Chondrocyte Adipogenic Differentiation in Softening Osteoarthritic Cartilage.

A chondrocyte-to-osteoblast lineage continuum exists in the growth plate. Adipogenic differentiation of chondrocytes in vivo should be investigated. Here, unilateral anterior crossbite (UAC), which can induce osteoarthritic lesions in the temporomandibular joint (TMJ), was applied to 6-wk-old C57BL/6 mice. Matrix loss in TMJ cartilage was obvious, as demonstrated by safranin O staining, and the condylar cartilage elastic modulus values, detected by using atomic force microscopy (AFM), were reduced, indicating cartilage softening that might be linked with loss of the highly charged proteoglycan

Development of Injectable Polydactyly-Derived Chondrocyte Sheets.

We are conducting a clinical study of the use of allogeneic polydactyly-derived chondrocyte sheets (PD sheets) for the repair of articular cartilage damage caused by osteoarthritis. However, the transplantation of PD sheets requires highly invasive surgery. To establish a less invasive treatment, we are currently developing injectable fragments of PD sheets (PD sheets-mini). Polydactyly-derived chondrocytes were seeded in RepCell™ or conventional temperature-responsive inserts and cultured. Cell counts and viability, histology, enzyme-linked immunosorbent assay (ELISA), quantitative real-time

Brd4 is required for chondrocyte differentiation and endochondral ossification.

Differentiation of multi-potent mesenchymal stromal cells (MSCs) is directed by the activities of lineage-specific transcription factors and co-factors. A subset of these proteins controls the accessibility of chromatin by recruiting histone acetyl transferases or deacetylases that regulate acetylation of the N-termini of H3 and H4 histone proteins. Bromodomain (BRD) proteins recognize these acetylation marks and recruit the RNA pol II containing transcriptional machinery. Our previous studies have shown that Brd4 is required for osteoblast differentiation in vitro. Here, we investigated the r

Rat Model of Intra-articular Chondrocyte Xenotransplantation.

Current treatment options are not providing an adequate solution for cartilage defects. Articular cartilage lesions in particular are not able to repair spontaneously and progressively degenerate with an arthrosic pattern. Aiming to solve this pressing medical need, xenotransplantation of porcine chondrocytes could be developed as a new therapeutic approach. Xenotransplantation is gaining much attention, thanks to the advances in animal genetic engineering and progress in the characterization of the rejection mechanisms that prevent long-term graft survival. In this regard, our team has identi

Mechanotransduction pathways in the regulation of cartilage chondrocyte homoeostasis.

Mechanical stress plays a critical role in cartilage development and homoeostasis. Chondrocytes are surrounded by a narrow pericellular matrix (PCM), which absorbs dynamic and static forces and transmits them to the chondrocyte surface. Recent studies have demonstrated that molecular components, including perlecan, collagen and hyaluronan, provide distinct physical properties for the PCM and maintain the essential microenvironment of chondrocytes. These physical signals are sensed by receptors and molecules located in the cell membrane, such as Ca2+ channels, the primary cilium and integrins,

Identification of chondrocyte-binding peptides by phage display.

As an initial step toward targeting cartilage tissue for potential therapeutic applications, we sought cartilage-binding peptides using phage display, a powerful technology for selection of peptides that bind to molecules of interest. A library of phage displaying random 12-amino acid peptides was iteratively incubated with cultured chondrocytes to select phage that bind cartilage. The resulting phage clones demonstrated increased affinity to chondrocytes by ELISA, when compared to a wild-type, insertless phage. Furthermore, the selected phage showed little preferential binding to other cell t

Systems genetics analysis of mouse chondrocyte differentiation.

One of the goals of systems genetics is the reconstruction of gene networks that underlie key processes in development and disease. To identify cartilage gene networks that play an important role in bone development, we used a systems genetics approach that integrated microarray gene expression profiles from cartilage and bone phenotypic data from two sets of recombinant inbred strains. Microarray profiles generated from isolated chondrocytes were used to generate weighted gene coexpression networks. This analysis resulted in the identification of subnetworks (modules) of coexpressed genes tha

Modulation of chondrocyte production of extracellular inorganic pyrophosphate.

PURPOSE OF REVIEW: Extracellular inorganic pyrophosphate (ePPi) both inhibits and promotes different forms of pathologic mineralization. Basic calcium phosphate (BCP) deposition results from depressed levels of ePPi while excess levels of ePPi leads to calcium pyrophosphate dihydrate crystal deposition (CPPD) disease. These crystals are also often identified in patients with osteoarthritis, the most prevalent form of arthritis causing significant morbidity. RECENT STUDIES: The two primary hypotheses for generation of ePPi, export of inorganic pyrophosphate through the multipass transmembrane

Autologous Chondrocyte Transplantation in Femoroacetabular Impingement Syndrome: Growth and Redifferentiation Potential of Chondrocytes Harvested from the Femur in Cam-Type Deformities.

OBJECTIVE: Cam-type femoroacetabular impingement (FAI) syndrome is one of the most frequent reasons for cartilage damage in the hip. Autologous chondrocyte transplantation has proven high success rates in the treatment of focal chondral defects; however, harvesting of chondrocytes in the hip has been reported but not specifically from the region of femoral cam lesions. Therefore, the goal of this study was to analyze the growth and redifferentiation potential of cartilage samples harvested from the cam deformities in patients with FAI. DESIGN: Cartilage samples were gained from 15 patients wi

Increased Chondrocytic Gene Expression Is Associated With Improved Repair Tissue Quality and Graft Survival in Patients After Autologous Chondrocyte Implantation.

BACKGROUND: Assays to quantitate the quality of autologous chondrocyte implants have recently become available. However, the correlation of the assay score with radiological and clinical outcomes has not been established. PURPOSE/HYPOTHESIS: The purpose was to assess the influence of cell identity (chondrocyte/synoviocyte gene expression ratio) and viability on patient-reported outcome measures, graft survival, and repair tissue quality. It was hypothesized that greater cell product quality as assessed through an identity assay and cell viability is associated with superior outcomes after aut

Chondrocyte-specific knockout of the G protein G(s)alpha leads to epiphyseal and growth plate abnormalities and ectopic chondrocyte formation.

UNLABELLED: G(s)alpha is a ubiquitously expressed G protein alpha-subunit that couples receptors to adenylyl cyclase. Mice with chondrocyte-specific ablation of the G(s)alpha gene had severe epiphyseal and growth plate abnormalities and ectopic cartilage formation within the metaphyseal region of the tibia. These results show that G(s)alpha negatively regulates chondrocyte differentiation and is the critical signaling mediator of the PTH/PTH-rP receptor in growth plate chondrocytes. INTRODUCTION: G(s)alpha is a ubiquitously expressed G protein alpha-subunit that mediates signaling through G p

BIOORTHOGONAL TUNING OF HYDROGEL STIFFNESS PROMOTES ZONAL REDIFFERENTIATION OF PASSAGED CHONDROCYTES.

Generating bioengineered cartilage that recapitulates the depth-dependent phenotype, structure, and function of native articular cartilage remains a challenge. While cartilage is rich in aggrecan and type II collagen, proper function depends on depth-dependent protein expression. Superficial zone chondrocytes (SZCs) secrete proteoglycan-4 (PRG4) to lubricate the cartilage surface. Deep zone chondrocytes produce type X collagen (COLX) to support compressive loading and load transfer to subchondral bone. We previously demonstrated that passaged full-thickness chondrocytes (FTCs) and zonal chondr

Anti-Inflammatory Effects of Artocarpin-Loaded Chitosan Microparticles on Inflammatory Chondrocytes.

Artocarpin is a promising bioactive compound with anti-inflammatory potential; however, its limited aqueous solubility may restrict its direct biological application. Therefore, artocarpin-loaded chitosan microparticles (CSPs/AE) were prepared as a biocompatible carrier to improve artocarpin delivery. This study aimed to evaluate the cytocompatibility, antioxidant activity, and anti-inflammatory effects of CSPs/AE in lipopolysaccharide (LPS)-stimulated chondrocytes. CSPs/AE maintained high cell viability (>80% at 0.125 and 0.5 mg/mL) and showed significantly lower cytotoxicity than free artoca

Energy crisis and cartilage collapse: metabolic reprogramming of chondrocytes in osteoarthritis.

Osteoarthritis (OA), a degenerative joint disease imposing a significant global disease burden, exhibits pathological mechanisms far more complex than mere "cartilage wear". Previous research has long centered on mechanical wear of articular cartilage and inflammatory responses. However, recent studies reveal that metabolic reprogramming and energy metabolism disorders constitute core metabolic alterations in OA. This review centers on the link between the energy crisis in articular chondrocytes and cartilage collapse. It systematically examines evidence suggesting that impaired oxidative phos

Expansion and Delivery of Human Chondrocytes on Gelatin-Based Cell Carriers.

Cartilage damage is common in sports injuries and cartilage-related diseases, such as degenerative joint and rheumatic disorders. Autologous chondrocyte implantation (ACI) is a widely used cell-based therapy for repairing cartilage damage in clinical practice. In this procedure, a patient's chondrocytes are isolated, cultured in vitro to expand the cell population, and then implanted into the damaged site. However, in vitro expansion of chondrocytes on standard 2D culture surfaces leads to dedifferentiation (loss of the chondrocyte phenotype), and the delivery of detached cells has proven to b

Exploring the Multifactorial Regulation of PIEZO1 in Chondrocytes: Mechanisms and Implications.

The mechanosensitive PIEZO1 ion channel plays a pivotal role in the regulation of chondrocyte function and is involved in various physiological and pathological processes, including cartilage degradation and osteoarthritis (OA). This review explores the regulatory mechanisms governing PIEZO1 activation and its interactions with mechanical stress, extracellular matrix (ECM) stiffness, inflammatory factors, and other ion channels. We discuss the role of PIEZO1 in calcium signaling, its modulation by ECM stiffness, and the implications for cartilage health, particularly under high mechanical load

Primary Isolation, Culture, and Phenotypic Verification of Rat Knee Articular Chondrocytes.

Cartilage damage resulting from trauma, aging, or inflammatory conditions remains a significant clinical challenge due to the tissue's inherently limited regenerative capacity. Chondrocytes, being the sole cellular component of cartilage, are responsible for maintaining extracellular matrix structure and function, making them essential for understanding cartilage physiology, pathology, and regeneration. The current study presents a comprehensive and standardized protocol for the isolation, culture, and phenotypic verification of primary rat articular chondrocytes from the knee joint. The proce

Aberrant anabolism hinders constructive metabolism of chondrocytes by pharmacotherapy in osteoarthritis.

Osteoarthritis (OA) is a highly prevalent and disabling disease with an unmet therapeutic need. The characteristic cartilage loss and alteration of other joint structures result from a complex interaction of multiple risk factors, with mechanical overload consistently playing a central role. This overload generates an inflammatory response in the cartilage due to the activation of the innate immune response in chondrocytes, which occurs through various cellular mechanisms. Moreover, risk factors associated with obesity, being overweight, and metabolic syndrome enhance the inflammatory response
