Characterization of Dye-Loaded Poly(lactic-co-glycolic acid) Nanoparticles by Comprehensive Two-Dimensional Liquid Chromatography Combining Hydrodynamic and Reversed-Phase Liquid Chromatography.

Analytical methods for the assessment of drug-delivery systems (DDSs) are commonly suitable for characterizing individual DDS properties, but do not allow determination of several properties simultaneously. A comprehensive online two-dimensional liquid chromatography (LC × LC) system was developed that is aimed to be capable of characterizing both nanoparticle size and encapsulated cargo over the particle size distribution of a DDS by using one integrated method. Polymeric nanoparticles (NPs) with encapsulated hydrophobic dyes were used as model DDSs. Hydrodynamic chromatography (HDC) was used

Separation of nine novel triterpene saponins from Camellia japonica seeds using high-performance countercurrent chromatography and reversed-phase high-performance liquid chromatography.

INTRODUCTION: Camellia japonica L. (Theaceae) is an evergreen shrub, which is cultivated as a popular ornamental tree in Korea, China, and Japan and its seeds have been used as a source of cooking oil, in cosmetics and as a traditional medicine. Intensive phytochemical works have revealed that oleanane-type saponins are the characteristic compounds of the seeds of C. japonica. OBJECTIVE: The purpose of the present study is to isolate and determine oleanane-type saponins from C. japonica using high-performance countercurrent chromatography (HPCCC) coupled with reversed-phase high-performance l

Potential and limitations of on-line comprehensive reversed phase liquid chromatography×supercritical fluid chromatography for the separation of neutral compounds: An approach to separate an aqueous extract of bio-oil.

On-line comprehensive Reversed Phase Liquid Chromatography×Supercritical Fluid Chromatography (RPLCxSFC) was investigated for the separation of complex samples of neutral compounds. The presented approach aimed at overcoming the constraints involved by such a coupling. The search for suitable conditions (stationary phases, injection solvent, injection volume, design of interface) are discussed with a view of ensuring a good transfer of the compounds between both dimensions, thereby allowing high effective peak capacity in the second dimension. Instrumental aspects that are of prime importance

Estimation of DNA Methylation in Wheat Using Reverse Phase Ultra-High-Performance Liquid Chromatography Coupled with Mass Spectrometry During Leaf Rust Pathogenesis.

Reverse phase ultra-high-performance liquid chromatography is considered the most reliable and sensitive technique to determine and separate analytes with small differences in their molecular weights. Leveraging this technique coupled with mass spectrometry, we developed a method for simultaneous determination of 2'-deoxycytidine and its methylated analogue, 5-methyl-2'-deoxycytidine, in wheat. The nonpolar C18 column was selected that greatly shortened the detection time to approximately 2 min. Formic acid was added to the mobile phase to achieve the complete separation. Thus, avoiding the si

Phosphorus-doped deep eutectic solvent-derived carbon dots-modified silica as a mixed-mode stationary phase for reversed-phase and hydrophilic interaction chromatography.

In this work, phosphorus-doped carbon dots (P-DESCDs) were successfully prepared using choline chloride/lactic acid type deep eutectic solvent and phosphoric acid as ingredients, and (3-aminopropyl) trimethoxysilane was used as a bridge to graft P-DESCDs onto the silica surface to obtain a new mixed-mode stationary phase (Sil-P-DESCDs) for reversed-phase and hydrophilic interaction liquid chromatography. The successful preparation of the stationary phase was confirmed by laser scanning confocal microscopy, elemental analysis, and Fourier transform infrared spectrometry. Interestingly, the dopi

[Prediction of n-octanol/water partition coefficient of strongly ionized compounds by ion-pair reversed-phase liquid chromatography with silica-based stationary phase].

The n-octanol/water partition coefficient (log P) is an important parameter to characterize the overall hydrophobicity of organic compounds. Reversed-phase liquid chromatography (RPLC) has been recommended as an effective method for the indirect determination of log P by the Organization for Economic Cooperation and Development (OECD). Using RPLC, most studies focus on the determination of log P or the apparent n-octanol/water partition coefficient (log D) of neutral compounds and weakly ionized compounds. However, the experimental log P or log D values of strongly ionized compounds have rarel

Enantiomeric separation of chiral pesticides by high performance liquid chromatography on cellulose tris-3,5-dimethyl carbamate stationary phase under reversed phase conditions.

Twenty chiral pesticides were tested, of which seven samples were directly separated by HPLC using cellulose tris-3,5-dimethyl carbamate (CDMPC) chiral stationary phase under RP conditions. The influence of mobile phase composition and column temperatures from 0 degrees C to 40 degrees C on the separations were investigated. The mobile phases were methanol/water or ACN/water at a flow rate of 0.8 mL/min with UV detection at 230 or 210 nm. Epoxiconazole, terallethrin, benalaxyl, and diclofopmethyl were observed to obtain the baseline separation under suitable conditions and other pesticides pyr

Characterization of Antisense Oligonucleotide Impurities by Ion-Pairing Reversed-Phase and Anion Exchange Chromatography Coupled to Hydrophilic Interaction Liquid Chromatography/Mass Spectrometry Using a Versatile Two-Dimensional Liquid Chromatography Setup.

Determination of phosphorothioate oligonucleotide purity and impurity profile is commonly performed by ion-pairing reversed-phase liquid chromatography (IPRP) with a mobile phase containing triethylamine (TEA) and hexafluoro-2-propanol (HFIP). However, ion-suppressing effects of TEA hamper mass spectrometry (MS) instrumentation sensitivity and HFIP can affect the robustness of the mass spectrometer due to its corrosive nature. Anion exchange chromatography (AEX) is an orthogonal separation mode to IPRP but typically cannot be directly coupled to MS. In this work, we developed a multiple heart-

Preparation of two ionic liquid bonded stationary phases and comparative evaluation under mixed-mode of reversed phase/ hydrophilic interaction/ ion exchange chromatography.

The present work describes the preparation of two ionic liquid and carboxyl acid silane reagents via photo-initiated thiol-ene click chemistry that have been bonded to silica to afford two mixed-mode stationary phases (Sil-C4Im-C9Co and Sil-C9Im-C4Co). The two stationary phases provided satisfactory retention repeatability and efficiencies. The influence of acetonitrile content, salt concentration and pH of the mobile phase was investigated to clarify the retention properties of the prepared stationary phases. The results showed that the prepared Sil-C4Im-C9Co and Sil-C9Im-C4Co undergo multipl

A rapid reversed-phase liquid chromatography method for the determination of individual monoclonal antibody content in a co-formulated drug product.

Co-formulations of therapeutic monoclonal antibodies (mAbs) offer distinct advantages to the patient and clinic compared to single mAb drug products. However, the quantification of individual mAb content within a co-formulation is complicated as most mAbs exhibit similar physiochemical properties. While other techniques have been reported to fit this purpose, they fall short in terms of resolution, reproducibility and/or timescale, limiting their applicability. To address these shortcomings, a reversed-phase liquid chromatography (RPLC) method was developed, utilizing a diphenyl column packed

Measurement by reversed-phase high-performance liquid chromatography of malondialdehyde in normal human urine following derivatisation with 2,4-dinitrophenylhydrazine.

A selective and sensitive method based on derivatisation with 2,4-dinitrophenylhydrazine (DNPH) and consecutive HPLC gradient separation is described for the determination of malondialdehyde (MDA) in urine. Preparation of urine samples involved a one-step derivatisation/extraction procedure. Separation was achieved using a Waters SymmetryC(18) column (3.9 x 150 mm) and linear gradient of acetonitrile in water (from 30% to 70% in 30 min). The overall detection limit of the method was 56 nM of MDA in urine. The recovery of MDA was 94.3+/-8.6%. MDA in urine of healthy volunteers, measured using t

Determination of ritonavir, a new HIV protease inhibitor, in biological samples using reversed-phase high-performance liquid chromatography.

A simple, accurate and precise high-performance liquid chromatographic method has been developed for measurement of ritonavir concentrations in human plasma. Ritonavir was partitioned from the plasma using liquid-liquid extraction with a mixture of ethyl acetate and hexane at neutral pH, with an average recovery >80%. Following two sequential washings of the reconstituted sample with hexane, chromatographic separation was accomplished on a C18 analytical column with a mobile phase containing acetonitrile, methanol and 0.01 M tetramethylammonium perchlorate in 0.1% aqueous trifluoroacetic acid

Immunochromatographic analysis of bovine growth hormone releasing factor involving reversed-phase high-performance liquid chromatography-immunodetection.

We have developed high-performance immunoaffinity chromatography (HPIAC) methods for the detection and quantitation of bovine growth hormone releasing factor (GHRF), which could also be applicable to its metabolites in biofluids. These approaches have involved a combination of IAC using immobilized antibody (Ab) to GHRF, together with reversed-phase high-performance liquid chromatography (RP-HPLC) separations of initially isolated and concentrated protein, followed by selective detection, involving on-line immunodetection (ID) schemes. ID methods involved HPIAC supports of the Ab, together wit

Accumulation of indoxyl sulfate, an inhibitor of drug-binding, in uremic serum as demonstrated by internal-surface reversed-phase liquid chromatography.

We quantified indoxyl sulfate in uremic serum by using internal-surface reversed-phase high-performance liquid chromatography. Its concentrations were markedly increased in chronic hemodialysis patients, and were significantly but weakly correlated with the concentrations of creatinine and beta 2-microglobulin in these patients' serum, and with the duration of their hemodialysis treatment. Indoxyl sulfate could not be removed effectively by conventional hemodialysis because of its strong binding to serum albumin. Equilibrium dialysis demonstrated that indoxyl sulfate inhibited the binding of s

Determination of lycopene, alpha- and beta-carotene and retinyl esters in human serum by reversed-phase high performance liquid chromatography.

A rapid specific, microdetermination of the major human blood carotenoids by high performance liquid chromatography (HPLC) separation and quantitation at 466 nm is detailed in this paper. Serum retinyl esters can also be quantified utilizing the same separation procedure but detected at 325 nm. One hundred microliters of deproteinated serum were extracted with chloroform and injected on a reverse-phase column. Separation occurred within 16 min for all compounds of interest employing a mobile solvent of MeOH/AcN/CHCl3 (47:47:6). All compounds were quantified at the wavelengths cited by integrat

Development, validation and application of an ion-pair reversed-phase liquid chromatography-tandem mass spectrometry method for the quantification of nusinersen.

Background: The fully phosphorothioate-modified oligonucleotide (OGN) nusinersen has low ionization efficiency in the negative ion mode, resulting in a low mass spectrometry response. There have been no relevant reports on developing a LC-MS method for the determination of nusinersen by optimizing mobile phase composition. Materials & methods: Mobile phase additives comprised of 15 mM triethylamine/25 mM 1,1,1,3,3,3-hexafluoro-2-propanol with a pH of 9.6. Nusinersen was extracted from plasma using Oasis® HLB solid-phase extraction (Waters, MA, USA). Results & conclusion: By adjusting the pH of

Monomer-mediated fabrication of microporous organic network@silica microsphere for reversed-phase/hydrophilic interaction mixed-mode chromatography.

Microporous organic networks (MONs) are promising in high performance liquid chromatography (HPLC) with large specific surface area, good hydrophobicity and stability. However, their superhydrophobic structures restrict MONs-based HPLC only in reversed-phase mode. To decrease the hydrophobicity of pristine MONs and to expand their broad application in HPLC, here we described the monomer-mediated fabrication of core-shell MON-2COOH@SiO2 microspheres for reversed-phase liquid chromatography (RPLC)/hydrophilic interaction liquid chromatography (HILIC) mixed-mode chromatography for the first time.
