Gastropods use pedal waves to transport thin fluid films at the body-substrate interface. Inspired by this strategy, we present a near-boundary pumping mechanism using magnetically actuated, prebuckled undulatory sheets. Through theoretical modeling and experimental validation, we systematically analyze how design, actuation, and fluid properties influence wave propagation and transport performance of a single unit under full and partial immersion. We then demonstrate that optimized temporal and spatial phase shifts among multiple units notably enhance fluid transport. Leveraging these insight

The ocular glymphatic system supports bidirectional fluid transport along the optic nerve, thereby removes metabolic wastes including amyloid-β. To better understand this biological process, we examined the distributions of intravitreally and intracisternally infused tracers in full-length optic nerves from different age groups of mice. Aging was linked to globally impaired ocular glymphatic fluid transport, similar to what has seen previously in the brain. Aging also reduced the pupillary responsiveness to light stimulation and abolished light-induced facilitation in anterograde ocular glymph

Organic anion and cation transporters (OATs and OCTs), members of the solute carriers (SLC)22 family, are critically involved in the transport of endogenous metabolites and xenobiotics across key barriers in the kidney, liver, and choroid plexus. While their role in small-molecule drug pharmacokinetics, drug-drug interactions, and clinical issues is well established, their potential interaction with nanomaterials remains a largely unexplored frontier. This is particularly significant given that the long-term retention of nanomaterials in vivo poses a major challenge to their clinical translati

The blood-brain barrier (BBB) and blood-cerebrospinal fluid barrier (BCSFB) represent major control checkpoints protecting the CNS, by exerting selective control over the movement of organic cations and anions into and out of the CNS compartment. In addition, multiple CNS cell types, e.g., astrocytes, ependymal cells, microglia, contribute to processes that maintain the status quo of the CNS milieu. To fulfill their roles, these barriers and cell types express a multitude of transporter proteins from dozens of different transporter families. Fundamental advances over the past few decades in ou

Urea transporters in bacteria are relatively rare. There are three classes, the ABC transporters such as those expressed by cyanobacteria and Corynebacterium glutamicum, the Yut protein expressed by Yersinia spp and the UreI expressed by gastric Helicobacter spp. This review focuses largely on the UreI proton-gated channel that is part of the acid acclimation mechanism essential for gastric colonization by the latter. UreI is a six-transmembrane polytopic integral membrane protein, N and C termini periplasmic, and is expressed in all gastric Helicobacter spp that have been studied but also in

Targeted drug delivery remains difficult because multiple biological barriers interfere with the stable transport of therapeutics to the site of action. Polymeric nanocarriers have gained broad attention as delivery platforms since their composition and surface properties can be adjusted to improve circulation behavior and cellular delivery. This review discusses the major biological barriers involved in targeted drug delivery and describes how polymeric nanocarriers are engineered to overcome them. Major carrier types, including polymeric nanoparticles and micelles, are considered with emphas

Retinopathy is a sight-threatening posterior eye disease involving microvascular complications. Diabetic retinopathy (DR) is the most prevalent form of this. Topical biologics (anti-VEGFs) have better therapeutic outcomes compared to steroids because of minimum adverse effects. However, only invasive methods have been approved, which are associated with serious issues related to patient compliance. Non-invasive administrations of anti-VEGFs have challenges, including stability and penetrability. Investigations on the use of cell-penetrating peptides as carriers for the delivery of biologics ha

The US Food and Drug Administration (FDA), European Medicines Agency (EMA), and Pharmaceuticals and Medical Devices Agency (PMDA) guidances on small-molecule drug-drug interactions (DDIs), with input from the International Transporter Consortium (ITC), recommend the evaluation of nine drug transporters. Although other clinically relevant drug uptake and efflux transporters have been discussed in ITC white papers, they have been excluded from further recommendation by the ITC and are not included in current regulatory guidances. These include the ubiquitously expressed equilibrative nucleoside

The glucose transporter family has an important role in the initial stage of glucose metabolism; Glucose transporters 2 (GLUTs, encoded by the solute carrier family 2, SLC2A genes) is the major glucose transporter in β-cells of pancreatic islets and hepatocytes but is also expressed in the small intestine, kidneys, and central nervous system; GLUT2 has a relatively low affinity to glucose. Under physiological conditions, GLUT2 transports glucose into cells and allows the glucose concentration to reach balance on the bilateral sides of the cellular membrane; Variation of GLUT2 is associated wit

Nucleoside transport is essential for maintaining intracellular nucleoside and nucleobase homeostasis for living cells. Here, we identified an uncharacterized GntR/HutC family transcriptional regulator, NagR2, renamed NupR (nucleoside permease regulator), that mainly controls nucleoside transport in the Bacillus thuringiensis BMB171 strain. The deletion or overexpression of nupR affected the bacteria's utilization of guanosine, adenosine, uridine, and cytidine rather than thymidine. We further demonstrated that zinc ion is an effector for the NupR, dissociating NupR from its target DNA. Moreov

The use of wild rodents as biological resources is hindered by the difficulties of live transport and primary culture contamination caused by skin bacteria. To address this, we developed a "tail-degloving" method to physically remove the contaminated external integument. Using the large Japanese field mouse (Apodemus speciosus), this method suppressed contamination compared to conventional biopsies, yielding cells within 5-7 days that were capable of subculturing and cryopreservation. The protocol withstood 4°C storage for up to 7 days and enabled contamination-free cell establishment from a l

Nanobiotechnology and nanomedicine are rapidly growing fields, in which nanomaterials (NMs) can lead to enhanced therapeutic efficacy by achieving efficient transport and drug delivery in vivo. The physicochemical properties of NMs have a great impact on their interactions with biological environments and hence determine their biological fates and drug delivery efficiency. Despite rapid advances in understanding the significance of NM properties, such as shape, size, and surface charge, there is a pressing need to engineer and discover how elasticity shapes NM transport. Recently, advances in

UNLABELLED: Recent clinical and experimental data demonstrate that thyroid stunning is caused by previous irradiation and may influence the efficacy of (131)I radiation therapy of thyroid cancer and possibly hyperthyroidism. To avoid stunning, many clinics have exchanged (131)I for (123)I for pretherapeutic diagnostic imaging and dose planning. Furthermore, recent in vitro studies indicate that (131)I irradiation reduces iodide uptake by downregulating the expression of the sodium iodide symporter (NIS). The rationale for this study was therefore to study effects on iodide transport and NIS me

The escalating global water scarcity poses an urgent demand for sustainable freshwater production technologies. Atmospheric water, representing over 12 900 billion tons, constitutes a ubiquitously distributed, abundantly available, and continuously renewable freshwater. Atmospheric water harvesting (AWH) has emerged as a promising technology due to its wide distribution and renewability. Numerous biological organisms offer a wealth of biological prototypes that serve as biomimetic templates for designing hierarchical structures in artificial water harvesting materials. The natural delivery properties of small extracellular vesicles (sEVs) can be harnessed and enhanced through engineering to create a new class of biotherapeutics, particularly for central nervous system (CNS) disorders. While evidence supports the ability of sEVs to cross biological barriers and deliver functional cargo to target cells, a limited understanding of their uptake and transport across the brain hinders their translational potential. In this study, we investigated either native and engineered sEVs, developed by us, using a novel modular engineering platform that employs a dual-targe

Biological olfactory perception relies on ionic transport, offering a promising alternative to conventional gas sensors that depend on electronic signal transmission, which often suffer from limitations such as limited sensitivity, high power consumption, and susceptibility to moisture. Inspired by biological olfactory ion channels, nanochannel-based ionic membranes incorporating 2D materials and ionic liquids have been developed. Through functional modifications, these membranes exhibit unique tremella-like structures that optimize gas diffusion pathways and provide effective gas interaction

BACKGROUND: The G subfamily of ABC (ATP-binding cassette) transporters of Leishmania include 6 genes (ABCG1-G6), some with relevant biological functions associated with drug resistance and phospholipid transport. Several studies have shown that Leishmania LABCG2 transporter plays a role in the exposure of phosphatidylserine (PS), in virulence and in resistance to antimonials. However, the involvement of this transporter in other key biological processes has not been studied.
METHODS: To better understand the biological function of LABCG2 and its nearly identical tandem-repeated transporter LA

Intermolecular charge transport plays an essential role in organic electronic materials and biological systems. To date, experimental investigations of intermolecular charge transport in molecular materials and electronic devices have been restricted to conjugated systems in which π-π stacking interactions are involved. Herein we demonstrate that the σ-σ stacking interactions between neighbouring non-conjugated molecules offer an efficient pathway for charge transport through supramolecular junctions. The conductance of σ-σ stacked molecular junctions formed between two non-conjugated cyclohex

PURPOSE OF REVIEW: Chylomicron biosynthesis plays a vital role in supplying essential lipids and lipid soluble vitamins to peripheral tissues for various functions. Despite this, the intracellular synthesis, trafficking, and secretion of chylomicrons remains only partly understood. The purpose of this review is to summarize the role of established proteins in this process and bring attention to recently identified proteins to provide an up-to-date model of chylomicron biosynthesis.
RECENT FINDINGS: Recently, several proteins have been shown to play a role in the initial formation and lipidati
