Effects of acute or prolonged administration of cabergoline on parkinsonism induced by MPTP in common marmosets.

The effects of a single treatment or chronic administration of cabergoline (1-[(6-allylergolin-8beta-yl)carbonyl]-1-[3-(dimethylamino)p ropyl]-3-ethyl-urea), a potent, long-lasting dopamine receptor agonist, on parkinsonism induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in common marmosets were studied. The administration of 0.2 mg/kg or a higer dose of cabergoline began to reverse parkinsonism-like symptoms 60 min after a subcutaneous injection, and showed steady and constant effects throughout the observation period. For prolonged administration, 0.2 mg/kg cabergoline was inj

Combined effects of cabergoline and L-dopa on parkinsonism in MPTP-treated cynomolgus monkeys.

The behavioral effects of L-dopa or cabergoline alone were compared with those of the joint administration of the two drugs in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-lesioned parkinsonian cynomolgus monkeys with attention to the induction of hyperactivity and dyskinesia. Cabergoline alone at 0.2 mg/kg or less improved in a dose-dependent fashion the parkinsonism without inducing hyperactivity and dyskinesia following a single subcutaneous injection. L-dopa alone improved the parkinsonism, but induced hyperactivity and dyskinesia, depending on the dose applied. Doses required for 5

Long-term primary pharmacotherapy of giant prolactinomas: A comparison of different cabergoline dosages.

OBJECTIVES: Dopamine agonists serve as first-line therapies for most giant prolactinomas. The objective of this study was to assess the differential impact of dopamine agonist dosing strategies on biochemical (prolactin normalisation) and radiological (tumour size reduction) responses in patients with giant prolactinomas. DESIGN: A single-centre retrospective real-life 23-year follow-up study. METHODS: Thirty-three patients with giant prolactinomas (≥ 4 cm) were treated with primary pharmacotherapy. We assessed pituitary function, the effect of therapy and different dosing regimens on prolac

Cabergoline monotherapy in polycystic ovary syndrome patients with elevated prolactin: a viable option?

PURPOSE: Cabergoline is widely used to treat hyperprolactinemia, but its effects on polycystic ovary syndrome (PCOS) remain unclear. Since hyperprolactinemia is present in nearly 30% of PCOS cases, this study aims to assess the impact of cabergoline on androgen levels and clinical outcomes in PCOS with elevated prolactin cases, discussing these findings with the results in prolactinoma cases. METHODS: A total of 66 women aged 18-40 were included in this retrospective cohort study, with 36 in the PCOS with elevated prolactin group (median 24.0 (22.0-27.5) years) and 30 in the prolactinoma grou

Brusatol Inhibits Tumor Growth and Increases the Efficacy of Cabergoline against Pituitary Adenomas.

Cabergoline (CAB) is the first choice for treatment of prolactinoma and the most common subtype of pituitary adenoma. However, drug resistance and lack of effectiveness in other pituitary tumor types remain clinical challenges to this treatment. Brusatol (BT) is known to inhibit cell growth and promote apoptosis in a variety of cancer cells. In our present studies, we investigate the effects of BT on pituitary tumor cell proliferation in vitro and in vivo. BT treatment resulted in an increase in Annexin V-expressing cells and promoted the expression of apoptosis-related proteins in rat and hum

Efficacy and Cerebrospinal Fluid Rhinorrhea after Cabergoline Treatment in Patients with Bioactive Macroprolactinoma.

Predicting dopamine agonist resistance in patients with macroprolactinoma is essential for clinicians to prevent treatment failure and subsequent complications such as medication-induced cerebrospinal fluid (CSF) rhinorrhea. We evaluated the features of patients with cabergoline resistance and CSF rhinorrhea in patients with prolactinomas with prolactin levels ≥1000 ng/mL. A total of 140 patients who were newly diagnosed with prolactinoma secreting only prolactin ≥1000 ng/mL and treated with cabergoline for the first time were included in this study. Based on the hormonal and radiologic respon

Cabergoline scavenges peroxynitrite enhanced by L-DOPA therapy in patients with Parkinson's disease.

Long-term or high-dose L-DOPA therapy in patients with Parkinson's disease (PD) may accelerate degeneration of dopaminergic neurons, possibly by increasing oxidative stress. To investigate the effects of cabergoline on peroxynitrite-mediated oxidative damage caused by L-DOPA, the concentration of 3-nitrotyrosine in cerebrospinal fluid (CSF) of 18 PD patients was compared with that in 20 normal controls. The concentration of 3-nitrotyrosine in patients following L-DOPA therapy was significantly higher than in untreated PD patients and controls. On the other hand, the concentration in PD patient

Cabergoline treatment rapidly improves gonadal function in hyperprolactinemic males: a comparison with bromocriptine.

This study evaluated the effects of chronic treatment with cabergoline (CAB), a new, potent and long-lasting ergoline-derived dopamine agonist, on seminal fluid parameters and sexual and gonadal function in hyperprolactinemic males in comparison with the effect of bromocriptine (BRC) treatment. Seventeen males with macroprolactinoma were treated with CAB at a dose of 0.5-1.5 mg/week (n = 7), or BRC at a dose of 5-15 mg/day (n = 10) for 6 months. Baseline prolactin (PRL) was 925.7 +/- 522.6 microg/l in the CAB-treated group and 1059.4 +/- 297.6 microg/l in the BRC-treated group. All the patient

Cabergoline-Resistant Recurrent Macroprolactinoma During Pregnancy: A Case Report and Review of the Literature.

We report a rare case of cabergoline-resistant recurrent macroprolactinoma during pregnancy. A 34-year-old woman, 17 weeks pregnant, presented with a four-week history of headache and a one-day history of visual disturbance. The medical history was significant for a macroprolactinoma with pituitary apoplexy, which was treated with an emergent pituitary resection approximately two years and four months before this presentation. At that time, postoperative prolactin levels improved but remained elevated despite gradually escalating cabergoline doses up to 6 mg per week. Our patient met the estab

Ultra-rapid prolactin normalization with cabergoline in a premenopausal woman with epilepsy and giant prolactinoma.

Giant prolactinomas are rare pituitary tumors characterized by extremely high prolactin levels and extensive extrasellar invasion. While dopamine agonists are usually effective, they may require months to normalize prolactin. We report a 47-year-old premenopausal woman presenting with new-onset temporal epilepsy and secondary amenorrhea. Assessments revealed marked hyperprolactinemia (mean serum prolactin concentration, 3902 ng/mL [SI: 169.8 nmol/L]) (reference range, 5.0-30.0 ng/mL [SI: 0.22-1.30 nmol/L]), and an invasive 47 × 43 × 50 mm adenoma extending into the right temporal lobe. Role of Tumor Volume and Prolactin Ratios in Differentiating Prolactinomas and Predicting Response to Cabergoline.

INTRODUCTION: Predicting resistance to cabergoline in prolactinomas remains a challenge. While prolactin (PRL)-based ratios help differentiate prolactinomas from non-functioning pituitary adenomas (NFPAs), their role in forecasting therapeutic response is unclear. OBJECTIVES: To assess the usefulness of PRL-based ratios and tumor volume in distinguishing between prolactinomas and NFPAs, as well as in predicting cabergoline resistance. METHODS: We conducted a retrospective study of 86 patients (46 NFPAs, 40 prolactinomas). Resistance was reclassified according to the updated 2023 Pituitary So

Successful response to cabergoline treatment in a non-diabetic cat with hypersomatotropism and pituitary macroadenoma.

CASE SUMMARY: A 12-year-old male neutered domestic shorthair cat was referred with severe polyphagia, polydipsia, polyuria, weight gain and signs of neurologic disease, including blindness, head tilt, and ataxia. Magnetic resonance imaging (MRI) identified a suprasellar mass consistent with a pituitary macroadenoma. Hypersomatotropism (HST) was diagnosed based on phenotypic changes compatible with acromegaly and an abnormally high serum insulin-like growth factor 1 (IGF-1) concentration (1253 ng/ml). Medical treatment with cabergoline monotherapy (10 μg/kg PO q24h) was prescribed. Progressive

Comparison of montelukast and cabergoline for prevention of ovarian hyperstimulation syndrome: in an experimental rat model.

Ovarian hyperstimulation syndrome (OHSS) is a serious iatrogenic complication that can occur during assisted reproductive techniques. The aim of this study is to investigate the effects of the leukotriene receptor antagonist (montelukast) treatment in prevention of OHSS and compare to cabergoline treatment. Twenty-four immature female Wistar rats were assigned to four groups. Group 1 was the control group. In the remaining three groups, OHSS was induced through ovarian stimulation with gonadotropins. No treatment was given to Group 2. Group 3 was administered a low-dose 100 mg/kg cabergoline t

Comparison of albumin and cabergoline in the prevention of ovarian hyperstimulation syndrome: A clinical trial study.

BACKGROUND: Ovarian hyperstimulation syndrome (OHSS) is the most serious and potentially life-threatening iatrogenic complication associated with ovarian stimulation during Assisted Reproductive Technology (ART) protocols. OHSS typically is a result of ovarian expression of vascular endothelial growth factor (VEGF) which increases vascular permeability. OBJECTIVE: Comparison of albumin and cabergoline in the prevention of OHSS. MATERIALS AND METHODS: 95 high risk infertile women for OHSS (more than 20 follicles in both ovaries at day of Human Chorionic Gonadotropin (HCG) injection) were rand

Efficacy of the combined cabergoline and octreotide treatment in a case of a dopamine-agonist resistant macroprolactinoma.

Prolactinomas in males can be voluminous macroadenomas invading the surrounding structures. Medical therapy with dopamine agonists (the treatment of choice for these tumours) may be ineffective in the case of pharmacological resistance. In such cases, even surgical and/or radiation therapy cannot be curative due to the invasive potential of the adenoma. Hence, the appropriate therapeutic approach for these tumours is still a relevant clinical problem for endocrinologists. We report the history of an adolescent male who was diagnosed with a large invasive macroprolactinoma in 2002. He had sever

Management of prolactinoma with cabergoline treatment in a pregnant woman during her entire pregnancy.

Management of prolactinoma in pregnancy is a big challenge for the treating obstetrician as prolactin levels are normally raised in pregnancy and this creates a possibility of missing the diagnosis of prolactinoma. Women with micro adenomas and intrasellar macro adenomas do not require serial magnetic resonance imaging (MRI) or visual field testing as required in macro adenomas with extrasellar extension. A strict and vigil monitoring during each trimester for any clinical signs and symptoms related to tumor will suffice for the diagnosis of enlarging prolactinoma and for any active interventi
