Brucella abortus strain 2308 produces brucebactin, a highly efficient catecholic siderophore.

Brucella abortus is known to produce 2,3-dihydroxybenzoate (2,3-DHBA) and to use this catechol as a siderophore to grow under iron-limited conditions. In this study a mutant (BAM41) is described that is deficient in siderophore production by insertion of Tn5 in the virulent B. abortus strain 2308. This mutant was unable to grow on iron-deprived medium and its growth could not be restored by addition of 2,3-DHBA. Production of catecholic compounds by both the Brucella mutant and parental strains under iron-deprivation conditions was assayed by TLC. Two catecholic substances were identified in t

Genotypes of catechol-O-methyltransferase and response to levodopa treatment in patients with Parkinson's disease.

A single nucleotide polymorphism at the nucleotide 1947 in the catechol-O-methyltransferase (COMT) gene encodes the high and low activity forms of the enzyme. We investigated COMT genotypes of 73 Korean patients with Parkinson's disease (PD), 29 with multiple system atrophy (MSA), and 49 controls, and analyzed the response to levodopa challenge in the PD patients. We found no significant difference in the distribution of the COMT genotypes among the three groups. The frequencies of the G- and A-alleles in the total population were 75 and 25%, respectively. The levodopa response was determined

Plasma metanephrines are markers of pheochromocytoma produced by catechol-O-methyltransferase within tumors.

This study examined whether the high sensitivity of plasma free metanephrines for diagnosis of pheochromocytoma may result from production of free metanephrines within tumors. Presence in pheochromocytomas of catechol-O-methyltransferase (COMT), the enzyme responsible for conversion of catecholamines to metanephrines, was confirmed by Western blot analysis, enzyme assay, and immunohistochemistry. Western blot analysis and enzyme assay indicated that membrane-bound and not soluble COMT was the predominant form of the enzyme in pheochromocytoma. Immunohistochemistry revealed colocalization of CO

Molecular origin of cancer: catechol estrogen-3,4-quinones as endogenous tumor initiators.

Cancer is a disease that begins with mutation of critical genes: oncogenes and tumor suppressor genes. Our research on carcinogenic aromatic hydrocarbons indicates that depurinating hydrocarbon-DNA adducts generate oncogenic mutations found in mouse skin papillomas (Proc. Natl. Acad. Sci. USA 92:10422, 1995). These mutations arise by mis-replication of unrepaired apurinic sites derived from the loss of depurinating adducts. This relationship led us to postulate that oxidation of the carcinogenic 4-hydroxy catechol estrogens (CE) of estrone (E1) and estradiol (E2) to catechol estrogen-3,4-quino

ESR identification of free radicals formed from the oxidation of catechol estrogens by Cu2+.

Catechol estrogens are genotoxic, indirectly through redox cycling mechanisms leading to oxidative DNA damage and directly by formation of quinone-DNA adducts. Previously, we demonstrated that Cu2+ can oxidize estradiol (E2) catechols, establishing a copper redox cycle leading to the formation of DNA strand breaks. The goal of this study was to use electron spin resonance techniques to identify the free radical intermediates formed. The 2- and 4-OH catechols of E2 and ethinyl estradiol (EE) were oxidized to semiquinone intermediates, stabilized by Mg2+, when incubated with Cu2+. The 4-OH-EE se

Effect of catechol on the discharge of muscle spindle afferents from the hind limb of the rat.

Previous work had shown that some of the effects of catechol could be via the fusimotor system. In order to determine the extent of fusimotor involvement, recordings have been made from muscle spindle afferents in split dorsal root filaments of anaesthetised rats. Catechol failed to excite de-efferented muscle spindles therefore eliminating many possible non-fusimotor effects. Over 80% of spindle afferents with intact efferents showed increased discharge frequency 1 min after injection, this increase often following a biphasic pattern with a pronounced pre-myoclonic burst and decline followed

Catechol-phytochemical conjugates potentiate quorum quenching and antivirulence response against Pseudomonas aeruginosa.

Development of drug hybrids with anti-quorum sensing (QS) and antivirulence properties has been widely regarded as a viable alternative for combating drug resistance in Pseudomonas aeruginosa. In this study, conjugates (1-3) were synthesized using three naturally-occurring phytochemicals: guaiacol, vanillin, and eugenol via pharmacophoric hybridization with catechol moiety. Among the synthesized conjugates that showed broad-spectrum antibacterial activity and iron-chelating potential, catechol-vanillin conjugate (2) demonstrated relatively higher degree of QS inhibition and significantly reduc

Functional characterization of Corynebacterium glutamicum catechol 2, 3-dioxygenase induced by aromatic compounds.

In bacteria, phenolic compounds are usually metabolized via the catechol pathway, in which catechol undergoes ring-opening reactions catalyzed by catechol 1, 2-dioxygenase (C12O, ortho cleavage) or catechol 2, 3-dioxygenase (C23O, meta cleavage), eventually entering central carbon metabolism. Corynebacterium glutamicum can utilize phenol as the sole carbon and energy source. The function of its C12O (encoded by ncgl2319) has been confirmed, whereas the putative C23O (encoded by ncgl2007, designated cgc23o) remains uncharacterized. In this study, we functionally characterized CgC23O, a protein

Multifunctional Mussel-Inspired Hydrogels and Films Formed via Catechol-VO2 Nanoparticle Coordination.

Marine mussels utilize DOPA-V metal coordination bonds as dynamic cross-links to fabricate tough and self-healing coatings and glues. Inspired by mussels, researchers have harnessed DOPA-metal coordination to create dynamic and mechanically responsive polymeric materials. Here, we harness the robust DOPA-V interactions observed in mussels combined with the temperature-responsive properties of vanadium dioxide (VO2) to develop a room-temperature process to produce thermochromic VO2 hydrogels and films. 4-arm polyethylene glycol (PEG) polymers functionalized with DOPA-catechol moieties were mixe

Ultrasmall catechol-PEG-anchored ferrite nanoparticles for highly sensitive magnetic resonance angiography.

Highly sensitive iron oxide nanoparticles with stable, safe and efficient surface functionalization, as potential substitutes for gadolinium-based contrast agents (GBCAs) with increasing biosafety concerns, exhibit great potential for high-performance magnetic resonance angiography (MRA). Herein, we developed ultrasmall catechol-PEG-anchored ferrite nanoparticles (PEG-UMFNPs) for highly sensitive MRA. The obtained nanoprobe has a high T1 relaxivity value (7.2 mM-1 s-1) due to its ultrasmall size and Mn doping. It has a suitable hydrodynamic size of 20 nm, which prevents rapid vascular extravas

Characterization of a unique catechol-O-methyltransferase as a molecular drug target in parasitic filarial nematodes.

BACKGROUND: Filarial nematodes cause severe illnesses in humans and canines including limb deformities and disfigurement, heart failure, blindness, and death, among others. There are no vaccines, and current drugs against filarial nematodes infections have only modest effects and are prone to complications. METHODOLOGY/PRINCIPAL FINDINGS: We identified a gene (herein called DiMT) encoding an S-adenosyl-L-methionine (SAM)-dependent methyltransferase with orthologs in parasite filarial worms but not in mammals. By in silico analysis, DiMT possesses catalytic sites for binding SAM and catecholam

Catechol-O-methyltransferase activity in individuals with substance use disorders: a case control study.

BACKGROUND: Impulsivity and substance use disorders (SUD) have been both associated with changes in dopaminergic processes. In this study, we intended to evaluate the dopaminergic function in imprisoned SUD offenders through the determination of s-COMT activity. METHODS: The study included 46 male individuals from a Portuguese penal institution. The participants were assessed through a battery of standardised instruments: Psychopathy Checklist-Revised (PCL-R), Barratt Impulsivity Scale Version 11 (BIS-11), and the European version of the Addiction Severity Index (EuropASI). In addition, s-COM

Thermofluor-Based Optimization Strategy for the Stabilization of Recombinant Human Soluble Catechol-O-Methyltransferase.

Catechol-O-methyltransferase (COMT) has been involved in a number of medical conditions including catechol-estrogen-induced cancers and a great range of cardiovascular and neurodegenerative diseases such as Parkinson's disease. Currently, Parkinson's disease treatment relies on a triple prophylaxis, involving dopamine replacement by levodopa, the use of aromatic L-amino acid decarboxylase inhibitors, and the use of COMT inhibitors. Typically, COMT is highly thermolabile, and its soluble isoform (SCOMT) loses biological activity within a short time span preventing further structural and functio

Catechol-O-methyltransferase gene expression in stress-induced and non-stress induced schizophrenia.

INTRODUCTION: As schizophrenia is a complex mental disorder and the outcome of gene-gene-environmental interactions, there are different possible pathophysiological mechanisms in different schizophrenia subtypes corresponding to various risk factors. This study was aimed at examining the impact of one of the most likely interactions, that is, 'dopamine and stress', in schizophrenia pathogenesis. METHODS: Here, we investigated the interaction between 'war-related psychological trauma' without brain trauma and catechol-O-methyltransferase gene. Using real-time PCR analysis we measured catechol-

Para-tertiary butyl catechol (PTBC), an industrial antioxidant induces human platelet apoptosis.

The catecholic derivative para-tertiary butyl catechol (PTBC) is a conventional antioxidant and polymerization inhibitor, which exhibits melanocytotoxic effects and contact dermatitis often leading to occupational leucoderma or vitiligo. Although numerous industrial workers will be in constant exposure to PTBC and its chances of getting entry into blood are most expected, its effect on blood components is still undisclosed. As platelets play a prominent role in dermatitis, inflammation, and immunity, in this study we have evaluated the effect of PTBC on human platelets in vitro. Exposure of pl

Structural insights into mechanisms for inhibiting amyloid β42 aggregation by non-catechol-type flavonoids.

The prevention of 42-mer amyloid β-protein (Aβ42) aggregation is promising for the treatment of Alzheimer's disease. We previously described the site-specific inhibitory mechanism for Aβ42 aggregation by a catechol-type flavonoid, (+)-taxifolin, targeting Lys16,28 after its autoxidation. In contrast, non-catechol-type flavonoids (morin, datiscetin, and kaempferol) inhibited Aβ42 aggregation without targeting Lys16,28 with almost similar potencies to that of (+)-taxifolin. We herein provided structural insights into their mechanisms for inhibiting Aβ42 aggregation. Physicochemical analyses reve

Oxidative modification of neurofilament-L and neuronal cell death induced by the catechol neurotoxin, tetrahydropapaveroline.

Tetrahydropapaveroline (THP), which is an endogenous neurotoxin, has been suspected to be associated with dopaminergic neurotoxicity of l-DOPA. In this study, we examined oxidative modification of neurofilament-L (NF-L) and neuronal cell death induced by THP. When disassembled NF-L was incubated with THP, protein aggregation was increased in a time- and THP dose-dependent manner. The formation of carbonyl compounds and dityrosine were observed in the THP-mediated NF-L aggregates. Radical scavengers reduced THP-mediated NF-L modification. These results suggest that the modification of NF-L by T

Application of gold nanoparticles/TiO2 modified electrode for the electrooxidative determination of catechol in tea samples.

A gold nanoparticles/TiO(2) composite modified Indium tin oxide (ITO) electrode has been constructed to study the electrochemical behaviour of catechol (CC) and hydroquinone (HQ) using cyclic voltammetry and differential pulse voltammetry (DPV). Increasing of separation of the oxidative peak potentials and peak current for CC and HQ in pH 6.0 phosphate buffer solution (PBS), make it suitable for selected determination of CC. After the optimization of the conditions, CC was determined by DPV and the linear range is from 1.0×10(-7) to 5.0×10(-4) mol L(-1) with a correlation coefficient of 0.999
