Isolation and Genomic Characterization of a Lytic Bacteriophage Against Multidrug-Resistant E. coli.

Antimicrobial resistance (AMR) has become a major concern in the treatment of bacterial infections, and bacteriophage therapy has emerged as a promising alternative to antibiotics. Bacteriophages are highly specific to their bacterial hosts; hence, isolating phages indigenous to a specific region offers a significant advantage against various pathogen strains. We have isolated a cocktail of bacteriophages against pathogenic E. coli from sewage water at a primary healthcare centre. Characterisation of the isolated phages demonstrated their stability across a broad pH and temperature range, stro

Systematic exploration of Escherichia coli phage-host interactions with the BASEL phage collection.

Bacteriophages, the viruses infecting bacteria, hold great potential for the treatment of multidrug-resistant bacterial infections and other applications due to their unparalleled diversity and recent breakthroughs in their genetic engineering. However, fundamental knowledge of the molecular mechanisms underlying phage-host interactions is mostly confined to a few traditional model systems and did not keep pace with the recent massive expansion of the field. The true potential of molecular biology encoded by these viruses has therefore remained largely untapped, and phages for therapy or other

Cloning of genes determining the production of vero cytotoxin by Escherichia coli.

Sequences encoding the production of a cytotoxin (VT) active on Vero cells were cloned in Escherichia coli K12 from a VT-determining phage that originated in E. coli strain H19 of serotype O26.H11. Subcloning resulted in the identification of a 2.5 kb fragment that still coded for VT production. Mutagenesis with transposon Tn1000 was used to map VT sequences and a 0.75 kb probe was developed. In colony hybridization tests with strains isolated from patients with haemolytic uraemic syndrome or diarrhoea, this probe derived from the H19 VT genes detected only some of the VT+ strains belonging to

The replication origin of pSC101: the nucleotide sequence and replication functions of the ori region.

The nucleotide sequence of a 770-bp ori region of plasmid pSC101 is presented. The sequence shows homologies to some parts of Escherichia coli oriC and phage G4 ori. Several other features are an 80-bp A + T-rich region overlapping a part of the region homologous to oriC, three direct repeats of an 18-bp sequence adjacent to the A + T-rich region, a typical promoter sequence just upstream of the longest open reading frame (ORF) and a long inverted repeat sequence overlapping the putative promoter region. Analysis of successive deletions by BAL31 exonuclease demonstrated that one of the regions

Location of the regulatory site for establishment of repression by bacteriophage lambda.

During the lysogenic response to infection by bacteriophage lambda, the phage-specified cII and cIII proteins provide for the coordinate establishment of repression and integration of the viral DNA. One critical regulatory function of cII/cIII is an activation of synthesis of the cI protein, the repressor that maintains lysogeny. The mechanism and site for regulation of the cI gene by cII/cIII have been a subject of controversy. The two principal hypotheses for cII/cIII action are: initiation of new RNA chains in the y region of lambda DNA just to the left of the cII gene; or antitermination o

Indirect potable water reuse to face drought events in Barcelona city. Setting a monitoring procedure to protect aquatic ecosystems and to ensure a safe drinking water supply.

The climate change and increasing anthropogenic pressures are expected to limit the availability of water resources. Hence, active measures must be planned in vulnerable regions to ensure a sustainable water supply and minimize environmental impacts. A pilot test was carried out in the Llobregat River (NE Spain) aiming to provide a useful procedure to cope with severe droughts through indirect water reuse. Reclaimed water was used to restore the minimum flow of the lower Llobregat River, ensuring a suitable water supply downstream for Barcelona. A monitoring was performed to assess chemical an

Elucidation of fecal inputs into the River Tagus catchment (Portugal) using source-specific mitochondrial DNA, HAdV, and phage markers.

Determining the source of fecal contamination in a water body is important for the application of appropriate remediation measures. However, it has been suggested in the extant literature that this can best be achieved using a 'toolbox' of molecular- and culture-based methods. In response, this study deployed three indicators (Escherichia coli (EC), intestinal enterococci (IE) and somatic coliphages (SC)), one culture-dependent human marker (Bacteroides (GB-124) bacteriophage) and five culture-independent markers (human adenovirus (HAdV), human (HMMit), cattle (CWMit), pig (PGMit) and poultry

Pathogen infection risk to recreational water users, associated with surface waters impacted by de facto and indirect potable reuse activities.

Water deficit, exacerbated by global population increases and climate change, necessitates the investigation of alternative non-traditional water sources to augment existing supplies. Indirect potable reuse (IPR) represents a promising alternative water source in water-stressed regions. Of high concern is the presence of pathogenic microorganisms in wastewater, such as enteric viruses, protozoa and bacteria. Therefore, a greater understanding of the potential impact to human health is required. The aim of this research was to use a quantitative microbial risk assessment (QMRA) approach to calc

Characterization of the New Bacteriophage Ec1-7 and Its Antibacterial Efficacy in Decontamination of Food Products and Stainless-Steel Surfaces.

Escherichia phage Ec1-7 was characterized and its efficacy on food products, as well as on stainless-steel surfaces, was evaluated. The bacteriophage with high adsorption rate and short latency period remained stable when exposed to various aggressive factors and demonstrated lytic activity against STEC strains. In addition, the phage did not contain antibiotic resistance, virulence, and toxin genes. In 24 h after application of the bacteriophage, E. coli concentration on lettuce leaves and on stainless-steel surfaces decreased by 98.2 and 90.5%, respectively. In the experiment with artificial

Prevention and treatment of infectious diarrhea. Speculations on the next 10 years.

The next decade should explode with exciting schemes and novel agents for the prevention and treatment of infectious diarrhea. The development of oral, nonabsorbed antibiotics will continue, but new antidiarrheal drugs, such as gastrointestinal hormone analogues and alpha-adrenergic agonists, will be added to our therapeutic armamentarium. Improved oral rehydration solutions, such as glycine in electrolyte solution, promise to revolutionize the management of diarrhea by diminishing diarrheal stool volume to the point where losses are too small to be clinically relevant. Infant formulas and adu

A cell-free assay measuring repair DNA synthesis in human fibroblasts.

Osmotic disruption of confluent cultured human fibroblasts that have been irradiated or exposed to chemical carcinogens allows the specific measurement of repair DNA synthesis using dTTP as a precursor. Fibroblasts similarly prepared from various xeroderma pigmentosum cell lines show the deficiencies of UV-induced DNA synthesis predicted from in vivo studies, while giving normal responses to methyl methanesulfonate. A pyrimidine-dimer-specific enzyme, T4 endonuclease V, stimulated the rate of UV-induced repair synthesis with normal and xeroderma pigmentosum cell lines. This system should prove

Ineffectiveness of rifampicin in inhibiting RNA synthesis in Escherichia coli and T(4)-infected Escherichia coli cells after exposure to ultraviolet radiation.

Resdiual RNA synthesis in Escherichia coli cells and T(4)-infected E. coli cells exposed to ultraviolet radiation is insensitive to rifampicin. On the other hand, residual RNA synthesis in these cells after gamma irradiation is further inhibited by rifampicin. The specific effect exerted by ultraviolet irradiation on transcription seems to arise from alterations in the DNA structure rather than in bacterial RNA polymerase since RNA synthesis in ultraviolet-irradiated E. coli cells infected with unirradiated T(4) is sensitive to rifampicin.

Immunosuppressive effects of macrophage antiserum.

Heterologous anti-macrophage serum (MAS) specifically destroys or damages macrophages in vitro. The treatment of mice by MAS results in various immunosuppressive effects: the latter are studied using phage ΦX174 and two pathogenic viruses (EMC virus and Yellow fever virus). It is shown that these effects depend both on the infectious capacity and on the dose of the particulate antigens injected. In the case of phage, MAS greatly inhibits the secondary response obtained by performing the antigenic challenge after a primary stimulation which is itself too weak for producing antibodies. In the ca

Rapidly measured indicators of recreational water quality and swimming-associated illness at marine beaches: a prospective cohort study.

INTRODUCTION: In the United States and elsewhere, recreational water quality is monitored for fecal indicator bacteria to help prevent swimming-associated illnesses. Standard methods to measure these bacteria take at least 24 hours to obtain results. Molecular approaches such as quantitative polymerase chain reaction (qPCR) can estimate these bacteria faster, in under 3 hours. Previously, we demonstrated that measurements of the fecal indicator bacteria Enterococcus using qPCR were associated with gastrointestinal (GI) illness among swimmers at freshwater beaches. In this paper, we report on r

Isolation and Characterization of a Lytic Phage vB_EcoS_GZMU_E2010: A Promising Candidate against Clinical Multidrug-Resistant Escherichia coli.

The global escalation of bacterial antimicrobial resistance, particularly in Escherichia coli, constitutes an urgent public health threat. Bacteriophages represent a promising therapeutic arsenal against multidrug-resistant pathogens, underscoring the necessity of acquiring additional phages to expand phage libraries. In response to multidrug-resistant E. coli, this study isolated a lytic phage, designated E. coli phage vB_EcoS_GZMU_E2010. This phage demonstrated effective lytic activity against pathogenic E. coli strains, that included Extended-Spectrum β-Lactamase-producing E. coli (ESBL-E.

From one host to another: adaptation of a phage infecting avian pathogenic Escherichia coli to the laboratory strain K-12.

Avian colibacillosis is an infectious disease that causes significant economic losses worldwide in the poultry industry. The bacterium avian pathogenic Escherichia coli (APEC) is the etiological agent of the disease and is primarily treated with antibiotics. Unfortunately, due to antibiotic resistance in APEC, therapeutic options for treating this disease are becoming increasingly limited. Bacteriophages-viruses that infect bacteria-represent promising therapeutic candidates; however, understanding how these viruses adapt is essential before considering them for commercial applications. This s

Development and clinical validation of a CRISPR/Cas9-engineered reporter phage cocktail for rapid detection of Escherichia coli in urine.

Urinary tract infections are one of the most common infectious diseases, with Escherichia coli as the predominant pathogen. Traditional diagnostic methods fail to meet clinical demands for rapid and specific detection. Here, we developed an efficient urine E. coli detection strategy via a reporter phage cocktail. Four reporter phages (T2::Nluc, T4::Nluc, T5::Nluc, T6::Nluc) were constructed by the CRISPR/Cas9 system combined with homologous recombination. One-step growth curves, optimal multiplicity of infection, and lytic efficiency showed that the Nluc gene block insertion exerted heterogene

Evaluation of a phage-inclusive multimodal strategy for recurrent urinary tract infections.

Urinary tract infections (UTIs) are among the most common bacterial infections worldwide and are associated with high recurrence rates and substantial morbidity. New therapeutic approaches are urgently needed, especially for recurrent UTIs (rUTIs), among which phage therapy represents a promising option. Here, we systematically evaluated the performance of six E. coli phages under urinary tract-relevant conditions and established a quantitative area-under-the-curve (AUC)-based framework for the rational selection of therapeutic phages. Phage lysis behavior was tested under oxic and anoxic cond
