Subgingival biofilm structure.

Periodontitis is an inflammatory disease of the oral cavity initiated by a microbial biofilm (or 'dental plaque'). Subgingival biofilms in periodontal pockets are not easily analyzed without the loss of structural integrity. These subgingival plaques are structured communities of microorganisms with great phylogenetic diversity embedded in a self-produced extracellular polymeric matrix. For almost three decades, knowledge of the structure of plaque located below the gingival margin has been limited to landmark studies from the 1970s that were unaware of the breadth of microbial diversity we ap

[Chronic prostatitis and biofilm].

Nowadays, inflammatory prostatic diseases represent one of the main social and economic problems that affect male gender in our country. The disease's prevalence is estimated at 11-14% in European countries, and males aged between 20 and 50 years are the most affected subjects. Bacterial prostatitis is documented at a very low prevalence (7-13% of prostatitis overall considered) due to either reduced utilization of viral and bacteriological tests (Meares test, urethral swab, bacterial and viral PCR) or to the possibility that individual patients show a scarce expression of bacterial disease in

Biofilm in nasal polyps.

INTRODUCTION: Bacterial biofilms are involved in many human bacterial infectious processes and in chronic rhinosinusitis as well. The aim of this study was to determine whether biofilm exists in nasal polyps, both in diffuse nasal polyposis (DNP) and antrochoanal polyps (ACP). METHOD: Tissue samples were taken from seven patients suffering from DNP and three patients suffering from ACP, based on the defined patient inclusion and exclusion criteria. After the preparation, the tissue samples were analyzed by means of scanning electron microscopy (SEM) for signs of biofilm formation. RESULTS: S

Nitroxides as anti-biofilm compounds for the treatment of Pseudomonas aeruginosa and mixed-culture biofilms.

A series of 23 nitroxides () was tested for biofilm modulatory activity using a crystal violet staining technique. 3-(Dodecane-1-thiyl)-4-(hydroxymethyl)-2,2,5,5-tetramethyl-1-pyrrolinoxyl () was found to significantly suppress biofilm formation and elicit dispersal events in both Pseudomonas aeruginosa and mixed-culture biofilms. Twitching and swarming motilities were enhanced by nitroxide , leaving the planktonic-specific swimming motility unaffected and suggesting that the mechanism of -mediated biofilm modulation is linked to the hyperactivation of surface-associated cell motilities. Biomass concentration and biofilm characteristics in high-performance fluidized-bed biofilm reactors.

Two laboratory scale fluidized-bed biofilm reactors (FBBRs) were used to investigate the biomass concentration and the biofilm characteristics in a high performance FBBR used for the denitrification of exceptionally high-nitrate wastewater (1000 mg-N/L). Reported correlations by other workers for predicting the biomass concentration in FBBR were examined for their validity in comparison with the experimental results of this study and the best set of applicable correlations was recommended. The effects of the two main operational parameters, the superficial velocity and nitrogen loading rate on

Acoustic signatures uncover multistage mechanical maturation of bacterial biofilms.

Bacterial biofilm formation is commonly viewed as a gradual increase in surface-bound biomass; however, the mechanical transitions that govern maturation remain insufficiently characterized. Here, we integrate quartz crystal microbalance with dissipation monitoring (QCM-D), field-emission scanning electron microscopy (FESEM), and bulk biofilm assays to investigate the multistage mechanical development of bacterial biofilms using a controlled heterologous expression platform for the Pseudomonas aeruginosa adhesin CdrA. This protein enhances biomass accumulation while accelerating structural and

Siderophore sharing protects clonal Pseudomonas aeruginosa biofilms from colistin.

Siderophores mediate cooperative iron acquisition and stress tolerance in polymicrobial communities. However, whether and how such social roles emerge within clonal populations of a single bacterial strain remain unclear. Here, we investigate whether and how such social roles emerge within clonal populations of a single bacterial strain. Using single-cell transcriptomics, we analyzed the response of clonal Pseudomonas aeruginosa PAO1 biofilms to colistin and identified 13 transcriptionally distinct subpopulations, with siderophore biosynthesis genes enriched in one subpopulation. Upon colistin

Rheological Characterization and 3D Fabrication of Artificial Bacterial Biofilms.

Biofilms are significantly involved in the progression of many diseases, such as cancer and upper respiratory infections, due to their ability to adhere to soft tissues. Factors influencing biofilm development have been extensively studied on planar substrates; however, there is limited understanding regarding biofilm growth and interactions within 3D matrices. Developing biofilm models that closely mimic natural bacterial communities' chemical and mechanical properties in soft tissues is essential for developing next-generation antibacterial compounds and therapeutics, as 3D biofilms are more

Polyproline peptide targets Klebsiella pneumoniae polysaccharides to collapse biofilms.

Hypervirulent Klebsiella pneumoniae is known for its increased extracellular polysaccharide production. Biofilm matrices of hypervirulent K. pneumoniae have increased polysaccharide abundance and are uniquely susceptible to disruption by peptide bactenecin 7 (bac7 (1-35)). Here, using confocal microscopy, we show that polysaccharides within the biofilm matrix collapse following bac7 (1-35) treatment. This collapse led to the release of cells from the biofilm, which were then killed by the peptide. Characterization of truncated peptide analogs revealed that their interactions with polysaccharid

Antimicrobial resistance: Biofilms, small colony variants, and intracellular bacteria.

Soft tissue and bone infections continue to be a serious complication in orthopedic and trauma surgery. Both can lead to a high burden for the patients and the healthcare system. Musculoskeletal infections can be induced by intraoperative contamination, bacterial contamination of open wounds or hematogenous bacterial spread. During the recent decades, advances were achieved in the understanding of pathogenesis and antibiotic resistance. Despite some progress in the diagnosis and advancing of therapeutic concepts, groundbreaking successful improvement of treatment concepts is still missing. Cur

Innovative Methodology for Antimicrobial Susceptibility Determination in Mycoplasma Biofilms.

Mycoplasma spp. are facultative pathogens that contribute to the pathogenesis of multiple bovine diseases, including the bovine respiratory disease complex, and have been shown to form biofilms. Biofilm formation is associated with increased antibiotic resistance in many organisms, but accurate determination of antimicrobial susceptibility in biofilms is challenging. In Mycoplasma spp., antimicrobial susceptibility is routinely determined using metabolic pH-dependent color change. However, biofilm formation can lead to reduced metabolism, making interpretation of metabolic readouts difficult.

Gastrointestinal Biofilms: Endoscopic Detection, Disease Relevance, and Therapeutic Strategies.

Gastrointestinal biofilms are matrix-enclosed, highly heterogenic and spatially organized polymicrobial communities that can cover large areas in the gastrointestinal tract. Gut microbiota dysbiosis, mucus disruption, and epithelial invasion are associated with pathogenic biofilms that have been linked to gastrointestinal disorders such as irritable bowel syndrome, inflammatory bowel diseases, gastric cancer, and colorectal cancer. Intestinal biofilms are highly prevalent in ulcerative colitis and irritable bowel syndrome patients, and most endoscopists will have observed such biofilms during

Intestinal biofilms: pathophysiological relevance, host defense, and therapeutic opportunities.

SUMMARYThe human intestinal tract harbors a profound variety of microorganisms that live in symbiosis with the host and each other. It is a complex and highly dynamic environment whose homeostasis directly relates to human health. Dysbiosis of the gut microbiota and polymicrobial biofilms have been associated with gastrointestinal diseases, including irritable bowel syndrome, inflammatory bowel diseases, and colorectal cancers. This review covers the molecular composition and organization of intestinal biofilms, mechanistic aspects of biofilm signaling networks for bacterial communication and

Coacervate Dense Phase Displaces Surface-Established Pseudomonas aeruginosa Biofilms.

For millions of years, barnacles and mussels have successfully adhered to wet rocks near tide-swept seashores. While the chemistry and mechanics of their underwater adhesives are being thoroughly investigated, an overlooked aspect of marine organismal adhesion is their ability to remove underlying biofilms from rocks and prepare clean surfaces before the deposition of adhesive anchors. Herein, we demonstrate that nonionic, coacervating synthetic polymers that mimic the physicochemical features of marine underwater adhesives remove ∼99% of Pseudomonas aeruginosa (P. aeruginosa) biofilm biomass

Antifungal therapy of Candida biofilms: Past, present and future.

Virtually all Candida species linked to clinical candidiasis are capable of forming highly resistant biofilms on different types of surfaces, which poses an additional significant threat and further complicates therapy of these infections. There is a scarcity of antifungal agents, and their effectiveness, particularly against biofilms, is limited. Here we provide a historical perspective on antifungal agents and therapy of Candida biofilms. As we reflect upon the past, consider the present, and look towards the future of antifungal therapy of Candida biofilms, we believe that there are reasons

Optimization of Phage-Antibiotic Combinations against Staphylococcus aureus Biofilms.

Phage therapy has gained attention due to the spread of antibiotic-resistant bacteria and narrow pipeline of novel antibiotics. Phage cocktails are hypothesized to slow the overall development of resistance by challenging the bacteria with more than one phage. Here, we have used a combination of plate-, planktonic-, and biofilm-based screening assays to try to identify phage-antibiotic combinations that will eradicate preformed biofilms of Staphylococcus aureus strains that are otherwise difficult to kill. We have focused on methicillin-resistant S aureus (MRSA) strains and their daptomycin-no

Effects of Novel Dental Composites on Streptococcus mutans Biofilms.

With the phase-out of amalgam and the increase in minimally invasive dentistry, there is a growing need for high-strength composite materials that can kill residual bacteria and promote tooth remineralization. This study quantifies how antibacterial polylysine (PLS) and re-mineralizing monocalcium phosphate monohydrate (MCPM) affect Streptococcus mutans biofilms and the strength of dental composites. For antibacterial studies, the MCPM-PLS filler percentages were 0-0, 8-4, 12-6, and 16-8 wt% of the composite filler phase. Composite discs were immersed in 0.1% sucrose-supplemented broth contain

Convergent Community Assembly among Globally Separated Acidic Cave Biofilms.

Acidophilic bacteria and archaea inhabit extreme geochemical "islands" that can tell us when and how geographic barriers affect the biogeography of microorganisms. Here, we describe microbial communities from extremely acidic (pH 0 to 1) biofilms, known as snottites, from hydrogen sulfide-rich caves. Given the extreme acidity and subsurface location of these biofilms, and in light of earlier work showing strong geographic patterns among snottite Acidithiobacillus populations, we investigated their structure and diversity in order to understand how geography might impact community assembly. Inhibition of bacterial biofilms by the snake venom proteome.

Snake venoms possess a range of pharmacological and toxicological activities. Here we evaluated the antibacterial and anti-biofilm activity against methicillin-susceptible and methicillin-resistant Staphylococcus aureus (MSSA and MRSA) of venoms from the Samar spitting cobra Naja samarensis and the Puff adder Bitis arietans. Both venoms prevented biofilm production by pathogenic S. aureus in a growth-independent manner, with the B. arietans venom being most potent. Fractionation showed the active molecule to be heat-labile and >10 kDa in size. Proteomic profiles of N. samarensis venom revealed
