Use of Copper as a Trigger for the in Vivo Activity of E. coli Laccase CueO: A Simple Tool for Biosynthetic Purposes.

Laccases are multi-copper oxidases that catalyze the oxidation of various electron-rich substrates with concomitant reduction of molecular oxygen to water. The multi-copper oxidase/laccase CueO of Escherichia coli is responsible for the oxidation of Cu+ to the less harmful Cu2+ in the periplasm. CueO has a relatively broad substrate spectrum as laccase, and its activity is enhanced by copper excess. The aim of this study was to trigger CueO activity in vivo for the use in biocatalysis. The addition of 5 mM CuSO4 was proven effective in triggering CueO activity at need with minor toxic effects

Perspectives of biotechnological production of L-ribose and its purification.

L-ribose is a non-natural and expensive sugar that can be used as an important intermediate for the synthesis of L-nucleoside analogues, which are used as antiviral drugs. In contrast to chemical production, biotechnological methods can produce L-ribose from biomass under environmentally friendly conditions. In this mini-review, various strategies for synthesizing L-ribose by applying microorganisms and their enzymes are discussed, including microbial biotransformation and biocatalysis by engineering bacteria. Furthermore, subsequent isolation-and-purification techniques, as an integral step i

Green and sustainable synthesis of chiral alcohols: the role of Daucus carota as a biocatalyst in organic chemistry.

Chiral alcohols are essential intermediates in pharmaceuticals, agrochemicals, and advanced materials. Conventional asymmetric reduction of ketones relies on costly metal catalysts with significant environmental impact. Biocatalysis, particularly whole-cell systems, offers a sustainable alternative, providing high regio- and stereoselectivity under mild conditions. Daucus carota (carrot) roots serve as a promising biocatalyst due to their broad substrate compatibility and natural cofactor recycling ability, reducing reliance on toxic reagents and energy-intensive processes, making them both en

Directed Evolution of Nonheme Iron Enzymes for Enantioselective Aminative Difunctionalization of Alkenes.

The direct aminative difunctionalization of alkenes offers a powerful strategy for synthesizing valuable amine-containing compounds. However, achieving this transformation enantioselectively remains a significant challenge in both synthetic chemistry and biocatalysis. In this study, we engineered a nonheme iron enzyme, quercetin 2,3-dioxygenase from Bacillus subtilis (BsQueD), to catalyze three distinct alkene aminofunctionalization reactions with high efficiency and enantiocontrol. Through directed evolution, we developed an optimized BsQueD variant capable of producing a wide array of chiral

Biocatalytic Phosphorylations of Metabolites: Past, Present, and Future.

Phosphorus is a key element for life that occurs in living cells as a structural part of many key biochemicals. Phosphorus plays important functional roles in biocatalysis, such as making metabolic reaction steps irreversible by transferring energy-rich groups, using phosphate groups as leaving groups, or selectively hydrolyzing phosphorus-oxygen or phosphorus-nitrogen bonds. The large number of biocatalysts known to catalyze the transfer of phosphorus-containing groups from donor molecules to acceptor molecules covers a wide variety of reaction classes. Bioprocessing technology can and should

[Laccase as a useful biotechnological tool in the synthesis of biologically active compounds].

The synthesis of organic compounds using oxidoreductive enzymes as biocatalysts is increasingly being considered as an environmentally friendly alternative to classical chemical synthesis. An example of such an enzyme, which exhibits low substrate specificity and operates under mild conditions of pH, pressure and temperature, is laccase, a versatile phenolic oxidase that uses oxygen as a natural reaction co-substrate. It can oxidise both phenolic derivatives and aromatic amines, which in homo- or heteromolecular reactions are coupled to form new organic compounds with unique properties and app

Transaminase-catalyzed sequential hydrodefluorinations of perfluorinated methylene groups.

Biocatalysis represents a particularly promising approach for the degradation of recalcitrant fluorochemicals. However, the enzymatic activation of perfluorinated methylene groups remains a challenge, primarily due to the increase in C-F bond strength that correlates with the number of fluorine atoms attached to a single carbon atom. Here, we describe a new-to-nature hydrodefluorination reaction catalyzed by engineered transaminases for the biodegradation of difluorinated fused-ring ketones. Remarkable defluorination efficiency was achieved in most cases by employing a slight excess of 2-PrNH2

Formaldehyde: An Essential Intermediate for C1 Metabolism and Bioconversion.

Formaldehyde is an intermediate metabolite of methylotrophic microorganisms that can be obtained from formate and methanol through oxidation-reduction reactions. Formaldehyde is also a one-carbon (C1) compound with high uniquely reactive activity and versatility, which is more amenable to further biocatalysis. Biosynthesis of high-value-added chemicals using formaldehyde as an intermediate is theoretically feasible and promising. This review focuses on the design of the biosynthesis of high-value-added chemicals using formaldehyde as an essential intermediate. The upstream biosynthesis and dow

Influence of Reaction Conditions on Enzymatic Enantiopreference: the Curious Case of HEwT in the Synthesis of THF-Amine.

Enzymatic enantiopreference is one of the key advantages of biocatalysis. While exploring the synthesis of small cyclic (chiral amines) such as 3-aminotetrahydrofuran (THF-amine), using the (S)-selective transaminase from Halomonas elongata (HEwT), inversion of the enantiopreference was observed at increasing substrate loadings. In addition, the enantiopreference could be altered by variation of the ionic strength, or of the co-solvent content in the reaction mixture. For example, using otherwise identical reaction conditions, the presence of 2 M sodium chloride gave (R)-THF-amine (14 % ee), w

Abiotic reduction of ketones with silanes catalysed by carbonic anhydrase through an enzymatic zinc hydride.

Enzymatic reactions through mononuclear metal hydrides are unknown in nature, despite the prevalence of such intermediates in the reactions of synthetic transition-metal catalysts. If metalloenzymes could react through abiotic intermediates like these, then the scope of enzyme-catalysed reactions would expand. Here we show that zinc-containing carbonic anhydrase enzymes catalyse hydride transfers from silanes to ketones with high enantioselectivity. We report mechanistic data providing strong evidence that the process involves a mononuclear zinc hydride. This work shows that abiotic silanes ca

Selective C18-Methyl Oxidation of Sclareol via an Engineered P450 Enables the Synthesis of Spiromyrrhene B.

While selective oxidation of unactivated C-H bonds has emerged as an effective tactic to reshape retrosynthetic strategies in the synthesis of natural products, the primary methyl C-H bonds remain inert and inaccessible to contemporary methods without preinstalled directing groups. Herein, we report a biocatalytic C-H oxidation strategy that leverages an engineered P450BM3 platform for the selective C18-methyl hydroxylation of sclareol. Through five rounds of directed evolution, variant Ox18M3-3 was developed, elevating the regioselectivity from 4% to >99% alongside a 2.2-fold increase in cata

Assessing biological degradation of non-hydrolysable synthetic polymers.

The discovery and engineering of enzymes capable of depolymerizing synthetic polymers with hydrolysable backbone linkages is now enabling biotechnological solutions for plastics recycling. Polyolefins and other non-hydrolysable polymers, however, have saturated carbon-carbon linkages that are more challenging to cleave biologically. Despite decades of research, the evidence that biocatalysts can appreciably deconstruct carbon-carbon-linked polymers remains ambiguous, often stemming from inconsistent assays and qualitative, low-resolution analytical methods and from the lack of proper identific

Recent advances in the chemoenzymatic synthesis of N- and O-glycans.

Protein N- and O-glycosylation are among the most common and essential post-translational modifications and are implicated in numerous physiological and pathological processes. The efficient assembly of structurally defined N- and O-glycans remains a major challenge due to their structural complexity and diversity. Integrating the flexibility of chemical synthesis and the high regio- and stereospecificity of enzymatic catalysis, chemoenzymatic synthesis represents a powerful strategy for constructing structurally defined N- and O-glycans. This review summarizes recent advances in the chemoenzy

Selective Reduction of α,β-Unsaturated Carbonyls with Ketoreductase.

Enzymatic catalysis promises the achievement of functional group reactivity selectivity with the conjunctional expansive acquirement of reactivity diversity through the conformal mutagenesis tuning of catalytic cavity. Herein, we report an enzymatic stringency-relaxation strategy for effecting reactivity diversification into the

Process-integrated engineered resting cells for biocatalytic production of rare natural sugars from sole methanol molecules.

The biocatalytic conversion of green methanol, a promising feedstock, into high value-added products is seen as an attractive way to establish sustainable biomanufacturing. Although natural and engineered microorganisms can utilize methanol, efficiently converting C1 carbon into valuable chemicals in growing cells remains challenging because non-native pathways often suffer from low carbon utilization and limited catalytic stability. Here, we report the integration of an engineered enzyme cascade into resting cells and its coupling with a screened alcohol oxidase enzyme to develop an efficient

An Enzymatic Platform for Late-Stage (Radio)isotope Labelling of Oligonucleotides With Methyltransferases.

The development of therapeutic nucleic acids has become a cornerstone of modern drug discovery. To support the clinical translation of these modalities, radiolabelled analogues are vital for absorption, distribution, metabolism and excretion (ADME) studies, particularly for quantitative whole-body autoradiography (QWBA). Access to such radio probes is limited because established methods for the preparation of 3H-labelled oligonucleotides are characterised by high costs, extensive lead times, low specific activity and the generation of substantial radioactive waste. To address these limitations

An artificial biocatalytic cascade for efficient synthesis of norepinephrine by combination of engineered L-threonine transaldolase with multi-enzyme expression fine-tuning.

Norepinephrine, a kind of β-adrenergic receptor agonist, is commonly used for treating shocks and hypotension caused by a variety of symptoms. The development of a straightforward, efficient and environmentally friendly biocatalytic route for manufacturing norepinephrine remains a challenge. Here, we designed and realized an artificial biocatalytic cascade to access norepinephrine starting from 3, 4-dihydroxybenzaldehyde and L-threonine mediated by a tailored-made L-threonine transaldolase PsLTTA-Mu1 and a newly screened tyrosine decarboxylase ErTDC. To overcome the imbalance of multi-enzymes

Recent Developments in the Biosynthesis of Aziridines.

Only 0.016 % of all known natural products contain an aziridine ring, but this unique structural feature imparts high reactivity and cytotoxicity to the compounds in which it is found. Until 2021, no naturally occurring aziridine-forming enzymes had been identified. Since 2021, the biosynthetic enzymes for ~10 % of known aziridine containing natural products have been identified and characterized. This article describes the recent advances in our understanding of enzyme-catalyzed aziridine formation in the context of historical methods for aziridine formation through synthetic chemistry.

Design of a synthetic enzyme cascade for the in vitro fixation of formaldehyde to acetoin.

Formaldehyde (FALD) has gained prominence as an essential C1 building block in the synthesis of valuable chemicals. However, there are still challenges in converting FALD into commodities. Recently, cell-free biocatalysis has emerged as a popular approach for producing such commodities. Acetoin, also known as 3-hydroxy-2-butanone, has been widely used in food, cosmetic, agricultural and the chemical industry. It is valuable to develop a process to produce acetoin from FALD. In this study, a cell-free multi-enzyme catalytic system for the production of acetoin using FALD as the substrate was de
