Cinnamic acid rescues behavioral deficits in a mouse model of traumatic brain injury by targeting miR-455-3p/HDAC2.

AIMS: Traumatic brain injury (TBI) not only induces physiological disabilities but also leads to cognitive impairment. However, no effective therapeutic approach for TBI-related memory decline exists. In this study, we treated TBI mice with cinnamic acid (CNA) to detect whether CNA is able to rescue the memory deficits induced by TBI and to explore the potential mechanisms. MAIN METHODS: Mice were divided into the following groups: the sham group, the TBI group, the TBI + CNA group and the CNA group. Basic physiological parameters, neurological severity score and brain water content were anal

Visible-Light-Assisted Enzyme-Catalyzed Cascade Decarboxylative Coupling Reaction of Benzoylformic Acids and Cinnamic Acids to Synthesize Chalcones.

A visible light-assisted enzyme-catalyzed biological cascade reaction for the synthesis of chalcones is established by continuous decarboxylative coupling reaction of aroylcarboxylic acids with cinnamic acids in water at 30 °C, and 30 chalcone derivatives are synthesized using this method, of which 5 were natural products. This is the first time that a visible-light-assisted system was introduced into the biological cascade reaction, which effectively promoted the in situ regeneration of coenzyme factors, thus realizing the catalytic cycle of the complete process. Vitamin B12 (VB12) showed a b

Endophytic bacterial's phenylalanine and trans-cinnamic acid as exogenous precursors involved in the modulating colchicine biosynthesis pathway in Gloriosa superba.

Unlocking the hidden power of endophytes reveals a novel pathway for boosting colchicine biosynthesis in Gloriosa superba. Metabolite analysis of endophyte culture filtrates identifies essential precursors i.e., phenyl ethyl alcohol, phenylalanine, trans-cinnamic acid, and benzoic acid participating in colchicine biosynthesis. Bacilli-treated Gloriosa superba plants demonstrated a remarkable increase in plant growth and colchicine content over untreated plants. Moreover, enhanced nutrient uptake (Fe, Zn, Cu, Mn) further induced the enzymatic activities critical for colchicine biosynthesis gene

Novel cinnamic acid-ribavirin conjugates: design, synthesis, antiviral activity against tobacco mosaic virus and mechanistic study.

BACKGROUND: Plant viral diseases represent a serious threat to global agricultural productivity and sustainable development. Most commercially available antiviral agents function primarily as protectants. However, the long-term and repetitive application of conventional broad-spectrum antiviral agents has led to the emergence of resistance, diminishing their field efficacy. To discover new antiviral drugs, we designed and synthesized 31 novel ribavirin derivatives incorporating cinnamic acid motifs via a lead optimization strategy and evaluated their anti-tobacco mosaic virus (TMV) activity.

Cinnamic acid promotes elongation of hair peg-like sprouting in hair follicle organoids via oxytocin receptor activation.

Considerable global demand exists for the development of novel drugs for the treatment of alopecia. A recent report demonstrated that oxytocin promotes hair growth activity in human dermal papilla (DP) cells; however, its application in drugs or cosmetic products is challenging because rapid degradation and relatively large molecular weight prevent long-term topical administration on the scalp. Here, we examined cinnamic acid, a small molecule activator for oxytocin receptor (OXTR) expression. Treatment with cinnamic acid led to upregulation of OXTR and trichogenic gene expression in human DP

A novel cinnamic and caffeic acid-conjugated peptide analogs with anticonvulsant and analgesic potency: Comparative analyses of trans/cis isomers.

The novel cinnamic acid (CA) (H4-CA, H5-CA, and H7-CA) and caffeic acid (KA) (H4-KA, H5-KA, and H7-KA) hemorphin analogs have recently been synthesized and their trans isomers have been tested for antiseizure and antinociceptive activity. In the present study, the cis forms of these compounds were tested and compared with their trans isomers in seizure and nociception tests in mice. The cis-H5-CA and H7-CA compounds showed efficacy against psychomotor seizures, whereas the trans isomers were ineffective. Both the cis and trans KA isomers were ineffective in the 6-Hz test. Activity of Cinnamic Acid Derivatives with 4-Chloro-2-mercaptobenzenesulfonamide Moiety against Clinical HLAR and VRE Enterococcus spp.

The rapid increase in strains that are resistant to antibiotics requires new active compounds to be found whose mechanism of action on bacteria is different to those that are currently known. Of particular interest are compounds that occur in plants as secondary metabolites. The focus of this study concerns the examination of the effects of synthetic cinnamic acid derivatives, with 4-chloro-2-mercaptobenzenesulfonamide moiety on Enterococcus spp. with HLAR (high-level aminoglycoside resistance) and VRE (vancomycin-resistant Enterococcus) mechanisms. Inhibition of starch digestion by phenolic acids with a cinnamic acid backbone: Structural requirements for the inhibition of α-amylase and α-glucosidase.

This study investigated the inhibition mechanism of cinnamic acid-based phenolic acids (cinnamic acid: CIA, 3,4-dimethoxy cinnamic acid: 3,4-mCIA, caffeic acid: CA, ferulic acid: FA) on starch digestion. CA, FA, and 3,4-mCIA contributed to reducing the rapidly digested starch content and increasing the resistant starch content. The enzyme activity inhibition results responded that the four phenolic acids inhibited α-amylase activity better than α-glucosidase. The order of IC50 against α-amylase and α-glucosidase was CA > FA > 3,4-mCIA > CIA. Phenolic acid's benzene ring formed conjugated Pi-sy

Liver function in protein-energy malnutrition measured by cinnamic acid tolerance and benzoic acid tolerance: effect of carnitine supplementation.

1. Rats fed on a protein-depleted diet for 8 weeks were repleted for 5 weeks on high-protein (HP), high-protein + 20 g DL-carnitine/kg (HP + C), or low-protein + 20 g DL-carnitine/kg (LP + C) diets. At 4 and 8 weeks of depletion, and 1 and 5 weeks of repletion, rats from each treatment group were given a benzoic acid tolerance test (BATT) or a cinnamic acid tolerance test (CATT) as a measure of liver function. 2. BATT and CATT measured the molar percentage of a test dose (1 mmol/kg body-weight) of benzoic acid or cinnamic acid excreted in the urine as hippuric acid within 24 h. Liver weight, l

Cinnamic Acid Attenuates Ethanol-Exacerbated Hepatocyte Pyroptosis in Type 2 Diabetes via AMPK-Mediated NLRP3/GSDMD Inhibition.

Type 2 diabetes mellitus (T2DM) is frequently accompanied by chronic ethanol (ET) consumption, which worsens metabolic dysregulation and organ damage. Cinnamic acid (CA), a natural compound with anti-diabetic and hepatoprotective effects, shows therapeutic potential, but its role in mitigating ET-exacerbated hepatocyte injury in diabetes remains unclear. This study thus investigated whether CA protects against ET-induced hepatocyte pyroptosis under diabetic conditions and explored the underlying mechanisms. In vitro, HepG2 cells cultured under high-glucose (HG) conditions were exposed to ET to

Design, synthesis, and biological evaluation of C-10-esterified dihydroartemisinin-cinnamic acid hybrids as anticancer agents against colorectal cancer.

Dihydroartemisinin (DHA) attracts considerable attention as a promising antineoplastic agent against colorectal cancer. However, its suboptimal antitumor efficacy and its C-10 configurational lability driven by hemiacetal-mediated epimerization collectively impede its clinical development. In this context, a library of 52 DHA-cinnamic acid (DHA-CA) hybrid derivatives with defined C-10 stereochemistry was constructed via stereocomplementary esterification: Mitsunobu conditions selectively afforded 10β-configured esters, whereas HATU-mediated acylation preferentially furnished 10α-isomers. Density Functional Theory, Molecular Docking Study, and In Vitro Antioxidant Activity of Cinnamic Acid Isolated From Piper betle Leaves.

Piper betle is an edible plant known for its potent antioxidant activity. Among its phenolic constituents, cinnamic acid has been identified as a key compound contributing to this bioactivity. Although cinnamic acid is a well-known molecule, this study is the first to report its isolation from P. betle leaves, contributing valuable insights into the chemotaxonomy and phytochemical profile of the species. The aim of this research is to isolate cinnamic acid from the methanol extract of betel leaves and evaluate its antioxidant activity using DPPH and nonenzymatic mimic superoxide dismutase (mSO

Alternative Naphthalene Metabolic Pathway Includes Formation of ortho-Phthalic Acid and Cinnamic Acid Derivatives in the Rhodococcus opacus Strain 3D.

Naphthalene, as a component of crude oil, is a common environmental pollutant. Biochemical and genetic aspects of naphthalene catabolism have been examined in most detail in the bacteria of Pseudomonas genus. In pseudomonads, the key intermediate in naphthalene degradation is salicylate. In this study, we investigated the ability of Rhodococcus opacus strain 3D to utilize naphthalene as a sole carbon and energy source. The characteristic feature of this strain is the inability to grow in the mineral medium supplemented with salicylate (typical intermediate of naphthalene degradation in Gram-ne

Cinnamic acid alleviates hypertensive left ventricular hypertrophy by antagonizing the vasopressor activity and the pro-cardiac hypertrophic signaling of angiotensin II.

BACKGROUND: Hypertension is the most common cause of pathological left ventricular hypertrophy, a condition causally associated with debilitating heart failure and cardiovascular events in hypertensive patients. It is well recognized that the disease burden of hypertension-linked heart failure remains unabated with existing treatments. New therapies controlling hypertensive left ventricular hypertrophy are thus required to decelerate or prevent the development of heart failure. Our previous study has demonstrated that cinnamic acid, a naturally occurring monocarboxylic acid, mitigates transver

Significant Increase of Cinnamic Acid in Metabolites of Chicks Infected with Infectious Bronchitis Virus and Its Remarkable Antiviral Effects In Vitro and In Vivo.

Avian infectious bronchitis virus (IBV) infection has caused significant economic losses to the poultry industry. Unfortunately, there is currently no effective cure for this disease. Understanding the pathogenic mechanism is crucial for the treatment of the disease. Studying the pathogenic mechanism of IBV based on metabolomics analysis is helpful for identifying antiviral drugs. However, studies on metabolomics analysis of IBV infection have been relatively limited, particularly without metabolomics analysis in sera after IBV infection. In this study, 17-day-old SPF chicks were infected with

Identification of Sulfonamide-Vinyl Sulfone/Chalcone and Berberine-Cinnamic Acid Hybrids as Potent DENV and ZIKV NS2B/NS3 Allosteric Inhibitors.

Dengue virus (DENV) and Zika virus (ZIKV) are flaviviruses transmitted by Aedes spp. mosquitoes, causing a spectrum of symptoms ranging from mild fevers and joint pain to severe damage to vital organs, including the kidneys, brain, and liver. Unfortunately, there are currently no specific treatments for these viruses. The NS2B/NS3 serine protease has been recognized as a crucial therapeutic target due to its pivotal role in viral replication. Herein, several molecular modeling techniques were employed to search for novel allosteric inhibitors against DENV and ZIKV NS2B/NS3 proteases from a set
