Copper-induced alteration of muscarinic binding in bovine adrenal medulla.

In microsomes of bovine adrenal medulla, there were two affinity binding sites for carbamylcholine with a high and a low affinity. Copper (1-10 microM) largely enhanced the affinity of carbamylcholine at the low affinity binding site, with a slight increase in the affinity at the high affinity binding site. On the other hand, copper slightly decreased the binding affinity of pirenzepine and atropine. Thus, low concentrations of copper modulate the muscarinic receptors in the adrenal medulla by selectively increasing agonist affinity.

Glucocorticoid treatment decreases muscarinic receptor expression in canine airway smooth muscle.

Corticosteroids upregulate the beta-adrenergic pathway, but little is known about corticosteroid regulation of muscarinic pathways. Basenji-greyhound (BG) dogs treated for 3 days with methylprednisolone (MPS) but not deoxycorticosterone (DOC) had decreased numbers of muscarinic receptors in airway smooth muscle homogenates as determined by radioligand binding with 1-[3H]quinuclidinyl benzilate (vehicle control, 578 +/- 53 fmol/mg protein; MPS, 290 +/- 22 fmol/mg protein; DOC, 565 +/- 141 fmol/mg protein). Competition radioligand binding with the M2-selective antagonist tripitramine showed a de

Methyl mercury during late gestation affects temporarily the development of cortical muscarinic receptors in rat offspring.

Pregnant Sprague-Dawley rats were treated by gavage with a single dose of 8 mg/kg of methyl mercury on gestational day 15. Offspring of control and treated rats were killed at 14, 21 and 60 days of age. The binding characteristics of muscarinic receptors labelled in cortical membrane preparation by 3H-L-quinuclidinyl benzilate were studied together with the assessment of mercury level in the same brain area. Furthermore, the performance in passive avoidance tasks was evaluated in 8 weeks old rats. Perinatal exposure to methyl mercury significantly reduced the maximum number of muscarinic recep

Changes in choline acetyltransferase activity and high-affinity choline uptake, but not in acetylcholinesterase activity and muscarinic cholinergic receptors, in rat somatosensory cortex after sciatic nerve injury.

Selected cholinergic markers (choline acetyltransferase, acetylcholinesterase, muscarinic acetylcholine receptor, high-affinity choline uptake) were studied in the hindlimb representation areas of the rat somatosensory cortex and within the visual cortex 1 to 63 days after unilateral transection of the sciatic nerve. In the contralateral somatosensory cortex, peripheral deafferentation resulted in a significant reduction of choline acetyltransferase activity (by 15%) 3 days after sciatic nerve injury, and in a significant reduction of high-affinity choline uptake (by 30%) 1 day after nerve tra

In vivo autoradiographic demonstration of beta-adrenergic binding sites in adult rat type II alveolar epithelial cells.

Adult male rats were injected intravenously with the muscarinic binding probe 3H-Quinuclidinyl benzilate (QNB) or the beta-adrenergic probe 3H-dihydroalprenolol (DHA). Other rats were pre-treated with an intraperitoneal injection of a 500-fold excess of L-isoproterenol prior to the DHA. Light microscopic autoradiography of 0.5 micron sections of lung from the QNB group demonstrated very little labelling even after 6 months of exposure. In contrast, trachealis smooth muscle from these animals contained substantial labelling. Autoradiographs of lung from rats injected with DHA demonstrated label

Phencyclidine: behavioral and biochemical evidence against the anticholinergic hypothesis.

Phencyclidine (PCP) is known to have anticholinergic effects in various in vitro test systems and to inhibit the binding of muscarinic antagonists to rat brain membranes. In order to verify the anticholinergic properties of PCP, its interaction with oxotremorine (OXO), a muscarinic agonist, was studied in mice. OXO (1 mg/kg) in combination with PCP (10 mg/kg) was lethal in 100% of the mice studied. The lethality of this combination was completely reversed by 3.3 mg/kg methyl atropine bromide (MA), a quarternary muscarinic antagonist. Therefore, PCP appears to act as a muscarinic agonist in som

Muscarinic receptor binding and behavioral effects of atropoine following chronic catecholamine depletion or acetylcholinesterase inhibition in rats.

Rats were subjected to one of two experimental treatments: (1) intraventricular infusion of the catecholamine neurotoxin 6-hydroxydopamine (6-OHDA), known to permanently reduce brain dopamine and norepinephrine levels, or (2) chronic administration of the irreversible acetylcholinesterase inhibitor diisopropylfluorophosphate (DFP). Both treatments are believed to produce relative overactivity of cholinergic systems and to suppress forward locomotion. The anticholinergic agent atropine sulfate yielded excessive forward walking in otherwise chronically akinetic 6-OHDA-treated rats, whereas atrop

[Muscarinic cholinergic receptor imaging of parkinsonian brain using 11C-NMPB and PET].

11C-NMPB is a useful ligand which is used to measure brain muscarinic cholinergic receptors (mAChRs) for its high affinity and high brain uptake. PET images of 11C-NMPB uptake had the highest accumulations in the cerebral cortex and striatum and the lowest in the cerebellum, both in patients with Parkinson's disease (PD) and normal subjects. 11C-NMPB uptake was homogeneous throughout the cerebral cortex in the normal subjects but greater in the frontal cortex of about one half of the PD patients as compared with other cerebral cortical areas. Quantitative analysis showed that the PD patients h

Healing of duodenal ulcer during treatment with an anticholinergic and an antacid.

Twenty-nine patients with endoscopically proven duodenal ulcers were treated with either placebo or benzilonium bromide (Ulcoban prolongatum) for 4 weeks. Antacid consumption was low in both groups. Healing after 4 weeks was verified endoscopically and was about equal for both treatments. Frequency of pain and other "dyspeptic" symptoms appeared to be less in patients treated with benzilonium. Lack of symptoms was not always associated with healing of the ulcers.

A Novel Method of Coupling In Situ Time-Resolved FTIR and Microwave Irradiation: Application to the Monitoring of Quinoxaline Derivatives Synthesis.

Traditional synthetic methods, often limited by inefficiency, are increasingly being replaced by sustainable alternatives. This study presents a green approach combining microwave irradiation with in situ FTIR spectroscopy for real-time monitoring and optimising nitrogen-heterocycle synthesis, focusing on quinoxalines. Although both microwave-assisted synthesis and time-resolved FTIR are established techniques, their combined application remains underexplored, limiting their synergistic potential. The methodology was applied to synthesising 2,3-diphenylquinoxaline, a compound of interest in me

Alterations of muscarinic acetylcholine receptors in atypical Pick's disease without Pick bodies.

BACKGROUND: Atypical Pick's disease without Pick bodies is a type of frontotemporal dementia characterised by semantic dementia and temporal dominant lobar atrophy with ubiquitinopathy. No neurochemical analyses have ever been reported in this condition. OBJECTIVE: To investigate muscarinic acetylcholine receptors (mAchR) and their subtypes (M1-M4) in atypical Pick's disease. SUBJECTS: Five cases of atypical Pick's disease were studied. They were compared with nine control cases, 11 cases of Alzheimer's disease, and seven cases of dementia with Lewy bodies. METHODS: A [(3)H]quinuclidinyl be

Muscarinic toxin-like proteins from Taiwan banded krait (Bungarus multicinctus) venom: purification, characterization and gene organization.

Two novel proteins, BM8 and BM14, were isolated from Bungarus multicinctus (Taiwan banded krait) venom using the combination of chromatography on a SP-Sephadex C-25 column and a reverse-phase HPLC column. Both proteins contained 82 amino acid residues including 10 cysteine residues, but there were two amino acid substitutions at positions 37 and 38 (Glu37-Ala38 in BM8; Lys37-Lys38 in BM14). CD spectra and acrylamide quenching studies revealed that the gross conformation of BM8 and BM14 differed. In contrast to BM8, BM14 inhibited the binding of [3H]quinuclidinyl benzilate to the M2 muscarinic

Accommodation and ciliary muscle muscarinic receptors after echothiophate.

Twice daily topical administration of echothiophate for 2 weeks to the eyes of living cynomolgus monkeys produced profound subsensitivity of the accommodative response to pilocarpine and an approximately 50% decrease in the number of specific binding sites for 3H-quinuclidinyl benzilate (3H-QNB) in the ciliary muscle without a change in their affinity. When echothiophate treatment was discontinued, functional cholinergic sensitivity and the number of QNB binding sites both returned to normal over a similar 4-8 week period. Most animals had a modest overshoot of both functional sensitivity and

Guanosine 5'-triphosphate converts some populations of propylbenzilylcholine mustard-sensitive muscarinic cholinoceptor sites to sites resistant to the drug in intestinal smooth muscle.

From functional studies with propylbenzilylcholine mustard (PrBCM), we reported that there coexist PrBCM-sensitive and PrBCM-resistant muscarinic cholinoceptor mechanisms in guinea pig taenia caecum. We investigated the interrelationship between these two cholinoceptor mechanisms using an in vitro receptor binding assay with [3H]quinuclidinyl benzilate (QNB) and [3H]PrBCM. Pretreatment of the muscle strips with 300 nM PrBCM (in vivo alkylation) for 10-50 min resulted in progressive decreases of the number of the maximum [3H]QNB binding sites. However, a prolongation of the period of in vivo al

Muscarinic receptor regulation of Ca2+ mobilization in a human salivary cell line.

We have studied receptor-mediated Ca2+ mobilization in an established exocrine epithelial cell line (HSG-PA) derived from a human submandibular gland. These cells possess a single class of high-affinity muscarinic cholinergic receptors identified using [3H]-quinuclidinyl-benzilate (Kd = 0.17 +/- 0.07 nmol/l; Bmax = 37 +/- 2 fmol/mg protein; n = 3). The muscarinic agonist carbachol elicits a concentration dependent increase of [3H]-inositol trisphosphate in HSG-PA cells (100 mumol/l; greater than 2 fold by 30 s). Carbachol also results in a rapid, approximately 5-fold increase in cytosolic [Ca2

Gallamine exerts biphasic allosteric effects at muscarinic receptors.

Although gallamine and a number of other compounds have been reported to slow the rate of dissociation of labeled ligands, especially [3H]N-methylscopolamine (NMS), from muscarinic receptors of heart and brain, there has been some dispute as to whether the dissociation of [3H]quinuclidinyl benzilate (QNB) is subject to such allosteric regulation. The present studies were intended to determine whether past discrepancies might be due to differences between tissues. We have found that gallamine modulates the dissociation of [3H]QNB from muscarinic receptors of the heart in a biphasic manner. Low

Muscarinic receptor binding and Ca2+ influx in the all-or-none response to acetylcholine of isolated smooth muscle cells.

Isolated smooth muscle cells from guinea pig taenia caecum were prepared by collagenase digestion. Isolated single smooth muscle cells showed an all-or-none response to acetylcholine (ACh) under our experimental conditions. A Scatchard plot of the specific binding of [3H]quinuclidinyl benzilate (QNB) to the cells gave a straight line, and the KD and Bmax values were calculated as 0.18 +/- 0.03 nM and 1.11 +/- 0.10 pmol/mg protein, respectively. ACh competitively inhibited the specific binding of [3H]QNB in a concentration-dependent manner. Thus, although the cells showed an all-or-none respons

Characterization of muscarinic cholinergic receptors in the brains of copper-deficient rats.

In order to assess a possible role for copper as a regulator of muscarinic receptors in vitro, the receptor was characterized in rats made copper deficient by a dietary regimen. In forebrain regions there was a decrease in both the affinity of the receptors for [3H]-1-quinuclidinyl benzilate and the density of receptors in the copper-deficient animals compared with control animals. Copper treatment in vitro of homogenates from deficient animals did not reverse the in vivo effects on antagonist binding but, rather, decreased receptor occupancy and ligand affinity in a manner similar to copper t
