BACKGROUND: Heme oxygenases, HO-1 (Hmox1) and HO-2 (Hmox2), regulate skeletal muscle homeostasis by degrading heme and generating carbon monoxide (CO), a bioactive signalling molecule. Although HO-1 is known to influence muscle fibre composition and mitochondrial function, the role of HO-2 in activity-dependent neuromuscular plasticity remains poorly understood. This study aimed to define the distinct contributions of each isoform and test whether CO could restore muscle function in HO-deficient states.
METHODS: We generated Hmox1/2 double-knockout mice (Hmox1/2-/-) and compared their skeleta

Synthetic chemistry within living systems offers a transformative strategy for precision medicine by enabling the targeted generation of therapeutic agents directly at disease sites. Carbon monoxide (CO), an endogenous gasotransmitter upregulated in many cancer phenotypes, represents a unique yet underexploited chemical feedstock for such intracellular synthesis. However, the intrinsically low reactivity of CO necessitates transition-metal mediation, which is typically compromised by rapid deactivation by intracellular thiols such as glutathione (GSH). Here, we present a biocompatible albumin-

Diverse bacteria and archaea use atmospheric CO as an energy source for long-term survival. Bacteria use [MoCu]-CO dehydrogenases (Mo-CODH) to convert atmospheric CO to carbon dioxide, transferring the obtained electrons to the aerobic respiratory chain. However, it is unknown how these enzymes oxidize CO at low concentrations and interact with the respiratory chain. Here, we use cryo-electron microscopy and structural modeling to show how Mo-CODHMs (CoxSML) from Mycobacterium smegmatis interacts with its partner, the membrane-bound menaquinone-binding protein CoxG. We provide electrochemical,

Carbon monoxide (CO) has long been considered a toxic gas but is now a recognized bioactive gasotransmitter with potent immunomodulatory effects. Although inhaled CO is currently under investigation for use in patients with lung disease, this mode of administration can present clinical challenges. The capacity to deliver CO directly and safely to the gastrointestinal (GI) tract could transform the management of diseases affecting the GI mucosa such as inflammatory bowel disease or radiation injury. To address this unmet need, inspired by molecular gastronomy techniques, we have developed a fam

OBJECTIVE: We report longitudinal changes in neuroimaging and neuropsychological measurements in a case of carbon monoxide (CO) poisoning showing reversible changes in hippocampal volume, which was closely linked to the degree of memory impairment.
METHODS: The current study presents a female in her early 20s, with a high school level of education. She became aware of headaches and drowsiness while working in a restaurant that operated charcoal braziers, and she was urgently transported to our hospital. Her high blood carboxyhemoglobin concentration led to a diagnosis of CO poisoning, but no

Carbon monoxide (CO) can occur in numerous situations and ambient conditions, such as fire smoke, indoor fireplaces, silos containing large quantities of wood pellets, engine exhaust fumes, and when using hookahs. Symptoms of CO poisoning are nonspecific and can range from dizziness, headache, and angina pectoris to unconsciousness and death. This guideline presents the current state of knowledge and national recommendations on the diagnosis and treatment of patients with CO poisoning. The diagnosis of CO poisoning is based on clinical symptoms and proven or probable exposure to CO. Negative c

Contamination of breathing gas is a risk for all divers. Some hydrocarbon contaminants will be sensed by the diver and the dive profile aborted. On the contrary, carbon monoxide may not be recognized by the diver and catastrophic consequences can result. Reported here is the fatal case of carbon monoxide poisoning while scuba diving, an event that has rarely been reported in the medical literature. A detailed review of other published cases of CO poisoning while scuba diving is included, attempting to identify causes in common and propose methods of prevention.

Carbon monoxide (CO) exposure is a prevalent cause of poisoning worldwide. The cardiac effects of CO poisoning are well described and can manifest as angina, myocardial ischemia or infarction, cardiogenic shock, and/or life-threatening arrhythmias. Atrial fibrillation has been associated with severe CO poisoning; however, few cases have described atrial fibrillation in acute CO poisoning with regard to hyperbaric oxygen (HBO2) therapy. Herein, we describe a case of severe CO poisoning that caused atrial fibrillation with successful conversion to sinus rhythm following HBO2 therapy and discuss

BACKGROUND: The fetus is uniquely susceptible to carbon monoxide (CO) exposure. We present a case of severe unintentional CO poisoning in the first trimester of pregnancy.
CASE: A 23-year-old G5P2022 female at 11 weeks' gestational age sat in a car with the engine idling. She was unaware that the vehicle's exhaust pipe was blocked with snow. She was found to be unresponsive, with an initial carboxyhemoglobin (COHb) concentration of 47.1%. She underwent emergent treatment with hyperbaric oxygen therapy. The remainder of her pregnancy was complicated by a diagnosis of myasthenia gravis. She del

A 47-year-old previously-well woman was found dead on the floor of a shower cubicle on a property in rural South Australia. The impression of the attending doctor and police was of collapse due to natural disease. Although there was significant stenosing coronary artery atherosclerosis found at autopsy, cherry pink discoloration of tissues prompted measurement of the blood carboxyhemoglobin level which was found to be 55%. The source of the gas was a poorly-maintained hot water heater that was mounted on the inside wall of the shower. Construction of the shower using an impermeable concrete ra

The clinical features of parkinsonism in carbon monoxide (CO) intoxication have been associated with striatal-related neuronal networks. As parkinsonian and neuropsychiatric features are both related to presynaptic dopaminergic integrity, the aim of this study was to explore the clinical significance of 99mTcTRODAT-1 in grading neurobehavioral scores and parkinsonian severity in CO intoxication. We enrolled 64 patients with CO intoxication, including 29 with parkinsonism (parkinsonism[+] group) and 35 without (parkinsonism[-] group). All of the patients received 99mTcTRODAT-1 neuroimaging eval

Suicide by inhalation of carbon monoxide is not uncommon and usually involves car exhausts or burning charcoal or defective boilers. We report a case of a 25-year-old man, who committed suicide by inhaling carbon monoxide gas inside a polythene bag in a bathroom. The open carbon monoxide cylinder found inside the polythene bag was purchased online by the deceased a few days earlier. He had stated that the gas would be used for his experiment on the environment. A suicide note recovered from his trouser pocket revealed his intention for a painless death.

Retrograde amnesia is a recognised neurological complication of carbon monoxide poisoning. This article describes the case of a female found dead in her bath where initial post-mortem findings and the surrounding circumstances raised strong suspicions of homicide, especially when there was contradictory evidence from her husband who was the only other person present. He was later diagnosed as having retrograde amnesia between his two visits to the bathroom to attend to his wife which caused him to merge them into one event, thus arousing suspicions of foul play. The discussion explores the cur

Over the last decade, studies have unraveled many aspects of endogenous production and physiological functions of carbon monoxide (CO). The majority of endogenous CO is produced in a reaction catalyzed by the enzyme heme oxygenase (HO). Inducible HO (HO-1) and constitutive HO (HO-2) are mostly recognized for their roles in the oxidation of heme and production of CO and biliverdin, whereas the biological function of the third HO isoform, HO-3, is still unclear. The tissue type-specific distribution of these HO isoforms is largely linked to the specific biological actions of CO on different syst

A case of unilateral sensorineural hearing loss of the left ear after carbon monoxide intoxication was presented. The diagnosis was based upon an interview, medical examinations and audiometric investigations. Results of diagnostic evaluations, clinical presentation and treatment were discussed. Hearing improvement was obtained after 6 days of treatment and normal hearing returned after 14 days. Patients who suffer from CO intoxication are at risk of hearing impairment, therefore, there is a need for audiometric follow up in these patients.

Carbon monoxide poisoning is a frequent occurrence in both developed and under developed countries of the world. Carbon monoxide can be produced in fires, automobile engine exhausts and the incomplete combustion of organic matter. It is a "silent killer" that initially produces a mild progressive frontal headache, drowsiness and sleep that is usually ignored as common place. Continued low-level CO exposure for a long period of time in a confined space is cumulative and these accidental deaths are frequent but should be avoidable. Several usual and unusual poisonings are reported to illustrate

UNLABELLED: In pediatrics, measurements of gaseous transfer in steady-state (SS) are easily applied. Nevertheless, interpretation of results is uncertain because predicted values depend on numerous metabolic and ventilatory parameters. In healthy subjects, the only invariant parameter of reference, beside the blood gas, is the CO uptake (VCO) when normalized by the reject of CO2 (VCO2). Theoretically, a deficit of VCO/VCO2 (VCOSpecifique = SpVCO) should not be observed on a young asthmatic in remission. Such a deficit should be due to either a circulatory impairment of the "gas-exchanging orga

INTRODUCTION: Skin lesions may occur in approximately 30 p. 100 of cases of carbon monoxide poisoning. Flushes, erythematous to violaceous blisters on pressure zones due to sweat gland necrosis can be observed in such case.
CASE REPORT: We report a case of acral (fingers, ears and nose) superficial cutaneous necrosis due to carbon monoxide poisoning.
DISCUSSION: Such a complication had never been reported as a cutaneous manifestation of acute carbon monoxide intoxication. Also rare, acral necrotic lesions in a comatose or obnubilated patient should suggest this diagnosis.
