Bisphosphonate treatment modifies canine bone mineral and matrix properties and their heterogeneity.

Bone loss and alterations in bone quality are major causes leading to bone fragility in postmenopausal women. Although bisphosphonates are well known to reduce bone turnover and prevent bone loss in postmenopausal osteoporosis, their effects on other bone properties are not fully characterized. Changes in bone mineral and matrix properties may contribute to the anti-fracture efficacy observed with bisphosphonate treatments. The aim of this work was to analyze the effect of a 1-year treatment with either alendronate or risedronate, at low and high doses, on spatially resolved bone material and

Effects of matrix metalloproteinase inhibitors on bone resorption and orthodontic tooth movement.

Matrix metalloproteinases are involved in the regulation of bone remodeling. The hypothesis that matrix metalloproteinase inhibitors may be useful for experimentally limiting orthodontic tooth movement, a process involving perturbations of normal bone remodeling, was tested. General matrix metalloproteinase inhibitors limited the resorption of bone slices by mouse marrow cultures stimulated by calcitriol, parathyroid hormone, and basic-fibroblast growth factor. Pre-coating dentin slices with short arginine-glycine aspartic acid (RGD) peptides, but not arginine-glycine-glutamic acid (RGE) contr

Bone morphogenetic protein-1/Tolloid-like proteinases process dentin matrix protein-1.

Bone morphogenetic protein-1 (BMP-1)/Tolloid-like metalloproteinases play key roles in formation of mammalian extracellular matrix (ECM), through the biosynthetic conversion of precursor proteins into their mature functional forms. These proteinases probably play a further role in formation of bone through activation of transforming growth factor beta-like BMPs. Dentin matrix protein-1 (DMP1), deposited into the ECM during assembly and involved in initiating mineralization of bones and teeth, is thought to undergo proteolysis in vivo to generate functional cleavage fragments found in extracts

Calcium signals in prostate cancer cells: specific activation by bone-matrix proteins.

Cancer of the prostate commonly metastasizes to bony sites where cells acquire an aggressive, rapidly proliferating, androgen-independent phenotype. The interaction between bone and prostate, thus, becomes a key factor in disease progression. Fluctuations in intracellular ionized Ca2+ [Ca2+]i are rapid, regulated signal transduction events often associated with cell proliferation. Hence, Ca2+ signals provide a convenient measure of early events in cancer cell growth. This study developed single cell fluorescent imaging techniques to visualize Ca2+ signals in Fura-2 loaded prostatic cancer cell

Phenotypic modulation of the swarm rat chondrosarcoma induced by morphogenetic bone matrix.

The recognized similarities between developing embryonic tissues and neoplastic cells have led to a number of experimental demonstrations which indicate that inductive microenvironments can alter the malignant phenotype. In postnatal life a morphogenetic cascade resembling endochondral bone development can be induced by s.c. implantation of demineralized diaphyseal bone matrix. We have examined the ability of this microenvironment to alter the phenotype of the transplantable Swarm rat chondrosarcoma in mixed implants. Neoplastic chondrocytes could be distinguished from host cells by nuclear mo

Non-collagen proteins of stapedial bone matrix in perilymph of otosclerotic patients.

Non-collagen proteins extracted from the otosclerotic stapes footplate, superstructure and temporal cortical bone were compared with the protein patterns of normal and otosclerotic perilymph by analytical isotachophoresis (ITP). Normal and otosclerotic perilymph yielded basically identical isotachophoretic subfractions. Thirteen subfractions were detected moving as anions at pH 9.6 in the normal perilymph, versus 16 subfractions of similar character in the otosclerotic perilymph. Of these 16 protein subfractions, one (which was not detected in the normal perilymph) could be found in the ITP-gr

Comparative Analysis of Different Methods in the Quantification of Bone Matrix in Irradiated Rats.

Histomorphometric analysis is crucial for evaluating tissue, aiding in disease diagnosis and prognosis. While automated image processing has advanced, its application in histomorphometry remains limited. This experimental study aimed to compare conventional and computational methods for analyzing histological images using double-threshold segmentation. To do this, researchers evaluated the effects of radiotherapy on bone repair in rats. Twenty-eight male Wistar rats had bone defects induced in the tibia and lower molars extracted, then were divided into four groups based on radiation dose: con

The Role of Matrix-Bound Extracellular Vesicles in the Regulation of Endochondral Bone Formation.

Matrix vesicles are key players in the development of the growth plate during endochondral bone formation. They are involved in the turnover of the extracellular matrix and its mineralization, as well as being a vehicle for chondrocyte communication and regulation. These extracellular organelles are released by the cells and are anchored to the matrix via integrin binding to collagen. The exact function and makeup of the vesicles are dependent on the zone of the growth plate in which they are produced. Early studies defined their role as sites of initial calcium phosphate deposition based on t

Influence of the degradation of the organic matrix on the microscopic fracture behavior of trabecular bone.

In recent years, the important role of the organic matrix for the mechanical properties of bone has become increasingly apparent. It is therefore of great interest to understand the interactions between the organic and inorganic constituents of bone and learn the mechanisms by which the organic matrix contributes to the remarkable properties of this complex biomaterial. In this paper, we present a multifaceted view of the changes of bone's properties due to heat-induced degradation of the organic matrix. We compare the microscopic fracture behavior (scanning electron microscopy; SEM), the topo

Influence of experimental diabetes and insulin on matrix-induced cartilage and bone differentiation.

The influence of streptozotocin-induced diabetes on discrete stages of matrix-induced endochondral bone formation has been investigated. Mesenchymal cell proliferation was inhibited in diabetic rats as evidenced by a 65% reduction of ornithine decarboxylase (ODC) activity and a 56% reduction of [3H]thymidine incorporation per microgram DNA compared to nondiabetic controls; the inhibition was prevented by insulin treatment. In diabetic animals, chondrogenesis on day 7 was reduced by 49% compared to control animals as assessed by 35SO4 incorporation. Exogenous insulin was stimulatory to cartilag

Static magnetic field enhances the bone remodelling capacity of human demineralized bone matrix in a rat animal model of cranial bone defects.

The regeneration of bone defects that exceed 2 cm is a challenge for the human body, necessitating interventional therapies. Demineralized bone matrices (DBM) derived from biological tissues have been employed for bone regeneration and possess notable osteoinductive and osteoconductive characteristics. Nevertheless, their efficiency in regenerating critically sized injuries is limited, and therefore additional signaling cues are required. Thanks to the piezoelectric properties of the bone, external physical stimulation is shown to accelerate tissue healing. We have implanted human DBM in criti

Hydrogel delivery of demineralized bone matrix augmented with ROS-triggered biomineralization and Trb3 activation for enhanced bone regeneration.

Cranial bone defects remain a significant clinical challenge due to limited intrinsic regenerative capacity and an adverse oxidative microenvironment that impairs osteogenesis. Demineralized bone matrix (DBM), a clinically used bone graft substitute, exhibits osteoinductive potential but suffers from inconsistent performance and poor retention at defect sites. Here, we report a DBM-loaded injectable dynamic hydrogel that enhances bone regeneration through coordinated redox modulation, reactive oxygen species (ROS)-triggered biomineralization, and Tribbles homolog 3 (Trb3)-mediated osteogenic s

Urinary Bladder Matrix as a Guide Bone Regeneration Barrier Membrane for Inhibiting Cell Invasion and Promoting Bone Formation.

Natural resorbable collagen membranes are widely used in guided bone regeneration (GBR) in oral implantology and prosthodontics. However, the rapid degradation and inadequate mechanical properties raise concerns about potentially compromising the ultimate bone regeneration efficacy. In this study, we developed a novel GBR membrane by coating a urinary bladder matrix (UBM) onto small intestinal submucosa (SIS), a commonly used acellular material, to improve the cell-barrier and bone-regeneration performances. The results showed that the UBM-SIS membrane exhibited superior tensile strength, high

rhBMP-2 with a demineralized bone matrix scaffold versus autologous iliac crest bone graft for alveolar cleft reconstruction.

BACKGROUND: Secondary alveolar cleft reconstruction using autologous iliac crest bone graft is currently the standard treatment for alveolar clefts. Although effective, harvesting autologous bone may result in considerable donor-site morbidity, most commonly pain and the potential for long-term sensory disturbances. In an effort to decrease patient morbidity, a novel technique using recombinant human bone morphogenetic protein (rhBMP)-2 encased in a demineralized bone matrix scaffold was developed as an alternative to autografting for secondary alveolar cleft reconstruction. METHODS: A chart
