Immunogenicity of a West Nile virus DIII-cholera toxin A2/B chimera after intranasal delivery.

West Nile virus (WNV) causes potentially fatal neuroinvasive disease and persists at endemic levels in many parts of the world. Despite advances in our understanding of WNV pathogenesis, there remains a significant need for a human vaccine. The domain III (DIII) region of the WNV envelope protein contains epitopes that are the target of neutralizing antibodies. We have constructed a chimeric fusion of the non-toxic cholera toxin (CT) CTA2/B domains to DIII for investigation as a novel mucosally-delivered WNV vaccine. Purification and assembly of the chimera, as well as receptor-binding and ant

Prenatal diagnosis and normal outcome of a 46,XX/46,XY chimera: a case report.

The phenotypic spectrum of 46,XX/46,XY chimeric patients is variable. It ranges from normal male or female genitalia to different degrees of ambiguous genitalia. Chimerism results from the amalgamation of two different zygotes in a single embryo, whereas mosaicism results from a mitotic error in a single zygote. Several other mechanisms resulting in a chimera have been discussed in the literature. Here, we report on a new case of chimerism (46,XX/46,XY) diagnosed at 17 weeks' gestation on amniocentesis performed because of advanced maternal age. Ultrasound examination revealed normal female ex

Persistent gestational trophoblastic disease after an androgenetic/biparental fetal chimera: a case report and review.

We present a case of a dichorionic/diamniotic twin pregnancy in which one twin presented with ultrasound findings suggestive of molar changes in the placenta. The placenta of twin A seemed to be grossly enlarged and cystic, and twin A was small for gestation. After an inevitable abortion, a detailed histological and genetic evaluation was performed on the fetus and placenta from twin A, including traditional cytogenetic techniques, microsatellite marker analysis, fluorescent in situ hybridization, and p57 immunostaining. It was determined that twin A was a chimera with a biparental XX cell lin

Autologous T cells control B-chronic lymphocytic leukemia tumor progression in human-->mouse radiation chimera.

B-chronic lymphocytic leukemia (B-CLL) is characterized by the clonal accumulation of CD5+ B cells. It has been suggested that CLL cells may be regulated by inhibitory and growth-promoting signals exerted by autologous T cells. We have recently described a model for human B-CLL in which peripheral blood mononuclear cells (PBMCs) are transplanted into the peritoneal cavity of lethally irradiated mice radioprotected with bone marrow from mice with severe combined immunodeficiency. In this model, adoptive transfer of low-stage PBMCs leads to marked engraftment of T cells or combined T and CLL cel

Bovine lactoferrin and chimera lactoferrin prevent and destroy Salmonella Typhimurium biofilms in Caco-2 cells.

Salmonellosis is a common foodborne disease caused by Salmonella bacteria. The emergence of multidrug-resistant (MDR) Salmonella serotypes, such as Typhimurium, and Salmonella's ability to form biofilms contribute to their resistance and persistence in host and non-host environments. New strategies are needed to treat or prevent Salmonella infections. This work aimed to determine the effect of the bovine lactoferrin (bLF) and lactoferrin chimera (LFchimera) in preventing or disrupting biofilms formed on abiotic surfaces or Caco-2 cells by S. Typhimurium ATCC 14028 or an MDR strain. The inhibit

Degradation of neurodegenerative disease-associated TDP-43 aggregates and oligomers via a proteolysis-targeting chimera.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) associated with TAR DNA-binding protein 43 (TDP-43) aggregation has been considered as a lethal and progressive motor neuron disease. Recent studies have shown that both C-terminal TDP-43 (C-TDP-43) aggregates and oligomers were neurotoxic and pathologic agents in ALS and frontotemporal lobar degeneration (FTLD). However, misfolding protein has long been considered as an undruggable target by applying conventional inhibitors, agonists, or antagonists. To provide this unmet medical need, we aim to degrade these misfolding proteins by designing a s

Altered Tau Kinase Activity in rTg4510 Mice after a Single Interfaced CHIMERA Traumatic Brain Injury.

Traumatic brain injury (TBI) is an established risk factor for neurodegenerative diseases. In this study, we used the Closed Head Injury Model of Engineered Rotational Acceleration (CHIMERA) to investigate the effects of a single high-energy TBI in rTg4510 mice, a mouse model of tauopathy. Fifteen male rTg4510 mice (4 mo) were impacted at 4.0 J using interfaced CHIMERA and were compared to sham controls. Immediately after injury, the TBI mice showed significant mortality (7/15; 47%) and a prolonged duration of loss of the righting reflex. At 2 mo post-injury, surviving mice displayed significa

Proteolysis targeting chimera technology: a novel strategy for treating diseases of the central nervous system.

Neurological diseases such as stroke, Alzheimer's disease, Parkinson's disease, and Huntington's disease are among the intractable diseases for which appropriate drugs and treatments are lacking. Proteolysis targeting chimera (PROTAC) technology is a novel strategy to solve this problem. PROTAC technology uses the ubiquitin-protease system to eliminate mutated, denatured, and harmful proteins in cells. It can be reused, and utilizes the protein destruction mechanism of the cells, thus making up for the deficiencies of traditional protein degradation methods. It can effectively target and degra

Biochemical and immunological characterization of an ETEC CFA/I adhesin cholera toxin B subunit chimera.

Surface-expressed colonization factors and their subunits are promising candidates for inclusion into a multivalent vaccine targeting enterotoxigenic Escherichia coli (ETEC), a leading cause of acute bacterial diarrhea in developing regions. However, soluble antigens are often poorly immunogenic in the absence of an adjuvant. We show here that the serum immune response to CfaE, the adhesin of the ETEC colonization factor CFA/I, can be enhanced in BALB/c mice by immunization with a chimeric antigen containing CfaE and pentameric cholera toxin B subunit (CTB) of cholera toxin from Vibrio cholera

BET protein proteolysis targeting chimera (PROTAC) exerts potent lethal activity against mantle cell lymphoma cells.

Bromodomain extraterminal protein (BETP) inhibitors transcriptionally repress oncoproteins and nuclear factor-κB (NF-κB) target genes that undermines the growth and survival of mantle cell lymphoma (MCL) cells. However, BET bromodomain inhibitor (BETi) treatment causes accumulation of BETPs, associated with reversible binding and incomplete inhibition of BRD4 that potentially compromises the activity of BETi in MCL cells. Unlike BETi, BET-PROTACs (proteolysis-targeting chimera) ARV-825 and ARV-771 (Arvinas, Inc.) recruit and utilize an E3-ubiquitin ligase to effectively degrade BETPs in MCL ce

Fluorescent protein-scorpion toxin chimera is a convenient molecular tool for studies of potassium channels.

Ion channels play a central role in a host of physiological and pathological processes and are the second largest target for existing drugs. There is an increasing need for reliable tools to detect and visualize particular ion channels, but existing solutions suffer from a number of limitations such as high price, poor specificity, and complicated protocols. As an alternative, we produced recombinant chimeric constructs (FP-Tx) consisting of fluorescent proteins (FP) fused with potassium channel toxins from scorpion venom (Tx). In particular, we used two FP, eGFP and TagRFP, and two Tx, OSK1 a

The use of low molecular weight protamine chemical chimera to enhance monomeric insulin intestinal absorption.

Although oral delivery of insulin offers a number of unmatched advantages, it nevertheless is beset by the poor permeability of insulin molecules through the epithelial cell membranes of the intestinal mucosal layer. We previously reported the development of low molecular weight protamine (LMWP) as a non-toxic yet potent cell-penetrating peptide, of which via covalent linkage was capable of translocating protein cargos through the membranes of almost all cell types. It is therefore hypothesized that LMWP could be practically employed as a safe and effective tool to deliver insulin across the i

Antigen-specific B and T cells in human/mouse radiation chimera following immunization in vivo.

Adoptive transfer of human peripheral blood mononuclear cells (PBMC) into mice with severe combined immunodeficiency (SCID) or into lethally irradiated BALB/c mice radioprotected with SCID bone marrow, leads to marked engraftment of human T and B cells. In such chimeras, human serum antibody responses can be stimulated readily by vaccination with recall antigens, but the detection of antigen-specific functional T or B cells has been extremely difficult. In the present study, we were able to detect by Elispot analysis high frequencies of immunoglobulin G (IgG)-secreting B cells and mitogen-resp

Proteolysis-targeting Chimeras induce ferroptosis in cancer: From Mechanism to clinical application.

Ferroptosis, an iron-dependent cell death driven by lipid peroxidation, is a crucial tumor suppression mechanism and promising therapeutic avenue, especially for resistant cancers. However, its clinical translation is hindered by complex regulatory networks, compensatory pathways, and a lack of selective modulators. The advent of Proteolysis-Targeting Chimeras (PROTACs) presents a groundbreaking opportunity to precisely manipulate this pathway. This review systematically explores leveraging PROTAC technology to induce ferroptosis. We first delineate the core regulatory axes, including GPX4, FS

Marker-Agnostic Tumor Anchoring Chimeras Enable pH-Gated Immune Engagement.

Targeted cancer immunotherapies typically depend on cell surface antigens to direct immune responses. However, many tumors either lack tumor-specific markers or downregulate their expression, limiting the applicability of antigen-directed strategies. In contrast, tumor acidosis represents a nearly universal hallmark of solid cancers, arising from dysregulated metabolism that acidifies the extracellular tumor microenvironment (TME). Here, we leverage this feature to develop Marker-Agnostic Tumor Anchoring Chimeras (MATRACs), pH-sensitive fusion proteins that mediate ternary complex formation be

Effect of Linker Sequence on the Interdomain Dynamics of Self-Sufficient CYP116B5-SOX Chimeras.

In this work, the design of CYP116B5-SOX-(PPII) is presented. This is an artificial self-sufficient enzyme in which the P450 116B5 peroxygenase and the H2O2-generating sarcosine oxidase (SOX) are linked together in a fusion complex. A polyproline type II helix (PPII) linker was used to connect the two enzymes, obtaining a rigid fusion protein. CYP116B5-SOX-(PPII) displayed 10-fold increased activity in terms of p-nitrophenol oxidation (k cat: 32.1 ± 0.5 min-1; K M: 150 ± 15 μM) compared to the isolated heme domain CYP116B5-hd, and more than 2-fold higher activity compared to CYP116B5-SOXa fus

Click-Activated Supramolecular Targeting Chimeras for Spatiotemporally Resolved Protein Degradation In Vivo.

Targeted protein degradation (TPD) holds enormous therapeutic potential, yet spatiotemporally controlled, tissue-selective protein degradation remains challenging. Herein, we report Click-activated Supramolecular TArgeting Chimeras (ClSupTAC), a multimodular platform leveraging bioorthogonal chemistry to realize spatially and temporally resolved TPD in vivo. ClSupTAC follows a two-step "click-to-activate" paradigm: self-assembled, functionally dormant tissue-targeting supramolecular nanoparticles presenting protein-binding ligands and trans-cyclooctene (TCO) moieties are locally activated upon

Endobody: Genetically Encodable Nanobody-CPP Chimeras for Degradation of Membrane and Extracellular Proteins.

Membrane protein degraders (MPDs) like LYTACs are emerging tools for targeting disease-relevant membrane proteins, but their reliance on specific endocytic receptors limits their applicability across diverse cell types. Here, we present a structurally concise and genetically encodable class of degraders, nanobody-cell-penetrating peptide (CPP) chimeras, termed endobodies. We demonstrated that a genetically fused CPP is sufficient to mediate the internalization and subsequent degradation of membrane or extracellular proteins recognized by the nanobody. Using this platform, we achieved targeted

Design and functional characterization of recombinant chimeras as vaccine candidates against Lawsonia intracellularis.

Proliferative enteropathy is a gastrointestinal disease caused by the intracellular bacterium Lawsonia intracellularis, affecting swine populations worldwide and leading to significant economic losses due to reduced growth performance, increased mortality, and treatment expenses. Current vaccines present limitations related to production processes, stability, and overall cost. This study aimed to design and evaluat chimeric proteins containing immunogenic epitopes of L. intracellularis. A recombinant chimera, designated Li_full, was rationally designed in silico by integrating predicted B-cell

Caged Glycan-Antibody Conjugates for Tumor-Selective Activation of Lysosome-Targeting Chimeras.

Lysosome-targeting chimeras (LYTACs) induce lysosomal degradation of extracellular and membrane proteins by bridging target proteins with lysosomal trafficking receptors. However, conventional LYTACs often suffer from off-tissue effects, whereas reported tissue-specific LYTACs typically display limited degradation efficiency. To address these challenges, we report a chemically activatable LYTAC platform that leverages tumor microenvironment-specific cues to achieve precise and safe protein degradation in vivo. We designed a glutathione (GSH)-responsive caged mannose-6-phosphate glycan, GSH-pM6
