Synthesis of PbS nanorods and other ionic nanocrystals of complex morphology by sequential cation exchange reactions.

We show that nanocrystals (NCs) with well-established synthetic protocols for high shape and size monodispersity can be used as templates to independently control the NC composition through successive cation exchange reactions. Chemical transformations like cation exchange reactions overcome a limitation in traditional colloidal synthesis, where the NC shape often reflects the inherent symmetry of the underlying lattice. Specifically we show that full or partial interconversion between wurtzite CdS, chalcocite Cu(2)S, and rock salt PbS NCs can occur while preserving anisotropic shapes unique t

Beyond Salt Resistance: Ion-Specific Modulation of Microstructure and Rheology of Sepiolite Dispersions.

Sepiolite, a fibrous clay mineral, is highly valued for its ability to form gel networks and control rheology in saline environments. However, the ion-specific effects governing this behavior are not fully understood. This study systematically investigates the influence of four common electrolytes (NaCl, KCl, CaCl2, and MgCl2) across a wide concentration range (10-4 to 1 mol/L) on the colloidal stability, microstructure, and rheological behaviors of 5 wt % sepiolite dispersions. While all dispersions remained macroscopically stable against sedimentation, their rheological properties, including

The Transient Receptor Potential Melastatin 7 (TRPM7) Inhibitors Suppress Seizure-Induced Neuron Death by Inhibiting Zinc Neurotoxicity.

Transient receptor potential melastatin 7 (TRPM7) is an ion channel that mediates monovalent cations out of cells, as well as the entry of divalent cations, such as zinc, magnesium, and calcium, into the cell. It has been reported that inhibitors of TRPM7 are neuroprotective in various neurological diseases. Previous studies in our lab suggested that seizure-induced neuronal death may be caused by the excessive release of vesicular zinc and the subsequent accumulation of zinc in the neurons. However, no studies have evaluated the effects of carvacrol and 2-aminoethoxydiphenyl borate (2-APB), b

Interaction of alpha-latroinsectotoxin from Latrodectus mactans venom with bilayer lipid membranes.

alpha-Latroinsectotoxin (LIT) from Latrodectus mactans venom increased the conductance of bilayer lipid membranes (BLM) by inducing channel like activity. The channels formed had a maximal single channel conductance of 5 pS in 10 mM CaCl2 solution. This process occurred more rapidly in symmetrical 10 mM CaCl2 solution than in equimolar KCl or NaCl. The LIT induced conductance showed pronounced rectification, that was dependent upon the face of the BLM to which the LIT was applied. This suggests that the LIT molecules incorporate into the bilayer lipid membrane in an oriented manner. The ion ch

Parametrized, Transferable Classical Density Functional Theory for Alkane/Alkene Separation in Cationic Zeolites.

We present a classical density functional theory (DFT) enhanced with a binary solid-fluid interaction parameter, ksi, for the accurate and efficient prediction of the adsorption of small alkane/alkene mixtures in cation-exchanged zeolites. The parameter ksi scales the van der Waals cross-interaction energies as a factor (1 - ksi) to the Berthelot combining rule, and it turns out to be transferable among zeolite structures. Focusing on sodium-exchanged Faujasite (FAU) and Linde Type A (LTA) zeolites, we have parametrized kZ,π and kNa,π to account for interactions between the propene double bond

Large-scale purification and in vitro characterization of the assembly of MreB from Leptospira interrogans.

BACKGROUND: Weil's syndrome is caused by Leptospira interrogans infections, a Gram negative bacterium with a distinct thin corkscrew cell shape. The molecular basis for this unusual morphology is unknown. In many bacteria, cell wall synthesis is orchestrated by the actin homolog, MreB. METHODS: Here we have identified the MreB within the L. interrogans genome and expressed the His-tagged protein product of the synthesized gene (Li-MreB) in Escherichia coli. Li-MreB did not purify under standard nucleotide-free conditions used for MreBs from other species, requiring the continual presence of A

Pleiotropic syndrome of dehydrated hereditary stomatocytosis, pseudohyperkalemia, and perinatal edema maps to 16q23-q24.

Dehydrated hereditary stomatocytosis (DHS) is a rare genetic disorder of red cell permeability to cations, leading to a well-compensated hemolytic anemia. DHS was shown previously to be associated in some families with a particular form of perinatal edema, which resolves in the weeks following birth and, in addition, with pseudohyperkalemia in one kindred. The latter condition was hitherto regarded as the separate entity, "familial pseudohyperkalemia." DHS and familial pseudohyperkalemia are thought to stem from the same gene, mapping to 16q23-q24. This study screened 8 French and 2 American f

Permeability characteristics of deoxygenated sickle cells.

This study investigated the effect of acute deoxygenation on membrane permeability characteristics of sickle cells. Measured fluxes of Na+ and K+ in ouabain-inhibited cells, of chloride and sulfate exchange in 4,4'-diisothiocyanostilbene-2,2'-disulfonate (DIDS)-inhibited and untreated cells, and of erythritol, mannitol, and arabinose in cytochalasin B-inhibited cells indicated that a deoxygenation-induced permeability change occurred in sickle cells only for cations and chloride. Monovalent cation permeabilities increased five-fold, and chloride influx into DIDS treated cells was enhanced near

Analyses of metal ions in spider venoms in relation to insecticidal activity of clavamine.

To elucidate insecticidal activity of spider toxins, metal ions in venoms and in the bodies were determined by thin layer chromatography, spark source mass spectrometry, ion chromatography, inductively coupled plasma emission spectrometry and atomic absorption spectrometry. Two kinds of spiders were used, Nephila clavata and Nephila maculata. Metals from their venom glands were extracted with hydrochloric acid and the metal concentrations were almost the same in the two species. Many kinds of metals, Fe, Zn, Pb, Cu, Ca, Mg, Na, P and S were found at higher levels in the venoms at concentration

Cation flux studies of the lesion induced in human erythrocyte membranes by the thermostable direct hemolysin of Vibrio parahaemolyticus.

Vibrio parahaemolyticus, an important agent of seafood-borne gastroenteritis, expresses several putative virulence factors that could account for the disease symptoms of infected humans, namely, diarrhea, nausea, and abdominal cramps. The pathogenicity of V. parahaemolyticus correlates well with the Kanagawa phenomenon (the hemolytic ability of strains grown on Wagatsuma blood agar), implicating the thermostable direct hemolysin (TDH) as the predominant toxin responsible for pathogenicity. TDH-induced hemolysis could be inhibited by the addition of the osmolyte sorbitol to the extracellular so

Magnesium and membrane function in vascular smooth muscle.

Since a tight electromechanical coupling exists in vascular smooth muscle, even small shifts of the membrane potential are sufficient to change the vascular lumen. The extracellular H+ and K+ concentrations are important effectors for the adjustment of the membrane potential. The ion concentrations in the immediate neighbourhood of the cell membrane can be influenced by the microdynamic binding properties of the basement membrane and the other vascular connective tissue. These structures are polyanionic macromolecules to which mono- and divalent cations are extensively bound, and which are sep

Peptoid-based antimicrobial strategies against polymyxin-resistant Gram-negative bacteria.

AIMS: Polymyxins remain the mainstay antibiotic for the treatment of infections caused by multidrug-resistant bacteria. However, with the increase in the use of polymyxins, the simultaneous rise of polymyxin-resistance cases has been another global threat, necessitating the need for novel therapeutic strategies. This work aimed to evaluate the antibacterial activity of cationic peptoids against polymyxin-resistant bacteria of global priority. METHODS AND RESULTS: Three paired polymyxin-sensitive/polymyxin-resistant strains were included, along with two clinical isolates and one reference stra

Gating pore current in an inherited ion channelopathy.

Ion channelopathies are inherited diseases in which alterations in control of ion conductance through the central pore of ion channels impair cell function, leading to periodic paralysis, cardiac arrhythmia, renal failure, epilepsy, migraine and ataxia. Here we show that, in contrast with this well-established paradigm, three mutations in gating-charge-carrying arginine residues in an S4 segment that cause hypokalaemic periodic paralysis induce a hyperpolarization-activated cationic leak through the voltage sensor of the skeletal muscle Na(V)1.4 channel. This 'gating pore current' is active at

Modulation of intracellular Na+ concentration by BmK AS, a scorpion toxin, in B104 cell line.

ABSL Effects of BmK AS, a toxin from scorpion Buthus mortensi Karsch and an activator of skeletal muscle RyRs, on the intracellular Na+ concentration have been investigated in the B104 neuroblastoma cell line by fluorescence digital imaging techniques. The intracellular Na+ concentration was elevated by 500 nM BmK AS significantly over a 30 min period, and the effect could be enhanced by addition of ouabain. In addition, BmK AS induced a biphasic modulation of [Na+]i in the dose-dependent range from 20 to 500 nM in the presence of ouabain. The results suggest that BmK AS may modulate intracell

Brain (Na+,K+)-ATPase. Opposite effects of ethanol and dimethyl sulfoxide on temperature dependence of enzyme conformation and univalent cation binding.

We examined effects of ethanol and dimethyl sulfoxide on the regulation and apparent thermodynamic properties of moderate affinity Na+ and K+ binding that regulates the K+-dependent phosphatase activity of (Na+,K+)-ATPase. Ethanol and other alcohols reduced the apparent affinity for Na+ and K+ at their moderate affinity sites and increased the negative delta H and delta S of cation binding. Dimethyl sulfoxide had the opposite effects. Inhibition by ethanol was favored by high temperature or low K+. Ethanol potentiated inhibition of K+ binding by ATP or Mg2+. Ethanol also shifted the equilibriu

Syntheses and crystal structures of the quaternary thio-germanates Cu4FeGe2S7 and Cu4CoGe2S7.

The quaternary thio-germanates Cu4FeGe2S7 (tetra-copper iron digermanium hepta-sulfide) and Cu4CoGe2S7 (tetra-copper cobalt digermanium hepta-sulfide) were prepared in evacuated fused-silica ampoules via high-temperature, solid-state synthesis using stoichiometric amounts of the elements at 1273 K. These isostructural compounds crystallize in the Cu4NiSi2S7 structure type, which can be considered as a superstructure of cubic diamond or sphalerite. The monovalent (Cu+), divalent (Fe2+ or Co2+) and tetra-valent (Ge4+) cations adopt tetra-hedral geometries, each being surrounded by four S2- anion

Identification of a cation transport pathway in Neisseria meningitidis PorB.

Neisseria meningitidis is the main causative agent of bacterial meningitis. In its outer membrane, the trimeric Neisserial porin PorB is responsible for the diffusive transport of essential hydrophilic solutes across the bilayer. Previous molecular dynamics simulations based on the recent crystal structure of PorB have suggested the presence of distinct solute translocation pathways through this channel. Although PorB has been electrophysiologically characterized as anion-selective, cation translocation through nucleotide-bound PorB during pathogenesis is thought to be instrumental for host ce

Molecular mechanism of calcium inhibition in viral channelrhodopsins.

Viral channelrhodopsins (VCR1s) are giant-virus-encoded light-gated channels permeable to monovalent and divalent cations, including Na+ and Ca2+ ions, and inhibited by millimolar Ca2+ concentrations. Here, we combine X-ray crystallography, time-resolved UV-vis spectroscopy, and ATR-FTIR spectroscopy to investigate molecular mechanisms of ion permeation and Ca2+-dependent inhibition in OLPVR1. An atomic resolution structure of OLPVR1 obtained in the presence of 10 mM CaCl2 and 900 mM NaCl reveals a transient intracellular Ca2+ binding site near T87 and T88, close to the retinal cofactor. Identification of the channel-forming domain of Clostridium perfringens Epsilon-toxin (ETX).

Epsilon-toxin (ETX) is a potent toxin produced by Clostridium perfringens strains B and D. The bacteria are important pathogens in domestic animals and cause edema mediated by ETX. This toxin acts most likely by heptamer formation and rapid permeabilization of target cell membranes for monovalent anions and cations followed by a later entry of calcium. In this study, we compared the primary structure of ETX with that of the channel-forming stretches of a variety of binding components of A-B-types of toxins such as Anthrax protective antigen (PA), C2II of C2-toxin and Ib of Iota-toxin and found
