Stichoposide C and Rhizochalin as Potential Aquaglyceroporin Modulators.

Aquaporins (AQPs) are a family of integral membrane proteins that selectively transport water and glycerol across the cell membrane. Because AQPs are involved in a wide range of physiological functions and pathophysiological conditions, AQP-based therapeutics may have the broad potential for clinical utility, including for disorders of water and energy balance. However, AQP modulators have not yet been developed as suitable candidates for clinical applications. In this study, to identify potential modulators of AQPs, we screened 31 natural products by measuring the water and glycerol permeabil

Permeability-Controlled Probe for Ratiometric Detection of Plasma Membrane Integrity and Late Apoptosis.

The destruction of plasma membrane integrity is closely related to immune response, neuronal injury, cell apoptosis, and other pathological events. However, the construction of ratiometric fluorescent probes capable of detecting plasma membrane integrity remains a significant challenge, hindering in-depth studies on related biomedical areas. Herein, a polarity-responsive fluorescent probe was constructed for the ratiometric detection of cell membrane integrity for the first time. The probe targeted intact plasma membranes in healthy cells and relocated into the cytoplasm to give significantly

Electrophysiology of mammalian gland cells.

The resting cell membrane potential varies from -40 to -70 mV according to type of gland cell and species. The RP depends mainly on the large transmembrane concentration gradient for K maintained by a pump mechanism extruding Na and accumulating K. Since the Na permeability (PNa) is much smaller than PK, the Na concentration gradient is less important. In addition to the dominant electrodiffusional control of RP the Na pump itself contributes since the active transport of Na (out) exceeds that of the active K uptake. Gland cells are generally electrically coupled--i.e., the junctional membrane

A bespoke microfluidic pharmacokinetic compartment model for drug absorption using artificial cell membranes.

Early prediction of the rate and extent of intestinal absorption is vital for the efficient development of orally administered drugs. Here we show a new type of pharmacokinetic compartment model that shows a threefold improvement in the prediction of molecular absorption in the jejunum than the current state-of-the-art in vitro technique, parallel artificial membrane permeability assays (PAMPA). Our three-stage pharmacokinetic compartment model uses microfluidic droplets and bespoke, biomimetic artificial cells to model the path of a drug proxy from the intestinal space into the blood via an e

Vasopressin-stimulated movement of drugs and uric acid across the toad urinary bladder.

Vasopressin is known to increase the permeability of the toad bladder, an analogue of the mammalian collecting duct, to water and hydrophilic solutes such as urea. In the present study, the effect of vasopressin on the permeability of a series of lipophilic compounds, including many commonly used drugs, has been determined. In all cases, permeability increased from 50 to 100%. The response to vasopressin was mediated by cyclic adenosine monophosphate (cAMP), and was generally not altered by phloretin, an agent that inhibits amide movement through the amide transport pathway. Evidence that thes

Triazine Linchpins for Selective Lys-Lys Stapling of Unprotected Peptides.

Peptide stapling exerts profound effects on key peptide properties, including metabolic stability, cell membrane permeability, and conformational constraint. Herein, we report a mild and efficient 2,4-dichloro-1,3,5-triazine-mediated stapling strategy for the selective cross-linking of lysine side-chain amines. This approach furnishes macrocyclic peptides of varying ring sizes. This method features broad functional group compatibility with most natural amino acid residues (His, Trp, Arg, Tyr, Ser and Thr), as well as N-terminal α-amino and C-terminal carboxylate moieties, whereas cysteine is n

Effect of calcium antagonist compound nisoldipine on transepithelial electrical parameters in the isolated frog cornea.

The new calcium antagonist, nisoldipine, inhibited short circuit current and transcorneal potential difference in the isolated frog cornea (a chloride transporting epithelium). Transepithelial resistance increased. The effects of nisoldipine were dose-dependent. Nisoldipine did not change the stimulation of short circuit current induced by epinephrine. Intracellular membrane potential across both the apical and basolateral cell membrane were decreased by nisoldipine. The data indicate that intracellular calcium seemed to modulate chloride transport of the cornea by changing cell membrane perme

Role of carbonic anhydrase IV in corneal endothelial HCO3- transport.

PURPOSE: Carbonic anhydrase activity has a central role in corneal endothelial function. The authors examined the role of carbonic anhydrase IV (CAIV) in facilitating CO(2) flux, HCO(3)(-) permeability, and HCO(3)(-) flux across the apical membrane. METHODS: Primary cultures of bovine corneal endothelial cells were established on membrane-permeable filters. Apical CAIV was inhibited by benzolamide or siRNA knockdown of CAIV. Apical CO(2) fluxes and HCO(3)(-) permeability were determined by measuring pH(i) changes in response to altering the CO(2) or HCO(3)(-) gradient across the apical membra

A physical interpretation of the phenomenological coefficients of membrane permeability.

A "translation" of the phenomenological permeability coefficients into friction and distribution coefficients amenable to physical interpretation is presented. Expressions are obtained for the solute permeability coefficient omega and the reflection coefficient sigma for both non-electrolytic and electrolytic permeants. An analysis of the coefficients is given for loose membranes as well as for dense natural membranes where transport may go through capillaries or by solution in the lipoid parts of the membrane. Water diffusion and filtration and the relation between these and capillary pore ra

Effect of inhibitors of cyclic nucleotide phosphodiesterases on electrical and contractile activity of smooth muscle cells.

Double sucrose gap experiments were carried out to study the effect of phosphodiesterase inhibitors and penetrating analogs of cyclic nucleotides on action potential and contraction of guinea pig ureteral smooth muscle cells. 3-Isobutyl-1-methylxanthine (10 microM) and dibutyryl-cAMP (20 microM) shortened the plateau of action potential and inhibited contraction of smooth muscle cells by increasing potassium permeability of their membrane. Vinpocetine (1 microM) and dibutyryl-cAMP (100 microM) strengthened contraction of smooth muscle cells and shortened action potentials by decreasing sodium

Identification of cyclic GMP-activated nonselective Ca2+-permeable cation channels and associated CNGC5 and CNGC6 genes in Arabidopsis guard cells.

Cytosolic Ca(2+) in guard cells plays an important role in stomatal movement responses to environmental stimuli. These cytosolic Ca(2+) increases result from Ca(2+) influx through Ca(2+)-permeable channels in the plasma membrane and Ca(2+) release from intracellular organelles in guard cells. However, the genes encoding defined plasma membrane Ca(2+)-permeable channel activity remain unknown in guard cells and, with some exceptions, largely unknown in higher plant cells. Here, we report the identification of two Arabidopsis (Arabidopsis thaliana) cation channel genes, CNGC5 and CNGC6, that are

Novel electrospun chitosan/PEO membranes for more predictive nanoparticle transport studies at biological barriers.

The design of safe and effective nanoparticles (NPs) for commercial and medical applications requires a profound understanding of NP translocation and effects at biological barriers. To gain mechanistic insights, physiologically relevant and accurate human in vitro biobarrier models are indispensable. However, current transfer models largely rely on artificial porous polymer membranes for the cultivation of cells, which do not provide a close mimic of the natural basal membrane and intrinsically provide limited permeability for NPs. In this study, electrospinning is exploited to develop thin c

Evaluation of the toxic potency of selected cadmium compounds on A549 and CHO-9 cells.

Cytotoxicity of cadmium sulphide, oxide and chloride was tested using A549 and CHO-9 cells. Metabolic activity of cells (MTT test) and cell membrane permeability (NRU test) were used as cytotoxicity endpoints. The results revealed unexpectedly low toxicity of cadmium sulphide as compared to chloride and oxide. This preliminary report does not provide any explanation for this effect, but the result may nevertheless be interesting for future studies of toxicity mechanisms of cadmium compounds. First cadmium compounds caused damage or change in the permeability of cell membranes, then inhibition

Novel Auristatins with High Bystander and Cytotoxic Activities in Drug Efflux-positive Tumor Models.

Auristatins, a class of clinically validated anti-tubulin agents utilized as payloads in antibody-drug conjugates, are generally classified by their membrane permeability and the extent of cytotoxic bystander activity on neighboring cells after targeted delivery. The drugs typically fall within two categories: membrane permeable monomethyl auristatin E-type molecules with high bystander activities and susceptibility to efflux pumps, or charged and less permeable monomethyl auristatin F (MMAF) analogs with low bystander activities and resistance to efflux pumps. Herein, we report the developmen

Myxoviruses do not induce non-specific alterations in membrane permeability early on in infection.

The permeability characteristics of cells infected with myxoviruses have been studied by measuring the concentrative uptake of nutrients, the concentration of intracellular K+, and the maintenance of the Na+ gradient across the plasma membrane. Cells either show no change at all (Sendai virus-infected BHK cells and measles virus-infected Vero cells) or they show a decreased ability to concentrate nutrients, while intracellular K+ and the Na+ gradient remain unchanged (Sendai and influenza virus-infected L-1210 cells, measles virus-infected lymphocytes and mumps virus-infected L-41 cells). Mechanisms for localization of radiopharmaceuticals in neoplasms.

Localization of a radiopharmaceutical agent in a "tumor" is best conceptualized in terms of the altered regional physiology attendant to the presence of the "tumor". Such localization should be expected to occur in association with other disease states characterized by similar altered regional physiology. Neoplasms, areas of inflammation, and certain phases of infarct development are characterized by increased permeability of their capillary beds to macromolecules. This is largely due to neovascularization and the large intercapillary pores associated with new growth of capillary beds in these

Folate-ClipTAC: Design and Proof of Concept Study.

OBJECTIVE: Proteolysis-targeting chimaeras (PROTAC) technology is a rapidly emerging and intensively investigated strategy in the field of new drug research and development. However, due to the tripartite structure of PROTAC molecules, their molecular weights are generally high, which results in poor cell membrane permeability and targeting. A folate-CLIPTAC system was proposed to overcome these limitations. METHODS: For proof of concept, the estrogen receptor (ER) was selected as the model target protein. Two clickable fragments (Part A and B) were synthesized first. Then, the stability, tar

Determination of volume and water permeability of plated cells from measurements of light scattering.

Measurements of cell membrane water osmotic permeabilities can be inaccurate because of the technical difficulties inherent to cell volume measurements and because of the presence of an unstirred water layer in contact with the cells. We detail here a method we have developed to quantify transient changes in cell volumes from the intensity of light scattered by cells. For this, we theorize how an unstirred layer originates in a perfusing chamber, and we calculate values for both cell membrane water osmotic permeability and unstirred layer thickness from time transient changes in scattered ligh
