UNLABELLED: The availability of a new vaccine is usually needed as an additional component to chemotherapy for control of schistosomiasis. Different strategies of different types of vaccines were assessed to decrease morbidity but did not give the best protection. The study assessed the efficacy of BAAP, SLAP and their combined preparations together with BCG adjuvant as an effective anti-schistosomal vaccine.
METHODOLOGY: Six groups of Swiss albino mice were used; (Gl) as a control, (G2) infected non immunized; (G3) infected and supported by Adj.; (G4) infected; vaccinated with BAAP and suppo

Schistosomiasis is an infectious disease infecting mammals as the definitive host and fresh water snails as the intermediate host. Understanding the molecular and biochemical relationship between the causative schistosome parasite and its hosts will be key to understanding and ultimately treating and/or eradicating the disease. There is increasing evidence that pathogens that have co-evolved with their hosts can manipulate their hosts' behaviour at various levels to augment an infection. Bacteria, for example, can induce beneficial chromatin remodelling of the host genome. We have previously s

An in vivo, non-invasive technique for gene silencing by RNA interference (RNAi) in the snail, Biomphalaria glabrata, has been developed using cationic polymer polyethyleneimine (PEI) mediated delivery of long double-stranded (ds) and small interfering (si) RNA. Cellular delivery was evaluated and optimized by using a 'mock' fluorescent siRNA. Subsequently, we used the method to suppress expression of Cathepsin B (CathB) with either the corresponding siRNA or dsRNA of this transcript. In addition, the knockdown of peroxiredoxin (Prx) at both RNA and protein levels was achieved with the PEI-med

Biomphalaria alexandrina were treated with sublethal concentrations (LC10) of dry powdered leaves Solanum nigrum or whole dry Ambrosia maritima. The two plants affected the development of schistosome parasite within snails by disturbing the glycolytic flux, the most important metabolic pathway for schistosome-infected snails. Attenuated cercariae released from treated snails were used to infect male albino mice to evaluate their pathogenicity compared to control cercariae shed from untreated snails. The mean number of worms established declined from about 70 +/- 17.6 worms/mouse infected with

Three calves experimentally infected with Schistosoma mansoni, and passing viable eggs in feces, as well as 5 normal calves (coming from a non-endemic area for schistosomiasis) kept as controls, were maintained in an enclosure (850 m2 in area). In this enclosure, a tank with water received 500 laboratory reared Biomphalaria glabrata. All the control calves were infected for a period ranging from 79 to 202 days after the beginning of the experiment, and afterwards presented viable S. mansoni eggs in feces. The mean worm recovery was 555. The snail population increased throughout the experimenta

BACKGROUND: Biomphalaria straminea, an intermediate host of Schistosoma mansoni, is originally native to Brazil but has invaded southern China since 1974. Nowadays, increasing human mobility raises the risk of S. mansoni dissemination. Therefore, this study aims to elucidate the genetic variation and structure of B. straminea in China and develop molecular tools for tracing its geographic origins, which could aid in schistosomiasis prevention and control.
METHODS: We collected 290 B. straminea individuals from Shenzhen City (GDSZ, n = 171), Dongguan City (GDDG, n = 65), and Hong Kong (HK, n =

Freshwater snails of the genus Biomphalaria serve as obligatory hosts for the digenetic trematode Schistosoma mansoni, the causative agent for the most widespread form of intestinal schistosomiasis. Within Biomphalaria, S. mansoni larvae multiply and transform into the cercariae form that can infect humans. Trematode development and proliferation is thought to be facilitated by modifications of host behavior and physiological processes, including a reduction of reproduction known as "parasitic castration." As neuropeptides participate in the control of reproduction across phylogeny, a neural t

Biomphalaria spp. snails are intermediary hosts of Schistosoma mansoni, etiologic agent of intestinal schistosomiasis, one of the most important neglected tropical diseases. Biomphalaria straminea is an important intermediary host that possess a different phenotype to parasite infection but shows a large geographic distribution and high capacity of new ecologic niche invasion. Our purpose was to characterize for the first time the differentially expressed proteome in B. straminea during two times intervals after primary and secondary exposure to S. mansoni. The hemolymph was collected at 1 and

The immune system of snails is highly sensitive to pollutants, which can suppress its immune response. We investigated the effects of exposure to the glyphosate-based herbicide Roundup® Original on the snail Biomphalaria glabrata infected by the platyhelminth Echinostoma paraensei by evaluating changes in the snail's internal defense system. Four cohorts were studied: control group, infected snails, snails treated with Roundup®, and snails infected and treated with Roundup®. The hemocyte viability was assessed, morphological differentiation of cells was observed and flow cytometry was performe

Schistosomiasis remains a major parasitic disease, endemic in large parts of South America. Five neotropical species of Biomphalaria have been found to act as intermediate hosts of Schistosoma mansoni in natural populations, while others have been shown to be susceptible in experimental infections, although not found infected in the field. Among these potential intermediate hosts, Biomphalaria peregrina represents the most widespread species in South America, with confirmed occurrence records from Venezuela to northern Patagonia. In this study, we report the southernmost record for the species

In Egypt, reclaiming portions of the desert using water from the Nile has resulted in large-scale invasion of Biomphalaria alexandrina in these regions. Studies exploring the local adaptation of Schistosoma mansoni to its snail host have been carried out to predict the extension of schistosomiasis to newly reclaimed areas. A meta-analysis of the relevant reports was conducted to compare the different biological characteristics of sympatric and allopatric Schistosoma mansoni and Biomphalaria alexandrina using different experimental designs. The results showed that there were no significant diff

In the present study, Biomphalaria snails collected from five Egyptian governorates (Giza, Fayoum, Kafr El-Sheikh, Ismailia and Damietta), as well as reference control Biomphalaria alexandrina snails from the Schistosome Biological Supply Center (SBSC) (Theodor Bilharz Research Institute, Egypt), were subjected to species-specific polymerase chain reaction (PCR) assays to identify the collected species. All of the collected snails were found to be B. alexandrina and there was no evidence of the presence of Biomphalaria glabrata. Randomly amplified polymorphic DNA (RAPD)-PCR assays showed diffe

A life table response experiment (LTRE) was used to quantify the population-level effects of continuous, multigeneration cadmium exposure on two strains of the freshwater gastropod, Biomphalaria glabrata: the parasite-resistant BS90 and parasite-susceptible NMRI strains. Snails were exposed to waterborne cadmium for three consecutive generations. Survival, growth, and reproduction were measured empirically and incorporated into a stage-based, deterministic population model. Cadmium significantly affected hatching success, time to maturity, and juvenile and adult survival in both strains. There

Susceptibility and compatibility experiments were carried out with 700 Biomphalaria tenagophila from the Paraná River basin exposed to infection with Schistosoma mansoni. Individual infection was performed with 10 miracidia of SJ2 strain from the Paraiba valley (Brazil) originally infective to B. tenagophila. These snails were laboratory-breed progeny of B. tenagophila collected from six localities of Argentina and one from Paraguay. From Argentina: Rincón de Vences (7%) and Posadas (11%) became infected with S. mansoni and the calculation of Frandsen's index (TCP/100) shows that they were Cla

This is the first report on occurrence of Biomphalaria straminea in the district of São José de Almeida (municipality of Jaboticatubas) State of Minas Gerais, Brazil. The presence of B. glabrata and B. tenagophila had already been reported in this area. Such municipality is part of the metropolitan region of Belo Horizonte and comprises 60% of the Tourist Complex of Serra do Cipó. Since the 1950s throughout the 1990s, a schistosomiasis prevalence ranging from 15 to 40% has been observed. Although no B. straminea specimen has been found naturally infected in the region, descendants of these sna

We have previously confirmed the presence of common antigens between Schistosoma mansoni and its vector, Biomphalaria glabrata. Cross-reactive antigens may be important as possible candidates for vaccine and diagnosis of schistosomiasis. Sera from outbred mice immunized with a soluble Biomphalaria glabrata antigen (SBgA) of non-infected B. glabrata snails recognized molecules of SBgA itself and S. mansoni AWA by Western blot. Recognition of several molecules of the SBgA were inhibited by pre-incubation with AWA (16, 30, 36, 60 and 155 kDa). The only specific molecule of AWA, inhibited by SBgA,

The importance of the glycolytic flux for the success of Biomphalaria-Schistosome sporocyst interaction was acertained in this study. Hexokinase (HK), pyruvate kinase(PK), glucose phosphate isomerase(GPI) and lactate dehydrogenase (LD) as four important glycolytic enzymes were markedly stimulated in trematode infected Biomphalaria alexandrina when measured two weeks post exposure to infection with Schistosoma mansoni miracidia. On the other hand phosphoenolpyruvate carboxy kinase (PEPCK), glucose-6-phosphatase (G6Pase) and fructose 1,6 diphosphatase(FDPase) as three gluconeogenic enzymes were

Adults of the African ampullariid snail Pila ovata were examined for their ability to control laboratory populations of the pulmonate snail Biomphalaria pfeifferi, a widespread, intermediate host of the human pathogen Schistosoma mansoni in sub-Saharan Africa. In a 6-week experiment conducted in large (100 x 60 x 60 cm) outdoor tanks containing floating macrophytes (Nymphaea caerula) and initially set up with one adult ampullariid for every three adult pulmonates, the numbers of B. pfeifferi egg masses were always about half those in similar tanks without P. ovata. Although, by week 6, the num

Selection of progenies from infected B.alexandrina and B.truncatus snails resulted in higher infection rates. This observation was consistent when the source of micardia were from either infected human or hamsters. Infection rate of B.alexandrina increased up to 88.2% while B.truncatus did not. Compatibility between the parasites and their intermediate hosts were also studied.
