World Health Organization Guidelines for Containment of Poliovirus Following Type-Specific Polio Eradication - Worldwide, 2015.

In 1988, the World Health Assembly of the World Health Organization (WHO) resolved to eradicate polio worldwide. Among the three wild poliovirus (WPV) types (type 1, type 2, and type 3), WPV type 2 (WPV2) has been eliminated in the wild since 1999, and WPV type 3 (WPV3) has not been reported since 2012. In 2015, only Afghanistan and Pakistan have reported WPV transmission. On May 25, 2015, all WHO Member States endorsed World Health Assembly resolution 68.3 on full implementation of the Polio Eradication and Endgame Strategic Plan 2013-2018 (the Endgame Plan), and with it, the third Global Act

National laboratory inventory as part of global poliovirus containment--United States, June 2002.

Since the initiation of the global poliomyelitis initiative in 1988 through 2001, the number of countries where polio is endemic decreased from 125 to 10, and the number of reported polio cases decreased >99%, from an estimated 350,000 to <1,000. The Global Commission for the Certification of the Eradication of Poliomyelitis, convened by the World Health Organization, will declare the world polio-free when all regions have documented the absence of wild poliovirus transmission for at least 3 consecutive years and when laboratories with wild poliovirus materials have implemented appropriate con

Establishment of a biosafe murine model of skeletal tuberculosis using Mycobacterium smegmatis.

BACKGROUND: Skeletal tuberculosis (TB) remains a persistent clinical and research challenge due to its chronic course, osteolytic destruction, and the limitations of existing animal models, which often require high-level biosafety containment or fail to replicate human skeletal pathology. METHODS: This study developed a biosafe, accessible, and versatile murine model of skeletal TB using Mycobacterium smegmatis, a fast-growing, nonpathogenic mycobacterial species with high genomic homology to Mycobacterium tuberculosis. Three infection routes-subperiosteal calvarial injection, intratibial inj

Evaluation and application of an innovative portable field endoscope: addressing battlefield and biosafety concerns.

BACKGROUND: At present, no commercially available endoscopic system is specifically designed for use in the battlefield, disaster relief, or unique environments with biosafety concerns. Therefore, this limitation stems from challenges such as limited portability, reliance on stable power, complex disinfection processes, and the risk of incomplete sterilization. To address these challenges, we developed a novel portable endoscopic system and evaluated its safety and effectiveness in both routine settings and specialized scenarios, including the global pandemic caused by a novel coronavirus, whi

A Cross-Border Biorisk Toolkit for Healthcare Professionals.

The COVID-19 pandemic posed significant challenges to public health, exposing first responders to high biosafety risks during medical assistance and containment efforts. The PANDEM-2 study aimed to address these critical biosafety issues by emphasising the importance of frequently updated, harmonised guidelines. This study reviewed scientific publications, lessons learned, and real-world experiences from the COVID-19 pandemic to identify biorisk gaps in three critical areas: (i) patient transportation and management, (ii) sample handling and testing, and (iii) data management and communication

Validated Methods for Effective Decontamination and Inactivation of Rift Valley Fever Virus.

The Rift Valley fever virus (RVFV) is an arthropod-borne, zoonotic, hemorrhagic fever virus that can cause severe diseases both in livestock and humans. The spread of RVFV in areas previously considered as non-endemic together with the absence of licensed vaccines for use in humans and animals poses a major health and economic threat worldwide. It is therefore crucial to make major progresses in our understanding and management of this virus and its zoonosis. RVFV is considered a bioterrorism pathogen, and, thus, only a few institutes, facilities, and personnel are legally authorized to detain

Burkholderia pseudomallei JW270 Is Lethal in the Madagascar Hissing Cockroach Infection Model and Can Be Utilized at Biosafety Level 2 to Identify Putative Virulence Factors.

Burkholderia pseudomallei, the causative agent of melioidosis, is classified by the CDC as a tier 1 select agent, and work involving it must be performed in a biosafety level 3 (BSL-3) laboratory. Three BSL-2 surrogate strains derived from B. pseudomallei 1026b, a virulent clinical isolate, have been removed from the CDC select agent list. These strains, Bp82, B0011, and JW270, are highly attenuated in rodent models of melioidosis and cannot be utilized to identify virulence determinants because of their high 50% lethal dose (LD50). We previously demonstrated that the Madagascar hissing cockro

Social Consequences of Ebola Containment Measures in Liberia.

INTRODUCTION: In the Ebola Virus Disease (EVD) outbreak in Liberia, two major emergency disease-control measures were cremation of bodies and enforcement of quarantine for asymptomatic individuals suspected of being in contact with a positive case. Enforced by State-related actors, these were promoted as the only method to curtail transmissions as soon as possible. However, as with other harsh measures witnessed by Liberian citizens, in many cases those measures elicited uncontrolled negative reactions within the communities (stigma; fear) that produced, in some cases, the opposite effect of t

Progress toward laboratory containment of poliovirus after polio eradication.

BACKGROUND: The first steps (phase 1) toward laboratory containment of poliovirus after eradication are a national survey of biomedical facilities and a global inventory of such facilities retaining wild poliovirus (WPV) infectious and potentially infectious materials. METHODS: We reviewed published reports on national laboratory surveys and inventories of WPV materials from each of the 3 polio-free World Health Organization (WHO) regions (the European Region, completed in 2006; the Western Pacific Region, completed in 2008; and the Region of the Americas, completed in 2010), as well as repor

CDC Report on the Potential Exposure to Anthrax.

The Centers for Disease Control and Prevention (CDC) conducted an internal review of an incident that involved an unintentional release of potentially viable anthrax within its Roybal Campus, in Atlanta, Georgia. On June 5, 2014, a laboratory scientist in the Bioterrorism Rapid Response and Advanced Technology (BRRAT) laboratory prepared extracts from a panel of eight bacterial select agents, including Bacillus anthracis (B. anthracis), under biosafety level (BSL) 3 containment conditions. These samples were being prepared for analysis using matrix-assisted laser desorption/ionization time-of-

Laboratory maintenance of pathogenic Leptospira.

Analysis of Leptospira requires use of specialized media for growth, maintenance, and storage of viable bacteria that can be used in experimental protocols in a predictable manner. However, pathogenic Leptospira are fastidious bacteria with unusual nutritional requirements. These problems make primary isolation, routine propagation, and storage of Leptospira difficult. Defined and complex media are available for routine growth of pathogenic Leptospira, and each medium has unique characteristics that favor the growth or isolation of some strains, while not supporting the growth of others. This

Medical surveillance for hazardous waste workers.

The efforts to clean up hazardous waste disposal sites have led to an increasing number of workers involved in these activities. Industrial hygiene surveys have demonstrated that these workers are potentially exposed to low levels of multiple contaminants during the work day and to high levels of some contaminants for very short time periods. Personal protective equipment and work practices are important components in protecting workers from these exposures. Medical monitoring for these workers is problematic. While monitoring for the potential health effects of these multiple exposures may be

Biosafety of rabies virus and related lyssaviruses: Risks, precautions, and knowledge gaps in diagnostic and research laboratories.

Lyssaviruses cause rabies in mammals, a viral encephalomyelitis that is almost always fatal once clinical symptoms develop and can be transmitted to humans. Given this high fatality rate, strict biosafety measures are essential during laboratory handling of potentially infectious materials. This review critically assesses current biosafety practices in rabies diagnostic and research laboratories by examining principal risk factors, transmission pathways, and the effectiveness of existing preventive measures. We conducted a comprehensive review of the literature on biosafety risks associated wi

Impact of inactivation methods on biosafety and antigen reactivity of Brucella melitensis from the perspective of Astral-DIA proteomics based on antibody immunoprecipitation mass spectrometry.

Effective Brucella inactivation is essential for safe vaccine development, diagnostics, and sample handling, particularly in resource-limited regions without high-level biosafety facilities. This study evaluated heat (80°C/95°C, 10-20 min) and formaldehyde (0.4%/0.6%, 48-72 h) inactivation using the Rev.1 vaccine strain and three Brucella melitensis field isolates from Gansu (GS-XG1, GS-SN2, GS-MQ3). All field isolates were collected from a single geographic region (Gansu, China), which should be considered when interpreting the broader geographic generalizability of the findings. Complete ina

Efficient Biosafe Inactivation of Classical Swine Fever Virus While Preserving Viral RNA for Molecular Diagnosis.

Classical swine fever (CSF) is a highly contagious transboundary animal disease that causes major economic losses to the global swine industry and remains notifiable to the World Organisation for Animal Health (WOAH). Handling and transport of infectious classical swine fever virus (CSFV)-positive samples require biosafety level 3 (BSL-3) containment, limiting diagnostic capacity and interlaboratory exchange in many regions. In this study, we evaluated the efficacy of PrimeStore® Molecular Transport Medium (PS-MTM) for eliminating detectable CSFV infectivity while preserving viral ribonucleic

Swinepox virus vector-based vaccines: attenuation and biosafety assessments following subcutaneous prick inoculation.

Swinepox virus (SPV) has several advantages as a potential clinical vector for a live vector vaccine. In this study, to obtain a safer and more efficient SPV vector, three SPV mutants, Δ003, Δ010, and ΔTK were successfully constructed. A virus replication experiment showed that these SPV mutants had lower replication abilities compared to wtSPV in 10 different host-derived cell lines. Animal experiments with mouse and rabbit models demonstrate that these three mutants and wtSPV did not cause any clinical signs of dermatitis. No fatalities were observed during a peritoneal challenge assay with

Rapid deployment of a mobile biosafety level-3 laboratory in Sierra Leone during the 2014 Ebola virus epidemic.

BACKGROUND: Ebola virus emerged in West Africa in December 2013. The high population mobility and poor public health infrastructure in this region led to the development of the largest Ebola virus disease (EVD) outbreak to date. METHODOLOGY/PRINCIPAL FINDINGS: On September 26, 2014, China dispatched a Mobile Biosafety Level-3 Laboratory (MBSL-3 Lab) and a well-trained diagnostic team to Sierra Leone to assist in EVD diagnosis using quantitative real-time PCR, which allowed the diagnosis of suspected EVD cases in less than 4 hours from the time of sample receiving. This laboratory was composed
