NFAT mediates pro-tumorigenic inflammation in cancer-associated fibroblasts in pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is characterized by a dense stroma, low immunogenicity, and resistance to therapy. Cancer-associated fibroblasts (CAFs) are key stromal cells within the tumor microenvironment (TME) that drive tumor progression. Interleukin-1 (IL-1) promotes fibrosis, pathogenic inflammation, and poor prognosis in PDAC. Using a single-cell multi-omic approach, we investigate the IL-1 signaling axis in human and mouse models of PDAC, identifying nuclear factor of activated T cells (NFAT) transcription factors as key mediators. IL1R1+ CAFs activate an inflammatory phenotyp

Brain metastasis-associated fibroblasts shape the tumour microenvironment to enhance cancer cell invasion.

Brain metastases (BrM) are a frequent and life-threatening complication of solid tumours, with lung cancer representing their most common source. While cancer-associated fibroblasts (CAFs) are well-established contributors to tumour progression in many extracranial malignancies, their presence and function within the brain tumour microenvironment, where fibroblasts have long been considered scarce or absent, remain poorly understood. Here, we isolated and characterised fibroblast-like cells from 13 human BrMs of diverse origins. These BrM-associated CAFs (BrM-CAFs) expressed canonical CAF mark

Targeting CTGF+ cancer-associated fibroblasts disrupts tumor-stroma crosstalk in pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is characterized by a heavily fibrotic tumor microenvironment (TME), primarily driven by cancer-associated fibroblasts (CAFs). Due to the high heterogeneity and diverse biological behaviors of CAFs, the current strategies targeting CAFs have limited benefits in clinic. This study identifies a subset of CAFs marked by high expression of connective tissue growth factor (CTGF), which enhances tumor cell migration and correlates with advanced PDAC stages. By combining in silico screening and functional assays, we identified DP as a first-in-class small-molec

[The Role of Tumor‒Draining Lymph Nodes in Photoimmunotherapy Targeting Cancer‒Associated Fibroblasts].

To effectively induce anti‒tumor immune responses, it is essential that tumor‒specific T cells are generated within the tumor microenvironment (TME), a process in which tumor‒draining lymph nodes (TDLNs) are thought to play a critical role. We have previously demonstrated that photoimmunotherapy targeting cancer‒associated fibroblasts (PIT‒CAF), which specifically targets the fibroblast activation protein (FAP), modulates the immune cell composition and function within the TME. In this study, we investigated the effects of CAFs and PIT‒CAF on tumor‒specific T cells in both the TME and TDLNs.

Stimuli-responsive polymer-dasatinib prodrug to reprogram cancer-associated fibroblasts for boosted immunotherapy.

The barriers from cancer-associated fibroblasts (CAFs) have diminished the clinical efficacy of immunotherapy for triple-negative breast cancer (TNBC). The obstacles from CAFs often result in poor drug penetration, constrained cytotoxic T lymphocyte infiltration, and an immunosuppressive microenvironment. Herein, chondroitin sulfate (CS) was engineered to conjugate dasatinib (DAS), a tyrosine kinase inhibitor, via the cathepsin B (CTSB)-responsive GFLG linker to produce CS-GFLG-DAS (CGD), which could be employed to reverse the CAF phenotype and regulate the biosynthesis of extracellular matrix

Snail1 as a key prognostic biomarker of cancer-associated fibroblasts in breast tumors.

Accurate cancer diagnosis is crucial for selecting optimal treatments, yet current classification systems often include non-responders who receive ineffective therapies. Cancer-associated fibroblasts (CAFs) play a central role in tumor progression, and CAF biomarkers are increasingly recognized for their prognostic value. Recent studies have revealed significant heterogeneity within CAF populations, with distinct subtypes linked to different tumors and stages of disease. In this review, we summarize recent findings from patient samples and mouse models of breast cancer, focusing on gene signat

Bile acids activate cancer-associated fibroblasts and induce an immunosuppressive microenvironment in cholangiocarcinoma.

Cholangiocarcinoma (CCA) is a highly lethal malignancy originating from the biliary tract and characterized with exposure to high levels of bile acids (BAs). Immunotherapies have demonstrated limited efficacy in CCA, but the underlying mechanism remains elusive. In this study, we reveal that the excessive BAs specifically activate GPBAR1 on cancer-associated fibroblasts (CAFs) to express high levels of CXCL10, enhancing epithelial-mesenchymal transition (EMT) and metastasis of CCA cells and creating an immunosuppressive tumor microenvironment (TME) by recruiting neutrophils in CCA. Cancer-associated fibroblasts in osteosarcoma: key players in immune escape and targeted therapy.

Osteosarcoma represents the most common principal malignant bone tumor that predominantly appears among teenagers and children. While multimodal treatment methods have greatly evolved with time, survival for recurrent or metastatic disease remains low due to the resistance that accumulates during treatment. Increasing evidence identifies the tumor microenvironment (TME), in particular cancer-associated fibroblasts (CAFs), as playing an important role in imposing immune suppression, enhancing tumor aggressiveness, and mediating resistance toward immunotherapy and chemotherapy. This article give

Brain metastasis-associated fibroblasts secrete fucosylated PVR/CD155 that induces breast cancer invasion.

Brain metastasis cancer-associated fibroblasts (bmCAFs) are emerging as crucial players in the development of breast cancer brain metastasis (BCBM), but our understanding of the underlying molecular mechanisms is limited. In this study, we aim to elucidate the pathological contributions of fucosylation (the post-translational modification of proteins by the dietary sugar L-fucose) to tumor-stromal interactions that drive the development of BCBM. Here, we report that patient-derived bmCAFs secrete high levels of polio virus receptor (PVR), which enhance the invasive capacity of BC cells. Mechan

Emerging Role of Cancer-Associated Fibroblasts in Progression and Treatment of Hepatocellular Carcinoma.

Hepatocellular carcinoma (HCC) is one of the most prevalent cancers worldwide and the fourth leading cause of cancer-related death globally. Tumor cells recruit and remodel various types of stromal and inflammatory cells to form a tumor microenvironment (TME), which encompasses cellular and molecular entities, including cancer-associated fibroblasts (CAFs), tumor-associated macrophages (TAMs), tumor-associated neutrophils (TANs), immune cells, myeloid-derived suppressor cells (MDSCs), immune checkpoint molecules and cytokines that promote cancer cell growth, as well as their drug resistance. Breast cancer-associated fibroblasts: their roles in tumor initiation, progression and clinical applications.

Breast cancer is the most common malignant tumor in women, and the incidence of this disease has increased in recent years because of changes in diet, living environment, gestational age, and other unknown factors. Previous studies focused on cancer cells, but an increasing number of recent studies have analyzed the contribution of cancer microenvironment to the initiation and progression of breast cancer. Cancer-associated fibroblasts (CAFs), the most abundant cells in tumor stroma, secrete various active biomolecules, including extracellular matrix components, growth factors, cytokines, prot

Targeting Cancer-Associated Fibroblasts with Small-Molecule Inhibitors: Molecular Mechanisms and Therapeutic Potential.

The multifaceted role of cancer-associated fibroblasts (CAFs) in orchestrating extracellular matrix remodeling, immune evasion, and therapeutic resistance positions these stromal cells as critical yet underexploited nodes in the tumor microenvironment (TME). While small-molecule agents offer practical advantages over biologics for stromal modulation, a systematic understanding of their mechanistic diversity and translational barriers remains incomplete. To address this gap, this review consolidates current knowledge on the principal signaling axes-TGF-β/STAT3, NF-κB, Notch, Hedgehog, PI3K/AKT,

NRF1 Induces ApoEhigh Cancer-Associated Fibroblasts to Promote Stemness of Renal Cell Carcinoma.

UNLABELLED: Cancer-associated fibroblasts (CAF) are abundant stromal cells in the tumor microenvironment (TME) that play a vital role in promoting tumor progression and drug resistance. The mechanisms regulating heterogeneity of CAFs in renal cell carcinoma (RCC) could represent potential targets for reprogramming the TME. In this study, we conducted single-cell RNA sequence and flow cytometry analyses that identified a CAF subset overexpressing apolipoprotein E (ApoE), which was correlated with poor survival in patients with RCC. Mechanistically, NRF1 activation in CAFs induced formation of A

YAP1-driven tumor-like cancer-associated fibroblasts modulate biomechanics of pancreatic ductal adenocarcinoma.

BACKGROUND: In pancreatic ductal adenocarcinoma (PDAC), activation of Yes-associated protein 1 (YAP1) is linked to increased extracellular matrix (ECM) stiffness, yet how YAP1 interacts with cancer-associated fibroblasts (CAFs) and their subtypes to promote stromal remodeling remains unclear. METHODS: Utilizing single-cell RNA sequencing (scRNA-seq) data to identify CAF subsets; validating the role of a specific subset, tumor-like CAFs (tCAFs), in YAP1-dependent promotion of ECM stiffness, epithelial-mesenchymal transition (EMT) combining the YAP1 inhibitor verteporfin with gemcitabine in KPC

FHL2 expression by cancer-associated fibroblasts promotes metastasis and angiogenesis in lung adenocarcinoma.

Cancer-associated fibroblasts (CAFs) contribute to the progression of lung cancer. Four and a half LIM domain protein-2 (FHL2) is a component of focal adhesion structures. We analyzed the function of FHL2 expressed by CAFs in lung adenocarcinoma. Expression of FHL2 in fibroblast subtypes was investigated using database of single-cell RNA-sequencing of lung cancer tissue. The role of FHL2 in the proliferation and migration of CAFs was assessed. The effects of FHL2 knockout on the migration and invasion of human lung adenocarcinoma cells and tube formation of endothelial cells induced by CAF-con

Eribulin normalizes pancreatic cancer-associated fibroblasts by simulating selected features of TGFβ inhibition.

BACKGROUND: Less than 11% of pancreatic cancer patients survive 5-years post-diagnosis. The unique biology of pancreatic cancer includes a significant expansion of its desmoplastic tumor microenvironment, wherein cancer-associated fibroblasts (CAFs) and their self-produced extracellular matrix are key components. CAF functions are both tumor-supportive and tumor-suppressive, while normal fibroblastic cells are solely tumor-suppressive. Knowing that CAF-eliminating drugs are ineffective and can accelerate cancer progression, therapies that "normalize" CAF function are highly pursued. Eribulin i

Simultaneously targeting cancer-associated fibroblasts and angiogenic vessel as a treatment for TNBC.

Fibrotic tumor stroma plays an important role in facilitating triple-negative breast cancer (TNBC) progression and chemotherapeutic resistance. We previously reported a rationally designed protein (ProAgio) that targets integrin αvβ3 at a novel site. ProAgio induces apoptosis via the integrin. Cancer-associated fibroblasts (CAFs) and angiogenic endothelial cells (aECs) in TNBC tumor express high levels of integrin αvβ3. ProAgio effectively induces apoptosis in CAFs and aECs. The depletion of CAFs by ProAgio reduces intratumoral collagen and decreases growth factors released from CAFs in the tu

Cancer-associated fibroblasts are a useful cytological finding for diagnosing pancreatic ductal adenocarcinoma.

INTRODUCTION: Cancer-associated fibroblasts (CAFs) are activated fibroblasts or myofibroblasts that play a crucial role in the invasiveness of pancreatic ductal adenocarcinoma (PDAC). In this study, the cytological features and diagnostic significance of CAFs based on pancreatic duct brushing cytology (PDBC) were evaluated. METHODS: The prevalence of fibrous stroma (FS) including CAFs on PDBC in 42 PDAC cases and 33 benign cases was retrospectively investigated. The average nuclear size of fibroblasts was compared between PDAC and benign cases to distinguish CAFs from normal FS. RESULTS: Ove

Loss of exosomal miR-3188 in cancer-associated fibroblasts contributes to HNC progression.

BACKGROUND: Head and neck cancer (HNC) is one of the most common deadly diseases worldwide. An increasing number of studies have recently focused on the malignant functions of cancer-associated fibroblasts (CAFs) in numerous cancers. However, the underlying mechanisms by which CAF-derived exosomes promote tumor progression need to be further elucidated. This study aims to determine whether the loss of specific miRNAs in CAF-derived exosomes may be involved in the malignant transformation of HNC. METHODS: MiRNA array and real-time PCR assays were used to analyze the differential expression of
