BACKGROUND: Variants in the patatin-like phospholipase domain containing 6 (PNPLA6) gene cause a broad spectrum of neurological disorders characterized by gait disturbance, visual impairment, anterior hypopituitarism, and hair anomalies. This review examines the clinical, cellular, and biochemical features found across the five PNPLA6-related diseases, with a focus on future questions to be addressed.
MATERIALS AND METHODS: A literature review was performed on published clinical reports on patients with PNPLA6 variants. Additionally, in vitro and in vivo models used to study the encoded prote

Based on of 10 personal observations, the authors discuss the modes of onset and the various clinical, functional and genetic aspects of primary degenerative choroidal atrophies. Central areolar atrophy may be the only lesion or is sometimes associated with other signs of a more extensive degeneration (fundus flavimaculatus, degeneration of the posterior pole). Among the diffuse forms a familial case is reported which can be interpreted as a sectorial hypoplasia, and a case similar to a choroideremia, but with recessive transmission.

In the past 10 y, there have been considerable advances in the mapping, isolation, and characterization of many genes for important ocular conditions: retinitis pigmentosa, Norrie disease, Waardenburg syndrome, choroideremia, aniridia, retinoblastoma, and others. The candidate gene approach has now supplemented classical linkage studies and positional cloning in the investigation of ocular disorders. Developmentally expressed genes and animal models have provided insights as to the etiology of other disorders. With this knowledge at hand, genetic counselling for heritable eye diseases has been

In outer retinal degenerative diseases such as retinitis pigmentosa, choroideremia, and geographic atrophy, 30% of the ganglion cell layer in the macula remains intact. With subretinal and epiretinal prostheses, these inner retinal cells are stimulated with controlled electrical current by either a microphotodiode placed in the subretinal area or a microelectrode array tacked to the epiretinal region. As the patient learns to interpret the resulting phosphene patterns created in the brain through special rehabilitation exercises, their orientation, mobility, and quality of life increase. Impla

The luminance threshold for the detection of 25 Hz flicker was measured in nine patients with retinal disorders under stimulus conditions that have been shown previously to involve an interaction between rod and cone systems. The disorders studied included congenital stationary nightblindness, X-linked juvenile retinoschisis, hereditary dominant optic atrophy, optic atrophy found in association with neurofibromatosis, retinitis pigmentosa, choroideremia, and an acquired diffuse photoreceptor disorder, all of which involve pathologic changes that are presumed to occur primarily at specific leve

Optical coherence tomography angiography (OCTA) is a novel, noninvasive imaging modality that allows depth-resolved imaging of the microvasculature in the retina and the choroid. It is a powerful research tool to study the pathobiology of retinal diseases, including inherited retinal dystrophies. In this review, we provide an overview of the evolution of OCTA technology, compare the specifications of various OCTA devices, and summarize key findings from published OCTA studies in inherited retinal dystrophies including retinitis pigmentosa, Stargardt disease, Best vitelliform macular dystrophy,

The availability of more and more reliable obstetrical echographies makes now possible to screen fetuses for microphtalmies and anophtalmies. More over, by mean of linkage studies with DNA markers within a family, we can identify the women who will transmit X-linked diseases, and realize a prenatal diagnosis. This technology can be applied to the following ophthalmological diseases: the X-linked retinoschisis, the choroideremia, the ocular albinism, the Noorie disease, and recently the retinoblastoma. From now on, the use of such a technology which makes it possible to detect ophthalmological

OBJECTIVE: Preimplantation genetic diagnosis (PGD) is an established application of genetic testing in the context of in vitro fertilization. PGD is an alternative method to prenatal diagnosis which aims to prevent the transmission of an inherited disorder to the progeny by implanting only embryos that do not carry genetic predisposition for a particular disease. The aim of this study is to provide an overview of eye disorders for which PGD has been carried out.
METHODS: The European literature search focused on best practices, ethical issues, risks and results of PGD for inherited eye disord

OBJECTIVES: Self-management interventions in chronic disease are frequently communicated as general wellness advice. Horne's Necessity-Concerns Framework showed adherence tracks perceived necessity (OR = 1.742) and ameliorated concerns (OR = 0.504); Miller found health literacy correlates more strongly with adherence for non-medication regimens (r = 0.20) than medication regimens (r = 0.09). Neither addresses the authority signals prescriptions carry and self-management advice lacks.
METHODS: Discussion paper grounded in conceptual analysis, integrating Horne's NCF, Miller's health literacy w

This retrospective follow-on study reviewed 25 consecutive patients with genetically confirmed choroideremia who were treated with subretinal AAV2-CAG-REP1 in dose-escalation gene therapy clinical trials conducted at Oxford University Hospitals NHS Foundation Trust and the University Eye Hospital Tübingen. Six patients in Oxford received 1010 vector genomes in 100 µL, while a further 19 patients across both centers received 1011 vector genomes in 100 µL. Although the primary safety outcomes of both investigator-led academic studies have previously been reported, retinal imaging changes were ob

BACKGROUND/OBJECTIVES: Inherited retinal diseases (IRDs) are a leading cause of blindness in working-age adults. Although artificial intelligence (AI) shows potential for disease classification, progress is limited by small datasets, reliance on labelled data and limited integration of multiple imaging modalities. RETFound, a foundation model pretrained on over 900,000 fundus photographs, may address these limitations. While RETFound is based on the transformer architecture, convolutional neural networks (CNNs), such as ResNet and EfficientNet_B0, have also demonstrated strong performance in c

Among the genetic diseases of vision, retinitis pigmentosa (alone or in combination with systemic afflictions like Usher syndrome, Kearns-Sayre syndrome) deserves special interest because it can be autosomal dominant, autosomal recessive, and X-linked. Leber's hereditary optic neuropathy, the first mitochondrial disease to be defined at the molecular level, manifests at mitochondrial DNA position 11778, but 10 other mutations have recently been found, all residing within mitochondrial DNA-encoded respiratory chain polypeptides. The analysis of Leber's optic neuropathy mutations has provided ne

Syndromic hearing impairment encompasses hundreds of phenotypes. We identified a young female patient affected by the unique combination of dysplasia of the auricular system, patent ductus arteriosus (PDA), choroideremia, and enamel hypoplasia. The patient was treated with PDA ligature and left exploratory tympanotomy. Impairment in all four systems suggests a correlation with the neural crest. It is presumed that all of the features result from the same origin, probably through autosomal recessive inheritance or a novel mutation during the embryonic period. When audio-dento-oculo-cardio syste
