The cannabinoid CB1 receptor antagonist SR141716A reduces appetitive and consummatory responses for food.

RATIONALE: The CB1 receptor antagonist SR141716A reduces food intake in rats. This effect is likely to depend on modulation of reward related processes. OBJECTIVE: To investigate the effects of SR141716A on responding for food under a second order instrumental task in which responding and consumption of food can be separated, and on Pavlovian responding for a stimulus predictive of food reward. METHODS: Instrumental responding and pellet consumption following administration of SR141716A (0-3 mg/kg) were recorded under an FI5 min FR5(5:S) operant schedule that incorporates both a 5 min initia

BP101 Peptide Promotes Female Sexual Receptivity in the Rat.

INTRODUCTION: Low sexual desire is a frequent sexual problem in women, with only one drug for the condition approved by the Food and Drug Administration. AIM: To evaluate the ability of a novel synthetic peptide, BP101, to facilitate sexual behavior after intranasal administration or infusion into certain brain areas in female rats. METHODS: Bilaterally ovariectomized female rats, primed with a suboptimal combination of estradiol benzoate (EB) and progesterone, were used as a model of low sexual motivation. Sexual behavior was tested with stud male rats after acute (experiment 1) or long-ter

Stimulation of mu opioid, but not GABAergic, receptors of the lateral habenula alters free feeding in rats.

Overconsumption, or eating beyond the point of homeostasis, is a key feature in the development of obesity. Although people are consuming beyond the point of homeostasis, they are not consuming constantly or indefinitely. Thus, there is likely a mechanism that acts to terminate periods of food intake at some point beyond satiation and prior to aversion, or the negative effects of extreme excess (nausea, bloating, etc.). The purpose of the present study was to assess the lateral habenula as a candidate region for such a mechanism, due to its connectivity to midbrain reward circuitry, sensitivit

Neuronal activity in female rat preoptic area associated with sexually motivated behavior.

Single unit activities were recorded from 31 neurons in the preoptic area (POA) of female rats engaging in sexual interactions. Concurrent videotape recordings were used to establish a relationship between neuronal activity and particular behavioral events. In 14 of the 31 neurons, the firing rate changed in association with bouts of sexual activity. The remaining 17 fired with more variability regardless of episodes of sexual interactions. Peri-event histograms identified four types of neurons: type 1 (n=4) increased their firing rate when the female rats initiated proceptive behavior; type 2

Activation of dopamine D4 receptors in the nucleus accumbens shell increases motivation for palatable food and homeostatic feeding, whereas systemic blockade reduces feeding behavior in rats.

OBJECTIVE: The nucleus accumbens shell is a key region involved in motivation and reinforcement behavior associated with the consumption of palatable foods. However, it has not been studied whether D4 dopaminergic receptors regulate homeostatic feeding and reward-dependent behaviors. The objective of this study was to evaluate the effects of activation of D4 dopaminergic receptors within nucleus accumbens shell and systemic D4 blockade on motivation for palatable food and homeostatic feeding in male Wistar rats. METHODS: Adult male Wistar rats were tested in operant conditioning paradigms usi

A stress-activated mid-insula to BNST pathway regulates susceptibility to abstinence-induced negative affect in female mice.

Stress is central to many neuropsychiatric conditions, including alcohol use disorder (AUD). Stress influences the initiation and continued use of alcohol, the progression to AUD, and relapse. Identifying the neurocircuits activated during stress, and individual variability in these responses is critical for developing new treatment targets for AUD, particularly to mitigate stress-induced relapse. Using a longitudinal approach, this study examined the relationship between sub-chronic stress exposure and negative affect during protracted abstinence following chronic ethanol exposure. Sub-chroni

Acute caffeine reverses the disruptive effects of chronic fluoxetine on the sexual behavior of female and male rats.

RATIONALE: Sexual side effects of chronic treatment with selective serotonin reuptake inhibitors (SSRIs) in humans include anorgasmia and loss of sexual desire and/or arousal which interferes with treatment compliance. There are few options at present to reduce these effects. Because orgasm and desire are mediated in part by activation of sympathetic arousal, we asked whether the sympathomimetic effects of acute caffeine treatment could reverse these effects. OBJECTIVE: The present study examined whether acute treatment with caffeine (CAF; 10 or 20 mg/kg, ip) versus vehicle could ameliorate t

Familiality of addiction and its developmental mechanisms in girls.

BACKGROUND: Drug use disorders (DUD) have been theorized to share sources of risk variation with other consummatory behaviors. We hypothesized that common mechanisms exist for familial risk for DUD, physiological maturation and nutritional status in girls. Whereas body fat content must exceed a threshold to enable adrenarche and gonadarche, nutritional status may also be a behavior risk indicator. Impaired psychological self-regulation associated with DUD risk may manifest in early overeating, which could in turn accelerate reproductive maturation, resulting in a greater likelihood of affiliat

Vaginocervical stimulation induces Fos in glutamate neurons in the ventromedial hypothalamus: attenuation by estrogen and progesterone.

Vaginocervical stimulation (VCS) induces the immediate-early gene product Fos in the ventromedial hypothalamus (VMH) of female rats. However, this induction is lower in ovariectomized rats that receive estradiol benzoate (EB) and progesterone (P) relative to an oil vehicle. We have observed that a substantial proportion of cells activated in the VMH by VCS stain for glutamate, and infusions of glutamate or its selective receptor agonists to the VMH inhibit both appetitive and consummatory sexual behaviors in females. This raises the possibility that VCS activates an inhibitory glutamate system

Exploring risky decision-making dynamics during antidepressant treatment in major depressive disorder: a computational modeling approach.

BACKGROUND: Patients with major depressive disorder (MDD) show greater loss sensitivity and higher delayed discounting rates during decision making, but findings regarding their risk preferences are inconsistent. Computational behavioral modeling provides a promising tool for the investigation of these latent factors underlying the decision-making behaviors. OBJECTIVES: Conduct both cross-sectional and longitudinal analyses to identify patterns and changes in risky decision-making over time, examine correlations between traditional CGT performance indicators and cognitive modeling parameters

From pleasure to punding: Distinct patterns of anhedonia and impulsivity linked to motivational disturbances in Parkinson disease.

INTRODUCTION: Apathy and impulse control disorders (ICD) are common comorbid motivational syndromes in Parkinson disease (PD). This study aimed to determine if patients with these motivational disturbances exhibit different patterns of anhedonia and trait impulsivity. METHODS: Sixty-four non-demented patients with PD completed questionnaires assessing apathy and ICD symptoms, which were used to classify participants into one of the following groups: apathy only, ICD only, both, and neither. Participants also completed multidimensional measures of anhedonia and trait impulsivity, which were co

An in vitro model of hippocampal sharp waves: regional initiation and intracellular correlates.

During slow wave sleep and consummatory behaviors, electroencephalographic recordings from the rodent hippocampus reveal large amplitude potentials called sharp waves. The sharp waves originate from the CA3 circuitry and their generation is correlated with coherent discharges of CA3 pyramidal neurons and dependent on activities mediated by AMPA glutamate receptors. To model sharp waves in a relatively large hippocampal circuitry in vitro, we developed thick (1 mm) mouse hippocampal slices by separating the dentate gyrus from the CA2/CA1 areas while keeping the functional dentate gyrus-CA3-CA1

Functional interaction between nociceptin/orphanin FQ and alpha-melanocyte-stimulating hormone in the regulation of feeding.

Nociceptin/orphanin FQ (N/OFQ), an endogenous agonist of the opioid N/OFQ (NOP) receptor, increases food intake when administered centrally. As N/OFQ is part of a larger neural network that governs consummatory behavior, presumably its orexigenic properties stem from interplay with other neuropeptidergic components of the feeding-related circuitry. One such peptide may be the ligand of the melanocortin-3 and -4 receptors, alpha-melanocyte-stimulating hormone (alpha-MSH), which is known to inhibit food intake. The aim of the present study was to establish whether there is a functional "interact

Differential regulation of female sexual behaviour by dopamine agonists in the medial preoptic area.

The medial preoptic area (mPOA) is a brain region critical in the control of male sexual behaviour, and the neurotransmitter dopamine (DA) plays an important role within it. However, both the roles of DA and the mPOA in female sexual behaviour are not fully understood, with few studies producing consistent data. The present study examined the function of DA within the mPOA on the full cascade of female sexual behaviour. Ovariectomized female rats were bilaterally cannulated into the mPOA and partially hormonally primed with estradiol benzoate (EB). Different doses of a nonselective DA receptor

Facilitation of sexual behavior in ovariectomized rats by estradiol and testosterone: A preclinical model of androgen effects on female sexual desire.

In the United States and Canada, there are no approved treatments for hypoactive sexual desire disorder in postmenopausal women. Testosterone improves female sexual desire in naturally- and surgically-menopausal women maintained on estrogen replacement therapy, and long-term safety data from randomized placebo-controlled clinical trials has yielded promising results. However, the mechanisms associated with its efficacy are not known, and could be addressed using preclinical rodent models; yet there is no systematic evaluation of the effects of estradiol and testosterone on female rat sexual be

Activation of anoctamin-1 calcium-activated chloride channels reduces voluntary alcohol consumption in rats.

Repeated episodes of binge drinking can lead to an alcohol use disorder, yet the underlying pharmacological mechanisms are still not fully understood. Nevertheless, emerging evidence indicates that Ca2+-dependent signaling effectively reduces alcohol consumption without affecting water intake. Therefore, activating anoctamin1 (ANO1), a Ca2+-activated chloride channel and a component of Ca2+-dependent signaling, can similarly decrease alcohol drinking while maintaining normal water intake. This study investigates how activation of ANO1 channels with EACT affects voluntary alcohol consumption in

Effect of ganaxolone and THIP on operant and limited-access ethanol self-administration.

Recent evidence suggests that GABA(A) receptor ligands may regulate ethanol intake via effects at both synaptic and extrasynaptic receptors. For example, the endogenous neurosteroid, allopregnanolone (ALLO) has a similar pharmacological profile as ethanol, and it alters ethanol intake in rodent models. Additionally, recent evidence suggests that δ-subunit-containing extrasynaptic GABA(A) receptors may confer high sensitivity to both ethanol and neurosteroids. The purpose of the present study was to determine the effects of ganaxolone (GAN; an ALLO analog) and gaboxadol (THIP; a GABA(A) recepto

Sexual desire and hypoactive sexual desire disorder in women. Introduction and overview. Standard operating procedure (SOP Part 1).

INTRODUCTION: Hypoactive sexual desire disorder (HSDD) is defined in Diagnostic and Statistical Manual of Mental Disorders Fourth Edition as persistent or recurrent deficiency (or absence) of sexual fantasies/thoughts, and/or desire for or receptivity to sexual activity, which causes personal distress. As a largely subjective experience, sexual desire may or may not be accompanied by externally observable changes in sexual behavior. AIM: Describe the models of understanding HSDD and the contributing factors to provide the basis for a diagnostic interview and guidance for care for healthcare p
