Mannan (mannosylated glycoproteins) in the outermost layer of the Candida cell wall may be the first molecules that interact with host dendritic cells (DCs) and activate immune responses that determine disease outcomes. However, little is known about how different mannan structures of common oral Candida species affect DC activation. The effects of heat-inactivated (HI) yeast cells and soluble mannan of Candida albicans, Candida parapsilosis, and Candida dubliniensis on bone marrow-derived DC (BMDC) responses were compared. HI Candida and the mannan exhibited different effects on BMDC activati

OBJECTIVE: Our purpose was to determine whether cultured human decidual cells produce chemokines in response to different strains of group B streptococci and purified bacterial cell wall components.
STUDY DESIGN: Human decidual cells were cultured from term placentas by standard techniques. Different strains of group B streptococci were isolated from neonates with early-onset group B streptococci sepsis. Confluent cell monolayers were incubated with these different strains of group B streptococci and various concentrations of purified bacterial cell wall components (including lipoteichoic aci

Lytic transglycosylases (LT) are enzymes involved in peptidoglycan (PG) remodeling. However, their contribution to cell-wall-modifying complexes and their potential as antimicrobial drug targets remains unclear. Here, we determined a high-resolution structure of the LT, an outer membrane lipoprotein from Neisseria species with a disordered active site helix (alpha helix 30). We show that deletion of the conserved alpha-helix 30 interferes with the integrity of the cell wall, disrupts cell division, cell separation, and impairs the fitness of the human pathogen Neisseria meningitidis during inf

BACKGROUND: Candida albicans is associated with high mortality among immunocompromised patients. Resistance to and toxic side effects of antifungal drugs require the development of alternative antifungal agents. AMP-17 is a novel antimicrobial peptide derived from Musca domestica that exerts excellent antifungal effects against the Candida species. In this article, we discuss the potential mechanism of AMP-17 against C. albicans from the perspective of affecting the latter's cell external structure.
METHODS: Recombinant AMP-17 was prepared by prokaryotic expression system, and its anti-C. alb

Glycosylphosphatidylinositol-anchored proteins were reported to be involved in many developmental progresses in Arabidopsis. Here I report that, a group of homologous glycosylphosphatidylinositol-anchored proteins from SKU5-Similar family regulate seedling root development of Arabidopsis through controlling cell polar expansion and cell wall synthesis. Due to the irregular expansion of root cells and the defective synthesis of cell walls, their knockout mutants generated shorter roots with irregularly shaped root cells, and thicker cell walls.

We have investigated a signal transduction system proposed to allow Streptomyces coelicolor to sense and respond to changes in the integrity of its cell envelope. The system consists of four proteins, encoded in an operon: sigmaE, an RNA polymerase factor; CseA (formerly ORF202), a protein of unknown function; CseB, a response regulator; and CseC, a sensor histidine protein kinase with two predicted transmembrane helices (Cse stands for control of sigma E). To develop a sensitive bioassay for inducers of the sigE system, the promoter of the sigE operon (sigEp) was fused to a reporter gene conf

The growing number of patients susceptible to invasive Candida albicans infections has intensified the need for effective antifungal targets in pathways essential for fungal growth and pathogenesis. Among these pathways, phosphate homeostasis has emerged as a significant determinant of virulence, yet how phosphate availability shapes cell wall structure in response to host-derived oxidative stress remains unclear. During commensal growth, C. albicans cells typically enjoy phosphate repletion and a neutral redox environment. Transitioning to invade host tissues, they simultaneously experience p

The effects of low (4 °C) versus ambient (25 °C) temperature on cell wall turnover, and redox homeostasis in 'Jin'Aofen' figs were investigated. Compared with 25 °C, storage at 4 °C reduced decay and respiration rates, delayed firmness loss and color deterioration. It also suppressed PG, PME and CEL activities, maintained higher contents of protopectin and cellulose; and slowed soluble pectin accumulation. Storage at 4 °C maintained higher activities of SOD, CAT, POD, APX and GR. It also preserved the levels of non-enzymatic antioxidants, including TPC, TFC, ASA GSH. Meanwhile, it reduced H2O2

Plant life is coordinated by several developmental and defense associated mechanisms and cell wall is actively important in these mechanisms. The plant cell wall also serves as an important component system that monitors plant growth and defense apart from only protecting the cell. However, resident sensors of the plant cell membrane and pattern recognition receptors (PRRs) also constitute major response mechanisms in plants while they are both as well developed for maintaining cell wall integrity (CWI). These PRRs are molecules such as ligands, peptides or glycans that are released for sensin

Streptococcus mutans, a cariogenic bacterium in humans, is associated with systemic disorders. Its cariogenic factors include glucosyltransferases (GTFs) and the glycosyltransferase rhamnose-glucose polysaccharide I (RgpI), which is involved in cell wall synthesis. However, the potential roles of these enzymes in systemic disorders remain unclear. We constructed a luciferase-tagged S. mutans UA159 mutant strain that lacked rgpI to explore the involvement of this enzyme in the systemic pathogenicity of S. mutans. We also employed the luciferase-tagged S. mutans UA159 variant, which exhibited re

Ice formation and growth during freezing in processed foods containing water can deteriorate food quality. Naturally derived antifreeze proteins and antifreeze polysaccharides are an attractive solution to this problem. Alkaline extracts from the basidiomycete Flammulina velutipes (enokitake) are known to inhibit ice crystal growth and are expected to maintain frozen food quality. In this study, polysaccharides/oligosaccharides (POS) were obtained from the readily available edible mushrooms F. velutipes, Hypsizygus marmoreus, Pleurotus eryngii, and Grifola frondosa. POS extracts were isolated

The widespread emergence of multidrug-resistant pathogenic bacteria across various environments, healthcare settings, and food industries, combined with the development of new methods to combat them, highlights the need for more precise, rapid, and cost-effective pathogen detection techniques. This is especially important for clinically relevant pathogens, as it allows treatment to begin as quickly as possible, enables more effective targeted therapies to be chosen, helps preserve the effectiveness of current antibacterial agents, and prevents infections from water- and foodborne bacterial pat

INTRODUCTION: Papillary renal cell carcinoma accounts for one tenth of all renal cell carcinomas. Compared to other renal cell carcinoma subtypes, it is more often localized at the time of diagnosis and rarely metastasizes to the skin. There are no previously reported cases of cutaneous papillary renal cell carcinoma localized to the abdominal wall which we present here.
CASE PRESENTATION: A 77 year-old female with multiple previous cancers, including a stage 1 left papillary renal cell carcinoma, treated with partial nephrectomy 32 months prior to presentation, was found to have a left upper

Gut microbiota affect progression of rheumatoid arthritis (RA). The present study aims at investigating the protective potential of Bifidobacterium longum cell wall lipoproteins (Lpps) shown to modulate the intestinal microbiome and prevent osteoarthritis. Arthritis was induced by collagen (CIA) or anti-collagen antibodies (CAIA) injection. Intake of 0.5 mg of Lpps/L, but not 0.25 and 1 mg of Lpps/L, significantly alleviated RA symptoms in CIA DBA/1OOaHsd mice. The arthritis index (AI) was also reduced in CAIA mice. In the CIA-protected group, colon Ligilactobacillus murinus, caecal Lactobacil

The Cdc14 phosphatase family is highly conserved in fungi. In Saccharomyces cerevisiae, Cdc14 is essential for down-regulation of cyclin-dependent kinase activity at mitotic exit. However, this essential function is not broadly conserved and requires only a small fraction of normal Cdc14 activity. Here, we identified an invariant motif in the disordered C-terminal tail of fungal Cdc14 enzymes that is required for full enzyme activity. Mutation of this motif reduced Cdc14 catalytic rate and provided a tool for studying the biological significance of high Cdc14 activity. A S. cerevisiae strain e

Covering: 2015 to 2022Staphylococcus aureus (S. aureus) is responsible for several community and hospital-acquired infections with life-threatening complications such as bacteraemia, endocarditis, meningitis, liver abscess, and spinal cord epidural abscess. In recent decades, the abuse and misuse of antibiotics in humans, animals, plants, and fungi and the treatment of nonmicrobial diseases have led to the rapid emergence of multidrug-resistant pathogens. The bacterial wall is a complex structure consisting of the cell membrane, peptidoglycan cell wall, and various associated polymers. The enz

Streptococcus pyogenes, or Group A Streptococcus (GAS), is a strictly human pathogen that causes a wide range of diseases, including skin and soft tissue infections, toxic shock syndrome and acute rheumatic fever. We have recently reported that Spy1094 and Spy1370 of S. pyogenes serotype M1 are N-acetylglucosamine (GlcNAc) deacetylases. We have generated spy1094 and spy1370 gene deletion mutants in S. pyogenes and gain-of-function mutants in Lactococcus lactis. Similar to other cell wall deacetylases, our results show that Spy1094 and Spy1370 confer lysozyme-resistance. Furthermore, deletion o

Staphylococcus aureus can complicate preceding viral infections, including influenza virus. A bacterial infection combined with a preceding viral infection, known as superinfection, leads to worse outcomes than a single infection. Most of the pulmonary infection literature focuses on the changes in immune responses to bacteria between homeostatic and virally infected lungs. However, it is unclear how much of an influence bacterial virulence factors have in single or superinfection. Staphylococcal species express a broad range of cell wall-anchored proteins (CWAs) that have roles in host adhesi

Fungal keratitis is one of the most common blindness-causing diseases, but clinical antifungal treatment remains a challenge. The fungal cell wall and biofilm matrix which severely confine the drug preparation are the critical obstructive factors to therapeutic effects. Herein, we report ethylenediaminetetraacetic acid (EDTA) modified AgCu2O nanoparticles (AgCuE NPs) to disrupt the cell wall and then eradicate C. albicans through the internal cascade synergistic effects of ion-released chemotherapy, chemodynamic therapy, photodynamic therapy, and mild photothermal therapy. AgCuE NPs exhibited
