Indoor allergens and endotoxins in relation to cockroach infestations in low-income urban homes.

BACKGROUND: Cockroach allergens are well recognized as important risk factors in the development and prevalence of allergic rhinitis and asthma in children, especially in low-income urban households. The German cockroach gut hosts a diverse community of highly abundant microbes, including gram-negative bacteria that shed large amounts of endotoxins in cockroach feces. OBJECTIVE: We sought to delineate the causal relationship between the presence of cockroaches in homes and levels of household endotoxins. METHODS: In laboratory assays, we measured the amount of endotoxin produced by cockroach

Screening and identification of antimicrobial peptides from the gut microbiome of cockroach Blattella germanica.

BACKGROUND: The overuse of antibiotics has led to lethal multi-antibiotic-resistant microorganisms around the globe, with restricted availability of novel antibiotics. Compared to conventional antibiotics, evolutionarily originated antimicrobial peptides (AMPs) are promising alternatives to address these issues. The gut microbiome of Blattella germanica represents a previously untapped resource of naturally evolving AMPs for developing antimicrobial agents. RESULTS: Using the in-house designed tool "AMPidentifier," AMP candidates were mined from the gut microbiome of B. germanica, and their a

Low-Cost Approaches in Neuroscience to Teach Machine Learning Using a Cockroach Model.

In an effort to increase access to neuroscience education in underserved communities, we created an educational program that utilizes a simple task to measure place preference of the cockroach (Gromphadorhina portentosa) and the open-source free software, SLEAP Estimates Animal Poses (SLEAP) to quantify behavior. Cockroaches (n = 18) were trained to explore a linear track for 2 min while exposed to either air, vapor, or vapor with nicotine from a port on one side of the linear track over 14 d. The time the animal took to reach the port was measured, along with distance traveled, time spent in

A steroid-resistant cockroach allergen model is associated with lung and cecal microbiome changes.

The pathogenesis of asthma has been partially linked to lung and gut microbiome. We utilized a steroid-resistant chronic model of cockroach antigen-induced (CRA) asthma with corticosteroid (fluticasone) treatment to examine lung and gut microbiome during disease. The pathophysiology assessment demonstrated that mucus and airway hyperresponsiveness were increased in the chronic CRA with no alteration in the fluticasone (Flut)-treated group, demonstrating steroid resistance. Analysis of mRNA from lungs showed no decrease of MUC5AC or Gob5 in the Flut-treated group. Furthermore, flow-cytometry in

Hematological and histopathological evaluation of meat-type quails fed Madagascar cockroach meal.

Aiming at the sustainability of meat production, insects can replace traditional ingredients in the diet of poultry. Studies evaluating performance in birds have emerged to ensure this ability, but few address the health parameters of animals. This study was carried out to evaluate the effects of the inclusion of Madagascar cockroach meal in traditional diets on hematological and histopathological traits of meat-type quails. The inclusion of Madagascar cockroach meal in the diet was evaluated in four levels: 0%, 6%, 12%, and 18%. Observations for hematological and histopathological traits from

Cross-reaction between Formosan termite (Coptotermes formosanus) proteins and cockroach allergens.

Cockroach allergens can lead to serious allergy and asthma symptoms. Termites are evolutionarily related to cockroaches, cohabitate in human dwellings, and represent an increasing pest problem in the United States. The Formosan subterranean termite (Coptotermes formosanus) is one of the most common species in the southern United States. Several assays were used to determine if C. formosanus termite proteins cross-react with cockroach allergens. Expressed sequence tag and genomic sequencing results were searched for homology to cockroach allergens using BLAST 2.2.21 software. Whole termite extr

Persistence of brodifacoum in cockroach and woodlice: implications for secondary poisoning during rodent eradications.

When planning rodent eradications, that normally involve the use of the anticoagulant poison brodifacoum, it is imperative to minimise impacts on other "non-target" species that dwell alongside the targeted rodents and may indeed be the intended beneficiaries of the eradication. Such impacts can arise either from primary poisoning when the non-target species ingest bait pellets containing toxicant or by secondary poisoning when the non-target species eats prey that has itself eaten brodifacoum. Cockroaches and woodlice, likely to scavenge bait pellets, are widely distributed on tropical and su

IgE-binding epitope analysis of Bla g 5, the German cockroach allergen.

Cockroach infestations have been linked with allergic diseases such as asthma in humans. Bla g 5, sigma class glutathione S-transferase (GST), is the major cockroach allergen which has the highest IgE response value of all cockroach allergens. Although several cockroach allergens have been identified and cloned, information regarding their B ell and T cell IgE-binding epitopes is limited. In order to analyze the IgE binding epitopes of Bla g 5, full-length and five peptide fragments (A, 1-100 amino acid residue; B, 91-200; Ba, 1-125; Bb, 1-150; Bc, 1-175) were expressed. Twelve (37.5%) of 32 s

Mediator release assay for assessment of biological potency of German cockroach allergen extracts.

BACKGROUND: Cockroach is an important allergen in inner-city asthma. The diagnosis and treatment of cockroach allergy has been impeded by the lack of standardized cockroach extracts. OBJECTIVE: We investigated the utility of a mediator release assay based on rat basophil leukemia (RBL) cells for comparing the potency of German cockroach extracts. METHODS: RBL cells (line 2H3) transfected with human FcepsilonRI were passively sensitized with sera from subjects with cockroach allergy and stimulated with serial dilutions of 3 commercial cockroach extracts (1:10 weight/volume). Molecular characterization and localization of the first tyramine receptor of the American cockroach (Periplaneta americana).

The phenolamines octopamine and tyramine control, regulate, and modulate many physiological and behavioral processes in invertebrates. Vertebrates possess only small amounts of both substances, and thus, octopamine and tyramine, together with other biogenic amines, are referred to as "trace amines." Biogenic amines evoke cellular responses by activating G-protein-coupled receptors. We have isolated a complementary DNA (cDNA) that encodes a biogenic amine receptor from the American cockroach Periplaneta americana, viz., Peatyr1, which shares high sequence similarity to members of the invertebra

[Cockroach allergy: a study of its prevalence using skin tests with commercial extracts].

Cockroaches have been increasingly recognized as an important source of indoor allergens. In this study we assessed the prevalence of cockroach sensitization among an outpatient population observed at our Department (155 patients with a mean age of 29 +/- 12 years), based on skin "prick" tests with four commercial cockroach extracts along with a common battery of standardized inhalant allergens. We found a positive wheal to at least one of these four extracts in 27 patients: 26 (96.2%) to Blatta orientalis, 10 (37%) both to Blatella germanica and 1 (3.7%) exclusively to Blatella germanica, wit

Purification and some properties of a lectin from the hemolymph of Periplaneta americana (American cockroach).

A lectin showing specificity for human A-type red blood cells was purified to homogeneity from the hemolymph of the American cockroach Periplaneta americana by affinity chromatography on bovine submaxillary gland mucin. This lectin was a huge molecule with molecular mass of about 1500 kDa, with a single subunit of 30-kDa protein, and required Ca2+ for expression of lectin activity. Electron microscopic examination showed that these molecules were rods with helical structure with an average length of 50.5 nm and width of 10 nm. The molecule was suggested to contain tandemly aligned basic units

A boric acid-rodenticide mixture used in the control of coexisting rodent-cockroach infestations.

Rodent bait combined with a commercially available boric acid compound was found in laboratory experiments to be completely effective in eliminating American cockroaches given the mixture as a food source. The presence of boric acid compound did not interfere with the rodenticidal capacity of the rodent bait.

Co-variation of insecticide resistance and host-microbe gene expression in geographically distant populations of German cockroaches (Blattodea: Blattellidae).

Insecticides can greatly limit the impacts of cockroaches on indoor environment and human health. Insecticide resistance, however, reduces the efficacy of insecticides and increases insecticide use by both residents and pest control professionals. This study examined the relationship between insecticide cross-resistance and associated gene expression across nine different German cockroach populations from the eastern U.S. Using vial bioassays, we identified significantly correlated resistance between three insecticides from different classes (indoxacarb, lambda-cyhalothrin, fipronil), with pot

Modulation of octopaminergic and cholinergic pathways induced by Caatinga tree Manilkara rufula chemical compounds in Nauphoeta cinerea cockroaches.

The entomotoxic potential of Manilkara rufula crude extract (CEMR) and its aqueous (AFMR) and methanolic (MFMR) fractions were evaluated against Nauphoeta cinerea cockroaches. The results point out to a direct modulation of octopaminergic and cholinergic pathways in insect nervous system. CEMR induced an anti-acetylcholinesterase (AChE) effect in cockroach brain homogenates. CEMR significantly decreased the cockroach heart rate in semi-isolated heart preparations. CEMR also caused a broad disturbance in the insect behavior by reducing the exploratory activity. The decreased antennae and leg gr

Multidrug-resistant pattern of food borne illness associated bacteria isolated from cockroaches in meal serving facilities, Jimma, Ethiopia.

INTRODUCTION: An increase in the emergence and spread of multidrug-resistant (MDR) bacteria in recent years is becoming worrisome. Domestic cockroaches can play a significant role in the dissemination of such bacteria between the environment and human beings. This study aimed at determining anti-microbial resistance pattern of food borne illness associated bacteria identified from cockroaches trapped in restaurants and cafeterias. METHODS: Trapped cockroaches were picked with surgical gloves, sealed in sterile plastic bags and transported to the Microbiology laboratory. Standard microbiologic

Rare occurrence of Blastocystis in sea turtles and insects (cockroaches, houseflies, and crickets) from several states in Peninsular Malaysia.

Blastocystis a single-celled eukaryotic protist, is known to inhabit the intestines of various hosts, including humans, and has been implicated in a wide spectrum of symptoms, ranging from gastrointestinal issues to skin disorders, thereby establishing its status as an emerging infectious agent. In this study, the prevalence of Blastocystis infection was investigated in insects, including cockroaches, houseflies, and crickets, as well as sea turtles. Additionally, the genotypic characteristics of the isolated Blastocystis strains were examined, and the evolutionary relationships between Blasto

Combined metabolic and target-site resistance mechanisms confer fipronil and deltamethrin resistance in field-collected German cockroaches (Blattodea: Ectobiidae).

Despite insecticide resistance issues, pyrethroids and fipronil have continued to be used extensively to control the German cockroach, Blattella germanica (L.) (Blattodea: Ectobiidae) for more than two decades. We evaluated the physiological insecticide resistance in five German cockroach populations collected from 2018 to 2020 and measured the extent of metabolic detoxification and target-site insensitivity resistance mechanisms. Topically applied doses of the 3 x LD95 of deltamethrin, fipronil, DDT, or dieldrin of a susceptible strain (UCR, Diagnostic Dose) failed to cause >23% mortality, an
