Tandem catalysis enables chlorine-containing waste as chlorination reagents.

Chlorinated compounds are ubiquitous. However, accumulation of chlorine-containing waste has a negative impact on human health and the environment due to the inapplicability of common disposal methods, such as landfill and incineration. Here we report a sustainable approach to valorize chlorine-containing hydrocarbon waste, including solids (chlorinated polymers) and liquids (chlorinated solvents), based on copper and palladium catalysts with a NaNO3 promoter. In the process, waste is oxidized to release the chlorine in the presence of N-directing arenes to afford valuable aryl chlorides, such

Three-component carboacylation of alkenes via cooperative nickelaphotoredox catalysis.

Various commercially available acyl chlorides, aldehydes, and alkanes were exploited for versatile three-component 1,2-carboacylations of alkenes to forge two vicinal C-C bonds through the cooperative action of nickel and sodium decatungstate catalysis. A wealth of ketones with high levels of structural complexity was rapidly obtained via direct functionalization of C(sp2)/C(sp3)-H bonds in a modular manner. Furthermore, a regioselective late-stage modification of natural products showcased the practical utility of the strategy, generally featuring high resource economy and ample substrate sco

Enhancement of Methane Catalysis Rates in Methylosinus trichosporium OB3b.

Particulate methane monooxygenase (pMMO), a membrane-bound enzyme having three subunits (α, β, and γ) and copper-containing centers, is found in most of the methanotrophs that selectively catalyze the oxidation of methane into methanol. Active sites in the pMMO of Methylosinus trichosporium OB3b were determined by docking the modeled structure with ethylbenzene, toluene, 1,3-dibutadiene, and trichloroethylene. The docking energy between the modeled pMMO structure and ethylbenzene, toluene, 1,3-dibutadiene, and trichloroethylene was -5.2, -5.7, -4.2, and -3.8 kcal/mol, respectively, suggesting

Noncovalent Interaction-Assisted Ferrocenyl Phosphine Ligands in Asymmetric Catalysis.

Noncovalent interactions are ubiquitous in nature and are responsible for the precision control in enzyme catalysis via the cooperation of multiple active sites. Inspired by this principle, noncovalent interaction-assisted transition metal catalysis has emerged recently as a powerful tool and has attracted intense interest. However, it is still highly desirable to develop efficient and operationally convenient ligands along this line with new structural motifs. Based on the specific nature of hydrogen bonding and ion pairing interactions, we developed a series of noncovalent interaction-assist

Selective Covalent Protein Modification by 4-Halopyridines through Catalysis.

We have investigated 4-halopyridines as selective, tunable, and switchable covalent protein modifiers for use in the development of chemical probes. Nonenzymatic reactivity of 4-chloropyridine with amino acids and thiols was ranked with respect to common covalent protein-modifying reagents and found to have reactivity similar to that of acrylamide, but could be switched to a reactivity similar to that of iodoacetamide upon stabilization of the positively charged pyridinium. Diverse, fragment-sized 4-halopyridines inactivated human dimethylarginine dimethylaminohydrolase-1 (DDAH1) through coval

Asymmetric Retro-Claisen Reaction by Chiral Primary Amine Catalysis.

The communication describes an enamine-based asymmetric retro-Claisen reaction of β-diketones by primary amine catalysis. The reaction proceeds via a sequence of stereoselective C-C formation, C-C cleavage, and a highly stereospecific enamine protonation to afford chiral α-alkylated ketones or macrolides with high yields and enantioselectivities. A detailed mechanism was explored on the basis of experimental evidence and computational studies to account for the observed stereocontrol.

Nickel Catalysis: Synergy between Method Development and Total Synthesis.

Nickel(0) catalysts have proven to be powerful tools for multicomponent coupling reactions in our laboratories over the past 15 years. This interest was originally sparked by the ubiquity of allylic alcohol motifs in natural products, such as (-)-terpestacin, which we envisioned assembling by the coupling of two π components (alkyne and aldehyde) with concomitant reduction. Mechanistic investigations allowed us to elucidate several modes of controlling the regioselectivity and stereoselectivity in the oxidative cyclization, and these insights enabled us to leverage combinations of alkenes and

Auto-Tandem Palladium Catalysis: From Isoxazole to 2-Azafluorenone.

An auto-tandem palladium catalysis from halogen-substituted isoxazoles and Michael acceptors is described. It involves two mechanistically distinct palladium-catalyzed reactions, a Heck reaction and a rearrangement, leading to 2-azafluorenones. It is the first example of palladium-catalyzed ring opening of isoxazoles and rearrangement of the β-imino ketone ring-opening product.

Biological catalysis regulated by cucurbit[7]uril molecular containers.

We report the synthesis of two-faced inhibitors 1-5 that contain both enzyme inhibitor and cucurbit[n]uril binding domains. The enzyme binding domains of 1-5 bind to the active sites of bovine carbonic anhydrase (BCA) or acetylcholinesterase (AChE) and inhibit their catalytic activities. Addition of CB[7] to BCA*1 and BCA*2 results in the transient formation of the BCA*1*CB[7] and BCA*2*CB[7] ternary complexes that undergo rapid dissociation to form free catalytically active BCA along with CB[7]*1 and CB[7]*2. The on-off cycle can be performed repetitively by the sequential addition of competi

Ascorbate sustains neutrophil NOS expression, catalysis, and oxidative burst.

Previous studies from this lab have demonstrated that in vitro ascorbate augments neutrophil nitric oxide (NO) generation and oxidative burst. The present study was therefore undertaken in guinea pigs to further assess the implication of ascorbate deficiency in vivo on neutrophil ascorbate and tetrahydrobiopterin content, NOS expression/activity, phagocytosis, and respiratory burst. Ascorbate deficiency significantly reduced ascorbate and tetrahydrobiopterin amounts, NOS expression/activity, and NO as well as free radical generation in neutrophils from scorbutics. Ascorbate and tetrahydrobiopt

Products of cyclooxygenase-2 catalysis regulate postoperative bowel motility.

Laparotomy involving manipulation of the small intestine causes injury, initiating an inflammatory cascade in the small bowel wall, which generates eicosanoids and proinflammatory cytokines. We have shown that ketorolac and salsalate, nonselective cyclooxygenase (COX) inhibitors, ameliorate postoperative small bowel ileus in a rodent model. Others have shown that interleukin-1 receptor antagonism improves postoperative gastric emptying. We examined whether inhibition of the proinflammatory cytokines, tumor necrosis factor alpha (TNFalpha) and interleukin-1 (IL-1), or selective blockade of cycl

Reductive dehalogenation by cytochrome P450CAM: substrate binding and catalysis.

Biological reductive dehalogenation reactions are important in environmental detoxification of organohalides. Only scarce information is available on the enzymology underlying these reactions. Cytochrome P450CAM with a known X-ray structure and well-studied oxygenase reaction cycle, has been studied for its ability to reduce carbon-halogen bonds under anaerobic conditions. The reductive reactions functioned with NADH and the physiological electron-transfer proteins or by using artificial electron donors to reduce cytochrome P450CAM. Halogenated methane and ethane substrates were transformed by

A Bioarmored Enzyme Cascade for Sustained Catalysis in Harsh Microenvironments.

Enzyme therapeutics operating in harsh physiological microenvironments are inherently constrained by rapid unfolding, proteolytic degradation, and acid-induced deactivation, especially in stomachs. These vulnerabilities severely limit catalytic longevity and practical efficacy, particularly for acetaldehyde detoxification in humans. Achieving durable enzymatic function in such hostile conditions requires a protective strategy that can physically armor enzymes while preserving molecular accessibility to substrates-an unresolved challenge in current biomaterial designs. Here, we introduce a bioa

Origin of Scission Selectivity for Phosphoranyl Radicals in Photoredox Catalysis.

Phosphoranyl radicals are versatile synthetic intermediates that can form downstream radicals through α- and β-scission. However, the unclear origin of scission selectivity hinders the rational design of new transformations using these elementary steps. Here we report a comprehensive study of phosphoranyl radical scission mechanisms for sulfonamide nucleophiles. Using a combination of experimental, computational, and machine-learning-driven strategies, the primary factors influencing selectivity in these transformations were uncovered. Computational and experimental studies revealed an unexpec

Photochemical Desaturation and Epoxidation with Oxygen by Sequential Flavin Catalysis.

Catalytic desaturations are important strategies for the functionalization of organic molecules. In nature, flavoenzymes mediate the formation of α,β-unsaturated carbonyl compounds by concomitant cofactor reduction. Contrary to many laboratory methods for these reactions, such as the Saegusa-Ito oxidation, no transition metal reagents or catalysts are required. However, a molecular flavin-mediated variant has not been reported so far. We disclose a photochemical approach for silyl enol ether oxidation, which leads to α,β-unsaturated ketones (13 examples) in very good yields. The flavin catalys

Organochalcogen ligands in catalysis of oxidation of alcohols and transfer hydrogenation.

Organochalcogen compounds have been used as the building blocks for the development of a variety of catalysts that have been studied comprehensively during the last two decades for several chemical transformations. Transfer hydrogenation (reduction of carbonyl compounds to alcohols) and oxidation of alcohols (conversion of alcohols to their respective ketones and aldehydes) are also among such chemical transformations. Some compilations are available in the literature on the development of catalysts, based on organochalcogen ligands, and their applications in Heck reaction, Suzuki reaction, an

Synthesis of Dendritic Glycoclusters and Their Applications for Supramolecular Gelation and Catalysis.

Glycoclusters with three, four, and six arms of glycosyl triazoles were designed, synthesized, and characterized. The self-assembling properties of these molecules and their catalytic activity as ligands in copper-catalyzed azide and alkyne cycloaddition (CuAAC) reactions were studied. The compounds with a lower number of branches exhibit excellent gelation properties and can function as supramolecular gelators. The resulting gels were characterized using optical microcopy and atomic force microscopy. The glycoconjugates containing six branches showed significant catalytic activity for copper

Catalysis-Based Total Syntheses of Pateamine A and DMDA-Pat A.

The marine natural product pateamine A (1) and its somewhat simplified designer analogue DMDA-Pat A (2) (DMDA = desmethyl-desamino) are potently cytotoxic compounds; most notably, 2 had previously been found to exhibit a promising differential in vivo activity in xenograft melanoma models, even though the ubiquitous eukaryotic initiation factor 4A (eIF4A) constitutes its primary biological target. In addition, 1 had also been identified as a possible lead in the quest for medication against cachexia, an often lethal muscle wasting syndrome affecting many immunocompromised or cancer patients. Overcoming product inhibition in catalysis of the intramolecular Schmidt reaction.

A method for carrying out the intramolecular Schmidt reaction of alkyl azides and ketones using a substoichiometric amount of catalyst is reported. Following extensive screening, the use of the strong hydrogen-bond-donating solvent hexafluoro-2-propanol was found to be consistent with low catalyst loadings, which ranged from 2.5 mol % for favorable substrates to 25 mol % for more difficult cases. Reaction optimization, broad substrate scope, and preliminary mechanistic studies of this improved version of the reaction are described.
