Cadmium toxicity on mouse pre-implantation zygotes in vitro: interactions of cadmium with manganese, zinc and calcium ions.

Simultaneous treatment with cadmium chloride (5 micrograms/ml medium) and manganese chloride (5 or 10 micrograms/ml medium) was less toxic to mouse pre-implantation zygotes in vitro than cadmium alone. Such a protective effect of manganese, resembling that of zinc, suggests that amelioration of cadmium-treated pre-embryos in vitro is not unique to zinc. The amelioration may require only the presence of another divalent metallic ion, such as zinc or manganese, which is capable of competing with cadmium for binding sites possibly involved in uptake. Under calcium-free conditions cadmium embryoto

The hepatoprotective effects of selenium against cadmium toxicity in rats.

Exposure to cadmium and other pollutants is a major environmental problem. Cadmium is causing acute liver injury but the mechanism of hepatotoxicity is poorly understood. The present study aimed to assess the possible reasons by which cadmium causes liver toxicity. Furthermore, the protective role of selenium against this toxicity was investigated. The hepatic level of lipid peroxidation (malondialdehyde, MDA), the antioxidant system (reduced glutathione, glutathione peroxidase, and thioredoxin reductase), as well as the levels of different lipid classes and the fatty acids pattern were determ

Optical studies on chemical bath deposited nanocrystalline CdS thin films.

Nanocrystalline cadmium sulphide thin films were prepared by the chemical bath (CB) deposition using a mixed aqueous solution of cadmium chloride, thiourea, and ammonium chloride. The XRD patterns showed that the CdS films were of hexagonal phase with preferred (002) orientation. From ellipsometric measurements, the film thickness was found to be in order of 66 nm and the optical band gap was estimated to be 2.4 eV.

Protective roles of glutathione in the toxicity of mercury and cadmium compounds to C6 glioma cells.

The neurotoxic effects of mercury (II) chloride, methylmercury (MeHg) and cadmium chloride on astrocytes were modelled using C6 glioma cell cultures. All three compounds were cytotoxic to these cells with an order of potency of cadmium > MeHg > mercurychloride. Addition of reduced glutathione (GSH) to the media protected the cells in all three cases, whereas depletion of GSH with l-buthionine-S,R-sulfoximine enhanced the toxicity of cadmium and mercury chloride but not MeHg. The effects of subcytotoxic concentrations of these compounds on intracellular GSH levels were assessed using chlorobima

Cadmium induces the expression of Grp78, an endoplasmic reticulum molecular chaperone, in LLC-PK1 renal epithelial cells.

To reveal the effects of cadmium exposure on the endoplasmic reticulum (ER) stress response, we examined the expression and function of 78-kDa glucose-regulated protein (Grp78) , an ER-resident molecular chaperone, in LLC-PK1 cells. In cells treated with 10 microM cadmium chloride, Grp78 protein levels increased after 6 hr and remained elevated at 24 hr. When cells were incubated with 1-20 microM CdCl2 for 6 hr, Grp78 increased in a dose-dependent manner. In addition, Grp78 mRNA levels were elevated in response to CdCl2 exposure. After exposure to 10 microM CdCl2, the levels of activating tran

Ionic mechanisms underlying spontaneous muscle contractions in the liver fluke, Fasciola hepatica.

Spontaneous contractions of liver fluke muscle were abolished in Ca(2+)-free saline and by 100 microM nifedipine and reduced by 5 mM cadmium chloride, suggesting that they are dependent on extracellular Ca(2+). Caffeine (5 mM) significantly increased contraction amplitude and frequency. Ryanodine (100 microM) failed to block the caffeine response but significantly reduced spontaneous contraction frequency, suggesting that intracellular stores have a functional role. Cyclopiazonic acid (5 microM) had no effect on the caffeine response or spontaneous activity. 3-Isobutyl-1-methylxanthine (IBMX),

Effect of Sub-Inhibitory Concentrations of Quaternary Ammonium Compounds and Heavy Metals on Antibiotic Resistance and Expression of Virulence Factors Among Staphylococcus spp. from Dairy Products.

Antimicrobial resistance is spreading rapidly throughout the world. The food chain can be one of the routes of transmission for microorganisms containing drug-resistance genes and thus serve as a channel for their transmission. Environmental stress and methods of preventing the spread of microorganisms trigger adaptive responses in bacterial cells. The aim of the present study was to determine the effect of the stress induced by sub-inhibitory concentrations (SICs) of cadmium chloride and benzalkonium chloride on antibiotic resistance and the expression of selected virulence factors in Staphyl

Restrictions on the applicability of mixed ligand chelate therapy (MLC) in acute cadmium intoxication.

An experimental study of the comparative effectiveness of single and mixed ligand chelates as antidotes for acute cadmium poisoning reveals few appreciable differences when the cadmium (as cadmium chloride or acetate) is administered ip. In each case the mixed ligand chelate system showed little or no improvement over the results obtained with the most effective of the individual components. This procedure, using mixed ligand chelate systems, may well be one which is limited to conditions more narrowly similar to those reported by Schubert and Derr (1978). It does not appear to possess the bro

Synthesis, crystal structure and thermal properties of catena-poly[[[bis-(2,3-di-methyl-pyrazine-κN 1)cadmium(II)]-di-μ-chlorido] monohydrate].

The title compound, {[CdCl2(C6H8N2)2]·H2O} n (C6H8N2 = 2,3-di-methyl-pyrazine) was prepared by the reaction of cadmium chloride with 2,3-di-methyl-pyrazine in water. The asymmetric unit consists of two crystallographically independent cadmium cations that are situated on a twofold rotation axis, two crystallographically independent chloride anions and 2,3-di-methyl-pyrazine ligands as well as one water mol-ecule that are all located in general positions. The cadmium cations are octa-hedrally coordinated by two terminally N-bonded 2,3-di-methyl-pyrazine ligands and four chloride anions and are

Suppression of Kupffer cell function prevents cadmium induced hepatocellular necrosis in the male Sprague-Dawley rat.

Exposure of humans to toxic metals and metalloids is a major environmental problem. Many metals, such as cadmium, can be hepatotoxic. However, the mechanisms by which metals cause acute hepatic injury are in many cases unknown. Previous reports suggest a major role for inflammation in acute cadmium induced hepatotoxicity. In initial experiments we found that a non-hepatotoxic dose of cadmium chloride (CdCl2; 2.0 mg/kg, i.v.) markedly increased the clearance rate of colloidal carbon from the blood, which is indicative of enhanced phagocytic activity by Kupffer cells (resident hepatic macrophage

Camphor Ameliorates Cadmium-induced Memory Deficits in Rats by Downregulating NF-κB and Enhancing Acetylcholine Signaling in the Hippocampus.

Cadmium (Cd) is a toxic heavy metal that can penetrate the blood-brain barrier, leading to harmful effects on the nervous system by disrupting the brain's antioxidant defense mechanisms. This research investigates how camphor, a natural compound recognized for its antioxidant effects, influences biochemical, molecular, and histological alterations in the hippocampus of rats exposed to cadmium. It also assesses its influence on the spatial memory abilities of these animals. Adult male Wistar rats were divided into four groups, each containing 10 rats: Control, Camphor, Cd, and Cd + Camphor. Ultrastructural study of the submandibular gland of the rat after 6-month exposure to cadmium and zinc in drinking water.

PURPOSE: Cadmium toxicity in the exposure of the general, professional and cigarette smoking populations has been well known. From the dental point of view, it is important to find out whether and how separate and joint exposures to cadmium and zinc affect the structure and function of the submandibular gland, which is the major saliva-releasing gland. Cadmium, a particularly active xenobiotic, damages cellular metabolism at the level of various enzymatic systems of the cell, which may disturb functioning of the salivary glands. Mutual interactions of cadmium and zinc suggest a protective role

Effects of heavy metals on structure, function, and metabolism of ciliated respiratory epithelium in vitro.

Tracheal explants were used to evaluate the relative ciliostatic and cytotoxic potential of heavy metal salts (cadmium chloride, chromium chloride, nickel chloride, and copper sulfate). Explants from hamster, rat, and guinea pig were all sensitive to the metals, though guinea pig explants showed the greatest difference between the untreated and metal treated tissues. Dosage levels were 50, 100, and 500 microM, for 24 to 148 h. Cadmium caused the greatest degree of ciliostasis and cell necrosis. Copper was less toxic, and nickel and chromium caused marginal damage when tested at 100 microM or l

Linking environmental cadmium to pterygium pathogenesis: Mechanistic insights from fibroblast proliferation and calcium antagonism.

Cadmium, a ubiquitous environmental toxicant, poses significant risks to human health, including ocular surface damage. This study investigates the link between cadmium exposure and pterygium, a common ocular fibrovascular proliferative disorder, using human conjunctival fibroblasts (HconFs) as an in vitro model. We demonstrated that low-dose cadmium chloride (CdCl2, 1 μmol/L) significantly enhanced HconFs proliferation and disrupted cell cycle progression by reducing the G₀/G₁ phase population and increasing G₂/M phase entry. Transcriptome analysis identified 724 differentially expressed gene
