Diverse in vitro liver models reveal comprehensive biotransformation pathways of tetrabromobisphenol A.

Tetrabromobisphenol A is a widely used brominated flame retardant. Its biotransformation in human liver has raised significant environmental and health concerns. This study investigated and compared the metabolism of TBBPA in three in vitro liver models-human liver microsome (HLM), human hepatoblastoma (HepG2) cell, and human normal hepatocyte (MIHA) cell systems. At the end of exposure, 92.4%, 13.6%, and 98.4% of TBBPA was metabolized in HLM, HepG2 cell, and MIHA cell systems, respectively. According to the nontarget analysis, a total of 21 metabolites of TBBPA, including 7 for HLM, 9 for Hep

Accumulation, Biotransformation, and Endocrine Disruption Effects of Fluorotelomer Surfactant Mixtures on Zebrafish.

As an alternative to perfluorooctanesulfonate (PFOS), novel fluorotelomer surfactants (6:2 fluorotelomer sulfonamide alkylbetaine (6:2 FTAB) and 6:2 fluorotelomer sulfonamide alkylamine (6:2 FTAA)) are widely used in aqueous film-forming foams and are frequently found to coexist in the environment. However, their potential toxicities remain unknown. Here, we investigated the chronic toxicity of 6:2 FTAB (65%) and 6:2 FTAA (35%) coexposure on adult zebrafish at doses of 0, 5, 50, or 500 μg/L using a flow-through exposure system for 180 days. Results showed that 6:2 FTAB was undetected in adult

Biotransformation of 8:2 polyfluoroalkyl phosphate diester in gilthead bream (Sparus aurata).

Polyfluoroalkyl phosphate esters (PAPs) are high production volume surfactants used in the food contact paper and packaging industry. PAPs may transform to persistent perfluoroalkyl carboxylic acids (PFCAs) under biotic conditions, but little is known about their fate and behavior in aquatic organisms. Here we report for the first time on the uptake, tissue distribution, and biotransformation of 8:2 polyfluoroalkyl phosphate diester (8:2 diPAP) in fish. Gilt-head bream (Sparus aurata) were dosed via the diet (8:2 diPAP at 29μg/g) for 7days, during which time 8:2 diPAP and its transformation pr

Biotransformation with cellulase, hemicellulase and Yarrowia lipolytica boosts health benefits of okara.

Okara (soybean residue) is a highly perishable food processing by-product from soymilk and tofu manufacture. It contains a large proportion of insoluble dietary fibre (40-60% on a dry basis), as well as digestion-resistant proteins, trypsin inhibitors and phytic acid. These factors contribute lead to the under-utilisation of okara. To improve the overall nutritional quality of okara, sequential saccharification of okara by Celluclast® 1.5L (cellulase) or Viscozyme® L (cellulase and hemicellulase) and fermentation by the yeast Yarrowia lipolytica were performed. The changes in the antioxidant c

Biotransformation and biological activity of N-(4-hydroxyphenyl)retinamide derivatives in rodents.

The metabolism and bioactivity of N-(4-hydroxyphenyl)-all-trans-retinamide (HPR) and of various O-alkyl and ester derivatives of HPR were investigated in rodents. The principal metabolite of HPR in tissues is N-(4-methoxyphenyl)-all-trans-retinamide. N-(4-methoxyphenyl)all-trans-retinamide is equipotent to HPR in reversing keratinization of retinoid-deficient hamster trachea in vitro. Another nonpolar metabolite of HPR is also present in tissue and (although not positively identified) is thought to be a long-chain fatty acid ester of HPR. HPR is excreted into rat bile as numerous polar retinam

Biotransformation of lithocholic acid by Mucor racemosus and the anti-neuroinflammatory activity of products.

The biotransformation of lithocholic acid (LCA) by Mucor racemosus CGMCC 3.20 resulted in the formation of four novel derivatives (compounds 1-4), characterised by 6β and/or 12β hydroxylation and acetylation, in addition to four previously recognised compounds. Comprehensive structural characterisation was achieved through detailed NMR spectroscopy and high-resolution elec-trospray ionisation mass spectrometry (HR-ESI-MS) analyses. M. racemosus exhibited catalytic proficiency in facilitating the structural modification of LCA, enabling the production of both 6β, 12β-hydroxylated and 6β, 12β-ac

Comparison of In Vitro Biotransformation of Olive Polyphenols Between Healthy Young and Elderly.

BACKGROUND: Olive leaves are a rich source of polyphenols, predominantly secoiridoids, flavonoids, and simple phenols, which exhibit various biological properties. Extracts prepared from olive leaves are associated with hypoglycemic, hypotensive, diuretic, and antiseptic properties. Upon ingestion, a substantial fraction of these polyphenols reaches the colon where they undergo extensive metabolism by the gut microbiota. Host characteristics, like age, can influence the composition of the gut microbiome, potentially affecting the biotransformation of these compounds. Therefore, it can be hypot

Biotransformation and its effects on anxiolytic activity and toxicity of Polygalae Radix saponins.

Saponins, accounting for over 2.0% (w/w) in Polygalae Radix extracts, act as both active and toxic constituents, with their glycosyl structures hypothesized to modulate efficacy and toxicity. This study aimed to modify the glycosyl structures of Polygalae Radix (PR) saponins via snail enzyme biotransformation and evaluate changes in the activity of inhibiting anxiety-like behavior in the EPM and gastrointestinal toxicity. The biotransformation process was optimized using one-way experiments and response surface methodology, identifying optimal conditions as 46 °C, pH 6.4, 72-h reaction time, a

Functional characterization of a novel Chlamydomonas reinhardtii hydrolase involved in biotransformation of chloramphenicol.

Microalgae-based biotechnology is one of the most promising alternatives to conventional methods for the removal of antibiotic contaminants from diverse water matrices. However, current knowledge regarding the biochemical mechanisms and catabolic enzymes involved in microalgal biodegradation of antibiotics is scant, which limits the development of enhancement strategies to increase their engineering feasibility. In this study, we investigated the removal dynamics of amphenicols (chloramphenicol, thiamphenicol, and florfenicol), which are widely used in aquaculture, by Chlamydomonas reinhardtii

Synthesis, fungal biotransformation, and evaluation of the antimicrobial potential of chalcones with a chlorine atom.

Chalcones are intermediate products in the biosynthesis of flavonoids, which possess a wide range of biological properties, including antimicrobial and anticancer activities. The introduction of a chlorine atom and the glucosyl moiety into their structure may increase their bioavailability, bioactivity, and pharmacological use. The combined chemical and biotechnological methods can be applied to obtain such compounds. Therefore, 2-chloro-2'-hydroxychalcone and 3-chloro-2'-hydroxychalcone were synthesized and biotransformed in cultures of two strains of filamentous fungi, i.e. Isaria fumosorose

Vitamin B12 as a source of variability in isotope effects for chloroform biotransformation by Dehalobacter.

Carbon and chlorine isotope effects for biotransformation of chloroform by different microbes show significant variability. Reductive dehalogenases (RDase) enzymes contain different cobamides, affecting substrate preferences, growth yields, and dechlorination rates and extent. We investigate the role of cobamide type on carbon and chlorine isotopic signals observed during reductive dechlorination of chloroform by the RDase CfrA. Microcosm experiments with two subcultures of a Dehalobacter-containing culture expressing CfrA-one with exogenous cobamide (Vitamin B12, B12+) and one without (to dri

Potential biotransformation pathways and efficiencies of ciprofloxacin and norfloxacin by an activated sludge consortium.

Fluoroquinolones (FQs), such as ciprofloxacin (CIP) and norfloxacin (NOR), are types of emerging trace pollutants that have attracted great attention. In this study, an activated sludge (AS) consortium with high bio-removal capability to CIP and NOR was obtained by acclimating with CIP and NOR for 10 d. Meanwhile, a CIP- and NOR- transforming bacterial strain (S5), which is highly homologous to the 16S rRNA gene sequence of Enterobacter sp., was isolated from the acclimated AS. The bio-removal efficiency of CIP under the acclimated AS consortium was better than that under the pure culture of E

Exploration of the biotransformation of phenanthrene degradation coupled with methanogensis by metabolites and enzyme analyses.

The ubiquitous environmental contaminants, polycyclic aromatic hydrocarbons (PAHs), can be aerobically biodegraded. Strategies for biodegradation of PAHs are needed for the persisted character of it in anoxic environments. In current study, we obtained a highly enriched anaerobic, PAHs-degrading co-culture DYM1, from petroleum-polluted soil. DYM1 significantly degrades a range of PAHs in 4 days without supplementary terminal electron acceptors.
