Studies on Effect of Certain Quinones: I. Electron Transport, Photophosphorylation, and CO(2) Fixation in Isolated Chloroplasts.

The effect of quinone herbicides and fungicides on photosynthetic reactions in isolated spinach (Spinacia oleracea) chloroplasts was investigated. 2,3-Dichloro-1,4-naphthoquinone (dichlone), 2-amino-3-chloro-1,4-naphthoquinone (06K-quinone), and 2,3,5,6-tetrachloro-1,4-benzoquinone (chloranil) inhibited ferricyanide reduction as well as ATP formation. Benzoquinone had little or no effect on these reactions. The two reactions showed a differential sensitivity to these inhibitors. Dichlone was a strong inhibitor of both photosystems I and II; photosystem I was more sensitive to 06K-quinone than

Sulfoquinovosyldiacylglycerol (SQDG) in chloroplast-poor tissues: What can lipidomics tell us?

Sulfoquinovosyldiacylglycerol (SQDG) is an anionic sulfolipid of plastid membranes, classically associated with chloroplasts and thylakoids. Its detection by lipidomics in chloroplast-poor tissues raises a central problem: how can a reported SQDG signal be validated, and what evidence is required to determine its plastid context and biological meaning? In this critical narrative review, we reassess evidence beyond chloroplast-dominated tissues and evaluate both the strength of evidence for SQDG occurrence or function and the steps required to test lipidomics-derived hypotheses. The literature

Chloroplast genome architecture and phylogenomics of the desert sand-fixing shrub Corethrodendron scoparium.

Corethrodendron scoparium (Fisch. & C.A.Mey.) Fisch. & Basiner is an ecologically vital sand-fixing shrub in arid northwestern China. However, its chloroplast genome structure and phylogenetic position within Corethrodendron remain poorly understood. This study reports the complete chloroplast genome of C. scoparium (123,115 bp , GC 34.5%), encoding 114 genes (80 protein-coding, 30 tRNA, 4 rRNA). It lacks inverted repeat (IR) regions, representing an IR loss type in Fabaceae. Phylogenomic analysis strongly supported C. scoparium as part of a monophyletic clade with C. fruticosum and C. multiju

Dynamic regulation of the chloroplast SUFBC2D complex by SUFB expression and CLP protease in Arabidopsis.

Chloroplast iron-sulfur (Fe-S) clusters, essential cofactors for Fe-S proteins, are assembled by the sulfur utilization factor (SUF) BC2D complex, but regulation of their biogenesis remains unclear. We show that during leaf senescence, SUFB transcription rapidly increases while SUFC/SUFD transcripts and the abundance of all three SUF proteins remain unchanged. Leaf necrosis with green color occurs solely when SUFB RNA interference (RNAi) is induced, and the decrease of SUFs is obviously faster in SUFB-RNAi than SUFC-RNAi lines. Furthermore, SUFB exhibits a higher turnover rate than SUFC/SUFD,

Chloroplastic protein PORC undergoes heat-induced condensation and enhances thermotolerance in Arabidopsis.

Chloroplast stress granules (cpSGs) are emerging as dynamic suborganellar condensates that play a crucial role in mediating stress response in plants. In this study, we demonstrate that the chlorophyll biosynthesis enzyme protochlorophyllide oxidoreductase C (PORC) localizes to cpSGs in response to acute and prolonged heat stress in Arabidopsis (Arabidopsis thaliana). While PORC promoter activity was developmentally regulated and remained unresponsive to heat, PORC protein re-localized from a diffuse chloroplast distribution into punctate structures under elevated temperatures. This condensati

Identification of chloroplast transit peptides inDunaliella salina and their application for heterologous astaxanthin production.

Dunaliella salinais a promising green cell factory, but efficient chloroplast targeting of exogenous proteins requires suitable chloroplast transit peptides (cTPs). In this study, six endogenous cTPs fromD. salinawere shown to direct mCherry import into the chloroplast, and downstream extensions beyond the cleavage site enhanced import efficiency and accuracy. To validate their utility, DAT-2 and DAT-3 were engineered to expressHaematococcus lacustrisβ-carotene hydroxylase and ketolase fused to the cTPs derived from light-harvesting complex II chlorophyll a/b binding protein 1 and tocopherol c

Comparative genomic and phylogenetic analyses of Crataegus chloroplast genomes: insights for evolution and identification.

Crataegus spp. plants are valuable horticultural crops because of their extensive use in Chinese herbal medications, cosmetics, food production, and other industries. However, the wide variety of species, similar morphological characteristics, inherent hybridization, apomixis, and polyploidy have led to confusion in terms of their taxonomic status. Herein, a total of 18 complete chloroplast genomes including 17 Crataegus species and 1 Mespilus species were newly sequenced and comprehensively analyzed for comparative genomics and phylogenetic relationships. The 18 chloroplast genomes possessed

6mA DNA methylation dynamics during chloroplast development in Agave angustifolia Haw.

N6-methyladenine (6mA) DNA accumulates during chloroplast maturation in Agave angustifolia Haw. and covaries with METTL4 protein abundance, identifying 6mA as a candidate epigenetic mark associated with plastid biogenesis. Chloroplast development requires precise coordination of nuclear and organellar genomes, yet the epigenetic mechanisms regulating this process remain poorly understood. Here, we provide the first description of N6-methyladenine (6mA), a dynamic DNA modification, during Agave angustifolia Haw. chloroplast biogenesis. Unveiling the Role of SlRNC1 in Chloroplast Development and Global Gene Regulation in Tomato Plants.

RNC1, a plant-specific gene, is known for its involvement in splicing group II introns within maize chloroplast. However, its role in chloroplast development and global gene expression remains poorly understood. This study aimed to investigate the role of RNC1 in chloroplast development and identify the genes that mediate its function in the development of entire tomato plants. Consistent with findings in maize, RNC1 silencing induced dwarfism and leaf whitening in tomato plants. Subcellular localization analysis revealed that the RNC1 protein is localized to both the nucleus and cytoplasm, in

Comparative chloroplast genomes: insights into the identification and phylogeny of rapid radiation genus Rhodiola.

Rhodiola L. is a genus exhibiting rapid radiation and represents a typical case for studying plastid gene adaptation in species that spread from high altitudes to low altitudes. In this study, 23 samples of 18 Rhodiola species were collected from the Qinghai-Tibetan Plateau and five scattered alpine areas, and the plastid genomes (plastomes) of these species were sequenced, annotated, and compared between high-altitude and widely distributed groups. The plastomes of Rhodiola were found to be highly conserved in terms of gene size, content, and order but highly variable in several lineage-speci

Chloroplast magnesium transporters play essential but differential roles in maintaining magnesium homeostasis.

Magnesium (Mg2+) is essential for photosynthesis in the chloroplasts of land plants and algae. Being the central ion of chlorophyll, cofactor and activator of many photosynthetic enzymes including RuBisCO, magnesium-deficient plants may suffer from leaf chlorosis symptoms and retarded growth. Therefore, the chloroplast Mg2+ concentration is tightly controlled by magnesium transport proteins. Recently, three different transporters from two distinct families have been identified in the chloroplast inner envelope of the model plant Arabidopsis thaliana: MGT10, MGR8, and MGR9. Here, we assess the

Disruption of Photomorphogenesis Leads to Abnormal Chloroplast Development and Leaf Variegation in Camellia sinensis.

Camellia sinensis cv. 'Yanlingyinbiancha' is a leaf-variegated mutant with stable genetic traits. The current study aimed to reveal the differences between its albino and green tissues, and the molecular mechanism underlying the variegation. Anatomic analysis showed the chloroplasts of albino tissues to have no intact lamellar structure. Photosynthetic pigment in albino tissues was significantly lower than that in green tissues, whereas all catechin components were more abundant in the former. Transcriptome analysis revealed most differentially expressed genes involved in the biosynthesis of p

A chloroplast tree for Viburnum (Adoxaceae) and its implications for phylogenetic classification and character evolution.

• Premise of the study: Despite recent progress, significant uncertainties remain concerning relationships among early-branching lineages within Viburnum (Adoxaceae), prohibiting a new classification and hindering studies of character evolution and the increasing use of Viburnum in addressing a wide range of ecological and evolutionary questions. We hoped to resolve these issues by sequencing whole plastid genomes for representative species and combining these with molecular data previously obtained from an expanded taxon sample.• Methods: We performed paired-end Illumina sequencing of plastid

A family of selfish minicircular chromosomes with jumbled chloroplast gene fragments from a dinoflagellate.

Chloroplast genes of several dinoflagellate species are located on unigenic DNA minicircular chromosomes. We have now completely sequenced five aberrant minicircular chromosomes from the dinoflagellate Heterocapsa triquetra. These probably nonfunctional DNA circles lack complete genes, with each being composed of several short fragments of two or three different chloroplast genes and a common conserved region with a tripartite 9G-9A-9G core like the putative replicon origin of functional single-gene circular chloroplast chromosomes. Their sequences imply that all five circles evolved by differ

Enhancement of the reductive activation of chloroplast fructose-1,6-bisphosphatase by modulators and protein perturbants.

To characterize the mechanism of chloroplast fructose-1,6-bisphosphatase activation, we have examined kinetic and structural changes elicited by protein perturbants and reductants. At variance with its well-known capacity for enzyme inactivation, 150 mM sodium trichloroacetate yielded an activatable chloroplast fructose-1,6-bisphosphatase in the presence of 1.0 mM fructose 1,6-bisphosphate and 0.1 mM Ca2+. Other sugar bisphosphates did not replace fructose 1,6-bisphosphate whereas Mg2+ and Mn2+ were functional in place of Ca2+. Variations of the emission fluorescence of intrinsic fluorophores

Expression of early genes in light-induced chloroplast differentiation of cultured plant cells.

The objective of this study was to identify genes, preferentially of the plastid, which are rapidly expressed during the initial phase of blue-light-induced chloroplast development in suspension-cultured cells (Chenopodium rubrum L.) and to analyse the encoded proteins. A cDNA library (lambda gt 10) was constructed using total RNA from plastid preparations of dark-grown cells exposed to blue light for 3, 6 and 12 h. By differential screening, at least three clones were identified which correspond to rapidly light-induced plastid genes. For these and a number of nuclear genes represented by oth
