Cohort profile: The Copenhagen Analgesic Study-The COPANA cohort.

BACKGROUND: Development of the gonads during fetal life is complex and vital for adult reproductive health. Cell and animal studies have shown an alarming effect of mild analgesics on germ cells in both males and females. More than 50% of pregnant women use mild analgesics during pregnancy, which potentially could compromise the reproductive health of the next generation. OBJECTIVES: We present a research protocol designed to evaluate the effect of prenatal exposure to mild analgesics and endocrine-disrupting chemicals on gonadal function in the offspring. POPULATION: Healthy, singleton preg

Implementing Immunoscore in colorectal cancer: lessons from cohort studies, limitations and barriers to adoption.

The Immunoscore (IS) quantifies the immune contexture of colorectal cancer (CRC) by measuring CD3+ and CD8+ T-cell densities in the tumour centre and invasive margin, providing superior prognostic performance compared with TNM staging and mismatch-repair (MMR) status alone. Large international cohort studies have validated its ability to predict benefit from adjuvant chemotherapy in stage III colon cancer. More recently, Immunoscore-IC - a refined version that incorporates PD-L1+ cell density and spatial interactions - has emerged as the first biomarker capable of identifying the subset of pat

Large cohort studies assessing sex-specific associations between anthropometric indices and liver cancer death.

BACKGROUND & AIMS: Sex-specific associations between adiposity and death from primary liver cancer (PLC) remain poorly characterized, particularly those related to emerging anthropometric indices and populations with high abdominal adiposity. METHODS: In two prospective cohort studies, the sex-specific associations between 16 anthropometric indices (six traditional, 10 emerging) and PLC death were examined in 72,691 women and 59,892 men in China. Cox proportional hazards regression models with restricted cubic spline functions were applied to evaluate the associations between adiposity indice

Association between handgrip strength and incident depression: a meta-analysis of prospective cohort studies.

A number of individual studies have suggested that higher handgrip strength is associated with a lower risk of depressive symptoms but the totality of the evidence remains inconclusive. This study aimed to investigate the association between handgrip strength and the incidence of depressive symptoms or depression and explore potential moderators of this association. We searched major databases (PubMed, Web of Science, PsycINFO, EMBASE, and SportDiscus) from inception to February 4, 2025, for prospective cohort studies evaluating the risk of incident depressive outcomes according to handgrip st

Outcomes and treatment patterns in germline BRCA1/2 carriers from two matched cohort studies.

BACKGROUND: Whether outcomes of germline BRCA1/2 (gBRCA1/2)-associated breast cancer differ compared with sporadic tumors is controversial. We explored the impact of gBRCA1/2 pathogenic variant (PV) status beyond established prognostic features. METHODS: We conducted 2 retrospective, matched cohort studies comparing gBRCA1/2 PV carriers and noncarriers with HER2-negative, stage I-III breast cancer (Clinical Outcomes Quality Database [COQD] cohort: 185 carriers, 555 noncarriers; Young Women's Breast Cancer Study [YWS] cohort: 113 carriers, 226 noncarriers). Chronic pain and risk of cognitive impairment: a meta-analysis of longitudinal cohort studies.

Chronic pain is increasingly recognized as a potential risk factor for cognitive decline, yet findings from observational studies are inconsistent. We conducted a meta-analysis to evaluate the long-term association between chronic pain and cognitive impairment. PubMed, Embase, and the Cochrane Library were searched from inception to January 2025 for longitudinal cohort studies assessing this relationship. Twenty-eight eligible cohorts comprising 7,914,407 participants were included. Adjusted odds ratios (ORs) were pooled using random-effects models; subgroup, sensitivity, and meta-regression a

Z-Drug Use and All-Cause Mortality: A Meta-Analysis of Observational Cohort Studies.

INTRODUCTION: Non-benzodiazepine hypnotic drugs known as z-drugs (including zolpidem, eszopiclone, zopiclone, zaleplon) are commonly prescribed for insomnia patients worldwide. Z-drugs have been thought to be safer than benzodiazepines; however, several studies have raised controversy over the side effects of z-drug. This meta-analysis was conducted to evaluate the association between z-drugs and all-cause mortality. METHODS: To evaluate the association between z-drugs and all-cause mortality, we conducted a meta-analysis of observational cohort studies identified through PubMed, Embase, and

Vasectomy and prostate cancer risk: a pooled of cohort studies and Mendelian randomization analysis.

BACKGROUND: The relationship between vasectomy and the risk of prostate cancer (PCa) remains unclear, with observational studies reporting inconsistent results. To clarify this ambiguity, we embarked on a comprehensive investigation comprising both a meta-analysis and a Mendelian randomization (MR) study. This dual approach aimed to thoroughly examine not only the association but also the causality between undergoing a vasectomy and the subsequent risk of PCa. METHODS: Our systematic review meticulously examined cohort studies published until January 2024, employing a random effects model for

Associations Between Adherence to the Mediterranean Diet and Incident Sarcopenia in Prospective Cohort Studies.

The loss of skeletal muscle mass and strength, known as sarcopenia, is prevalent in older adults and linked to an increased risk of disability, frailty, and early mortality. Muscle health is crucial for the functionality and independence of older adults. As the aging population continuously grows, finding cost-effective strategies for preventing and treating sarcopenia is an important public health priority. While nutrition is recognized as a key factor in the development of sarcopenia, its role in preventing and treating the condition is still under investigation. In recent decades, nutrition
