The cells of origin of the incertofugal projections to the tectum, thalamus, tegmentum and spinal cord in the rat: a study using the autoradiographic and horseradish peroxidase methods.

Efferent projections of the zona incerta were examined in the rat using the autoradiographic and horseradish peroxidase methods, with special reference to the cytoarchitectonic structure of the zona incerta. Autoradiographic experiments showed that the incertofugal fiber systems reach ipsilaterally to the thalamus (lateral dorsal, central lateral, ventral lateral geniculate, parafascicular, subparafascicular and reuniens nuclei, and posterior nuclear complex), to the hypothalamus (dorsal, lateral and posterior hypothalamic areas), to the tectum (medial pretectal area, deep pretectal and pretec

Thalamic reticular nucleus in Alligator mississippiensis: Soma and dendritic morphology.

The thalamic reticular nucleus (TRN) is a critical structure influencing information transfer to the forebrain. In crocodilians, the TRN shares many features with its mammalian counterpart. One area that has not been explored is how individual neurons in the crocodilian TRN compare with those found in mammals. In mammals, TRN neurons are aligned parallel to the external border of the dorsal thalamus, have their dendrites oriented perpendicular to the fibers in the internal capsule, have fine, filamentous dendritic appendages, are either bipolar or multipolar, and are commonly considered to be

Variability of laminar patterns in the human lateral geniculate nucleus.

The structure of the human lateral geniculate nucleus has been studied on serial Nissl stained sections from 57 human brains. Most of the brains were from neurologically normal individuals and were obtained during routine autopsy procedures. The laminar arrangement within the human nucleus is surprisingly variable. It is always possible to recognize a small segment with two layers (the monocular segment), one with four layers and one with six layers. Often an 8-layered segment can also be seen. The posterior half of the nucleus, within which central vision is represented, is made up mainly of

Construction and simulation-experimental characterization of a fluorescence image-based finite element model for cell mechanics.

OBJECTIVE: A finite element (FE) model was developed to characterize the three-dimensional (3D) heterogeneous structure features of cells, and the influence mechanism of cytoskeleton remodeling on the mechanical properties of cells and the intracellular stress transmission behavior was analyzed. METHODS: Human lung adenocarcinoma A549 cells were used in this study, and the cytoskeleton model was constructed by using cytochalasin D (Cyto-D) at concentrations of 0, 0.1, and 1.0 μmol/L. Based on the 3D fluorescence image stacks obtained by confocal laser scanning microscopy (CLSM), combined with

[Effect of the intermittent hypoxic training on the functioning of peptidergic neurons of the paraventricular hypothalamic nucleus and brain stem neurons in rats].

Internittent hypoxic training (IHT) increased the quantity and secretory activity of peptidergic neurons of the paraventricular hypothalamic nucleus (PHN) and activated neurons of the dorsal motor nucleus of n.vagus. These structures seem to take part in realisation of the IHT activating effect on condition of the pancreatic delta-cells. The effect involves insulin-stimulating and insuloprotective effects realised via hypothalamic and neuro-conducting ways of regulation of the endocrine pancreas with a direct participation of hypothalamic neuropeptides.

Immunohistochemical labeling of the mu opioid receptor carboxy terminal splice variant mMOR-1B4 in the mouse central nervous system.

The mu opioid receptor gene Oprm is alternatively spliced into many variants, providing for the multiplicity of mu opioid receptor subtypes. One of the mouse variants, mMOR-1B4, is unique in that it displays high affinity towards a wide range of mu opioid receptor antagonists, but poor affinity towards most classical mu opioid agonists. The present study examined the immunohistochemical distribution of the mMOR-1B4 variant in mouse brain and spinal cord. mMOR-1B4-like immunoreactivity (mMOR-1B4-LI) was enriched in many regions of the brain, spinal cord and in the dorsal root ganglia. Gene expression in rat vestibular and reticular structures during and after space flight.

Space flight produces profound changes of neuronal activity in the mammalian vestibular and reticular systems, affecting postural and motor functions. These changes are compensated over time by plastic alterations in the brain. Immediate early genes (IEGs) are useful indicators of both activity changes and neuronal plasticity. We studied the expression of two IEG protein products [Fos and Fos-related antigens (FRAs)] with different cell persistence times (hours and days, respectively) to identify brainstem vestibular and reticular structures involved in adaptation to microgravity and readaptat

Expression of corticotropin-releasing hormone transgenes in neurons of adult and developing mice.

The DNA sequences important for cell-specific expression and developmental regulation of corticotropin-releasing hormone (CRH) were analyzed in transgenic mice. A construct containing 0.5 kb of CRH 5' flanking DNA linked to the chloramphenicol acetyltransferase reporter gene was expressed in many brain regions and in several ectopic peripheral sites, suggesting that this portion of the CRH gene contains basal promoter activity but lacks DNA elements necessary for appropriate tissue specificity. Cell specificity of transgene expression was examined with a CRH-beta-galactosidase reporter constru

Apoptosis of odontoclasts under physiological root resorption of human deciduous teeth.

This study was designed to establish the apoptosis of odontoclasts during physiological root resorption of human deciduous teeth. Deciduous teeth were fixed, decalcified, and embedded in paraffin for immunohistochemical (IHC) observations and in Epon for transmission electron microscopy (TEM). Apoptotic cells were identified by terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-digoxigenin nick-end labeling (TUNEL), and then tartrate-resistant acid phosphatase (TRAP) activity was determined on the same sections. Epon-embedded specimens were sectioned serially into 0.5-microm semithin se

An efficient autometallography approach to localize lead at ultra-structural levels of cultured cells.

Understanding the precise intracellular localization of lead (Pb) is a key in deciphering processes in Pb-induced toxicology. However, it is a great challenge to trace Pb in vitro, especially in cultured cells. We aimed to find an innovative and efficient approach to investigate distribution of Pb in cells and to validate it through determining the subcellular Pb content. We identified its ultra-structural distribution with autometallography under electron microscopy in a choroidal epithelial Z310 cell line. Electron microscopy in combination with energy-dispersive X-ray spectroscope (EDS) was

Cerebellar and thalamic stimulation treatment for epilepsy.

The present chapter describes the most important available experimental and clinical evidence on the role of electrical stimulation of the cerebellum or the thalamus in the control of epilepsy. Cerebellum serves as an integrator of sensory information and regulator of motor coordinating and training. The sole output of the cerebellum is inhibitory Purkinje cell projections to deep cerebellar nuclei in the brainstem. Cerebellar stimulation in animal models of epilepsy has given mixed results. Nevertheless, more than 130 epileptic patients have been subjected to cerebellar stimulation and the re

Basal forebrain amnesia: does the nucleus accumbens contribute to human memory?

OBJECTIVE: To analyse amnesia caused by basal forebrain lesions. METHODS: A single case study of a patient with amnesia after bleeding into the anterior portion of the left basal ganglia. Neuropsychological examination included tests of attention, executive function, working memory, recall, and recognition of verbal and non-verbal material, and recall from remote semantic and autobiographical memory. The patient's MRI and those of other published cases of basal forebrain amnesia were reviewed to specify which structures within the basal forebrain are crucial for amnesia. RESULTS: Attention a

The Role of (Nuclear) Lipid Droplets in the Pathogenesis of Metabolic Syndrome.

Metabolic syndrome (MetS) is a widespread multi-component pathological condition characterized by meta-inflammation and cellular dysfunction. MetS and other metabolic diseases (metabolic dysfunction-associated steatotic liver disease (MASLD), cardiovascular-kidney-metabolic syndrome (CKMS)) stem from the disorder of energy metabolism and changes in the structure and function of specialized organelles such as lipid droplets, endoplasmic reticula, mitochondria, and nuclei. The discovery of lipid droplets within the nucleus and the investigation of their functions across various cell types in bot

Genomic analysis of intracranial and subcortical brain volumes yields polygenic scores accounting for variation across ancestries.

Subcortical brain structures are involved in developmental, psychiatric and neurological disorders. Here we performed genome-wide association studies meta-analyses of intracranial and nine subcortical brain volumes (brainstem, caudate nucleus, putamen, hippocampus, globus pallidus, thalamus, nucleus accumbens, amygdala and the ventral diencephalon) in 74,898 participants of European ancestry. We identified 254 independent loci associated with these brain volumes, explaining up to 35% of phenotypic variance. We observed gene expression in specific neural cell types across differentiation time p

The origin of nuclear bodies: a study of the undifferentiated epithelial cells of the equine small intestine.

During an electron and light microscopic study of the equine intestinal epithelium, it was observed that some secretory granules of the undifferentiated crypt epithelium were incorporated into the nucleus during mitosis. A study was made of the chemical nature of the granules, using standard histochemical techniques: PAS-Alcian blue, Deamination-PAS, and Ninhydrin-Schiff reactions. The granules contained a neutral protein-polysaccharide complex with many terminal amino groups, possibly an antibody (IgA). The intranuclear granules underwent coalescence and degeneration during differentiation. T

Multicharged Phthalocyanines as Selective Ligands for G-Quadruplex DNA Structures.

The stabilization of G-Quadruplex DNA structures by ligands is a promising strategy for telomerase inhibition in cancer therapy since this enzyme is responsible for the unlimited proliferation of cancer cells. To assess the potential of a compound as a telomerase inhibitor, selectivity for quadruplex over duplex DNA is a fundamental attribute, as the drug must be able to recognize quadruplex DNA in the presence of a large amount of duplex DNA, in the cellular nucleus. By using different spectroscopic techniques, such as ultraviolet-visible, fluorescence and circular dichroism, this work evalua

Delivery Characterization of SPL84 Inhaled Antisense Oligonucleotide Drug for 3849 + 10 kb C- > T Cystic Fibrosis Patients.

Recent advances in the therapeutic potential of RNA-related treatments, specifically for antisense oligonucleotide (ASO)-based drugs, have led to increased numbers of ASO regulatory approvals. In this study, we focus on SPL84, an inhaled ASO-based drug, developed for the treatment of the pulmonary disease cystic fibrosis (CF). Pulmonary drug delivery is challenging, due to a variety of biological, physical, chemical, and structural barriers, especially when targeting the cell nucleus. The distribution of SPL84 throughout the lungs, penetration into the epithelial cells and nucleus, and structu

Hepatoblastoma in a cat.

Hepatoblastomas are neoplasms that originate from putative pluripotential stem cells of the liver. A hepatic mass from an 8-year-old Abyssinian cat was composed of cords and sheets of neoplastic cells, with scattered rosettes and small ductal structures. Most neoplastic cells had a pale eosinophilic cytoplasm and a round to ovoid nucleus. The tumor also had short spindle cells with an oval nucleus. Immunohistochemically, neoplastic cells were weakly positive for embryonic hepatocellular markers, such as alpha-fetoprotein and cytokeratin (CK) 8/18, but negative for the hepatocellular marker Hep

Long-term cinemicrography of erythroblasts from a patient with congenital dyserythropoietic anemia type III: direct observation of dysplastic erythroblast formation.

Erythroblasts from a patient with congenital dyserythropoietic anemia type III (CDA III) were observed by time-lapse phase-contrast cinemicrography. The erythroblasts were compressed with a new mechanical device; this procedure enhanced the visualization of their structures. Soret band micrography was carried out simultaneously to assess cytoplasmic hemoglobinization. The proliferation and differentiation course of the majority of CDA III erythroblasts was similar to that of normal erythroblasts. The formation of dysplastic erythroblasts showed two patterns of progression: (1) Usually, the pro
