[Cognitive advantage in atypical development: enhanced procedural learning in Tourette syndrome].

Tourette syndrome is a neurodevelopmental disorder that is characterized by motor and vocal tics and by alterations in the cortico-basal ganglia-thalamo-cortical (CBGTC) circuitry. The CBGTC circuitry plays an important role in procedural learning, in the acquisition of skills and habits. Tics and habits are similar phenomenologically since tics can be described as overlearned habits. Based on these characteristics, prior studies proposed enhanced pro - ce dural learning, i.e., procedural hyperfunctioning in Tourette syndrome. A growing body of evidence supports this notion. The focus of the p

Pharmacokinetic and pharmacodynamic of the cognitive enhancer modafinil: Relevant clinical and forensic aspects.

Modafinil is a nonamphetamine nootropic drug with an increasingly therapeutic interest due to its different sites of action and behavioral effects in comparison to cocaine or amphetamine. A review of modafinil (and of its prodrug adrafinil and its R-enantiomer armodafinil) chemical, pharmacokinetic, pharmacodynamic, toxicological, clinical and forensic aspects was performed, aiming to better understand possible health problems associated to its unconscious and unruled use. Modafinil is a racemate metabolized mainly in the liver into its inactive acid and sulfone metabolites, which undergo prim

Non-invasive brain stimulation to enhance cognitive rehabilitation after stroke.

Stroke is a main cause for long-term disability. Stroke symptoms cover various domains, e.g., motor, sensory, language, or other cognitive functions. In clinical practice and rehabilitation research, especially motor impairment attracts much attention. However, also cognitive impairments are common after stroke, effecting approximately two-thirds of stroke patients in the acute phase. Although 30% of stroke patients spontaneously recover from their cognitive impairments, large amounts of patients remain cognitively impaired. These patients have more problems reintegrating in personal and profe

Exercise-enhanced neuroplasticity targeting motor and cognitive circuitry in Parkinson's disease.

Exercise interventions in individuals with Parkinson's disease incorporate goal-based motor skill training to engage cognitive circuitry important in motor learning. With this exercise approach, physical therapy helps with learning through instruction and feedback (reinforcement) and encouragement to perform beyond self-perceived capability. Individuals with Parkinson's disease become more cognitively engaged with the practice and learning of movements and skills that were previously automatic and unconscious. Aerobic exercise, regarded as important for improvement of blood flow and facilitati

Rat prefrontal dopamine and cognitive control: impaired and enhanced conflict performance.

It has been proposed that the cognitive dysfunction observed in patients with schizophrenia reflects a failure in cognitive control, in particular an inability to use task-relevant information to guide behavior. Using a prefrontal-dependent task designed to reflect aspects of cue and response conflict seen in human cognitive paradigms, we examined the influence of prefrontal dopamine receptor manipulation on the contextual control of response conflict. Rats were trained on two biconditional discrimination tasks, one auditory and one visual, in two discriminably different contexts. At test, aud

The effectiveness of enhanced cognitive behavioural therapy for eating disorders: an open trial.

The aim of this study was to examine the effectiveness of Enhanced Cognitive Behaviour Therapy (CBT-E) for eating disorders in an open trial for adults with the full range of eating disorders found in the community. The only previously published trial of CBT-E for eating disorders was a randomised controlled trial (RCT) conducted in the U.K. for patients with a BMI ≥ 17.5. The current study represents the first published trial of CBT-E to include patients with a BMI<17.5. The study involved 125 patients referred to a public outpatient clinic in Perth, Western Australia. Patients attended, on a

Enhanced motor cortex facilitation in patients with vascular cognitive impairment-no dementia.

Data on Transcranial Magnetic Stimulation (TMS) derived measures of cortical excitability and intracortical circuits in age-related white matter changes are scarce. We aimed to assess early changes of motor cortex excitability in nondemented elderly patients with subcortical ischemic vascular disease (SVD). Ten SVD elderly and ten age-matched controls underwent paired-pulse TMS for the analysis of intracortical inhibition (ICI) and facilitation (ICF). All subjects performed neuropsychological assessment and brain magnetic resonance imaging. SVD patients showed abnormal executive control functi

Intensive practice of a cognitive task is associated with enhanced functional integration in schizophrenia.

BACKGROUND: There is increasing evidence that the frequently reported working memory impairments in schizophrenia might be partly due to an alteration in the functional connectivity between task-relevant areas. However, little is known about the functional connectivity patterns in schizophrenia patients during learning processes. In a previous study, Koch et al. [Neuroscience (2007) 146, 1474-1483] have demonstrated stronger exponential activation decreases in schizophrenia patients during overlearning of short-term memory material. The question arises whether these differential temporal patte

Enhanced stimulus strength improves visual cognition in aging and Alzheimer's disease.

Deficits in visual cognition in Alzheimer's disease (AD) arise from neuropathological changes in higher-order association areas of the cortex and from defective input from lower-level visual processing areas. We investigated whether enhanced signal strength may lead to improvement of visual cognition in AD. We tested 35 individuals with probable AD, 35 age-matched elderly control (EC) and 58 young control (YC) adults on letter identification, word reading, picture naming, discrimination of unfamiliar faces, and pattern completion. The contrast sensitivity step-difference across an independent

Drug dependence study of the new cognition-enhancing agent nefiracetam in rats.

The new cognition-enhancing agent nefiracetam (N-(2,6-dimethylphenyl)-2-(2-oxo-1-pyrrolidinyl) acetamide, DM-9384, CAS 77191-36-7) was assessed for dependence liability in rats using the DAF (drug admixed food) method and an intravenous self-administration system. In the physical dependence test, nefiracetam and codeine phosphate were administered to rats mixed with food for 43 days in a gradually increasing dosage schedule, followed by feeding a drug-free normal diet to detect signs of withdrawal. After the withdrawal, rats in the nefiracetam treated groups showed no withdrawal symptoms (e.g.

Enhancing cognitive performance with fronto-parietal transcranial alternating current stimulation.

Executive cognitive functions are essential for human life, and their age- or disease-related decline significantly impacts quality of life. Non-invasive brain stimulation (NIBS) targeting the multiple-demand or fronto-parietal network (FPN) has emerged as a promising intervention for enhancing various executive cognitive functions. However, variations in cognitive tasks and stimulation protocols across studies led to mixed results, rendering findings and conclusions incomparable. In this study, we synchronized oscillations over two main areas of the frontoparietal cognitive network, right dor

Intranasal GHK peptide enhances resilience to cognitive decline in aging mice.

Brain aging and cognitive decline are aspects of growing old. Age-related cognitive impairment entails the early stages of cognitive decline, and is extremely common, affecting millions of older people. Investigation into early cognitive decline as a treatable condition is relevant to a wide range of cognitive impairment conditions, since mild age-related neuropathology increases risk for more severe neuropathology and dementia associated with Alzheimer's Disease. Recent studies suggest that the naturally occurring peptide GHK (glycyl-L-histidyl-L-lysine) in its Cu-bound form, has the potentia

Zunyimycin C enhances immunity and improves cognitive impairment and its mechanism.

This study aimed to explore the efficacy of zunyimycin C in the immunological enhancement of hypoimmune mice and improvement of cognitive impairment in a mice model of Alzheimer's disease (AD). Zunyimycin C was administered intranasally to interfere with AD mouse models or gavage to hypoimmune animals. Results of the Morris water maze (MWM) showed that zunyimycin may improve the learning and memory abilities of the AD mice model. The results of differential expression analysis of mRNA levels of inflammatory factors and pathways in brain tissues of the AD mouse model suggested that differential

Music in premature infants enhances high-level cognitive brain networks.

Neonatal intensive care units are willing to apply environmental enrichment via music for preterm newborns. However, no evidence of an effect of music on preterm brain development has been reported to date. Using resting-state fMRI, we characterized a circuitry of interest consisting of three network modules interconnected by the salience network that displays reduced network coupling in preterm compared with full-term newborns. Interestingly, preterm infants exposed to music in the neonatal intensive care units have significantly increased coupling between brain networks previously shown to b

Sodium valproate enhances doxorubicin-induced cognitive dysfunction in Wistar rats.

BACKGROUND: Increasing number of scientific reports have highlighted the role of histone acetylation/deacetylation in neurodegenerative conditions, including chemotherapy-induced cognitive dysfunction (also known as chemobrain). Multiple sources state that increased activity of histone deacetylases (HDACs) play a detrimental role in chemobrain. In the present study, sodium valproate, a well-known HDAC inhibitor, was explored for its neuroprotective potential against chemobrain development. METHODS: Doxorubicin (DOX), a chemotherapeutic agent, was used to induce chemobrain in experimental anim
