BACKGROUND: The centrosome is a critical regulator of cortical development, orchestrating microtubule dynamics, cell cycle progression, and neuronal migration. Disruptions in centrosome-associated proteins have been associated with a range of neurodevelopmental disorders. CEP170, a microtubule-binding protein localized to the subdistal appendages (SDA) of centrioles, has been implicated in centrosome function, yet its role in corticogenesis remains poorly defined.
METHODS: We analyzed CEP170 expression in mouse cortex using western blotting, qPCR, scRNA-seq, and spatial transcriptomics, and e

Chemoresistance is a major challenge in effective chemotherapy for breast cancers. Unfortunately, the precise molecular mechanisms that confer resistance remain elusive so far. A remarkable feature in a multitude of breast cancers is the presence of clustered supernumerary centrosomes, which is a dysregulated condition that arises primarily through centrosome over-duplication. Normally, microtubule motor proteins (MiMos) maintain the integrity of centrosomes via regulating cohesion, separation and positioning of centrosomes. In the recent years, several MiMos have been reported to be different

Preeclampsia is a serious pregnancy-related disorder that poses significant health risks to the mother and offspring. A key aspect of preeclampsia is impaired endothelial cell migration, which is essential for the repair and integrity of the vasculature. Preeclamptic endothelial progenitor cells (EPC) exhibit defects in migration, although the precise molecular mechanisms underlying these defects remain unclear. Cellular spatial organization, such as centrosome orientation, regulates the migration process by influencing cell polarity and movement. Epigenetic changes in preeclampsia may disrupt

Centrosomal proteins play pivotal roles in orchestrating microtubule dynamics, and their dysregulation leads to disorders, including cancer and ciliopathies. Understanding the multifaceted roles of centrosomal proteins is vital to comprehend their involvement in disease development. Here, we report novel cellular functions of CEP41, a centrosomal and ciliary protein implicated in Joubert syndrome. We show that CEP41 is an essential microtubule-associated protein with microtubule-stabilizing activity. Purified CEP41 binds to preformed microtubules, promotes microtubule nucleation and suppresses

Alström syndrome (ALMS) is a very rare autosomal-recessive disorder, causing a broad range of clinical defects most notably retinal degeneration, type 2 diabetes, and truncal obesity. The ALMS1 gene encodes a complex and huge ∼0.5 MDa protein, which has hampered analysis in the past. The ALMS1 protein is localized to the centrioles and the basal body of cilia and is involved in signaling processes, for example, TGF-β signaling. However, the exact molecular function of ALMS1 at the basal body remains elusive and controversial. We recently demonstrated that protein complex analysis utilizing end

Centrosome amplification is a phenomenon frequently observed in human cancers, so centrosome depletion has been proposed as a therapeutic strategy. However, despite being afflicted with a lack of centrosomes, many cancer cells can still proliferate, implying there are impediments to adopting centrosome depletion as a treatment strategy. Here, we show that TFEB- and TFE3-dependent autophagy activation contributes to acentrosomal cancer proliferation. Our biochemical analyses uncover that both TFEB and TFE3 are novel PLK4 (polo like kinase 4) substrates. Centrosome depletion inactivates PLK4, re

Mutations in PKD1 (encoding for polycystin-1 [PC1]) are found in 80%-85% of patients with autosomal dominant polycystic kidney disease (ADPKD). We tested the hypothesis that changes in actin dynamics result from PKD1 mutations through dysregulation of compartmentalized centrosomal RhoA signaling mediated by specific RhoGAP (ARHGAP) proteins resulting in the complex cellular cystic phenotype. Initial studies revealed that the actin cytoskeleton was highly disorganized in cystic cells derived from patients with PKD1 and was associated with an increase in total and centrosomal active RhoA and ROC

Tripartite motif (TRIM) proteins mediate antiviral host defences by either directly targeting viral components or modulating innate immune responses. Here we identify a mechanism of antiviral restriction in which a TRIM E3 ligase controls viral replication by regulating the structure of host cell centrosomes and thereby nuclear lamina integrity. Through RNAi screening we identified several TRIM proteins, including TRIM43, that control the reactivation of Kaposi's sarcoma-associated herpesvirus. TRIM43 was distinguished by its ability to restrict a broad range of herpesviruses and its profound

Hepatic cystogenesis in polycystic liver diseases is associated with abnormalities of cholangiocyte cilia. Given the crucial association between cilia and centrosomes, we tested the hypothesis that centrosomal defects occur in cystic cholangiocytes of rodents (Pkd2(WS25/-) mice and PCK rats) and of patients with polycystic liver diseases, contributing to disturbed ciliogenesis and cyst formation. We examined centrosomal cytoarchitecture in control and cystic cholangiocytes, the effects of centrosomal abnormalities on ciliogenesis, and the role of the cell-cycle regulator Cdc25A in centrosomal

CONTEXT: Microcephalic primordial dwarfism (MPD) is a rare, severe form of human growth failure in which growth restriction is evident in utero and continues into postnatal life. Single causative gene defects have been identified in a number of patients with MPD, and all involve genes fundamental to cellular processes including centrosome functions.
OBJECTIVE: The objective of the study was to find the genetic etiology of a novel presentation of MPD.
DESIGN: The design of the study was whole-exome sequencing performed on two affected sisters in a single family. Molecular and functional studi

The centrosome directs chromosomal migration by a complex process of tubulin-chromatin binding. In this contribution centrosomal abnormalities, including centrosomal amplification, were explored in Chinese hamster ovary (CHO) and normal human mammary epithelial cells (NHMECs) exposed to the antiretroviral drug zidovudine (3'-azido-3'-deoxythymidine, AZT). Centrosomal amplification/fragmentation was observed in both cell types and kinetochore positive micronuclei were found in AZT-exposed CHO cells in correlation with dose. Normal human mammary epithelial cell (NMHEC) strain M99005, previously

A novel centrosome-related protein CrpF46 was detected using a serum F46 from a patient suffering from progressive systemic sclerosis. We identified the protein by immunoprecipitation and Western blotting followed by tandem mass spectrometry sequencing. The protein CrpF46 has an apparent molecular mass of ~60 kDa, is highly homologous to a 527 amino acid sequence of the C-terminal portion of the protein Golgin-245, and appears to be a splice variant of Golgin-245. Immunofluorescence microscopy of synchronized HeLa cells labeled with an anti-CrpF46 monoclonal antibody revealed that CrpF46 local

CEP68 is best known for its role at the centrosome linker, but its functions beyond the centrosome remain largely unexplored. Here, we show that CEP68 also localizes to the Golgi apparatus and nucleus, where it contributes to the stress response. CEP68 associates with stress response proteins, modulating eIF2α phosphorylation and stress granule (SG) formation during oxidative stress. In the nucleus, CEP68 forms liquid-like nuclear condensates adjacent to nuclear speckles (NSs), influencing their protein dynamics. The small heat shock protein HSP27, which translocates to NSs during stress, inte

In Drosophila testis, germline stem cells (GSCs) typically divide asymmetrically, keeping one daughter cell within the niche for self-renewal and placing the other daughter cell outside the niche to undergo differentiation. This is the primary mechanism for maintaining a stable number of stem cells in the niche. However, it has been shown that small fraction of GSCs continuously leave the niche, necessitating an additional mechanism to compensate for this loss and maintain a constant number of GSCs. Dedifferentiation and symmetric renewal have been shown to replenish lost GSCs during regenerat

Autosomal dominant polycystic kidney disease (ADPKD) is a monogenic disorder accounting for approximately 5% of patients with renal failure, yet therapeutics for the treatment of ADPKD remain limited. ADPKD tissues display abnormalities in the biogenesis of the centrosome, a defect that can cause genome instability, aberrant ciliary signaling, and secretion of pro-inflammatory factors. Cystic cells form excess centrosomes via a process termed centrosome amplification (CA), which causes abnormal multipolar spindle configurations, mitotic catastrophe, and reduced cell viability. However, cells w

Centrosome asymmetry has been implicated in stem cell fate maintenance in both flies and vertebrates, but the underlying molecular mechanisms are incompletely understood. Here, we report that loss of CG7337, the fly ortholog of WDR62, compromises interphase centrosome asymmetry in fly neural stem cells (neuroblasts). Wdr62 maintains an active interphase microtubule-organizing center (MTOC) by stabilizing microtubules (MTs), which are necessary for sustained recruitment of Polo/Plk1 to the pericentriolar matrix (PCM) and downregulation of Pericentrin-like protein (Plp). The loss of an active MT

BACKGROUND: Centrosome amplification (CA) has been reported in nearly all types of human cancer and is associated with deleterious clinical factors such as higher grade and stage. However, previous reports have not shown how CA affects cellular differentiation and clinical outcomes in breast cancer.
METHODS: We analyzed centrosomes by immunofluorescence and compared to ploidy and chromosomal instability (CIN) as assessed by 6-chromosome FISH in a cohort of 362 breast cancers with median clinical follow-up of 8.4 years. Centrosomes were recognized by immunofluorescence using antibodies for per

Chromosome instability (CIN) is a major contributor to drug resistance and recurrence. As a crucial mechanism of CIN, centrosome clustering has emerged as a promising therapeutic strategy. However, the roles and regulatory mechanisms of centrosome clustering in gastric cancer (GC) remain unclear. BATF2 was previously identified as a key modulator of multidrug resistance (MDR) in GC. To examine the involvement of centrosome clustering in the mechanism by which BATF2 reverses MDR in GC, adriamycin (ADR)- and vincristine (VCR)-resistant cell lines, NCI-N87/ADR and NCI-N87/VCR, were used for inves

Centrosome amplification is a hallmark of colorectal cancer (CRC), yet its molecular origins remain elusive. Protein Kinase A (PKA), anchored to centrosomes, regulates a number of mitotic processes, though its role in maintaining centrosome integrity remains poorly understood. Here, we show that PKA is central to a signaling cascade involving the G protein-coupled estrogen receptor GPER1, which is essential for centrosome integrity in colon cancer cells. Activation of GPER1 by estrogens or a specific agonist triggers Gαs protein-mediated stimulation of adenylyl cyclase, elevating cAMP levels a

Anaplastic Large Cell Lymphoma (ALCL) is an aggressive T-cell lymphoma affecting children and young adults. About 30% of patients develop therapy resistance therefore new precision medicine drugs are highly warranted. Multiple rounds of structure-activity optimization of Caffeic Acid Phenethyl Ester have resulted in CM14. CM14 causes upregulation of genes involved in oxidative stress response and downregulation of DNA replication genes leading to G2/M arrest and subsequent apoptosis induction. In accordance with this, an unbiased proteomics approach, confocal microscopy and molecular modeling
