In vitro hepatic metabolism of mefloquine using microsomes from cats, dogs and the common brush-tailed possum (Trichosurus vulpecula).

Feline infectious peritonitis (FIP) is a systemic, fatal, viral-induced, immune-mediated disease of cats caused by feline infectious peritonitis virus (FIPV). Mefloquine, a human anti-malarial agent, has been shown to inhibit FIPV in vitro. As a first step to evaluate its efficacy and safety profile as a potential FIP treatment for cats, mefloquine underwent incubation in feline, canine and common brush-tailed possum microsomes and phase I metabolism cofactors to determine its rate of phase I depletion. Tramadol was used as a phase I positive control as it undergoes this reaction in both dogs

Feline respiratory tract disease in Louisiana.

An investigation of feline respiratory tract disease (FRD) was made involving 65 affected cats (group 1) and 65 control cats (group 2) matched for age, sex, and residence. Feline calicivirus strains (FCV), feline herpesvirus-1 (FHV-1), or both, were isolated from 59% of group 1 cats and from 7% of group 2 cats. Isolation of these viruses showed a significantly greater (P < 0.05) relative risk for group 1 cats. Virus isolations were three times more frequent from males than from females older than 1 year (both groups), but were equally distributed between males and females younger than 1 year.

Administration of porcine surfactant in the treatment of a fatal case of acute respiratory distress syndrome in a cat with calicivirus pneumonia.

CASE SUMMARY: A 1-year-old, male neutered Persian cat presented to the emergency room with acute, severe wheezing and coughing after the introduction of a new kitten to the household. The patient progressed to hypoxemic respiratory failure consistent with acute respiratory distress syndrome (ARDS) and required mechanical ventilation. RT-PCR testing from a conjunctival and deep pharyngeal swab confirmed feline calicivirus infection. Because of refractory hypoxemia and declining lung compliance (2.4 ml/cmH2O), novel therapy with intratracheal porcine surfactant (Curosurf) was attempted. A dose o

Monitoring Seroprevalence of Infectious Diseases in the Florida Panther (Puma concolor coryi).

Infectious diseases can have detrimental effects on wildlife populations, particularly those that persist at small sizes, have low genetic diversity, and are affected by fragmented habitat. One such example is the endangered Florida panther (Puma concolor coryi), which has been intensively managed since the early 1980s, with the current population ranging between 120 and 230 individuals. For more than three decades, panthers have been captured, demographics recorded, and blood samples collected to monitor for multiple infectious diseases; however, an updated comprehensive study of many of thes

Investigation of the epidemiology of calicivirus infection of cats using molecular and virus isolation techniques.

Feline calicivirus (FCV) an important and widely detected upper respiratory system agent in cats. Being genetically diverse, FCV can cause different symptoms, such as pneumonia, oral lesions, conjunctivitis, arthritis, and, recently, virulent systemic disease. The present study first determined the presence/prevalence of FCV infection in sampled vaccinated/unvaccinated cats with suspected FCV and/or clinically healthy. Second, it compared PCR and virus isolation (VI) in detecting FCV in these cats. It also aimed to diagnose FCV, and evaluate the advantages/disadvantages of the region and prime

Using a gradient boosted model for case ascertainment from free-text veterinary records.

Case ascertainment for prevalence and incidence studies from veterinary clinical data poses a major challenge because medical notes are not consistently structured or complete. Using natural language processing (NLP) and machine learning, this study aimed to obtain accurate case recognition for feline upper respiratory tract infections (primarily caused by viruses such as feline herpes virus (FHV-1) and feline calici virus (FCV), and bacteria such as Chlamydophila felis, Mycoplasma felis and Bordetella bronchiseptica using retrospective electronic veterinary records from the Royal Society for

The Efficacy of Commercial Surface Sanitizers against Norovirus on Formica Surfaces with and without Inclusion of a Wiping Step.

Commonly used surface sanitizers often lack activity against human noroviruses (hNoVs). The impact of inactivation versus removal when these products are applied via wiping is poorly characterized. The purpose of this work was to assess the anti-hNoV efficacy of various surface sanitizer chemistries, as applied to a laminate material commonly used for restaurant tabletops, using standard surface assays (ASTM E1053-11) and a newly developed wiping protocol. Four commercially available products with different active ingredient(s) (i.e., ethanol [EtOH], acid + anionic surfactant [AAS], quaternary

Molecular characterization of feline caliciviruses isolated from several adult cats with atypical infection showing severe flu-like symptoms on a remote island in Ehime, Japan.

In November 2020, a volunteer group reported an outbreak of an infectious disease with a high fatality rate and flu-like symptoms among stray cats in Aoshima, a remote island in Ehime, Japan. Nine adult cats with severe symptoms were hospitalized. Feline calicivirus (FCV) was isolated from pharyngeal swabs of six hospitalized cats. An outbreak of virulent systemic FCV (VS-FCV) infection was initially suspected because of obvious flu-like symptoms in adult cats; however, no symptoms typically associated with VS-FCV, such as skin ulcers on the limbs, edema, or viremia, were observed. Notably, tw

Ability of vaccine strain induced antibodies to neutralize field isolates of caliciviruses from Swedish cats.

BACKGROUND: Feline calicivirus (FCV) is a common cause of upper respiratory tract disease in cats worldwide. Its characteristically high mutation rate leads to escape from the humoral immune response induced by natural infection and/or vaccination and consequently vaccines are not always effective against field isolates. Thus, there is a need to continuously investigate the ability of FCV vaccine strain-induced antibodies to neutralize field isolates. METHODS: Seventy-eight field isolates of FCV isolated during the years 2008-2012 from Swedish cats displaying clinical signs of upper respirato

POSTVACCINAL VIRAL DISEASE PRESENTATION IN TWO LITTERS OF CHEETAHS (ACINONYX JUBATUS).

Protective antibody titers against core vaccines have not been standardized for cheetahs (Acinonyx jubatus) under human care. Vaccine-induced disease has been suspected after administration of modified live virus vaccine (MLVV), but it has not been confirmed as the causative agent. MLVV and killed virus vaccines (KVV) elicit humoral response in cheetahs; however, the use of both vaccines for initial immunization in cheetah cubs <6 months old within the same population has not been reported. The current case series describes viral disease presentation in two cheetah litters after using both vac

Viral infections of the feline urinary tract.

The exact cause of hematuria, dysuria, and urethral obstruction remains unknown in a large percentage of naturally occurring cases of feline lower urinary tract disease (FLUTD). One attractive hypothesis implicates viruses as the cause of some idiopathic forms of FLUTD; supporting this hypothesis is the fact that a gamma herpesvirus, a calicivirus, and a retrovirus have been isolated from urine and tissues obtained from cats with this type of disease. Although the clinical course and laboratory findings of cats with idiopathic FLUTD are suggestive of an infectious cause, the question of whethe

Herpesvirus and calicivirus infection in a black-footed cat (Fels nigripes) family group following vaccination.

Preventive healthcare recommendations for zoo felids include vaccination against primary viral diseases that affect domestic felids. Although associated with a more-substantial immune response in their domestic counterparts, use of modified live virus vaccines (MLV) in nondomestic carnivores can result in vaccine-induced viral disease. This case report details a feline herpesvirus and calicivirus outbreak in two black-footed cat (Felis nigripes) kittens and their dam following use of an MLV in the kittens. Clinical signs included anorexia, nasal discharge, sneezing, and tachypnea. Patient hist

Efficient eukaryotic expression and potent antiviral activity of a long-acting recombinant feline interferon-ω2-Fc fusion protein against major feline viruses.

Feline interferon-ω2 (FeIFN-ω2) holds potential as a therapeutic agent against feline viral infections. However, its clinical application is limited by rapid clearance and suboptimal antiviral effectiveness. Thus, in this study, an Fc-fused construct, FeIFN-ω2-Fc, was engineered to improve antiviral potency and pharmacokinetic properties both in vitro and in vivo. Three recombinant constructs-native FeIFN-ω2, FeIFN-ω2-Dimer, and FeIFN-ω2-Fc-were expressed in Chinese Hamster Ovary cells. All showed strong antiviral activity (106.74-108.42 IU/mg) and effectively activated downstream interferon s

Structural elucidation and virucidal properties of resveratrol octamers isolated from Vatica albiramis.

Two highly condensed resveratrol octamers, vateriaphenol A and a newly discovered analogue, vaticanol Q, were isolated from the stem of Vatica albiramis. Vaticanol Q represents the first naturally occurring resveratrol octamer bearing a rare ortho-quinone moiety, as unambiguously established through comprehensive spectroscopic analyses, including low-temperature NMR and circular dichroism. The absolute configuration of vaticanol Q was deduced based on biosynthetic considerations and chiroptical data, further supported by micro-electron diffraction and synchrotron X-ray crystallography of its b

Limited efficacy of topical recombinant feline interferon-omega for treatment of cats with acute upper respiratory viral disease.

Despite a lack of controlled studies confirming its efficacy, recombinant feline interferon-omega (rfeIFN-ω) is used in the treatment of feline upper respiratory tract disease (FURTD), which is usually caused by feline calicivirus (FCV) or feline herpesvirus-1 (FHV-1). The aims of the present study were to investigate whether administration of rfeIFN-ω improves clinical signs in cats with acute FURTD and whether this treatment reduces shedding of FCV. Thirty-seven cats affected with acute FURTD were recruited into a prospective, randomised, placebo-controlled, double-blinded clinical trial. Th
