Efficient pretreatment of Phragmites australis biomass using glutamic acid for bioethanol production by a hybrid hydrolysis and fermentation strategy.

Microbial fermentation of renewable lignocellulosic biomass to produce biofuels presents significant environmental advantages. The conversion of cellulose and hemicellulose into fermentable sugars provides essential carbon sources for microbial metabolism. However, the recalcitrance of biomass limits enzymatic accessibility. In this study, mild L-glutamic acid (GA) pretreatment was applied to Phragmites australis residues (reed straw) to fractionate lignin and polysaccharides for enhancing enzymatic hydrolysis. Pretreatment with 0.20 mol/L GA at 180 °C for 50 min (logRo = 4.1) achieved glucan

Influence of titer, rate, yield, and scale on the cost and life-cycle emissions of biomanufacturing.

Building a thriving bioeconomy requires microbial hosts that efficiently convert diverse carbon sources at high titer, rate, and yield (TRY). Yet, the impacts of TRY on biofuel production cost and carbon footprint across different stages of strain optimization remain unclear. This study presents a systematic analysis of the individual and combined impacts of TRY and scale for the production of multiple bio-based products, including medium-chain (C13-C15) methyl ketones, bisabolene, and ethanol, from lignocellulosic biomass hydrolysates. The results show nonlinear behavior in both cost and CO2-

Operando 1H nuclear magnetic resonance (NMR) activity assay for pyruvate decarboxylase.

The biological conversion of pyruvate by thiamine pyrophosphate (TPP)-linked enzymes, such as pyruvate decarboxylase (Pdc), is a critical step in microbial production of biofuels such as ethanol. Traditionally, the activity of these enzymes is measured using UV-Vis spectrophotometric coupled assays. However, these assays typically only measure one chemical species. Here, we describe an Operando 1H Nuclear Magnetic Resonance (NMR) activity assay that provides a robust, non-destructive alternative for monitoring Pdc kinetics in real time. This quantitative NMR (qNMR) method allows for the simult

Lipidomic Analysis: From Archaea to Mammals.

Lipids are among the most important organic compounds found in all living cells, from primitive archaebacteria to flowering plants or mammalian cells. They form part of cell walls and constitute cell storage material. Their biosynthesis and metabolism play key roles in faraway topics such as biofuel production (third-generation biofuels produced by microorganisms, e.g. algae) and human diseases such as adrenoleukodystrophy, Zellweger syndrome, or Refsum disease. Current lipidomic analysis requires fast and accurate processing of samples and especially their characterization. Because the number

The potential of bio-methane as bio-fuel/bio-energy for reducing greenhouse gas emissions: a qualitative assessment for Europe in a life cycle perspective.

Anaerobic digestion is a well known process that (while still capable of showing new features) has experienced several waves of technological development. It was "born" as a wastewater treatment system, in the 1970s showed promise as an alternative energy source (in particular from animal waste), in the 1980s and later it became a standard for treating organic-matter-rich industrial wastewater, and more recently returned to the market for its energy recovery potential, making use of different biomasses, including energy crops. With the growing concern around global warming, this paper looks at

A multi-centre observational study of HIV, tuberculosis and risk of chronic lung disease in urban West Africa.

OBJECTIVE: HIV and tuberculosis (TB) are risk factors for non-communicable chronic lung disease (CLD). Despite the high prevalence of these infections in West Africa, there are no studies that compare CLD between people with HIV and HIV-negative populations in this setting. This study sought to quantify the contribution of HIV and TB infection in addition to conventional CLD risk factors, such as tobacco and biofuel exposure, to CLD in urban West Africa. DESIGN: A multi-centre cross-sectional study was conducted in three community clinics in Lagos, Nigeria between 2018 and 2019. METHODS: Spi

Quantitative Photo-Reforming of Biomass to CO Spurred by In Situ Protonation of Carbon Dioxide Radical Anions.

Heterogeneous photocatalysis has emerged as a green solution for converting renewable biomass into biofuels and chemicals, though it typically suffers from unsatisfactory carbon utilization efficiency. Here, we present a "two-in-one" photoreduction strategy enabled by electron-rich Cd sites, both remote from and adjacent to sulfur vacancies (Vs). This strategy is highly selective for the reduction of O2 and carbon dioxide radical anions (•CO2 -) in situ formed on the Bi2S3 composition (hole) through water oxidation and oxalic acid (OA) oxidative C-C cleavage, respectively. The shell-yolk Z-sch

Development of Saccharomyces cerevisiae isobutanol production strain from osmotolerant and ethanol-producing industrial isolated yeast.

Isobutanol is a promising advanced biofuel alternative to ethanol for advanced biofuels and can be converted into sustainable aviation fuel (SAF) via the alcohol-to-jet (ATJ) process. Recent studies attempted to improve isobutanol production from S. cerevisiae by metabolic engineering; nevertheless, engineered strains, mostly derived from laboratory strains, often encountered difficulties in industrial-scale production. Herein, this present study aims to develop an industrially viable isobutanol-producing strain of S. cerevisiae D3C (isolate G2-3-2, identified in the previous study) with minim

Reprogramming ligand-receptor interactions via a self-powered molecular engine for proton-responsive regulation of cellular motility.

Manipulation of ligand-receptor interactions (LRIs) in a user-defined manner is one of the effective ways to regulate cell motility and disease progression. Molecular engines orchestrate LRIs to regulate cell motility by harnessing various forms of energy within physiological surroundings. However, engineering chemical engines remains largely unexplored, primarily due to the insufficient chemical cue density, uncertain conformational changes of scaffolds, and a limited repertoire of environment-responsive switchable scaffolds. Enzymatic biofuel cells (EBFCs) are electrochemical devices that co

Magnetically recoverable solid acid photocatalyst activated by white LED for sustainable high-yield synthesis of 5-ethoxymethylfurfural from biomass.

The transition toward renewable energy demands innovative pathways for upgrading biomass into high-value biofuels with minimal environmental impact. Among various biofuel candidates, 5-ethoxymethylfurfural (EMF) has emerged as a superior green fuel additive due to its high energy density, low volatility, and compatibility with existing fuel infrastructure. However, achieving high yields from renewable carbohydrate sources remains challenging, particularly under environmentally benign conditions. Herein, we present a magnetically recoverable solid acid photocatalyst, Fe3O4@TiO2-diamine-SO3H, en

Clostridium beijerinckii displays a soluble [FeFe]-hydrogenase/formate dehydrogenase enzyme complex that links H2 and CO2 metabolism.

The production of biofuels by bacterial fermentation receives sustained attention due to the need to develop novel circular and sustainable technologies. Clostridium beijerinckii produces both hydrogen (H2) and carbon-based biofuels acetone, butanol and ethanol (ABE solvents). H2 metabolism in C. beijerinckii is complex and mostly unexplored. Seven hydrogenase genes are contained in the genome, but their exact physiological role is unknown. Here, we report on the characterisation of a novel heterotetrameric soluble enzyme complex composed of an [FeFe]-hydrogenase component stably bound to a fo

Microwave-assisted conversion of lignocellulosic biomass into furans in ionic liquid.

Production of 5-hydroxymethylfurfural (HMF) and furfural from lignocellulosic biomass was studied in ionic liquid in the presence of CrCl(3) under microwave irradiation. Corn stalk, rice straw and pine wood treated under typical reaction conditions produced HMF and furfural in yields of 45-52% and 23-31%, respectively, within 3 min. This method should be valuable to facilitate energy-efficient and cost-effective conversion of biomass into biofuels and platform chemicals.

Sustainable bioethanol production by solid-state fermentation: a systematic review.

The escalation of the global population has accelerated the demand for sustainable energy sources such as bioethanol. Traditionally, bioethanol was obtained by the fermentation of sugar from agricultural crops and grains. However, this technique creates serious threats on the global food supplies, thus hindering the commercial production of bioethanol. Therefore, there is a need to develop new technologies and low-cost raw materials in order to ensure that bioethanol is economically comparable to the first generation of bioethanol. Solid-state fermentation (SSF) has been in the limelight withi

Biodiesel production potential of mixed microalgal culture grown in domestic wastewater.

In this study, a mixed microalgal culture grown in secondarily treated domestic wastewater effluent was investigated for biodiesel production using in situ transesterification method with conventional heating. The total lipid content of the mixed culture was found as 26.2% ± 0.6 by weight of dry biomass, and 74% of the lipids were contributed by total glycerides. In situ transesterification with conventional heating process under acidic conditions produced higher biodiesel yield with chloroform as the co-solvent (82.1% ± 3.9) compared to hexane (55.3% ± 3.9) under the same reaction conditions.

Expanding the horizon of bio-naphtha beyond gasoline blend: property characterization and conversion opportunity assessment through techno-economic and life-cycle analyses.

Bio-naphtha, a common by-product of biorefineries, is expected to experience substantial growth in supply due to increasing demands for renewable diesel and synthetic aviation fuel (SAF). However, demand for bio-naphtha itself as a gasoline blendstock is limited because of the electrification of light-duty vehicles. Although various bio‑naphtha upgrading routes have been explored, a systematic techno-economic and life-cycle comparison across different bio‑naphtha types and upgrading pathways remains missing. This work integrated detailed processing modeling, techno-economic and life-cycle anal

A systematic review on utilization of biodiesel-derived crude glycerol in sustainable polymers preparation.

With the rapid development of biodiesel, biodiesel-derived glycerol has become a promising renewable bioresource. The key to utilizing this bioresource lies in the value-added conversion of crude glycerol. While purifying crude glycerol into a pure form allows for diverse applications, the intricate nature of this process renders it costly and environmentally stressful. Consequently, technology facilitating the direct utilization of unpurified crude glycerol holds significant importance. It has been reported that crude glycerol can be bio-transformed or chemically converted into high-value pol
