Functional characterization of VaMAPK9 in enhancing cold tolerance of Vitis amurensis.

Mitogen-activated protein kinases (MAPKs) play pivotal roles in response to low-temperature stress, but their functional characterization under low-temperature stress in Vitis amurensis (V. amurensis) remains elusive. This study identified eleven MAPK genes through sequence analysis in V. amurensis. Phylogenetic tree analysis revealed that VaMAPK9 shares the closest evolutionary relationships with AtMAPK18 and OsMAPK9. Transcriptional profiling demonstrated significant upregulation of VaMAPK9 in 'Zuoshan 1' under low-temperature stress. Subcellular localization analysis confirmed that the VaMA

Phytohormone-Assisted Bioprocess Engineering for Enhanced Omega Fatty Acid Production in Marine Thraustochytrids Under Cold Stress Conditions.

Microbial platforms are now recognized as sustainable sources of long-chain polyunsaturated fatty acids (PUFAs). This study presents a novel bioprocessing approach that integrates abiotic stress with exogenous phytohormones, indole-3-acetic acid (IAA), salicylic acid (SA), and abscisic acid (ABA) to enhance the biosynthesis of PUFAs in marine Thraustochytrium sp. BM2. While individual stress strategies are known to either enhance lipid accumulation or modulate oxidative responses. The combined effect of phytohormones and multiple treatments of cold stress conditions (4 °C) enhanced the PUFAs f

Genome-wide identification of WRKY gene family reveals its regulatory role in ABA/CBF-mediated cold adaptation in king grass (Pennisetum purpureum × P. americanum).

BACKGROUND: King grass is a fast-growing C4 perennial with high biomass potential, yet its productivity is limited by cold stress. WRKY transcription factors (TFs) regulate abiotic stress responses, but their roles in king grass cold adaptation remain unclear. METHODS: WRKY TFs were identified genome-wide using BLASTP and HMMER, followed by phylogenetic classification, transcriptome profiling, promoter cis-element analysis, weighted gene co-expression network analysis (WGCNA), molecular docking, and yeast heterologous expression assays. RESULTS: Sixty-two PsiWRKY genes were identified and cl

Txnip deficiency causes a susceptibility to acute cold stress with brown fat dysfunction in mice.

Mammals adaptively regulate energy metabolism in response to environmental changes such as starvation and cold circumstances. Thioredoxin-interacting protein (Txnip), known as a redox regulator, serves as a nutrient sensor regulating energy homeostasis. Txnip is essential for mice to adapt to starvation, but its role in adapting to cold circumstances remains unclear. Here, we identified Txnip as a pivotal factor for maintaining nonshivering thermogenesis in mice. Txnip protein levels in brown adipose tissue (BAT) were upregulated by the acute cold exposure. Txnip-deficient (Txnip-/-) mice accl

Interaction of R2R3-MYB transcription factor EgMYB111 with ABA receptors enhances cold tolerance in oil palm.

The oil palm is a prominent tropical oil crop with holds considerable economic value. MYB transcription factors are key regulators in growth and plant stress adaptation mechanisms in plants. However, the roles and operational mechanisms of MYB genes in oil palm are not yet well understood. In this study, EgMYB111 was cloned from oil palm, and its behavior under cold stress was examined in genetically engineered tobacco and oil palm embryoids. Physiological and biochemical analysis demonstrated that genetically engineered lines exhibited substantially greater cold tolerance than control plants.

Trans-Ferulic Acid-4-β-Glucoside Alleviates Cold-Induced Oxidative Stress and Promotes Cold Tolerance.

Trans-ferulic acid-4-β-glucoside (C16H20O₉, TFA-4β-G) is a monomer extracted from the Chinese medicine called radix aconiti carmichaeli (Fuzi). To date, research on this substance is lacking. Here, we found that trans-ferulic acid-4-β-glucoside effectively promoted cold acclimatization in mice via increased heat production and alleviation of oxidative stress in a cold environment. Thus, our work indicates that ferulic acid-4-β-glucoside is a potential therapeutic candidate for prevention and treatment of cold stress injury.

Nitric oxide reduces freezing stress damage on leaf pigments, photosynthetic apparatus, and chlorophyll fluorescence of broad bean (Vicia faba L.).

Freezing stress is one of the abiotic stresses limiting the growth and development of fall crops, which are an important part of sustainable food production worldwide. To investigate the effect of foliar spraying of nitric oxide (NO) on reducing freezing stress damage in broad bean plants, an experiment was conducted under a controlled environment in 2022. NO at three levels (control [distilled water], 50, and 100 µM) and freezing stress at five levels (0, -4, -8, -12, and -16 °C; 2 h) were considered as the experimental treatments. The results showed that freezing stress reduced plant surviva

The effects of acute cold exposure on morphology and gene expression in the heart of neonatal chicks.

Cold exposure induces an increase in blood flow and blood pressure, and long-term exposure to cold causes cardiac hypertrophy. Neonatal chicks (Gallus gallus domesticus) are highly sensitive to cold exposure, because their capacity for thermogenesis is immature until 1 week after hatching. Hence, we hypothesized that the heart of chicks at around 1 week of age acutely responds to cold environment. To investigate the effect of acute (24 h) and long-term (2 weeks) cold on the heart of chicks, 7-day-old chicks were exposed to cold temperature (4 °C) or kept warm (30 °C). Chicks exposed to the col

Deciphering the Cold Adaptive Mechanisms in Pseudomonas psychrophila MTCC12324 Isolated from the Arctic at 79° N.

Psychrophiles, host of cold environments, have been successfully undergoing the process of evolution by which they have acquired innate adaptations to withstand the unfavorable effects of low temperature. Psychrophiles renders immense opportunity to explore the underlying mechanisms of cold adaptation. The present study focused to explore the cold adaptive mechanisms of Pseudomonas psychrophila MTCC12324, a facultative psychrophilic bacterium isolated from the Ny-Alesund, an island in the Svalbard Archipelago (79°55' N, 11°56' E) in the Arctic. Cold-shock proteome of myoblasts reveals role of RBM3 in promotion of mitochondrial metabolism and myoblast differentiation.

Adaptation to hypothermia is important for skeletal muscle cells under physiological stress and is used for therapeutic hypothermia (mild hypothermia at 32  °C). We show that hypothermic preconditioning at 32  °C for 72 hours improves the differentiation of skeletal muscle myoblasts using both C2C12 and primary myoblasts isolated from 3 month and 18-month-old mice. We analyzed the cold-shock proteome of myoblasts exposed to hypothermia (32  °C for 6 and 48 h) and identified significant changes in pathways related to RNA processing and central carbon, fatty acid, and redox metabolism. The analy

O-GlcNAcylation of SIRT1 Protects against Cold Stress-Induced Skeletal Muscle Damage via Amelioration of Mitochondrial Homeostasis.

Cold stress disturbs cellular metabolic and energy homeostasis, which is one of the causes of stress-induced illnesses. O-GlcNAcylation is a nutrient-sensing pathway involved in a myriad of cellular processes. It plays a key role in metabolic homeostasis. Nevertheless, a specific sensing mechanism linking skeletal muscle to O-GlcNAcylation in cold stress is unknown. In this study, O-GlcNAcylation of SIRT1 was targeted to explore the mechanism of skeletal muscle adaptation to cold stress. Ogt mKO aggravated skeletal muscle fibrosis induced by cold stress. At the same time, Ogt gene deletion acc

Recovering body temperature from acute cold stress is associated with delayed proinflammatory cytokine production in vivo.

A poor outcome of whole-body hypothermia often results from a late complication, rather than from acute effects of hypothermia. A low body (cell) temperature or the increase in the concentrations of the stress hormones cortisol, epinephrine, and norepinephrine in response to acute cold stress have been proposed as potent proinflammatory cytokine suppressant. In the current study, we tested the hypothesis that the recovery of body temperature from a whole-body intermittent cold-water immersion (CWI, at 13-14 °C for a total 170 min) is associated with a delayed response of proinflammatory cytoki

Altered growth, pigmentation, and antimicrobial susceptibility properties of Staphylococcus aureus due to loss of the major cold shock gene cspB.

An insertional mutation made in the major cold shock gene cspB in Staphylococcus aureus strain COL, a methicillin-resistant clinical isolate, yielded a mutant that displayed a reduced capacity to respond to cold shock and many phenotypic characteristics of S. aureus small-colony variants: a growth defect at 37 degrees C, a reduction in pigmentation, and altered levels of susceptibility to many antimicrobials. In particular, a cspB null mutant displayed increased resistance to aminoglycosides, trimethoprim-sulfamethoxazole, and paraquat and increased susceptibility to daptomycin, teicoplanin, a

ZNF334 truncation mutation drives cold-induced autoinflammation.

The role of zinc finger protein 334 (ZNF334) in immunological processes remains unknown. We identified a ZNF334 truncation mutation (p.Thr399fs) in a rare case of late-onset cold-induced autoinflammatory disease with elevated TNF-α, IL-1β, IL-6, and extracellular heat shock protein 90 (eHsp90) plasma levels and progressive sensorineural hearing loss. Using patient-derived monocytes and CRISPR/Cas9-edited THP-1 monocytes with a ZNF334 truncation mutation, we discovered that the mutation reduced the interaction between ZNF334 and Hsp90, diminished the endogenous levels of the cold stress regulat

Reduced cerebral perfusion on sudden immersion in ice water: a possible cause of drowning.

INTRODUCTION: Near-drowning incidents and drowning deaths after accidental immersion in open waters have been linked to cold shock response. It consists of inspiratory gasps, hyperventilation, tachycardia, and hypertension in the first 2-3 min of cold-water immersion. This study explored the immediate changes in cerebral blood flow velocity (Vmean) during cold-water immersion since cold shock induced hyperventilation may diminish Vmean and lead to syncope and drowning. METHODS: There were 13 male volunteers who were lowered into a 0 degrees C immersion tank for 30 s. Vmean in the middle cereb

Cold-induced heat shock protein expression in rat aorta and brown adipose tissue.

Recent reports indicate that Heat Shock Proteins (HSPs) are induced in mammalian tissues as part of a homeostatic response to environmental stressors. Administration of sympathomimetic drugs and neuroendocrine stress hormones has been shown to evoke an HSP response in unstressed animals indicating that cell signaling events exists that couple specific neurotransmitter/hormone-receptor interactions with HSP expression in mammalian tissues. Herein, we demonstrate that exposure of rats to a cold ambient temperature (6 degrees C) results in increased expression of constitutive and inducible member

Cold-responsive interaction between MdRAD23D1 and MdMYB15 confers cold stress tolerance via the CBF pathway in apple (Malus domestica).

Low temperature is a major environmental factor that impairs plant growth and development, posing a significant threat to crop yield and quality. RAD23 (RADIATION SENSITIVE23) proteins belong to the UBL-UBA (Uiquitin-like-ubiquitin associated) family and function as shuttle factors in the UPS (ubiquitin proteasome system). Although UBL-UBA proteins are known regulators of plant stress responses, the function and mechanism of RAD23 in apple under cold stress are poorly understood. Here, we demonstrated that MdRAD23D1 is induced by 4 °C and positively regulates cold tolerance. Silencing MdRAD23D

Physiological and transcriptomic analyses reveal HuBBX1 activating the transcription of COR genes to enhance pitaya cold tolerance.

Low-temperature stress (LTS) inhibits the growth and yield of pitaya. HuBBX1, a cold-tolerance transcription factor, was identified based on the physiological and transcriptomic analyses between two pitaya cultivars. HuBBX1 enhances plant cold tolerance via the activation of cold-regulated (COR) genes. Low-temperature stress (LTS) affects fruit quality and yields. Pitaya plants are sensitive to LTS; however, the molecular mechanisms underlying LTS in pitaya remain poorly understood. In this study, the chlorophyll fluorescence, photosynthetic system, cold tolerance-associated physiological inde

CpCBF6-Mediated Activation of Antioxidant Defense and Osmotic Adjustment Enhanced Cold Tolerance in Cerasus pseudocerasus Lindl.

Low-temperature stress restricts plant growth, development, and geographical distribution. The C-repeat binding factor (CBF) is a core transcriptional regulator in plant cold stress responses, but its role in the thermophilic "Manaohong" cherry (Cerasus pseudocerasus Lindl.) remains elusive. Here, a cold-induced transcription factor gene, CpCBF6, was isolated and characterized from this species. Phylogenetic analysis showed its protein shares the highest homology with orthologs from Prunus persica and Prunus dulcis in Rosaceae. CpCBF6 localized to the nucleus, and its transactivation function
