Rheology of CHO Cell Suspensions and Its Effects on High-Density Cultivation Process and Bioreactor Design.

With the rapidly growing demand for monoclonal antibodies (mAbs) worldwide, optimizing the high-density and ultra-high-density cultivation processes of Chinese hamster ovary (CHO) cells has become crucial for enhancing production efficiency. Shear stress and mass transfer have always been the vital operating parameters for the bioreactor in creating a suitable microenvironment for cell growth and antibody production. However, researchers have not actively focused on the rheology of CHO cell suspensions and its impact on these parameters in bioreactors. The factors influencing the rheology of s

Without a trace: multiple knockout of CHO host cell hydrolases to prevent polysorbate degradation in biologics.

Enzymatic degradation of polysorbates (PS) in biologic drug formulations is often caused by hydrolytic host cell proteins (HCPs) and can lead to particle formation and reduced shelf-life. Here, we present a host cell line-engineering approach by removing nine Chinese hamster ovary (CHO) host cell hydrolases, which have previously been confirmed to degrade PS. Strikingly, the sequential genomic knockout (KO) of these hydrolases, including the genetic removal of two entire gene clusters of unprecedented size, yielded viable CHO host cell line variants. This novel host cell line was further optim

Enhancing CHO cell recombinant protein production using a perfusion-directed host evolution approach.

Clonally derived cell lines generated from Chinese hamster ovary (CHO) cells encounter numerous stressors when cultured in high-intensity perfusion bioreactors leading to poor process performance. To circumvent this, the ability of CHO cells to adapt to different culture environments was exploited. Here host cells were selected in the presence of physical and chemical stressors associated with a perfusion environment by culturing at a high cell density in a perfusion bioreactor for 30 days. Following recovery and expansion, the performance of the resulting perfusion-evolved host was evaluated

Glycoprofile Comparison of the SARS-CoV-2 Spike Proteins Expressed in CHO and HEK Cell Lines.

Coronavirus SARS-CoV-2 spike protein remains a key focus of research due to a continued need for diagnostic and therapeutic tools to monitor and respond to new variants. Glycosylation of the spike protein is critical for the protein's functions in viral attachment and host cell entry. For scalable and cost-effective production of the spike protein, expression system-driven divergence in glycosylation patterns on recombinant spike proteins needs to be fully understood. This study assessed the N-glycosylation profiles of a full-length trimeric spike protein expressed in either Human Embryonic Ki

Addition of valproic acid to CHO cell fed-batch cultures improves monoclonal antibody titers.

Improving the productivity of a biopharmaceutical Chinese hamster ovary (CHO) fed-batch cell culture can enable cost savings and more efficient manufacturing capacity utilization. One method for increasing CHO cell productivity is the addition of histone deacetylase (HDAC) inhibitors to the cell culture process. In this study, we examined the effect of valproic acid (VPA, 2-propylpentanoic acid), a branched-chain carboxylic acid HDAC inhibitor, on the productivity of three of our CHO cell lines that stably express monoclonal antibodies. Fed-batch shake flask VPA titrations on the three differe

[Characteristics of Vibrio cholerae Cef (CHO cell elongating factor): bioinformatics analysis and experimental data].

Bioinformatics analysis of the primary and secondary structure of the Vibrio cholerae Cef (CHO cell elongating factor) protein was conducted. Similarity with triacylglycerol lipases and cytotonic toxins of other bacterial species was observed. Cef was predicted to be a heat-tolerant serine lipase with the Kunitz domain and leucine zipper. These data were confirmed experimentally. The Cefs of the two biotypes of V. cholerae O1, as well as O139 and nonO1/nonO139 serogroups, were purified from the recombinant Escherichia coli strains carrying corresponding cloned genes, and their physicochemical

Establishment of a stable CHO cell line with high level expression of recombinant porcine IFN-β.

A CHO cell clone (CHO-PoIFN-β) with stable porcine IFN-β expression under control of CMV promoter was selected under G418 pressure. In a 25cm(2) cell culture flask (5 ml culture medium), the cumulative protein yield of recombinant PoIFN-β reached 2.3×10(6) IU/ml. This cells clone maintained stable expression for at least 20 generations even in the absence of G418 selection pressure. The expressed recombinant PoIFN-β could induce the expression of porcine Mx protein in PK15 cells, and activate the chicken Mx promoter-controlled luciferase reporter gene expression, confirming that the recombinan

Towards a cleaner CHO chassis: systematic knockout of host cell proteins for efficient biopharmaceutical manufacturing.

Host cell proteins (HCPs) are major process-related impurities in biopharmaceutical manufacturing, imposing metabolic burden on production hosts and complicating downstream purification, thereby impacting cost, product stability, and patient safety. Here, we engineered Chinese hamster ovary (CHO) cells toward a streamlined production chassis through multiplex CRISPR-mediated knockout of abundant and difficult-to-remove HCPs. Proteomic profiling across multiple clones, products, and downstream purification steps identified a core set of highly abundant proteins that contributed disproportionate

Effect of CHO cell line constructed with CMAH gene-directed integration on the recombinant protein expression.

Chinese hamster ovary (CHO) cells are the most widely used platform for recombinant therapeutic protein (RTP) production. Traditionally, the development of CHO cell lines has mainly depended on random integration of transgenes into the genome, which is not conducive to stable long-term expression. Cytidine monophosphate N-acetylneuraminic acid hydroxylase (CMAH) is expressed in CHO cells and produces N-hydroxyacetylneuraminic acid, which may cause a human immune response. However, the effects of transgene integration at the CMAH site on RTP expression in CHO cells remain unclear. In this study

[Development of an intensified fed-batch process to enhance recombinant antibody productivity of CHO cells based on cell cycle regulation].

Chinese hamster ovary (CHO) cells are the most widely used hosts for biopharmaceutical production, yet their low recombinant antibody expression efficiency remains one of key bottlenecks in industrial applications. To enhance anti-EGFRv III antibody expression in CHO cells, an intensified fed-batch process was successfully developed for Cell A and Cell B cell strains based on cell cycle regulation. First, flow cytometry was used to establish the relationship between cell cycle distribution and specific antibody production rate (Qp). The results showed that the proportions of cells in the G0/G1

Sensitivity of bulk electrical impedance spectroscopy (bio-capacitance) probes to cell and culture properties: Study on CHO cell cultures.

Bulk electrical impedance spectroscopy (bio-capacitance) probes, hold significant promise for real-time cell monitoring in bioprocesses. Focusing on Chinese hamster ovary (CHO) cells, we present a sensitivity analysis framework to assess the impact of cell and culture properties on the complex permittivity spectrum, εmix, and its associated parameters, permittivity increment, Δε, critical frequency, fc, and Cole-Cole parameter, α, measured by bio-capacitance probes. Our sensitivity analysis showed that Δε is highly sensitive to cell size and concentration, making it suitable for estimating bio

Purine mutants of mammalian cell lines: III. Control of purine biosynthesis in adenine phosphoribosyl transferase mutants of CHO cells.

Spontaneous and mutagen-induced 2,6-diaminopurine-resistant mutants of Chinese hamster ovary (CHO-K1) cells were isolated. Such mutants fell into two classes: spontaneous and ethylmethane-sulfonate-induced mutants had approximately 5% wild-type adenine phosphoribosyl transferase (APRT) activity, whereas ICR-170G-induced mutants had barely detectable APRT activity. Since it has been reported that human hypoxanthine-guanine phosphoribosyl transferase (HGPRT) (Lesch-Nyhan syndrome) and APRT mutants over-produce purines, we examined the control and rate of purine biosynthesis in the Chinese hamste

Differential effects of N-acetylmannosamine on O-glycan sialylation of therapeutic Fc-fusion glycoproteins in CHO and HEK293 cells.

Sialylation is a critical quality attribute of therapeutic glycoproteins, yet strategies to specifically modulate O-glycan sialylation remain underdeveloped compared to N-glycosylation. While N-acetylmannosamine (ManNAc) is widely utilized as an established enhancer of N-glycan sialylation, its efficacy in modulating O-glycan structures has remained largely unexplored across different mammalian hosts. In this study, we investigated ManNAc supplementation as a practical approach for modulating O-glycan sialylation in recombinant Chinese hamster ovary (CHO) cells. Using etanercept and erythropoi

High yield process for the production of active human α-galactosidase a in CHO-K1 cells through lentivirus transgenesis.

Fabry disease is an X-linked recessive disorder caused by a deficiency in lysosomal α-Galactosidase A. Currently, two enzyme replacement therapies (ERT) are available. However, access to orphan drugs continues to be limited by their high price. Selection of adequate high-expression systems still constitutes a challenge for alleviating the cost of treatments. Several strategies have been implemented, with varying success, trying to optimize the production process of recombinant human α-Galactosidase A (rhαGAL) in Chinese hamster ovary (CHO-K1) cells. Herein, we describe for the first time the a

Ubiquitous Chromatin Opening Elements (UCOEs) effect on transgene position and expression stability in CHO cells following methotrexate (MTX) amplification.

The requirement for complex therapeutic proteins has resulted in mammalian cells, especially CHO cells, being the dominant host for recombinant protein manufacturing. In creating recombinant CHO cell lines, the expression vectors integrate into various parts of the genome leading to variable levels of expression and stability of protein production. This makes mammalian cell line development a long and laborious process. Therefore, with the intention to accelerate process development of recombinant protein production in CHO systems, UCOEs are utilized to diminish instability of production by ma

Comparative study of the repair kinetics of chromosomal aberrations and DNA strand breaks in proliferating and quiescent CHO cells.

Repair kinetics observable at the level of exchange-type chromosomal aberrations (dicentric chromosomes), using fractionation and delayed-plating techniques, have been compared with repair kinetics of radiation-induced DNA double-strand breaks, measured with PFGE, and with repair kinetics of all strand breaks, measured with the alkali-unwinding technique. Only data from quiescent or proliferating CHO K1 cells obtained in the same laboratory were used. We determined repair kinetics in terms of the time constant tau (equal to half-time/log(e)2). The repair kinetics (tau approximately 11-14 min)

Valeric acid induces cell cycle arrest at G1 phase in CHO cell cultures and improves recombinant antibody productivity.

To find a more effective chemical reagent for improved monoclonal antibody (mAb) production, eight chemical reagents (curcumin, quercein, DL-sulforaphane, thymidine, valeric acid, phenyl butyrate, valproic acid, and lithium chloride) known to induce cell cycle arrest were examined individually as chemical additives to recombinant CHO (rCHO) cell cultures producing mAb. Among these chemical additives, valeric acid showed the best production performance. Valeric acid decreased specific growth rate (μ), but increased culture longevity and specific mAb productivity (qmAb ) in a dose-dependent mann

Efficient production of recombinant mAbs mediated by a MAR-enhanced transposon vector combined with blasticidin selection in CHO cells.

Recombinant antibodies, primarily produced in Chinese hamster ovary (CHO) cells, are widely used to treat various diseases. For industrial production, a rapid and efficient method to screen stable, high-expressing clones is essential. However, conventional screening based on random integration is often cumbersome and labor intensive. This study establishes a novel strategy for generating stable, high-yielding clones by combining a MAR-based piggyBac (PB) transposon semitargeted integration system with blasticidin (BSD) selection. Compared to the random integrated vector pMAR-mAb, the MAR-PB sy
