[Importance of the immotile cilia syndrome in E.N.T. field].

Associated with nasal secretion, the ciliary activity establish the first line of defense of respiratory mucosa. The primary pathology of the ciliary structures is not well known. Correlated with testing of the mucociliary transport, only the ultrastructural study could open this very interesting field. With a short review of the present status of the ineffective mucociliary clearance mechanisms, 9 new cases of immotile cilia syndrome, with or without situs inversus, are studied. Actually, the following anomalies seems to be significant : lack of one or two dyneïn arms, confusion of the centra

Genetical and ultrastructural aspects of the immotile-cilia syndrome.

The immotile-cilia syndrome is a congenital disorder characterized by all cilia in the body being either immotile or showing an ineffective beating pattern. Most symptoms, not unexpectedly, come from the ciliated epithelia, but two further symptoms are: (1) male sterility caused by the spermatozoa being unable to swim progressively (the sperm tail has the same structure as a cilium), and (2) situs inversus in 50% of the cases possible caused by an inability of embryonic cilia to shift the heart to the left side. By electron microscopy, one can see directly which of the many ciliary components

Effect of trichloroethylene on cilia activity in rabbit trachea.

The ciliotoxicity of trichloroethylene was studied in terms of the time required for ciliostasis in specimens of trachea from 35 male rabbits following exposure of the specimens to different vapour concentrations. The highest vapour concentration was linked with the most rapid ciliostasis but even in the lowest concentration tested, 5.000 p.p.m. (26 g/m3), hastened ciliostasis. Specimens from the upper region of the trachea underwent ciliostasis more slowly than did those from the lower region. The absolute uptake of trichloroethylene was lower in the former than in the latter specimen. The di

Kaempferol Restores Cilia Length Through Axonemal Microtubule Stabilization.

Ciliary shortening disrupts cilium-dependent signaling and tissue homeostasis and contributes to diverse ciliopathies and acquired disorders. However, the underlying mechanisms and pharmacological strategies for restoring ciliary length remain poorly understood. Here, we identify Kaempferol (Kae) as a previously unrecognized small-molecule regulator of ciliary length. Transcriptomic profiling showed that Kae-responsive genes were enriched in cytoskeleton- and microtubule-related processes, including microtubule organization and motor protein activity, suggesting that cytoskeletal regulation ma

Structural diversity of axonemes across mammalian motile cilia.

Reproduction, development and homeostasis depend on motile cilia, whose rhythmic beating is powered by a microtubule-based molecular machine called the axoneme. Although an atomic model of the axoneme is available for the alga Chlamydomonas reinhardtii1, structures of mammalian axonemes are incomplete1-5. Furthermore, we do not fully understand how molecular structures of axonemes vary across motile-ciliated cell types in the body. Here we use cryoelectron microscopy, cryoelectron tomography and proteomics to resolve the 96-nm modular repeat of axonemal doublet microtubules (DMTs) from both sp

Methods to assess neuronal primary cilia electrochemical signaling.

Primary cilia are polymodal sensory organelles which project from the apical side of polarized cells. They are found in all brain hemispheres but are most pronounced in neurons which comprise the granular layers of the hippocampus and cerebellum. Pathogenic variants in genes which encode primary cilia components are responsible for neuronal ciliopathies- a group of central nervous system disorders characterized by neurodevelopmental conditions such as intellectual disability, seizure, ataxia, and sensory deficits. In the hippocampus, neuronal primary cilia form chemical synapses with axons and

Primary Cilia Mediate TSH-Regulated Thyroglobulin Endocytic Pathways.

Primary cilia are sensory organelles with a variety of receptors and channels on their membranes. Recently, primary cilia were proposed to be crucial sites for exocytosis and endocytosis of vesicles associated with endocytic control of various ciliary signaling pathways. Thyroglobulin (Tg) synthesis and Tg exocytosis/endocytosis are critical for the functions of thyroid follicular cells, where primary cilia are relatively well preserved. LRP2/megalin has been detected on the apical surface of absorptive epithelial cells, including thyrocytes. LRP2/megalin on thyrocytes serves as a Tg receptor

Cilia, ciliopathies and hedgehog-related forebrain developmental disorders.

Development of the forebrain critically depends on the Sonic Hedgehog (Shh) signaling pathway, as illustrated in humans by the frequent perturbation of this pathway in holoprosencephaly, a condition defined as a defect in the formation of midline structures of the forebrain and face. The Shh pathway requires functional primary cilia, microtubule-based organelles present on virtually every cell and acting as cellular antennae to receive and transduce diverse chemical, mechanical or light signals. The dysfunction of cilia in humans leads to inherited diseases called ciliopathies, which often aff

Intrahepatic bile duct primary cilia in biliary atresia.

AIM: The etiopathogenesis of non-syndromic biliary atresia (BA) is obscure. The primary aim was to investigate intrahepatic bile duct cilia (IHBC) in BA at diagnosis and its correlation with clinical outcome. The secondary aim was to analyze IHBC in routine paraffin-embedded liver biopsies using conventional scanning electron microscopy (SEM). METHODS: Surgical liver biopsies taken at diagnosis from 22 BA infants (age range, 39-116 days) and from eight children with non-BA chronic cholestasis (age range, 162 days -16.8 years) were evaluated for IHBC by immunofluorescence (IF) and SEM. A minim

ATR promotes cilia signalling: links to developmental impacts.

Mutations in ATR(ataxia telangiectasia and RAD3-related) cause Seckel syndrome (ATR-SS), a microcephalic primordial dwarfism disorder. Hitherto, the clinical manifestation of ATR deficiency has been attributed to its canonical role in DNA damage response signalling following replication fork stalling/collapse. Here, we show that ATR regulates cilia-dependent signalling in a manner that can be uncoupled from its function during replication. ATR-depleted or patient-derived ATR-SS cells form cilia of slightly reduced length but are dramatically impaired in cilia-dependent signalling functions, in

[Primary cilia control different steps of brain development].

The role of primary cilia in adult neurons remains elusive, however their developmental functions during brain morphogenesis have been recently highlighted thanks to mouse models. Unmistakably, they are needed for Hedgehog (Hh)-dependent patterning in the forebrain. Not only for Hh reception itself, but most importantly for a downstream event in the Hh transduction pathway, independent of Hh ligand: the Gli3 processing. Indeed, phenotypes due to cilia disruption in the developing brain, such as early patterning, olfactory bulb or corpus callosum formation, can be rescued by reintroducing Gli3-

Primary cilia regulate Gli/Hedgehog activation in pancreas.

Previous studies have suggested that defects in pancreatic epithelium caused by activation of the Hedgehog (Hh) signaling pathway are secondary to changes in the differentiation state of the surrounding mesenchyme. However, recent results describe a role of the pathway in pancreatic epithelium, both during development and in adult tissue during neoplastic transformation. To determine the consequences of epithelial Hh activation during pancreas development, we employed a transgenic mouse model in which an activated version of GLI2, a transcriptional mediator of the pathway, is overexpressed spe

Cultured peritoneal mesothelial cells exhibit apical primary cilia.

This study examined primary cilia on cultured human and rabbit peritoneal mesothelial cells (PMC) and investigated factors that influence ciliary expression. Primary cilia were examined with indirect immunocytochemistry, laser scanning confocal microscopy and scanning electron microscopy. Ciliary expression was evaluated in cultures with or without l-cysteine (0.25 mM) or exposure to Ca(2+)-free Krebs-Ringer solution supplemented with EGTA, 0.5 mM. This treatment disrupted cell monolayer integrity. Cilia were counted and normalized to total cell counts using NIH image. Primary cilia were ident

Short nasal respiratory cilia and impaired mucociliary function.

A 48-year-old man was examined because of anosmia. He was otherwise healthy except for mild arterial hypertension. He had stopped smoking 20 years ago. Previously he had been exposed to sulfuric acid gases for 3 years in his work. Clinical examination revealed no findings to explain his anosmia. The patient was fertile, indicating normal sperm/cilia motility. Nasal mucociliary function was examined by radioactive tracer and found to be markedly and constantly impaired. Ciliary ultrastructure in cross-sections was normal. However, in longitudinal sections the length of the cilia varied from 0.6

Situs inversus in homozygous mice without immotile cilia.

Mice homozygous for the mutation situs inversus, iv, are characterized by situs inversus in 50 percent of the individuals. We have investigated the ultrastructure and motility of tracheal cilia and sperm tails in homozygous individuals with and without situs inversus. We found no abnormalities of ciliary structure. Therefore, ciliary immotility is not an essential correlate of situs inversus.

Primary cilia: sensors of biliary homeostasis and disease.

The primary cilium (plural 'cilia') is a highly specialized organelle found on nearly every vertebrate cell. It functions as a sensor, cell cycle regulator and coordinator of multiple signalling transduction pathways. In cholangiocytes, primary cilia extend from the cell surface into the bile duct lumen, acting as chemosensors, mechanosensors and osmosensors in response to changes in bile composition and flow. This primary-cilium-mediated sensing regulates the biliary microenvironment and is crucial to maintaining homeostasis and promoting biliary regeneration. Moreover, alterations in the str

Primary Cilia of Astrocytes in the Retina in Development and Disease.

PURPOSE: Defects in primary cilia and ciliary signaling are associated with an array of neurodegenerative diseases. Primary cilia are sensory organelles that transmit and regulate various cellular communication pathways, including the significant Sonic hedgehog and Wnt signal pathways. Extensive studies have shown that neuronal cilia in the central nervous system contribute to early patterning, neuronal maturation, and survival. The behavior of cilia on astrocytes in glaucoma and aging remains poorly understood. This study used rodent and human retinal astrocytes to examine primary cilia and t

Primary cilia as mechanosensors in musculoskeletal homeostasis and disease.

The musculoskeletal system relies on precise mechanical signal transduction to maintain physiological homeostasis, yet the underlying mechanisms remain incompletely characterized. Primary cilia (PC), membrane-bound organelles that protrude into the extracellular matrix, acting as cellular sensors to integrate biomechanical forces into cellular responses. Moreover, their dynamic assembly and disassembly are tightly coupled to mechanical stress perception, which also suggests a pivotal role in regulating musculoskeletal tissue function. This review systematically synthesizes current evidence on
