Dynamic control of irregular bursting in an identified neuron of an oscillatory circuit.

In the oscillatory circuits known as central pattern generators (CPGs), most synaptic connections are inhibitory. We have assessed the effects of inhibitory synaptic input on the dynamic behavior of a component neuron of the pyloric CPG in the lobster stomatogastric ganglion. Experimental perturbations were applied to the single, lateral pyloric neuron (LP), and the resulting voltage time series were analyzed using an entropy measure obtained from power spectra. When isolated from phasic inhibitory input, LP generates irregular spiking-bursting activity. Each burst begins in a relatively stere

Serotonergic modulation of post-synaptic inhibition and locomotor alternating pattern in the spinal cord.

The central pattern generators (CPGs) for locomotion, located in the lumbar spinal cord, are functional at birth in the rat. Their maturation occurs during the last few days preceding birth, a period during which the first projections from the brainstem start to reach the lumbar enlargement of the spinal cord. Locomotor burst activity in the mature intact spinal cord alternates between flexor and extensor motoneurons through reciprocal inhibition and between left and right sides through commisural inhibitory interneurons. By contrast, all motor bursts are in phase in the fetus. The alternating

[Embryonic origin of temperament: notochord and early signaling].

It is proposed that the origin of temperament is rooted in events of the third week of gestation, when gastrulation and the notochord organize body axes and morphogenetic gradients, integrated through intracellular pathways with downstream effects on circuit architecture and locomotion. The aim of this article is to propose a theoretical mechanistic model that articulates how gastrulation, the notochord, and prenatal epigenetic programming could contribute—probabilistically rather than deterministically—to the origin of human temperament. A three-phase mechanistic flow is proposed: Phase 1, in

Activity-dependent neuromodulation and calcium homeostasis cooperate to produce robust and modulable neuronal function.

Neurons rely on two interdependent mechanisms - homeostasis and neuromodulation - to maintain robust and adaptable functionality. Calcium homeostasis stabilizes neuronal activity by adjusting ionic conductances, whereas neuromodulation dynamically modifies ionic properties in response to external signals carried by neuromodulators. Combining these mechanisms in conductance-based models often produces unreliable outcomes, particularly when sharp neuromodulation interferes with calcium-homeostatic tuning. This study explores how a biologically inspired neuromodulation controller can harmonize wi

The transient potassium outward current has different roles in modulating the pyloric and gastric mill rhythms in the stomatogastric ganglion.

The crustacean stomatogastric nervous system is a classic model for understanding the effects of modulating ionic currents and synapses at both the cell and network levels. The stomatogastric ganglion in this system contains two distinct central pattern generators: a slow gastric mill network that generates flexible rhythmic outputs (8-20 s) and is often silent, and a fast pyloric network that generates more consistent rhythmic outputs (0.5-2 s) and is always active in vitro. Different ionic conductances contribute to the properties of individual neurons and therefore to the overall dynamics o

Muscle group dependent responses to stimuli in a grasshopper model for tonic immobility.

Tonic Immobility (TI) is a prolonged immobile condition exhibited by a variety of animals when exposed to certain stimuli, and is thought to be associated with a specific state of arousal. In our study, we characterize this state by using the reliably inducible TI state of the grasshopper (Hieroglyphus banian) and by monitoring abdominal pulsations and body movements in response to visual and auditory stimuli. These pulsations are present during the TI and 'awake', standing states, but not in the CO2 anesthetized state. In response to the stimuli, animals exhibited a suppression in pulsation a

Dedicated Hippocampal Inhibitory Networks for Locomotion and Immobility.

Network activity is strongly tied to animal movement; however, hippocampal circuits selectively engaged during locomotion or immobility remain poorly characterized. Here we examined whether distinct locomotor states are encoded differentially in genetically defined classes of hippocampal interneurons. To characterize the relationship between interneuron activity and movement, we used in vivo, two-photon calcium imaging in CA1 of male and female mice, as animals performed a virtual-reality (VR) track running task. We found that activity in most somatostatin-expressing and parvalbumin-expressing

A model of the basal ganglia in voluntary movement and postural reactions.

A basal ganglia central pattern generator (CPG) is developed and its role in voluntary movements on the ground and postural reactions on a disturbed platform are studied and analysed by simulation. Biped dynamics and platform kinematics are utilised. The effects of agonist-antagonist muscular co-activation and joint stiffness are formulated. The implementation of the necessary counter-manoeuvres for maintaining balance and postural stability is studied. A control strategy, applicable to large systems, is formulated. The biped manoeuvres and transitions terminate in pre-specified intervals of t

Probing the Human Spinal Locomotor Circuits by Phasic Step-Induced Feedback and by Tonic Electrical and Pharmacological Neuromodulation.

The mammalian lumbar spinal cord experimentally isolated from supraspinal and afferent feedback input remains capable of expressing some basic locomotor function when appropriately stimulated. This ability has been attributed to spinal neural circuits referred to as central pattern generators (CPGs). In individuals with a severe spinal cord injury, rhythmic activity in paralyzed leg muscles can be generated by phasic proprioceptive feedback during therapist- or robotic-assisted stepping on a motorized treadmill. Here, we critically review to what extent the resulting motor output represents lo

Influence of periaqueductal gray on other salience network nodes predicts social sensitivity.

The intrinsic connectivity of the salience network (SN) plays an important role in social behavior, however the directional influence that individual nodes have on each other has not yet been fully determined. In this study, we used spectral dynamic causal modeling to characterize the effective connectivity patterns in the SN for 44 healthy older adults and for 44 patients with behavioral variant frontotemporal dementia (bvFTD) who have focal SN dysfunction. We examined the relationship of SN effective connections with individuals' socioemotional sensitivity, using the revised self-monitoring

Voluntary Movement Frequencies in Submaximal One- and Two-Legged Knee Extension Exercise and Pedaling.

Understanding of behavior and control of human voluntary rhythmic stereotyped leg movements is useful in work to improve performance, function, and rehabilitation of exercising, healthy, and injured humans. The present study aimed at adding to the existing understanding within this field. To pursue the aim, correlations between freely chosen movement frequencies in relatively simple, single-joint, one- and two-legged knee extension exercise were investigated. The same was done for more complex, multiple-joint, one- and two-legged pedaling. These particular activities were chosen because they c

From Fin to Limb: Orientational Shift and Evolution of Diagonal-Couplet Gait in Tetrapods.

The transition from fins to limbs represents a pivotal evolutionary shift that enabled vertebrates to engage terrestrial environments through coordinated transformations in appendicular structure and neuromuscular control. Here, we propose that this transition can be understood as a biomechanical reorganization of force transmission across multi-joint musculoskeletal linkages. Rather than a simple rotational modification transforming fins directly into forelimbs, this process involved progressive reorganization of sarcopterygian appendages, including axial rotation, directional force productio

An analytical formulation of the law of intersegmental coordination during human locomotion.

The law of intersegmental coordination is a kinematic law that describes the coordination patterns among the elevation angles of the lower limb segments during locomotion (Borghese et al. in J Physiol 494:863-879, 1996). This coordination pattern reduces the number of degrees of freedom of the lower limb to two, i.e. the elevation angles covary along a plane in angular space. The properties of the plane that constrains the time course of the elevation angles have been extensively studied, and its orientation was found to be correlated with gait velocity and energy expenditure (Bianchi et al. i

Spatial and functional architecture of the mammalian brain stem respiratory network: a hierarchy of three oscillatory mechanisms.

Mammalian central pattern generators (CPGs) producing rhythmic movements exhibit extremely robust and flexible behavior. Network architectures that enable these features are not well understood. Here we studied organization of the brain stem respiratory CPG. By sequential rostral to caudal transections through the pontine-medullary respiratory network within an in situ perfused rat brain stem-spinal cord preparation, we showed that network dynamics reorganized and new rhythmogenic mechanisms emerged. The normal three-phase respiratory rhythm transformed to a two-phase and then to a one-phase r

Coupling of minor motor events and epileptiform discharges with arousal fluctuations in NFLE.

PURPOSE: We recently demonstrated that in nocturnal frontal lobe epilepsy (NFLE) highly stereotyped minor motor events (MMEs, in the form of short-lasting stereotyped movements involving the limbs, the axial musculature, and/or the head), could occur in either the presence or absence of an epileptiform discharge (ED). In lack of a systematic analysis, both MMEs and EDs were frequently observed to occur in association with arousal fluctuations. Hereby, in the same group of refractory NFLE subjects, we report a methodical neurophysiolgical investigation set out to investigate whether, and how, t

Modulating reflexes enables speed control in simulated human walking and running.

Human locomotion is characterized by remarkable versatility, enabling smooth speed modulation, acceleration, deceleration, and transitions between gaits. This versatility is thought to emerge from interactions between the musculoskeletal system, spinal circuits—including reflexes and central pattern generators—and supraspinal control. The relative contributions of these components remain a subject of ongoing debate. In this study, we explore the potential role of reflex modulation in achieving the observed flexibility of human locomotion. Using a neuromusculoskeletal model controlled by reflex

Hop Mice Display Synchronous Hindlimb Locomotion and a Ventrally Fused Lumbar Spinal Cord Caused by a Point Mutation in Ttc26.

Identifying the spinal circuits controlling locomotion is critical for unravelling the mechanisms controlling the production of gaits. Development of the circuits governing left-right coordination relies on axon guidance molecules such as ephrins and netrins. To date, no other class of proteins have been shown to play a role during this process. Here, we have analyzed hop mice, which walk with a characteristic hopping gait using their hindlimbs in synchrony. Fictive locomotion experiments suggest that a local defect in the ventral spinal cord contributes to the aberrant locomotor phenotype. Ho

A Descending Circuit Derived From the Superior Colliculus Modulates Vibrissal Movements.

The superior colliculus (SC) is an essential structure for the control of eye movements. In rodents, the SC is also considered to play an important role in whisking behavior, in which animals actively move their vibrissae (mechanosensors) to gather tactile information about the space around them during exploration. We investigated how the SC contributes to vibrissal movement control. We found that when the SC was unilaterally lesioned, the resting position of the vibrissae shifted backward on the side contralateral to the lesion. The unilateral SC lesion also induced an increase in the whiskin
