CITF1 interacts with FIT and regulates copper-iron crosstalk in Arabidopsis.

Iron (Fe) and copper (Cu) are essential yet potentially toxic metals with interconnected metabolic pathways; however, the mechanisms underlying Fe-Cu crosstalk remain poorly defined. Here, we show that CITF1 (COPPER DEFICIENCY INDUCED TRANSCRIPTION FACTOR 1), a Cu homeostasis regulator in Arabidopsis thaliana, physically interacts with FIT (FER-LIKE IRON DEFICIENCY-INDUCED TRANSCRIPTION FACTOR), the central Fe homeostasis regulator, forming a nutrient-responsive transcriptional module. Under Cu deficiency, the CITF1-FIT complex accumulates and promotes expression of the Cu uptake genes COPT2 (

8-Hydroxyquinoline Glycoconjugates Containing Sulfur at the Sugar Anomeric Position-Synthesis and Preliminary Evaluation of Their Cytotoxicity.

One of the main factors limiting the effectiveness of many drugs is the difficulty of their delivery to their target site in the cell and achieving the desired therapeutic dose. Moreover, the accumulation of the drug in healthy tissue can lead to serious side effects. The way to improve the selectivity of a drug to the cancer cells seems to be its conjugation with a sugar molecule, which should facilitate its selective transport through GLUT transporters (glucose transporters), whose overexpression is seen in some types of cancer. This was the idea behind the synthesis of 8-hydroxyquinoline (8

Dietary copper supplementation enhances lipolysis in Rex rabbits.

BACKGROUND: Copper is an important regulator of lipid metabolism in mammals, as a cofactor of many enzymes and is involved in the lipolysis. Copper deficiency has been considered as a significant factor in human diseases related to abnormal lipid metabolism, while adding copper to the diet seems to be the simplest and most effective way to prevent copper deficiency. AIMS: The aim of this study was to investigate the effects of dietary copper level on lipid metabolism in Rex Rabbits. METHODS: A total of 120 90-d-old Rex Rabbits were randomly allotted into three treatments, with 40 replicates

Organelle-Targeted, ROS-Gated Prochelators for Inhibition of Triple-Negative Breast Cancer via Depleting Exchangeable Copper.

The dyshomeostasis of copper has been implicated in a variety of human diseases. However, the selective manipulation of copper flux at the subcellular organelle level remains challenging, which hinders the development of mechanism-guided therapeutic interventions for related diseases. We report a modular prochelator platform that integrates organelle-targeting ligands with stimuli-cleavable masking groups to unmask a conserved dithiocarbamate (DTC) chelating motif in situ, enabling precise depletion of exchangeable copper within the targeted subcellular compartment. In triple-negative breast c

Sodium butyrate induces cuproptosis by regulating the HDAC1-SLC31A1 axis in hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is the primary type of liver malignancy, which stands as the third leading contributor to cancer-related death worldwide. Sodium butyrate (NaB), a gut microbiota-derived short-chain fatty acid with known anticancer activity, remains poorly understood in HCC. This study aimed to elucidate the effects and underlying mechanisms of NaB-induced cuproptosis in HCC cells. 200 mg/kg NaB inhibits HCC tumor growth by 73.9% and induced a 1.83- and 6.83-fold increase in the Cu2+ content and SLC31A1 protein expression, respectively. Furthermore, 5 mmol/L NaB inhibits cell via

The human cytochrome c oxidase assembly factors SCO1 and SCO2 have regulatory roles in the maintenance of cellular copper homeostasis.

Human SCO1 and SCO2 are metallochaperones that are essential for the assembly of the catalytic core of cytochrome c oxidase (COX). Here we show that they have additional, unexpected roles in cellular copper homeostasis. Mutations in either SCO result in a cellular copper deficiency that is both tissue and allele specific. This phenotype can be dissociated from the defects in COX assembly and is suppressed by overexpression of SCO2, but not SCO1. Overexpression of a SCO1 mutant in control cells in which wild-type SCO1 levels were reduced by shRNA recapitulates the copper-deficiency phenotype in

Ctr1 drives intestinal copper absorption and is essential for growth, iron metabolism, and neonatal cardiac function.

The trace element copper (Cu) is a cofactor for biochemical functions ranging from energy generation to iron (Fe) acquisition, angiogenesis, and free radical detoxification. While Cu is essential for life, the molecules that mediate dietary Cu uptake have not been identified. Ctr1 is a homotrimeric protein, conserved from yeast to humans, that transports Cu across the plasma membrane with high affinity and specificity. Here we describe the generation of intestinal epithelial cell-specific Ctr1 knockout mice. These mice exhibit striking neonatal defects in Cu accumulation in peripheral tissues,

Copper transporters promote the malignant progression of glioblastoma by regulating the immunosuppressive tumour microenvironment via the SLC31A1-WNT5A axis.

BACKGROUND: The prognosis for patients with glioblastoma (GBM) remains extremely poor, a challenge largely attributable to the complex nature of its malignant progression and an insufficient understanding of the dynamic plasticity and spatial heterogeneity inherent in the tumour microenvironment during invasion. In recent years, copper has emerged as a trace element of considerable interest due to its implicated role in tumour initiation and progression. This study was therefore designed to systematically investigate the involvement of copper transporter genes in the malignant evolution of gli

Opto-electrochemical planar wave-guide sensor for copper (II) ion.

An opto-electrochemical sensor for copper (II) ion was developed. The sensor consists of a planar conductive indium-tin-oxide (ITO) glass support, which was coated by a polymeric copper (II) sensitive membrane. The sensing membrane is made of plasticized polyurethane matrix containing Zincon colorimetric reagent immobilized as ion pair with tetraoctylammonium ion. The instrumentation used commonly for wave-guide sensor development was extended with a potentiostat that made possible the control of the electric potential of the conductive surface. In this way the transport rate of ionic species

Spectroscopic and Small-angle X-ray scattering analysis of binding between Copper(II) -1-allylimidazole complex, a potential anti-tumor agent, and bovine serum albumin.

Interactions between transport proteins and compounds with therapeutic potential are pharmacologically important. In this study, using fluorescence, circular dichroism (CD), and small-angle X-ray Scattering (SAXS), we investigated the interaction between bovine serum albumin (BSA) and a copper(II)-1-allylimidazole complex with potential anti-cancer properties. The results revealed dynamic fluorescence quenching of the model carrier protein BSA by the copper(II) complex. The enthalpy change (ΔH), free energy (ΔG), and entropy change (ΔS) were calculated to be 108 kJ/mol, -16.47 kJ/mol, and 419

Tumor CTR1 Expression and Systemic Copper Dynamics Converge on a Copper Axis in High-Grade Triple-Negative Breast Cancer.

UNLABELLED: Copper (Cu) metabolism represents a unique vulnerability in cancer. CTR1 (SLC31A1) mediates cellular Cu uptake, but its relationship with tumor aggressiveness and neoadjuvant response is unclear. We tested whether tumor SLC31A1 expression and systemic Cu dynamics reflect a coordinated tumor-systemic Cu axis in breast cancer. Baseline tumor SLC31A1 gene expression was analyzed in a retrospective cohort of 1,632 neoadjuvant-treated patients with breast cancer, who were subsequently stratified by pathologic response across molecular subtypes and tumor grades. These findings were exten

Vitamin B6 metabolism influences the intracellular accumulation of cisplatin.

Vitamin B6 metabolism influences the adaptive response of non-small lung carcinoma (NSCLC) cells to distinct, potentially lethal perturbations in homeostasis, encompassing nutrient deprivation, hyperthermia, hypoxia, irradiation as well as the exposure to cytotoxic chemicals, including the DNA-damaging agent cisplatin (CDDP). Thus, the siRNA-mediated downregulation of pyridoxal kinase (PDXK), the enzyme that generates the bioactive form of vitamin B6, protects NSCLC cells (as well as a large collection of human and murine malignant cells of distinct histological derivation) from the cytotoxic

GSH-responsive self-assembled nanoplatform synergistically enhances cuproptosis through metabolic reprogramming and oxidative stress amplification.

Cuproptosis, an emerging copper-dependent regulated cell death pathway, demonstrates significant potential for overcoming therapeutic resistance in oncology. However, its clinical translation remains constrained by the poor bioavailability of copper ionophores and intrinsic resistance mechanisms in tumor cells. Here, we developed a tumor microenvironment-responsive nanoparticle platform (PEMA) co-loading an MPC1 (mitochondrial pyruvate carrier 1) overexpression plasmid and the copper ionophore Elesclomol to establish a synergistic "metabolic reprogramming-oxidative stress amplification" strate

Blocking lactate regulation of the Grhl2/SLC31A1 axis inhibits trophoblast cuproptosis and preeclampsia development.

PURPOSE: Abnormal cell death due to superficial trophoblast dysfunction caused by placental hypoxia plays a vital role in the development of preeclampsia (PE). Lactic acid stimulates gene transcription in chromatin through lactate modification of histone lysine. Nevertheless, the content and function of lactate in PE development remains largely unclear. METHODS: The contents of lactic acid and copper in 30 PE and 30 normal placentas were determined by kit colorimetry. Real-time quantitative fluorescent PCR (qRT-PCR) and Western blot were used to detect the expression of SLC31A1 in cells and t

Curcumin may impair iron status when fed to mice for six months.

Curcumin has been shown to have many potentially health beneficial properties in vitro and in animal models with clinical studies on the toxicity of curcumin reporting no major side effects. However, curcumin may chelate dietary trace elements and could thus potentially exert adverse effects. Here, we investigated the effects of a 6 month dietary supplementation with 0.2% curcumin on iron, zinc, and copper status in C57BL/6J mice. Compared to non-supplemented control mice, we observed a significant reduction in iron, but not zinc and copper stores, in the liver and the spleen, as well as stron

Matrigel-based organoid culture of malignant mesothelioma reproduces cisplatin sensitivity through CTR1.

Organoids are a three-dimensional (3D) culture system that simulate actual organs. Therefore, tumor organoids are expected to predict precise response to chemotherapy in patients. However, to date, few studies have studied the drug responses in organoids of malignant mesothelioma (MM). The poor prognosis of MM emphasizes the importance of establishing a protocol for generating MM-organoid for research and clinical use. Here, we established murine MM organoids from p53+/- or wild-type C57BL/6 strain by intraperitoneal injection either with crocidolite or carbon nanotube. Established MM-organoid

The mitochondrial metallochaperone SCO1 maintains CTR1 at the plasma membrane to preserve copper homeostasis in the murine heart.

SCO1 is a ubiquitously expressed, mitochondrial protein with essential roles in cytochrome c oxidase (COX) assembly and the regulation of copper homeostasis. SCO1 patients present with severe forms of early onset disease, and ultimately succumb from liver, heart or brain failure. However, the inherent susceptibility of these tissues to SCO1 mutations and the clinical heterogeneity observed across SCO1 pedigrees remain poorly understood phenomena. To further address this issue, we generated Sco1hrt/hrt and Sco1stm/stm mice in which Sco1 was specifically deleted in heart and striated muscle, res

Self-Assembly of the Second Transmembrane Domain of hCtr1 in Micelles and Interaction with Silver Ion.

Human copper transporter 1 (hCtr1) transports copper and silver by a homotrimer. The protein contains three transmembrane domains in which the second transmembrane domain (TMD2) is a key component lining the central pore of the trimer. The MXXXM motif in the C-terminal end of TMD2 plays a significant role in the function of hCtr1. In this study, we characterized the structure and assembly of isolated TMD2 of hCtr1 in sodium dodecyl sulfate (SDS) micelles and the interaction of the micelle-bound peptide with silver ion using nuclear magnetic resonance, circular dichroism, isothermal titration c
