The drug discovery and therapeutic nano-strategies targeting cellular senescence.

Cellular senescence is a primary driver of aging, where damaged or senescent cells (SnCs) continuously accumulate in the body, altering the local tissue environment and causing various pathologies in different organs or tissues, offering brand-new diagnostic and therapeutic possibilities for aging-related disease. This review begins by discussing the multiple molecular mechanisms of cellular senescence. We mainly focus on the therapeutic strategies and their clinical applications targeting cellular senescence, where senolytic and senomorphic drugs are currently two main categories of anti-agin

Prx1/PHB2 axis mediates mitophagy in oral leukoplakia cellular senescence.

BACKGROUND: Oral leukoplakia (OLK) is the most common oral potentially malignant disorder (OPMD), which can be malignantly transformed into oral squamous cell carcinoma (OSCC). Peroxiredoxin1(Prx1) has been predicted to bind to Prohibitin2 (PHB2), which confers to affect OLK progression; however, the mechanism of Prx1/PHB2 mediated mitophagy involved in OLK remains unclear. METHODS: This study aimed to explore the mechanism of the Prx1/PHB2 axis on senescence in OLK through mediating mitophagy. The positive rate of Ki67 and the expression of p21, p16, PHB2, and LC3 in human normal, OLK, and O

Mapping epidermal and dermal cellular senescence in human skin aging.

Single-cell RNA sequencing and spatial transcriptomics enable unprecedented insight into cellular and molecular pathways implicated in human skin aging and regeneration. Senescent cells are individual cells that are irreversibly cell cycle arrested and can accumulate across the human lifespan due to cell-intrinsic and -extrinsic stressors. With an atlas of single-cell RNA-sequencing and spatial transcriptomics, epidermal and dermal senescence and its effects were investigated, with a focus on melanocytes and fibroblasts. Photoaging due to ultraviolet light exposure was associated with higher b

Therapeutic approaches targeting aging and cellular senescence in Huntington's disease.

Huntington's disease (HD) is a devastating neurodegenerative disease that is manifested by a gradual loss of physical, cognitive, and mental abilities. As the disease advances, age has a major impact on the pathogenic signature of mutant huntingtin (mHTT) protein aggregation. This review aims to explore the intricate relationship between aging, mHTT toxicity, and cellular senescence in HD. Scientific data on the interplay between aging, mHTT, and cellular senescence in HD were collected from several academic databases, including PubMed, Google Scholar, Google, and ScienceDirect. The search ter

Cellular Senescence in Craniofacial Tissue Regeneration: Inducers, Biomarkers, and Interventions.

Craniofacial defects and dental tissue loss have significant negative impacts on the structure and function of jaws and face, often resulting in psychological issues in patients, emphasizing the urgent need for effective craniofacial tissue reconstruction. Unfortunately, natural regeneration of these tissues is limited. Dental-derived mesenchymal stem cells (MSCs) have emerged as a promising resource for tissue engineering-based therapeutic approaches. However, the clinical outcomes of MSC-based transplantation have not met expectations due to various complex reasons, and cellular senescence i

Inflammaging, cellular senescence, and cognitive aging after traumatic brain injury.

Traumatic brain injury (TBI) is associated with mortality and morbidity worldwide. Accumulating pre-clinical and clinical data suggests TBI is the leading extrinsic cause of progressive neurodegeneration. Neurological deterioration after either a single moderate-severe TBI or repetitive mild TBI often resembles dementia in aged populations; however, no currently approved therapies adequately mitigate neurodegeneration. Inflammation correlates with neurodegenerative changes and cognitive dysfunction for years post-TBI, suggesting a potential association between immune activation and both age- a

Nicotinamide Improves Skin Photoaging in Mice by Delaying Cellular Senescence and Suppressing the Senescence-Associated Secretory Phenotype.

Nicotinamide (NAM), a precursor of nicotinamide adenine dinucleotide (NAD+), and NAD+ are integral to a variety of cellular processes. NAM supplementation has been shown to have benefits for cellular senescence. However, the mechanism by which NAM improves skin photoaging remains unclear. In this study, the multi-omics analysis revealed that insufficient nicotinamide metabolism may be associated with a decrease in NAD+ synthesis during skin aging. Importantly, we found that NAM has an ameliorating effect on the skin photoaging in mice. Supplementation with NAM restored the expression of the sa

Targeted apoptosis of senescent cells by valproic acid alleviates therapy-induced cellular senescence and lung aging.

BACKGROUND: Accumulation of senescent cells in tissues and their downstream effect programs have emerged as key drivers of aging and age-associated pathologies. Recent progresses in senotherapeutics indicated that either selectively killing senescent cells with senolytics or suppressing the senescence-associated secretory phenotype (SASP) secretion using senomorphics contributes to extending of the healthy lifespan and alleviating numerous age-related disorders in mice. PURPOSE: However, the potential side-effects and long-term cytotoxicity of the above novel compounds have not yet been deter

Effects and Therapeutic Strategies of Intra-Hepatic Cellular Senescence and Regeneration.

The liver constitutes a vital organ characterized by remarkable regenerative capacity, wherein intra-hepatic cellular senescence and regeneration exert pivotal regulatory functions in maintaining hepatic homeostasis and in developing therapeutic interventions for liver diseases. Intra-hepatic cellular aging is typically characterized by diminished proliferative capacity, genomic instability, mitochondrial dysfunction, and exacerbated inflammatory responses, all of which contribute to hepatic functional impairment and the pathogenesis of associated diseases. Liver regeneration depends upon the

Neuroendocrine-associated epigenetic factors in cellular senescence: mechanisms and therapeutic implications.

Ageing is a progressive biological process causing a reduction in tissue and cellular function due to accumulated molecular damage over time. Cellular senescence is a stable cellular state characterized by irreversible cell-cycle arrest accompanied by distinct molecular, epigenetic, and secretory alterations. Moreover, neuroendocrine signalling and epigenetic regulation have had a great impact on controlling ageing and senescence. As a part of the neuroendocrine system, the hypothalamic-pituitary-adrenal (HPA) axis and insulin-like growth factor-1 (IGF-1) mediate stress responses, circadian re

Cellular Senescence in Skin Wound Repair: Dual Roles and Senotherapeutic Interventions.

Skin wound repair is a complex process involving numerous signalling pathways in various cell types. Cellular senescence (CS), described as a proliferative limit, has dual implications in this process. In acute wounds, transient cellular senescence acts as a beneficial mechanism that facilitates tissue repair through the controlled secretion of the senescence-associated secretory phenotype (SASP), promoting immune cell recruitment, myofibroblast differentiation and angiogenesis. In contrast, chronic wound settings, which are frequently associated with pathological conditions including type 2 d

Orbital hybridization-mediated nanozymes reverse cellular senescence for aged bone regeneration.

The regeneration of aged bone is severely compromised by a deteriorating microenvironment characterized by excessive reactive oxygen species (ROS) and consequential endoplasmic reticulum stress (ERS). Herein, we report a biomimetic nanozyme engineered through synergistic p-d orbital hybridization and youthful membrane camouflage to precisely reverse this degenerative cascade. The designed Cu-Sn dual-atom core exhibits exceptional multi-enzymatic activity, scavenging superoxide anions with a 2.5-fold higher efficiency than Cu single-atom control. Mechanistic studies confirm that p-d hybridizati

High-Salt Diet-Induced Endothelial Dysfunction Is Mediated by Cellular Senescence.

BACKGROUND: High salt (HS) intake is a known cardiovascular risk factor, yet the mechanisms linking salt intake to endothelial dysfunction remain unclear. We investigated whether HS induces vascular senescence and dysfunction, and whether targeting senescent cells could prevent these effects. METHODS: Male C57BL/6J mice were fed a high-salt diet (HSD [8% NaCl]) for 14 or 28 days. Vascular function was assessed in thoracic aortae and mesenteric arteries using wire myography. Senescence was evaluated by p21, p16, and interleukins (IL) 6 and 1β expression in endothelial cells and whole vessels.

Disordered differentiation and cellular senescence in pediatric Hodgkin Reed-Sternberg cells.

Hodgkin lymphoma (HL) is characterized by inflammatory lesions with relatively few (typically <1%) pathogenic Hodgkin Reed-Sternberg (HRS) cells. The scarcity of malignant HRS cells in HL has historically limited characterization of these disease drivers, and pathogenic mechanisms underlying HL are incompletely defined. Using multiparameter flow cytometry to purify HRS cells and immune cell populations from pediatric HL lesions and control tonsils, we sought to define transcriptional programs in pediatric HRS cells. HRS cells are thought to arise from transformed germinal center B cells. Inter

Cellular Senescence and SASP in Bladder Ageing: Mechanisms and Emerging Interventions.

Bladder dysfunction is a prevalent age-related condition that severely impairs quality of life in older adults and commonly manifests as lower urinary tract symptoms (LUTS), including frequency, urgency, incontinence, and nocturia. Cellular senescence, characterized by irreversible cell-cycle arrest accompanied by persistent metabolic activity, is increasingly recognized as a contributor to age-associated tissue degeneration. Senescent cells secrete a diverse array of inflammatory cytokines, chemokines, growth factors, and matrix-remodeling enzymes, collectively termed the senescence-associate

Cellular sensor DAP5 decodes Betacoronaviral NSP5 to drive virus-induced senescence.

BACKGROUND: Viral infection induces host cells to enter a state of "virus-induced senescence (VIS)", which provides a stable cellular environment for viral replication. However, it is unclear about the molecular mechanism of this process. Here, we identified cellular protein DAP5 and its N-terminal fragment DAP51-451 as sensors to viral infection. METHODS: Upon SARS-CoV-2 infection, cellular apoptosis and senescence levels were assessed. This led to the identification of DAP5 as a pivotal proteolytic substrate that links viral protease activity to host cell fate determination. The specific cl

BACKGROUND: Alzheimer's disease (AD) is a progressive neurodegenerative disorder in the elderly, characterized by extracellular amyloid β‑ (Aβ) plaque deposition and intracellular neurofibrillary tangles (NFTs). Impaired autophagy, the cellular pathway for degrading damaged organelles and misfolded proteins, and cellular senescence, permanent cell cycle arrest with proinflammatory secretions, have emerged as key contributors to AD pathogenesis. METHODS: We performed a narrative review of recent mechanistic and preclinical studies investigating (1) autophagic flux and its role in Aβ and tau cl

Cellular senescence in adult pulmonary hypertension: current state and future challenges.

Pulmonary hypertension (PH) is a life-threatening disease increasingly being diagnosed in the elderly population, marked by vascular injury, excessive vasoconstriction and progressive remodelling of the pulmonary arteries (PAs). These lead to sustained elevation of PA pressure and subsequent development of right ventricular failure. Despite the beneficial effects on disease progression and quality of life, current treatments do not cure PH, highlighting the need for a deeper understanding of the underlying mechanisms. Recently, cellular senescence has gained much attention as a stress response
