestinal tract (necrotizing enterocolitis and anorectal abscesses) was the most common source of infection. Prior antibiotic therapy was the most frequent host condition before bacteremia, followed by cancer chemotherapy, neutropenia, thrombocytopenia and hypoproteinemia. Septic shock occurred only in seve

Bacteroides fragilis when inoculated intragastrically induced abscess formation in the mice of which the intestinal microbial flora had been disturbed by the prior intake of streptomycin and erythromycin in their drinking water. The lesion usually developed at the left axillary lymph node one week after inoculation and persisted for at least two months. Without the antibiotic treatment, B. fragilis did not induce such abscess formation. Although B. fragilis was isolated from blood after inoculation, intravenous administration of B. fragilis into mice did not induce the abscess formation. There

Current problems of antibiotic resistance in the Bacteroides fragilis group are reviewed. The original susceptibility (before 1976) of this group of strict anaerobic microorganisms to some beta-lactams, tetracyclines and lincosamides is presently severely damaged, and new mechanisms of resistance, such the enzymatic inactivation of chloramphenicol, seem to arise. Conjugative transfer of chromosomal resistance genes appears to be the main strategy for the spread of resistance and in various cases plasmids are involved. The origin of such resistance genes remains obscure. There is a possibility

Over the past eight years, a Wistar rat model for intraabdominal sepsis has been developed and used to document the role of obligate anaerobes in this infections. The ability of Bacteroides fragilis alone to provoke abscesses in this model system is due to a species-specific capsular polysaccharide. It has been shown that active immunization of rats with capsular polysaccharide of B. fragilis protects these animals against the development of abscesses after intraperitoneal challenge with this species. Passive transfer of hyperimmune globulin provided protection against B. fragilis bacteremia i

Electron transport components involved in H2 oxidation were studied in membranes from Rhizobium japonicum bacteroids. Hydrogen oxidation in membranes was inhibited by antimycin A and 2-n-heptyl-4-hydroxyquinoline-N-oxide with Ki values of 39.4 and 5.6 microM, respectively. The inhibition of H2 uptake by cyanide was triphasic with Ki values of 0.8, 9.9, and 93.6 microM. This result suggested that three cyanide-reactive components were involved in H2 oxidation. H2-reduced minus O2-oxidized absorption difference spectra showed peaks at 551.5 and 560 nm, indicating the involvement of c- and b-type

Carbenicillin has been advocated for treatment of infections caused by Bacteroides fragilis and other anaerobic bacteria. Wide-scale use of the drug in this setting could result in a substantial increase in carbenicillin-resistant Pseudomonas aeruginosa, an effect that would have serious implications. Thirty-four strains of B. fragilis, one-half from bacteremic infections, were tested in vitro, and penicillin G was found to be twice as active as carbenicillin on an equal weight basis; 94% of the strains were inhibited by 32 microgram of penicillin/ml, a level easily achieved therapeutically. Oxygen uptake and reduction of C2H2 by bacteroids was found to be inhibited by low concentrations of cyanide and azide. However, oxygen uptake was not completely suppressed even by 10(-3) M KCN. Cyanide-resistant respiration was not inhibited by salicyl-hydroxamic acid, and seemed to be accomplished at the account of autoxidable flavo-proteins. A small light-reversible inhibition of respiration by carbon monoxide was found only in the bacteroids with a high rate of nitrogen fixation. Rotenone, antimycin A, and tenoyltrifluoroacetone inhibited oxygen uptake and methylene reduction. Nitrogen fix

BACKGROUND: Colorectal cancer (CRC) progression is closely associated with gut microbiota. Bacteroides thetaiotaomicron (B.theta), a key species in the human cecal and colonic microbiota, plays an incompletely defined role in CRC progression.
METHODS: CRC animal models were established utilizing immunocompetent C57BL/6 mice and immunodeficient BALB/C nude mice. Histopathological changes were assessed by H&E staining. Immune cell infiltration was evaluated by IHC. CD8+ T cells and cancer cells were treated with B.theta culture supernatant. Cytokine secretion was measured by ELISA. Surface prot

Gut dysbiosis has been implicated in epilepsy, yet probiotic efficacy and mechanisms remain unclear. Here, we identify that Bacteroides fragilis (B. fragilis) is markedly reduced in children with epilepsy and show that oral B. fragilis administration suppresses seizures in both pentylenetetrazole- and kainic-acid-induced mouse models. Mechanistically, B. fragilis activates colonic choline acetyltransferase-positive (ChAT+) cells and enhances gut-vagus-brain cholinergic signaling, as demonstrated by vagal recordings, pharmacological blockade, and chemogenetic manipulation, identifying a colonic

Mucinous cancers are characterized by tumor gland cells secreting copious amounts of mucus that forms gel-like structures. Pseudomyxoma peritonei (PMP), a rare yet prototypical mucinous malignancy, exemplifies this pathology. Biologically inert mucus coats tumor cells, promoting colonization, immune evasion, and chemoresistance. Pathological mucus accumulation leads to intra-abdominal hypertension, refractory abdominal pain, abdominal adhesions, and intestinal obstruction because of the lack of rapid, efficient, and safe clinical strategies for mucin clearance. Current management relies on ext

Bacteriodes fragilis is an anaerobic bacterium found in the human intestinal flora. In this study, BfEno was targeted with a structure-based drug design approach because inhibition of this enzyme may prevent both the aerobic and anaerobic pathways due to its role in the glycolytic pathway. First, the gene encoding BfEno was cloned, expressed and the protein produced over 95% purity. The Km and Vmax values of BfEno were determined as 314.9 µM and 256.2 µmol/min.mg, respectively. Drug-like chemicals were retrieved from the ZINC database for high-throughput virtual screening analyses. As a result

Three strains of anaerobic Gram-stain-negative, short to longer rod-shaped bacteria isolated from the caecum of chicken in Indonesia were studied using a polyphasic taxonomic approach. These strains belonged to the genus Bacteroides, based on sequence analysis of 16S rRNA and hsp60 (groEL) genes, with similarities of 93.2-94.1 and 89.8-90.8 %, respectively, to the closest recognized species, Bacteroides coprocola JCM 17929T. Sugar fermentation and enzyme characteristics, cellular fatty acid profiles, menaquinone profiles and metabolic end products were also investigated. Furthermore, DNA-DNA h

The Gram-negative anaerobic bacterium B. fragilis is a member of the commensal flora of the human intestine, but is also frequently found in severe intra-abdominal infections. Several B. fragilis virulence factors have been implicated in the development of these infections. A B. fragilis protein of circa 60-kDa was identified as a putative plasminogen binding protein (Pbp). The corresponding gene was located, cloned, sequenced and the subcellular localization of the protein was investigated. Pbp was both determined in the outer membrane of B. fragilis and of E. coli that expressed the cloned p

Microbial therapy has emerged as a promising strategy for cancer treatment, driven by advances in synthetic biology that enable precise engineering of probiotic functions. Engineered bacteria offer several therapeutic advantages, including localized drug delivery, direct tumor killing, and immune modulation. Bacteria-based therapies offer promise for colorectal cancer treatment, yet safety issues of chassis cells and poor targeting remain key challenges. Here, we engineered a novel bacterial system based on the gut symbiont Bacteroides thetaiotaomicron, a naturally abundant and safe resident o
