Combined effects of adrenalectomy and noise exposure on compound action potentials, endocochlear potentials and endolymphatic potassium concentrations.

The effects of removal of endogenous corticosteroids via bilateral adrenalectomy in combination with noise exposure (30 min at 100 dB) were determined by recording compound action potential (CAP) and endocochlear potentials (EP), and by measuring potassium concentrations (K+e) within the endolymph. Thirty-eight Long-Evans rats were divided into groups according to experimental treatments: adrenalectomy (ADX) or non-ADX and noise exposure or non-noise exposure. CAP thresholds, EP and K+e values were subjected to repeated-measures analysis of variance with group and time as factors classifying t

Effects of intra-cochlear perfusion of salicylates on cochlear microphonic and other auditory responses in the guinea pig.

The ototoxic action of salicylate was investigated in the guinea pig by perfusion of both salicylate and bromosalicylate through scala tympani. The results qualitatively confirmed experiments using intravenous administration in cats (Stypulkowski, 1990), showing dose-dependent elevations in compound action potential (CAP) thresholds, increases in cochlear microphonics (CM) and level-dependent reductions in 2f1-f2 acoustic distortion products. The endocochlear potential was not significantly affected and iontophoretic injection of salicylate into scala media had no measurable effect on CAP thre

Modifications of the nonlinearity of the cochlear microphonic responses produced by noise exposure in the guinea pig.

Cochlear microphonics (CM) were recorded with differential electrodes from several locations in the guinea pig cochlea. Input-output curves and amplitude-frequency curves were plotted before and after short exposures to intense noise. In addition to amplitude losses, important changes in the intensity functions resulted in a decrease of the nonlinearity and in a modification of the frequency response. The mechanisms producing these alterations are discussed.

Effects of noise on cochlear potentials and endolymph potassium concentration recorded with potassium-selective electrodes.

Guinea pig cochleas were exposed to either broad-band noise at intensities between 95 and 115 dBA or octave-band noise centered at 380 Hz or 4.2 kHz at intensities between 115 and 125 dB SPL. Cochlear microphonics (CM), summating potentials (SP) and action potentials (AP) were recorded from differential electrodes in the perilymphatic scalae between successive 20-min periods of noise exposure. The endocochlear potential (EP) and endolymph potassium concentration [Kendo+] were recorded continuously from scala media using double-barreled potassium-sensitive electrodes. It was found that the init

[Influence of a CNS pathology on the electrocochleography response].

This study analyzed 73 electrocochleographic recordings made in children with a normal hearing threshold, selected retrospectively from 1563 recordings made between 1973 and 1990. The aim of the study was to check the original findings for any correlation between the various response parameters which might be indicative of a pathological condition. Compound action potential (AP) latency and amplitude, presynaptic summation potential (SP) and cochlear microphonic (CM) amplitudes and AP rapid adaptation behavior were calculated and recordings were associated with clinical information on aetiolog

Comparison of an Implantable Middle Ear Microphone and Conventional External Microphone for Cochlear Implants: A Clinical Feasibility Study.

OBJECTIVES: All commercially available cochlear implant (CI) systems use an external microphone and sound processor; however, external equipment carries lifestyle limitations. Although totally implantable devices using subcutaneous microphones have been developed, these are compromised by problems with soft tissue sound attenuation, feedback, and intrusive body noise. This in vivo pilot study evaluates a middle ear microphone (MEM) that aims to overcome these issues and compares hearing performance with that of an external CI microphone. DESIGN: Six adult participants with an existing CI were

Effects of high-frequency sound on electrochemical potential using the single-barrel method in guinea pigs.

There are only a few studies investigating the effects of ultrasound on hearing. This paper reports on an experimental investigation on the effect of the long-time exposure to a moderate sound pressure level, high-frequency sound of 16 kHz on the guinea pig inner ear using electrophysiological methods. A decrease of the absolute value of negative potential was observed in the endocochlear potential (EP). The effects of high-frequency sound on EP and electrochemical potential could be explained by a change in the K+ permeability of hair cells.

Air- and Bone-Conducted Sources of Feedback With an Active Middle Ear Implant.

OBJECTIVES: Active middle ear implants (AMEI) have been used to treat hearing loss in patients for whom conventional hearing aids are unsuccessful for varied biologic or personal reasons. Several studies have discussed feedback as a potential complication of AMEI usage, though the feedback pathway is not well understood. While reverse propagation of an acoustic signal through the ossicular chain and tympanic membrane constitutes an air-conducted source of feedback, the implanted nature of the device microphone near the mastoid cortex suggests that bone conduction pathways may potentially be an

Parents' experiences of remote microphone systems for children with hearing loss.

PURPOSE: Remote microphone systems improve intelligibility in difficult conditions when the performance of hearing aids/cochlear implants is insufficient. The purpose of this study was to explore parents' experiences with remote microphone systems for their children with hearing loss and to determine the advantages and disadvantages as perceived by parents. MATERIALS AND METHODS: Data were collected by means of semi-structured interviews with parents of children with moderate to profound bilateral hearing loss (2-19 years old). The open coding method and thematic analysis were used. The final

Effects of aromatic solvents on acoustic reflexes mediated by central auditory pathways.

From previous in vivo investigations, it has been shown that toluene can mimic the effects of cholinergic receptor antagonists and may thereby modify the response of protective acoustic reflexes. The current study aimed to define the relative effects of aromatic solvents on the middle ear and inner ear acoustic reflexes. Toward this end, the cochlear microphonic (CMP) elicited with a band noise centered at 4 kHz, and the compound action potential (CAP) elicited with 4-kHz tone pips was measured in rats. Both potentials were recorded before, during, and after triggering the protective reflexes

Does BAPTA leave outer hair cell transduction channels closed?

The calcium chelator BAPTA was iontophoresed into the scala media of the second turn of the guinea pig cochlea. This produced a reduction in low frequency cochlear microphonic (CM) measured in scala media and an elevation of the cochlear action potential (CAP) threshold that lasted for the duration of the experiment. Using two pipettes, one filled with KCl and the other KCl and BAPTA (50, 20 and 5 mM) it was possible to observe the effect of passing current through one electrode while measuring the endolymphatic potential (EP) with the other. The results demonstrated that current passed via th

Experimental congenital cytomegalovirus labyrinthitis and sensorineural hearing loss.

Cytomegalovirus is a leading cause of human congenital viral infection and hearing loss. The pathogenesis of human congenital cytomegalovirus infection is poorly understood. We have developed a reproducible model of congenital cytomegalovirus-induced sensorineural hearing loss in guinea pigs. This report reviews our previously published results and provides additional new information about this model.

[What effect does ligation of the large neck arteries have on cochlear potentials?].

In guinea-pigs the course of summating potentials (SP) is observed. After the potentials have been identified by their typical form, their sensitivity to a reduction of oxygen, damage by streptomycin and adaptation to excessive noise, the quantity of the SP after ligature of the main neck arteries is recorded. After ligature of the external carotid arteries, the internal carotid arteries and the vertebral arteries on both sides, the SP remain active. This is presumably caused by anastomoses between the external carotid and subclavian arteries, the subclavian and spinal arteries, the external c

[Medium-latency acoustically evoked brain potentials used for examination of the auditory pathway (author's transl)].

Potentials of the 10-15 ms latency range evoked by acoustic clicks and Gauss-shaped tone bursts habe been investigated in normal hearing adults, 20 patients with cochlear damages, and 6 cases of temporal lobe processes. Methods and results are compared to those of brain stem audiometry. Mean and standard latency ranges are calculated for the different peaks (Fig. 1). Amplitudes should be used only for side difference evaluation in the same patient, because of their big interindividual variation. In cases of profound high frequency hearing loss (Fig. 2) medium-latency potentials yield true thre

Laser scanning confocal microscopy of the hearing organ: fluorochrome-dependent cellular damage is seen after overexposure.

In order to combine laser confocal microscopy with physiological measurements, a number of conditions have to be met: the dye must not be toxic to the cells the laser light itself must not damage the cells; and the excitation of the fluorochrome during imaging must not generate products with toxic effects. We have investigated these conditions the hearing organ of the guinea pig. Two dyes were used, namely, calcein-AM, which is metabolized in vital cells to a fluorescent product in the cytoplasm, and a lipophilic membrane dye. The effect of the dyes on cell function was tested in the intact he

[Changes in endocochlear potential induced by potassium-channel blockers].

The effect of various potassium-channel blockers, 4-aminopyridine (4-AP), tetraethylammonium (TEA) and quinine, on the endocochlear potential (EP) was studied in perfused guinea pig inner ears. The fast K(+)-channel blocker, 4-AP, did not alter EP but changed its response model to intense noise exposure. While TEA and quinine significantly reduced the amplitude of negative component of EP (N-EP), comparing with a relatively smaller increase in general EP (G-EP). The results indicated the existence of different K(+)-channels with different physiological functions.

Electrocochleographic study of low-tone hearing loss without vertigo.

Auditory nerve action potential (AP), cochlear microphonics (CM) and summating potential (SP) were recorded from 6 patients with low-tone hearing loss without vertigo. All these cases showed high AP and -SP amplitude and satisfactory CM response. These findings resembled the electrocochleographic findings of type 1, which is an early stage of Ménière's disease. Hearing returned to normal range in half of the cases but remained hardly changed in the other half. The difference between these two groups could not be clarified electrocochleographically. On the other hand, low-tone hearing loss due

[Early and late reaction of the guinea pig cochlea to impulse noise].

Guinea pigs (GP) were exposed to 10 impulses of 164 dB (SPL). Measuring of cochlear microphonics (CM) at frequencies between 0.5 and 10 kHz and morphological examination of the cochlea by the surface preparation technique followed 2 hours, 2 and 6 weeks after exposure to impulse noise. During the 2 hours following noise exposure the amplitudes of CM decreased in all tested frequencies, while recovery of CM never could be observed at this time. Subsequent morphological changes in the structure of the organ of Corti could be found. They varied considerably between the tested animals. 2 and 6 wee

Ultrasonic electrocochleography in guinea pig.

In order to obtain information about how ultrasonic stimuli are perceived (USP) in man, guinea pig cochleae were stimulated by bone conduction with frequencies (98.8 and 143.5 kHz) above the normal auditory field of this animal. The cochlear potentials recorded consisted of CM, SP and AP originating from the basal turn of the cochlea, and were found to be influenced by asphyxia, administration of ethacrynic acid, hypothermia and change of interstimulus interval. In addition, in kanamycin-treated animals the mean AP amplitude decreased to about one fourth of the normal value, and the mean AP la
