Bystander senescence in human peritoneal mesothelium and fibroblasts is related to thrombospondin-1-dependent activation of transforming growth factor-β1.

Senescence bystander effect refers to a phenomenon in which senescent cells elicit the development of senescence phenotype in their nearby young counterparts. In this paper we examined the mechanism of senescence bystander effect triggered by senescent human peritoneal mesothelial cells (HPMCs) in proliferating HPMCs and peritoneal fibroblasts (HPFBs). The results showed that conditioned medium (CM) derived from senescent HPMCs elicited a senescence response (growth inhibition coupled with increased expression of senescence-associated β-galactosidase and accumulation of histone γ-H2A.X) in eit

TGFbeta type II receptor signaling controls Schwann cell death and proliferation in developing nerves.

During development, Schwann cell numbers are precisely adjusted to match the number of axons. It is essentially unknown which growth factors or receptors carry out this important control in vivo. Here, we tested whether the type II transforming growth factor (TGF) beta receptor has a role in this process. We generated a conditional knock-out mouse in which the type II TGFbeta receptor is specifically ablated only in Schwann cells. Inactivation of the receptor, evident at least from embryonic day 18, resulted in suppressed Schwann cell death in normally developing and injured nerves. Notably, t

Computational investigation of mechanical mechanisms underlying diverse cerebellar lobular morphologies.

The mammalian cerebellum is a densely folded structure composed of lobules separated by deep fissures, while individual lobules display striking diversity in shape and size. Although multicellular processes such as granule cell proliferation and migration drive cerebellar morphogenesis, mechanical mechanisms that generate diverse lobular morphologies after initial folding remain poorly understood. Spatially heterogeneous cortical growth arising from multicellular dynamics is considered critical for the formation of characteristic lobular morphologies. In this study, we employed mathematical mo

Interleukin-24 in type 2 immune diseases.

Interleukin (IL)-24, a member of the IL-20 cytokine family, is secreted by multiple cell types, such as immune cells (T cells, B cells, NK cells, macrophages), and non-immune cells (epithelial cells and fibroblasts). IL-24 and its downstream signaling pathways mediate vital biological processes, including processes governing cell growth, fate determination, cell death, and inflammation, albeit with effects that are context-dependent across disease states. In this review, we present comprehensive summary and recent breakthroughs in IL-24 characterization and its emerging pathogenic functions in

Cell-wall recycling and synthesis in Escherichia coli and Pseudomonas aeruginosa - their role in the development of resistance.

The bacterial cell-wall that forms a protective layer over the inner membrane is called the murein sacculus - a tightly cross-linked peptidoglycan mesh unique to bacteria. Cell-wall synthesis and recycling are critical cellular processes essential for cell growth, elongation and division. Both de novo synthesis and recycling involve an array of enzymes across all cellular compartments, namely the outer membrane, periplasm, inner membrane and cytoplasm. Due to the exclusivity of peptidoglycan in the bacterial cell-wall, these players are the target of choice for many antibacterial agents. Our c

Developmental immunohistochemistry of growth inhibitory factor in normal brains and brains of patients with Down syndrome.

Growth inhibitory factor, a new metallothioneinlike protein, was investigated at postmortem examination in the brains of 18 patients with Down syndrome ranging in age from 18 weeks gestation to 50 years of age and in 20 age-matched normal controls by developmental immunohistochemistry. In the frontal cortex of both Down syndrome patients and controls, growth inhibitory factor immunoreactivity was localized in the cell bodies and processes of protoplasmic astrocytes from 18 weeks gestation, and these immunoreactive processes formed so dense a meshwork in the gray matter that they outlined neuro

[Is it possible to individualize microvillous large-cell lymphoma among sinusoid lymphomas?].

Microvillous non-hodgkin lymphomas are large cell malignant lymphomas with a sinus growth pattern and the presence of cytoplasmic processes detected by electron microscopy. Microvillous non-hodgkin lymphomas appear to be of B cell lineage. Neoplasms considered in the differential diagnosis include anaplastic large cell Ki-1 lymphomas, malignant histiocytosis and metastatic carcinoma or malignant melanoma. A panel of markers are usually sufficient to recognize all of these neoplasms except microvillous lymphomas. We report a case of microvillous non-hodgkin lymphoma characterized by a histologi

Intercellular calcium waves mediate preferential cell growth toward the wound edge in polarized hepatic cells.

During liver regeneration, hepatocytes sense the damage and initiate proliferation of the quiescent cells through poorly understood mechanisms. Here, we have used cultured hepatic cells to study the roles played by intercellular calcium in mediating wound-healing processes. Well-differentiated and polarized Hep-G2 cells repaired an experimentally induced wound by induction of cell divisions. The resulting cellular growth did not occur evenly across the healing cell lawn; instead, proliferations were three times more active within 150-200 microm from the wound edge than further away; this periw

WAVE2-Abi2 complex controls growth cone activity and regulates the multipolar-bipolar transition as well as the initiation of glia-guided migration.

Glia-guided migration (glia-guided locomotion) during radial migration is a characteristic yet unique mode of migration. In this process, the directionality of migration is predetermined by glial processes and not by growth cones. Prior to the initiation of glia-guided migration, migrating neurons transform from multipolar to bipolar, but the molecular mechanisms underlying this multipolar-bipolar transition and the commencement of glia-guided migration are not fully understood. Here, we demonstrate that the multipolar-bipolar transition is not solely a cell autonomous event; instead, the inte

Proliferation, apoptosis and expression of non-collagenous proteins: differences between the upper and the lower jaw bone in vitro.

One of the effects observed during several screening studies for osteocompatibility in vitro was that cells derived from the upper and lower jaw exhibited distinct differences regarding proliferation. Therefore, the aim of this study was to examine systematically whether a single osteoblast possesses abilities which are specific to the upper or lower jaw. Both human maxillary and mandibular bone samples without any clinical or radiographic evidence of pathology were obtained from 4 male donors aged between 40 and 45 years. Cells were cultured for up to 25 days to investigate in vitro developme

Stability analysis of a simplified yet complete model for chronic myelogenous leukemia.

We analyze the asymptotic behavior of a partial differential equation (PDE) model for hematopoiesis. This PDE model is derived from the original agent-based model formulated by Roeder (Nat. Med. 12(10):1181-1184, 2006), and it describes the progression of blood cell development from the stem cell to the terminally differentiated state. To conduct our analysis, we start with the PDE model of Kim et al. (J. Theor. Biol. 246(1):33-69, 2007), which coincides very well with the simulation results obtained by Roeder et al. We simplify the PDE model to make it amenable to analysis and justify our appr

Selective fasciculation as a mechanism for the formation of specific chemical connections between Aplysia neurons in vitro.

Selective fasciculation of growth cones along preestablished axon pathways expressing matching or complementary adhesion molecules is thought to be an important strategy in axon guidance. Growth cone inhibiting factors also appear to influence pathfinding decisions. We have used identified Aplysia neurons in vitro to explore the hypothesis that similar mechanisms could be involved in target selection. Co-cultures of L10 neurons with RB neuron targets or R2 neurons with RUQ neuron targets reliably formed chemical connections. In contrast, co-cultures of L10 with RUQ targets usually failed to fo

Role of ferroptosis and autophagy in pulmonary diseases.

Programmed cell death is regulated by genes that not only play important roles in the growth and development of the body and the homeostasis of tissues and organs, but also participate in various pathological processes. Besides apoptosis, programmed cell death includes autophagy, ferroptosis and pyroptosis. Recent studies have found that ferroptosis and autophagy are involved in the occurrence and development of various diseases. Here, we mainly discuss the role and relationship between ferroptosis and autophagy in various lung diseases, such as lung injury, pulmonary fibrosis, asthma, chronic

S-trityl-L-cysteine derivative induces caspase-independent cell death in K562 human chronic myeloid leukemia cell line.

Effect of CF(3)-STLC, a potent kinesin spindle protein (KSP) inhibitor, on K562 human CML cell line was investigated. Treatment with CF(3)-STLC induced mitotic arrest of the cell cycle with the appearance of characteristic monoastral spindles, subsequent apoptotic cell death and cleavage of PARP-1, caspase-3, and 4E-BP1. The wide ranging caspase inhibitor z-VAD fmk prevented the cleavage of caspase-3 and 4E-BP1, but failed to attenuate PARP-1 cleavage or cell death triggered by CF(3)-STLC. These results suggest that CF(3)-STLC can induce apoptotic cell death in a caspase-independent manner, an

Conditional ablation of integrin alpha-6 in mouse epidermis leads to skin fragility and inflammation.

Hemidesmosomes (HDs) are essential anchorage junctions which mediate the firm attachment of epithelia to the underlying basement membranes, of which one main component is the integrin α6β4. These specific junctions are also able to trigger signalling pathways, via the recruitment and interactions of signalling molecules with HD components such as the cytoplasmic tail of the β4 integrin or the plakin plectin. HDs must also assemble and disassemble depending on the tissue context for example during tissue remodelling. Alterations of HD components or their loss result in skin blistering disorders

VEGF ligands and receptors: implications in neurodevelopment and neurodegeneration.

Intensive research in the last decade shows that the prototypic angiogenic factor vascular endothelial growth factor (VEGF) can have direct effects in neurons and modulate processes such as neuronal migration, axon outgrowth, axon guidance and neuronal survival. Depending on the neuronal cell type and the process, VEGF seems to exert these effects by signaling via different receptors. It is also becoming clear that other VEGF ligands such as VEGF-B, -C and -D can act in various neuronal cell types as well. Moreover, apart from playing a role in physiological conditions, VEGF and VEGF-B have be

Patterns of intercellular connectivity in the mammalian cumulus-oocyte complex.

Electron and fluorescence microscopic techniques have been used in a complementary fashion to study the patterns of follicle cell-oocyte interactions within cumulus-oocyte-complexes of various mammals. The principal findings are: (1) two distinct types of transzonal processes exist that are distinguishable on the basis of cytoskeletal composition; (2) in some of the species examined (pig, goat, primate), corkscrew-shaped processes rich in tubulin, traverse the zona pellucida and are invaginated into the oocyte cortex; (3) actin-rich processes either ramify as a network at the outer surface of

CRISPR screens unveil nutrient-dependent lysosomal and mitochondrial nodes impacting intestinal tissue-resident memory CD8+ T cell formation.

Nutrient availability and organelle biology direct tissue homeostasis and cell fate, but how these processes orchestrate tissue immunity remains poorly defined. Here, using in vivo CRISPR-Cas9 screens, we uncovered organelle signaling and metabolic processes shaping CD8+ tissue-resident memory T (TRM) cell development. TRM cells depended on mitochondrial translation and respiration. Conversely, three nutrient-dependent lysosomal signaling nodes-Flcn, Ragulator, and Rag GTPases-inhibited intestinal TRM cell formation. Depleting these molecules or amino acids activated the transcription factor T

UV-A mediated induction of carotenoid accumulation in Dunaliella bardawil with retention of cell viability.

The effect of adding UV-A radiation (320-400 nm) to photosynthetically active radiation (PAR, 400-700 nm) during growth of the photosynthetic marine microalga Dunaliella bardawil was investigated in this work in terms of cell growth and carotenoid production. Although signs of slow cell growth (slight reduction of chlorophyll and protein content) were observed after 24 h of cell exposure to UV-A (40 micromol photons m(-2) s(-1) and 70 micromol photons m(-2) s(-1)) plus 140 micromol photons m(-2) s(-1) PAR , 84 h exposure to these UV-A conditions slightly stimulated cell growth and increased th

Regenerative perfusion bioreactor for biologics production with lower COGs.

Perfusion bioreactor systems enable intensified bioprocessing with high cell densities and volumetric productivities in monoclonal antibody (mAb) manufacturing. However, their widespread adoption remains constrained by the high consumption of chemically defined cell culture media. In this study, we introduce a multi-stage electrokinetic waste separation process that regenerates spent media, recovering up to 87.5% of the spent-medium volume for reuse. Using a mock-perfusion model, the regeneration-recycle processes were shown to sustain high-density Chinese hamster ovary cell growth, compared w
