The solubility profiles of theobromine, theophylline, and caffeine at 25 degrees were examined in binary solvent systems including dioxane-formamide, water-polyethylene glycol 400, and glycerin-propylene glycol. A number of rifamycin non-producing UV-mutants derived from Nocardia mediterranei strains N813 (rifamycin B producer) and A10 (aro--mutant excreting shikimate derived from strain N813) were found to accumulate an identical complex of aromatic components instead of rifamycin B. The main component of this aromatic complex, product P8/1-OG, was isolated from six of these P--mutant strains and identified spectroscopically as a very early precursor in the biosynthesis of rifamycins.

The activity coefficient (gamma) and the partition coefficient n-octanol/water (P) of various groups of substances of pharmaceutical and biological interest (anaesthetic gases, primary n-alcohols and barbiturates) were compared in order to obtain a linear relationship. For a particular form (amorphous phase) of physical acting substances the water solubility, in conditions very close to the in vivo ones, has been compared with corresponding biological activities. The reciprocal of the above parameter shows a good correlation with various physico-chemical, biochemical and pharmacological parameters of xanthinic compounds, analgesic alkaloids, barbiturates, sulphonamides. The solubility of the crystalline phase cannot be correlated with the biological activity: the ratio between these two solubilities shows this inability.

This study was undertaken to evaluate the efficacy of ursodeoxycholic acid (UDCA) in the treatment of primary biliary cirrhosis (PBC). Three patients with PBC (stage II) have been treated with UDCA (Ursofalk and Falk Pharma, Freiburg, Germany)--of 10 mg/kg daily dosage in the course of three years. We report a refined potential energy function for the ground electronic state of CS2 based on a least-squares fitting to several low-lying experimental vibrational frequencies. Energy levels up to 20,000 cm(-1) have been obtained on this empirical potential using the Lanczos algorithm and potential optimized discrete variable representation. Diuretics have been empirically developed. Inorganic mercury was used as diuretics from 16C. Guy's Hospital Pills (containing the equal part of Hg2Cl2, digitalis, and squill) was well known in the 19 C. Discovery of the diuresis of merbaphen has led to develop many organomercurial diuretics and to give suggestion of the drug design of ethacrynic acid. Diuresis in patients given sulfonamide was discovered in 1938. Inhibition of carbonic anhydrase by sulfonamide was related to its diuresis. Drug design aimed to enhance the inhibition of this enzyme has obtained acetazolamide, then chlorothazide.
